1A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. 2Schroth exercises improve health-related quality of life and radiographic parameters in adolescent idiopathic scoliosis patients显示文摘Background:Finding an optimal treatment strategy for adolescent idiopathic scoliosis(AIS)patients remains challenging because of its intrinsic complexity.For mild to moderate scoliosis patients with lower skeletal growth potential(Risser 3-5),most clinicians agree with observation treatment;however,the curve progression that occurs during puberty,the adolescent period,and even in adulthood,remains a challenging issue for clinicians.The aim of the study is to investigate the efficacy of Schroth exercise in AIS patients with lower skeletal growth potential(Risser 3-5)and moderate scoliosis(Cobb angle 20°-40°).Methods:From 2015 to 2017,data of 64 patients diagnosed with AIS in Peking University Third Hospital were reviewed.Forty-three patients underwent Schroth exercise were classified as Schroth group,and 21 patients underwent observation were classified as observation group.Outcomes were measured by health-related quality of life(HRQOL)and radiographic parameters.HRQOL was assessed using the visual analog scale(VAS)scores for back,Scoliosis Research Society-22(SRS-22)patient questionnaire.Radiographic spinopelvic parameters were obtained from anteroposterior and lateral X-rays.The pre-treatment and post-treatment HRQOL and radiographic parameters were tested to validate Schroth exercise efficacy.The inter-rater reliability of the radiographic parameters was tested using the interclass correlation coefficient(ICC).The pairedt test was used to examine HRQOL and radiographic parameters.Clinical relevance between C2-C7 sagittal vertical axis(SVA)and thoracic kyphosis was analyzed using Spearman correlation.Results:In Schroth group,VAS back score,SRS-22 pain,and SRS-22 self-image domain were significantly improved from pre-treatment 3.0±0.8,3.6±0.5,and 3.5±0.7 to post-treatment 1.6±0.6(t=5.578,P=0.013),4.0±0.3(t=-3.918,P=0.001),and 3.7±0.4(t=-6.468,P<0.001),respectively.No significant improvements of SRS-22 function domain(t=-2.825,P=0.088)and mental health domain(t=-3.174,P=0.061)were observed.The mean Cobb angle decreased from 28.9±5.5°to 26.3±5.2°at the final follow-up,despite no statistical significance was observed(t=1.853,P=0.102).The mean C2-C7 SVA value decreased from 21.7±8.4 mm to 17.0±8.0 mm(t=-1.224P=0.049)and mean T1 tilt decreased from 4.9±4.2°to 3.5±3.1°(t=2.913,P=0.011).No significant improvement of radiographic parameters and HRQOL were observed in observation group.Conclusions:For AIS patients with a Risser 3-5 and a Cobb angle 20°-40°,Schroth exercises improved HRQOL and halted curve progression during the follow-up period.Both cervical spine alignment and shoulder balance were also significantly improved after Schroth exercises.We recommend Schroth exercises for patients with AIS.Ang Gao Jun-Yu Li Rui Shao Tong-Xuan Wu Yong-Qiang Wang Xiao-Guang Liu Miao Yu 2021Chinese Medical Journal2021,,21:10
3Second-harmonic generation using d(33)in periodically poled lithium niobate microdisk resonators显示文摘A fabrication process allowing for the production of periodically poled lithium niobate(PPLN)photonic devices with any domain pattern and unit size down to 200 nm is developed by combining semiconductor fabrication techniques and piezo-force-microscopy tips polarization.Based on this fabrication process,PPLN microdisk resonators with quality factors of 8×10~4 were fabricated from a Z-cut lithium niobate film.Second-harmonic generation(SHG)utilizing d(33)in the whole cavity was demonstrated in a PPLN microdisk with a 2μm-spatialperiod radial domain pattern.The SHG conversion efficiency was measured to be 1.44×10^(-5)m W^(-1).This work paves the way to fabricate complex PPLN photonic devices and to obtain efficient nonlinear optical effects that have wide applications in both classical and quantum optics.ZHENZHONG HAO LI ZHANG WENBO MAO ANG GAO XIAOMEI GAO FENG GAO FANG BO GUOQUAN ZHANG JINGJUN XU 2020Photonics Research2020,8,3:6

