| 1 | Two quantitative trait loci, Dw1 and Dw2, are primarily responsible for rootstock-induced dwarfing in apple显示文摘The apple dwarfing rootstock‘Malling9’(‘M9’)has been used worldwide both to reduce scion vigour and as a genetic source for breeding new rootstocks.Progeny of‘M9’segregate for rootstock-induced dwarfing of the scion,indicating that this trait is controlled by one or more genetic factors.A quantitative trait locus(QTL)analysis of a rootstock population derived from the cross between‘M9’בRobusta5’(non-dwarfing)and grafted with‘Braeburn’scions identified a major QTL(Dw1)on linkage group(LG)5,which exhibits a significant influence on dwarfing of the scion.A smaller-effect QTL affecting dwarfing(Dw2)was identified on LG11,and four minor-effect QTLs were found on LG6,LG9,LG10 and LG12.Phenotypic analysis indicates that the combination of Dw1 and Dw2 has the strongest influence on rootstock-induced dwarfing,and that Dw1 has a stronger effect than Dw2.Genetic markers linked to Dw1 and Dw2 were screened over 41 rootstock accessions that confer a range of effects on scion growth.The majority of the dwarfing and semi-dwarfing rootstock accessions screened carried marker alleles linked to Dw1 and Dw2.This suggests that most apple dwarfing rootstocks have been derived from the same genetic source. | Toshi M Foster Jean-Marc Celton David Chagné D Stuart Tustin Susan E Gardiner | 2015 | Horticulture Research2015,2,1: | 10 |
| 2 | New anti-proliferative agent,MK615,from Japanese apricot “Prunus mume” induces striking autophagy in colon cancer cells in vitro显示文摘AIM: To investigate the anti-neoplastic effects of MK615, an extract from the Japanese apricot (Prunus mume), against colon cancer cells. METHODS: Three colon cancer cell lines, SW480, COLO, and WiDr, were cultured with MK615. Growth inhibition was evaluated by cell proliferation assay and killing activity was determined by lactate dehydrogenase assay. Induction of apoptosis was evaluated by annexin Ⅴ flow cytometry. Morphological changes were studied by light and electron microscopy, and immunofluorescence staining with Atg8. RESULTS: MK615 inhibited growth and lysed SW480, COLO and WiDr cells in a dose-dependent manner. Annexin Ⅴ flow cytometry showed that MK615 induced apoptosis after 6 h incubation, at which point the occurrence of apoptotic cells was 68.0%, 65.7% and 64.7% for SW480, COLO, and WiDr cells, respectively. Light and electron microscopy, and immunofluorescence staining with Atg8 revealed that MK615 induced massive cytoplasmic vacuoles (autophagosomes) in all three cell lines. CONCLUSION: MK615 has an anti-neoplastic effect against colon cancer cells. | 3 | Edaravone inhibits apoptosis caused by ischemia/reperfusion injury in a porcine hepatectomy model显示文摘AIM:To investigate the effect of E3-methyl-1-phenyl-2-pyrazolin-5-one (Edr) on hepatic ischemia-reperfusion (I/R) injury and liver regeneration in a porcine hepatectomy model.METHODS:One hour ischemia was induced by occluding the vessels and the bile duct of the right and median lobes.A 40% left hepatectomy was performed after reperfusion.Six animals received Edr (3 mg/kg per hour) intravenously and six control animals received saline just before reperfusion.Remnant liver volume,hemodynamics,aspartate aminotransferase (AST),alanine aminotransferase,lactate dehydrogenase and lactic acid,were compared between the groups.The expression of transforming growth factor (TGF1) and toll-like receptor (TRL) mRNA in hepatic tissues was examined using reverse transcription polymerase chain reaction.Apoptosis was demonstrated by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining,respectively.RESULTS:Serum AST (P = 0.029),and toll like receptor 4 level (P = 0.043) were significantly lower after 3 h in animals receiving Edr.In addition,TUNEL staining in Edr-treated pigs showed significantly fewer hepatocytes undergoing apoptosis compared with control pigs.After 1 mo,all factors were non-significantly different between the two groups.CONCLUSION:Edr is considered to reduce hepatic injury in the early stage of I/R injury in a porcine model. | Mitsugi Shimoda Yoshimi Iwasaki Toshie Okada Keiichi Kubota | 2012 | World Journal of Gastroenterology2012,18,27: | 8 |
| 4 | MK615 inhibits pancreatic cancer cell growth by dual inhibition of Aurora A and B kinases显示文摘AIM:To investigate the anti-neoplastic effect of MK615, an anti-neoplastic compound isolated from Japanese apricot, against human pancreatic cancer cells in vitro. METHODS: Three human pancreatic cancer cell lines PANC-1, PK-1, and PK45H were cultured with MK615 at concentrations of 600, 300, 150, and 0 μg/mL. Growth inhibition was evaluated by cell proliferation assay, and killing activity was determined by lactate dehydrogenase (LDH) assay. Expression of Aurora A and B kinases was detected by real-time polymerase chain reaction (PCR) and Western blotting. Cell cycle stages were evaluated by flow cytometry. RESULTS: The growth inhibitory rates of MK615 at 150, 300, and 600 μg/mL were 2.3% ± 0.9%, 8.9% ± 3.2% and 67.1% ± 8.1% on PANC1 cells, 1.3% ± 0.3%, 8.7% ± 4.1% and 45.7 ± 7.6% on PK1 cells, and 1.2 ± 0.8%, 9.1% ± 2.1% and 52.1% ± 5.5% on PK45H cells, respectively (P <0.05). The percentage cytotoxicities of MK615 at 0, 150, 300, and 600 μg/mL were 19.6% ± 1.3%, 26.7% ± 1.8%, 25.5% ± 0.9% and 26.4% ± 0.9% in PANC1 cells, 19.7% ± 1.3%, 24.7% ± 0.8%, 25.9% ± 0.9% and 29.9% ± 1.1% in PK1 cells, and 28.0% ± 0.9%, 31.2% ± 0.9%, 30.4% ± 1.1% and 35.3 ± 1.0% in PK45H cells, respectively (P < 0.05). Real-time PCR and Western blotting showed that MK615 dually inhibited the expression of Aurora A and B kinases. Cell cycle analysis revealed that MK615 increased the population of cells in G2/M phase. | 5 | MK615 decreases RAGE expression and inhibits TAGE-induced proliferation in hepatocellular carcinoma cells显示文摘AIM:To investigate the proliferative effect of advanced glycation end-products(AGEs) and the role of their cellular receptor(RAGE) on hepatocellular carcinoma(HCC) cells,and the inhibitory effects of MK615,an extract from Japanese apricot,against AGEs were also evaluated.METHODS:Two HCC cell lines,HuH7 and HepG2,were used.Expression of RAGE was investigated by poly-merase chain reaction,Western blotting,and flow cytemetry(FACS).The effect of MK615 on RAGE expression was also evaluated by FACS.The proliferative effects of a control(unglycated bovine serum albumin),glucosederived AGEs(Glc-AGE),and glyceraldehyde-derived AGEs(Glycer-AGE),and the anti-proliferative effect of MK615 against AGEs,were evaluated using MTT assays.RESULTS:Expression of RAGE was confirmed at both the mRNA and protein levels in both HuH7 and HepG2.FACS revealed that the level of RAGE expression was higher in HuH7 than in HepG2.Treatment with 0.1 μg/mL MK615 decreased the expression level of RAGE from 24.3% to 3.7% in HuH7 and from 6.2% to 4.8% in HepG2.The growth indices for the control,Glc-AGE,and Glycer-AGE were 1.06 ± 0.08,0.99 ± 0.04,and 1.38 ± 0.05,respectively,in HuH7(P = 0.037),and were 1.03 ± 0.04,1.04 ± 0.03,and 1.07 ± 0.05,respectively,in HepG2(P > 0.05).When the cells were cultured simultaneously with Glycer-AGE and MK615,MK615 abrogated the proliferative effect of Glycer-AGE in HuH7.CONCLUSION:Only Glycer-AGE has a proliferative effect on HuH7,which expresses a higher level of RAGE.MK615 suppresses the proliferative effect of GlycerAGE on HuH7 by decreasing the expression of RAGE. | Yuhki Sakuraoka Tokihiko Sawada Toshie Okada Takayuki Shiraki Yoshikazu Miura Katsuya Hiraishi Tatsushi Ohsawa Masakazu Adachi Jun-ichi Takino Masayoshi Takeuchi Keiichi Kubota | 2010 | World Journal of Gastroenterology2010,16,42: | 6 |

| 7 | Apple dwarfing rootstocks exhibit an imbalance in carbohydrate allocation and reduced cell growth and metabolism显示文摘Apple dwarfing rootstocks cause earlier shoot termination and reduced root and shoot mass.To identify physiological factors responsible for rootstock-induced growth restriction,we compared vascular-enriched gene expression between two dwarfing rootstocks(‘M27’and‘M9’)and the vigorous rootstock‘M793’using RNA sequencing and quantitative reverse transcriptase PCR.Differentially expressed genes common to both dwarfing rootstocks belonged to five main biological processes:(1)primary metabolism,(2)cell wall synthesis and modification,(3)secondary metabolism,(4)hormone signalling and response and(5)redox homeostasis.Genes promoting the biosynthesis of amino acids,lipids and cell walls were downregulated in dwarfing rootstocks,whereas genes promoting the breakdown of these compounds were upregulated.The only exception to this trend was the upregulation of starch synthesis genes in dwarfing rootstocks.Non-structural carbohydrate analysis demonstrated that starch concentrations in‘M9’roots,stems and grafted‘Royal Gala’(‘RG’)scions were double that of equivalent tissues from‘RG’homo-grafted trees(‘RG’/‘RG’).Fructose and glucose concentrations were much lower in all three tissues of the‘RG’/‘M9’trees.Together,these data indicate that dwarfing rootstocks are in a state of sugar depletion and reduced cellular activity despite having large starch reserves.Another significant finding was the over-accumulation of flavonoids and the downregulation of auxin influx transporters MdAUX1 and MdLAX2 in dwarfing rootstocks.We propose that both factors reduce polar auxin transport.The results of this study contribute novel information about the physiological state of dwarfing rootstocks. | Toshi M Foster Peter A McAtee Chethi N Waite Helen L Boldingh Tony K McGhie | 2017 | Horticulture Research2017,4,1: | 4 |