5Analysis of the Phenotype and the Restriction Enzyme Mapping Level of Mutations Induced by the New Mutagen Glycidyl Methacrylate显示文摘Glycidyl methaerylate (GMA) is a recently recognized chemical mutagen. In order to explore the mutagenicity and mutagenic process of GMA, plasmid pBR322 was used for in vitro binding, mutant screening, and restriction enzyme mapping. The binding between GMA and DNA in vitro has been verified by means of a spectrophotometric method. When pBR322 and GMAbound pBR322 were used to transform Eschenchia coli HB101, the following results were obtained: (1) The transformation efficiency of GMA-bound pBR322 was much lower than that of pBR322 alone. (2) GMA-bound pBR322 induced phenotype changes in competent cells (i.e., tetracycline-resistance inactivation or ampicillin-resistance inactivation). There were two mutants of pBR322, Ap^RTc^S and Ap^STc^R, in the transformants and a deductive mutant Ap^STc^S in the nontranstormants. (3) All of the selected mutants were stable and heritable. (4) When restriction enzyme maps were used to analyze the mutant Ap^RTc^S, four of seven maps were changed. some sites were shifted to other resistant gene regions, for example, sites of Bgll, EcoRl, Ilindlll. Hinclll, etc., and there was a new recognition site for Hindi (252). We did not observe any DNA fragment insertion or deletion on any maps. Our results suggest that when GMA is covalently linked to the plasmid DNA, it gives rise to a premutagenic lesion of DNA that is converted in vivo into a point mutation. 6Infection recurrence following minimally invasive treatment in patients with infectious pancreatic necrosis显示文摘BACKGROUND In recent decades,an increasing number of patients have received minimally invasive intervention for infected pancreatic necrosis(IPN)because of the benefits in reducing postoperative multiple organ failure and mortality.However,there are limited published data regarding infection recurrence after treatment of this patient population.AIM To investigate the incidence and prediction of infection recurrence following successful minimally invasive treatment in IPN patients.METHODS Medical records for 193 IPN patients,who underwent minimally invasive treatment between February 2014 and October 2018,were retrospectively reviewed.Patients,who survived after the treatment,were divided into two groups:one group with infection after drainage catheter removal and another group without infection.The morphological and clinical data were compared between the two groups.Significantly different variables were introduced into the correlation and multivariate logistic analysis to identify independent predictors for infection recurrence.Sensitivity and specificity for diagnostic performance were determined.RESULTS Of the 193 IPN patients,178 were recruited into the study.Of them,9(5.06%)patients died and 169 patients survived but infection recurred in 13 of 178 patients(7.30%)at 7(4-10)d after drainage catheters were removed.White blood cell(WBC)count,serum C-reactive protein(CRP),interleukin-6,and procalcitonin levels measured at the time of catheter removal were significantly higher in patients with infection than in those without(all P<0.05).In addition,drainage duration and length of the catheter measured by computerized tomography scan were significantly longer in patients with infection(P=0.025 and P<0.0001,respectively).Although these parameters all correlated positively with the incidence of infection(all P<0.05),only WBC,CRP,procalcitonin levels,and catheter length were identified as independent predictors for infection recurrence.The sensitivity and specificity for infection prediction were high in WBC count(≥9.95×109/L)and serum procalcitonin level(≥0.05 ng/mL)but moderate in serum CRP level(cut-off point≥7.37 mg/L).The catheter length(cut-off value≥8.05 cm)had a high sensitivity but low specificity to predict the infection recurrence.CONCLUSION WBC count,serum procalcitonin,and CRP levels may be valuable for predicting infection recurrence following minimally invasive intervention in IPN patients.These biomarkers should be considered before removing the drainage catheters.Chong-Chong Gao Jia Li Feng Cao Xiao-Hui Wang Ang Li Zhe Wang Fei Li 2020World Journal of Gastroenterology2020,26,22:5
7Periodically poled lithium niobate whispering gallery mode microcavities on a chip显示文摘In this study, we investigate the fabrication of periodically poled lithium niobate(PPLN) microdisk cavities on a chip. These resonators are fabricated from a PPLN film with a 16 μm poling period on insulator using conventional microfabrication techniques.The quality factor of the PPLN microdisk resonators with a 40-μm radius and a 700-nm thickness is 6.7×10~5. Second harmonic generation(SHG) with an efficiency of 2.2×10^(-6) mW(-1) is demonstrated in the fabricated PPLN microdisks. 9Dectin-1 expression at early period of Aspergillus fumigatus infection in rat's corneal epithelium显示文摘AIM:To investigate the expression of dendritic cell-associated C-type lectin-1 (dectin-1) at the early period of Aspergillus fumigatus infection in rat's corneal epithelium. ·METHODS:A total of 72 Wistar rats were randomly divided into three groups:A, B and C. The right eyes were chosen as experimental eyes. Group A was control group. Rats in group B were not inoculated with Aspergillus fumigatus. Group C was taken as Aspergillus fumigatus keratitis model. Rats in group B and C (six from each group) were executed randomly at 4, 8, 16 and 24 hours after experimental model being established to assess the expression of dectin-1 mRNA through real-time PCR. Another six rats in group B and C were executed randomly at 24 hours to assess the expression of dectin-1 protein through immunohistochemistry. ·RESULTS:The results of real-time PCR indicated that dectin-1 mRNA expression was low in corneal epithelium of normal rats'. There was no significantly difference of dectin-1 mRNA expression in group A and B (P >0.05). The expression of Aspergillus fumigatus infected corneal epithelium increased gradually after 8 hours in group C. The synchronous expression of group A and C had significant difference (P <0.01). Immunohistochemisty discovered that dectin-1 receptor existed in normal rat's corneal epithelium . Dectin-1 protein increased after 24 hours in group C. There was a significant difference of synchronous expression in group B and C(P<0.01). · CONCLUSION:Dectin-1 exists in rat's cornealepithelium and its expression significantly increases at the early period of Aspergillus fumigatus infection. 11Determination of time-and size-dependent fine particle emission with varied oil heating in an experimental kitchen显示文摘Particulate matter(PM) from cooking has caused seriously indoor air pollutant and aroused risk to human health.It is urged to get deep knowledge of their spatial-temporal distribution of source emission characteristics,especially ultrafine particles(UFP < 100 nm) and accumulation mode particles(AMP 100-555 nm).Four commercial cooking oils are auto dipped water to simulate cooking fume under heating to 255℃ to investigate PM emission and decay features between 0.03 and 10 μm size dimension by electrical low pressure impactor(ELPI) without ventilation.Rapeseed and sunflower produced high PM_(2.5) around5.1 mg/m^3,in comparison with those of soybean and corn(5.87 and 4.55 mg/m^3,respectively)at peak emission time between 340 and 450 sec since heating oil,but with the same level of particle numbers 6-9 × 10~5/cm^3.Mean values of PM_(1.0)/PM_(2.5) and PM_(2.5)/PM_(10) at peak emission time are around 0.51-0.55 and 0.23-0.29.After 15 min naturally deposition,decay rates of PM_(1.0),PM_(2.5) and PM_(10) are 13.3%-29.8%,20.1%-33.9%and 41.2%-54.7%,which manifest that PM_(1.0) is quite hard to decay than larger particles,PM_(2.5) and PM_(1.0).The majority of the particle emission locates at 43 nm with the largest decay rate at 75%,and shifts to a larger size between137 and 555 nm after 15 min decay.The decay rates of the particles are sensitive to the oil type.Shuangde Li Jiajia Gao Yiqing He Liuxu Cao Ang Li Shengpeng Mo Yunfa Chen Yaqun Cao 2017Journal of Environmental Sciences2017,29,1:3