| 9 | Recurrence of autoimmune hepatitis after liver transplantation without elevation of alanine aminotransferase显示文摘It is controversial whether steroid therapy should be continued to prevent the recurrence of autoimmune hepatitis(AIH)in patients who have undergone liver transplantation(LTx)due to AIH.We report a case of recurrent autoimmune hepatitis after LTx despite a persistently normal range of alanine aminotransferase(ALT).A 50-year-old woman was admitted to our hospital because of jaundice and severe liver dysfunction,where she was diagnosed with liver failure due to AIH.Steroid therapy was not effective enough and the patient received living-donor LTx in 1999.Following the operation,the level of ALT was maintained within a normal range and anti-nuclear antibody(ANA)became negative,however,the serum level of IgG gradually elevated and ANA became positive,while platelets decreased.A liver biopsy performed 6 years after LTx showed histological findings of AIH and she was diagnosed with recurrent AIH.A recurrence of AIH may occur after LTx even if the level of ALT remains within a normal range.We consider that a protocol liver biopsy should be performed in patients who undergo LTx due to AIH to decide the indication for steroid therapy. | Huaiqi Yao Kojiro Michitaka Yoshio Tokumoto Yosuke Murata Toshie Mashiba Masanori Abe Yoichi Hiasa Norio Horiike Morikazu Onji | 2007 | World Journal of Gastroenterology2007,13,10: | 3 |

The far-reaching contributions of bud sport mutants to horticulture and plant biology显示文摘A bud sport is a lateral shoot,inflorescence or single flower/fruit with a visibly different phenotype from the rest of the plant.The new phenotype is often caused by a stable somatic mutation in a single cell that is passed on to its clonal descendants and eventually populates part or all of a meristem.In many cases,a bud sport can be vegetatively propagated,thereby preserving the novel phenotype without sexual reproduction.Bud sports provide new characteristics while retaining the desirable qualities of the parent plant,which is why many bud sports have been developed into popular cultivars.We present an overview of the history of bud sports,the causes and methods of detecting somaclonal variation,and the types of mutant phenotypes that have arisen spontaneously.We focus on examples where the molecular or cytological changes causing the phenotype have been identified.Analysis of these sports has provided valuable insight into developmental processes,gene function and regulation,and in some cases has revealed new information about layer-specific roles of some genes.Examination of the molecular changes causing a phenotype and in some cases reversion back to the original state has contributed to our understanding of the mechanisms that drive genomic evolution. | Toshi M.Foster Maria JoséAranzana | 2018 | Horticulture Research2018,5,1: | 3 |

| 13 | Hepatocellular carcinoma with chronic B-type hepatitis complicated by autoimmune hemolytic anemia:A case report显示文摘A 57-year-old man consulted a local hospital because of a persistent slight fever. At the age of 37 years he was diagnosed having B-type hepatitis,but left the liver dysfunction untreated. Twenty years later,he was diagnosed having chronic hepatitis B,hepatocellular carcinoma (HCC) and macrocytic anemia,and referred to our hospital for further investigation. A HCC with a maximum diameter of 5.2 cm was detected in segment 8. Results of blood tests included 1.8 mg/dL serum total bilirubin,0.9 mg/dL bilirubin,less than 10 mg/dL haptoglobin,7.9 g/dL hemoglobin,130 fL MCV,and 14.5% reticulocytes. A bone marrow sample showed erythroid hyperplasia. The direct Coombs test gave a positive result. We diagnosed the anemia as autoimmmune hemolytic anemia (AIHA),for which prednisolone could not be administered due to positivity for HBsAg and HBeAg. After preparation of washed blood cells for later transfusion,the patient underwent systematic resection of segment 8. The cut surface of the resected specimen demonstrated an encapsulated yellow-brownish tumor measuring 52 mm×40 mm which was diagnosed pathologicaly as moderately differentiated HCC. On the 9th postoperative day,the patient's temperature rose to 38℃,and exacerbated hemolysis was observed. The maximum total bilirubin value was 5.8 mg/dL and minimum hemoglobin level was 4.6 g/dL. He tolerated this period without blood transfusion. Currently he is being followed up as an outpatient,and shows no signs of HCC recurrence or symptoms of anemia.