12A programmed surface on polyetheretherketone for sequentially dictating osteoimmunomodulation and bone regeneration to achieve ameliorative osseointegration under osteoporotic conditions显示文摘Polyetheretherketone(PEEK)is a desirable alternative to conventional biomedical metals for orthopedic implants due to the excellent mechanical properties.However,the inherent bioinertness of PEEK contributes to inferior osseointegration of PEEK implants,especially under pathological conditions of osteoporosis.Herein,a programmed surface is designed and fabricated on PEEK to dictate osteoimmunomodulation and bone regeneration sequentially.A degradable hybrid coating consisting of poly(lactide-co-glycolide)and alendronate(ALN)loaded nano-hydroxyapatite is deposited on PEEK and then interleukin-4(IL-4)is grafted onto the outer surface of the hybrid coating with the aid of N_(2) plasma immersion ion implantation and subsequent immersion in IL-4 solution.Dominant release of IL-4 together with ALN and Ca^(2+) during the first few days synergistically mitigates the early acute inflammatory reactions and creates an osteoimmunomodulatory microenvironment that facilitates bone regeneration.Afterwards,slow and sustained delivery of ALN and Ca^(2+) in the following weeks boosts osteogenesis and suppresses osteoclastogenesis simultaneously,consequently ameliorating bone-implant osseointegration even under osteoporotic conditions.By taking into account the different phases in bone repair,this strategy of constructing advanced bone implants with sequential functions provides customizable and clinically viable therapy to osteoporotic patients.Yanyan Zheng Ang Gao Jiaxiang Bai Qing Liao Yuzheng Wu Wei Zhang Min Guan Liping Tong Dechun Geng Xin Zhao Paul K.Chu Huaiyu Wang 2022Bioactive Materials2022,7,8:2
13Rational integration of defense and repair synergy on PEEK osteoimplants via biomimetic peptide clicking strategy显示文摘Polyetheretherketone(PEEK)has been widely used as orthopedic and dental materials due to excellent mechanical and physicochemical tolerance.However,its biological inertness,poor osteoinduction,and weak antibacterial activity make the clinical applications in a dilemma.Inspired by the mussel adhesion mechanism,here we reported a biomimetic surface strategy for rational integration and optimization of anti-infectivity and osteo-inductivity onto PEEK surfaces using a mussel foot proteins(Mfps)-mimic peptide with clickable azido terminal.The peptide enables mussel-like adhesion on PEEK biomaterial surfaces,leaving azido groups for the further steps of biofunctionalizations.In this study,antimicrobial peptide(AMP)and osteogenic growth peptide(OGP)were bioorthogonally clicked on the azido-modified PEEK biomaterials to obtain a dual-effect of host defense and tissue repair.Since bioorthogonal clicking allows precise collocation between AMP and OGP through changing their feeding molar ratios,an optimal PEEK surface was finally obtained in this research,which could long-term inhibit bacterial growth,stabilize bone homeostasis and facilitate interfacial bone regeneration.In a word,this upgraded mussel surface strategy proposed in this study is promising for the surface bioengineering of inert medical implants,in particular,achieving rational integration of multiple biofunctions to match clinical requirements.Meng Li Jiaxiang Bai Huaqiang Tao Li Hao Weiling Yin Xiaoxue Ren Ang Gao Ning Li Miao Wang Shiyuan Fang Yaozeng Xu Liang Chen Huilin Yang Huaiyu Wang Guoqing Pan Dechun Geng 2022Bioactive Materials2022,7,2:2

15GPU-accelerated phase field simulation of directional solidification显示文摘The phase field simulation has been actively studied as a powerful method to investigate the microstructural evolution during the solidification.However,it is a great challenge to perform the phase field simulation in large length and time scale.The developed graphics processing unit(GPU)calculation is used in the phase filed simulation,greatly accelerating the calculation efficiency.The results show that the computation with GPU is about 36 times faster than that with a single Central Processing Unit(CPU)core.It provides the feasibility of the GPU-accelerated phase field simulation on a desktop computer.The GPU-accelerated strategy will bring a new opportunity to the application of phase field simulation.GAO Ang HU YanSu WANG ZhiJun MU DeJun LI JunJie WANG JinCheng 2014Science China(Technological Sciences)2014,57,6:1
