Protective effects of Jiayan Kangtai granules on autoimmune thyroiditis in a rat model by modulating Th17/Treg cell balance显示文摘OBJECTIVE: To investigate the protective effects of Jiayan Kangtai(JYKT) granules, consisting of 9 Chinese herbs, in a rat model of autoimmune thyroiditis(AIT), and the possible underlying mechanism.METHODS: Female Lewis rats(6-8 weeks) were randomly apportioned to 5 groups of 10, including a normal control. AIT was induced in the untreated AIT-model group, and rats treated subsequently with daily low, medium, or high dose JYKT granules. After 12 weeks, plasma levels of thyroid autoantibodies and morphological changes in the thyroid were detected by enzyme-linked immunosorbent assay and histological examination, respectively. The presence of interleukin(IL)-6,IL23 p19, and IL-2 in thyroid tissue was assessed by immunohistochemical staining. The percentages of T helper(Th)17 cells and regulatory T cells(Tregs) in the peripheral blood were analyzed by flow cytometry. Relevant levels of cytokines and proteins were examined via bead-based multiplex flow cytometry and ELISA, respectively. Expressions of genes and proteins regulated by Th17 cells and Tregs were shown by real-time PCR and Western blot.RESULTS: Compared to the control, AIT-model rats had higher plasma concentrations of thyroid autoantibodies. The high-dose JYKT rats showed significantly lower levels of thyroid autoantibodies compared with the AIT model group. Rats in the AIT-JYKT groups also had fewer thyroid lesions and less lymphocytic infiltration, a lower percentage of Th17 cells, and a higher percentage of Tregs, compared with the AIT-model. Rats given high-dose JYKT had a significantly lower Th17/Treg ratio compared with the AIT model. Differences in plasma cytokine concentrations and relevant gene and protein expressions in the spleens of JYKT-treated rats and the AIT group suggested an association between JYKT treatment and lower Th17 cell percentage and higher Treg activity.CONCLUSION: JYKT treatment appeared to be protective against AIT in rats, possibly via the regulation of the Th17 cell/Treg imbalance in AIT. | Hou Yi Wang Tieshan Guo Xiangyu Sun Wen Guo Xuan Wu Lili Qin Lingling Zhang Chengfei Liu Tonghua | 2018 | Journal of Traditional Chinese Medicine2018,38,3: | 9

Predictor-based neural dynamic surface control for distributed formation tracking of multiple marine surface vehicles with improved transient performance显示文摘In this paper, we investigate the distributed formation tracking problem of multiple marine surface vehicles with model uncertainty and time-varying ocean disturbances induced by wind, waves, and ocean currents. The objective is to achieve a collective tracking with a time-varying trajectory, which can only be accessed by a fraction of follower vehicles. A novel predictor-based neural dynamic surface control design approach is proposed to develop the distributed adaptive formation controllers. We use prediction errors, rather than tracking errors, to construct the neural adaptive laws, which enable the fast identification of the vehicle dynamics without incurring high-frequency oscillations in control signals. We establish the stability properties of the closed-loop network via Lyapunov analysis, and quantify the transient performance by deriving the truncated L2 norms of the derivatives of neural weights, which we demonstrate to be smaller than the classical neural dynamic surface control design approach. We also extend the above result to the distributed formation tracking using the relative position information of vehicles, and the advantage is that the velocity information of neighbors and leader are required. Finally, we give the comparative studies to illustrate the performance improvement of the proposed method. | Zhouhua PENG Dan WANG Tieshan LI | 2016 | Science China(Information Sciences)2016,59,9: | 5
Suburbanization and Subcentering of Population in Beijing Metropolitan Area:A Nonparametric Analysis显示文摘This study focuses on the suburbanization and subcentering of population and examines the nature of spatial restructuring in terms of the population distribution in the Beijing metropolitan area.Instead of the classic density function approach,we employ the nonparametric analysis to characterize the spatial pattern of population densities in the Beijing metropolitan area and identify the suburban subcenters. Our findings suggest that the population has spread with rapid urban growth in the Beijing metropolitan area, and the compact urban form has been replaced by a more dispersed polycentric spatial distribution. However, compared with the decentralization of western cities, the spatial extent of the decentralization of population in the Beijing metropolitan area is quite limited. The rapid growth of population in the near suburbs has expedited the sprawl of the central city, with a larger central agglomeration of population dominating the metropolitan area.In this sense, the spatial pattern of the Beijing metropolitan area is still characterized by the continuous compactness. However, our findings do provide the evidence that the city has been turning to a polycentric structure. We find significant population subcenters have emerged in the suburbs of Beijing since the 1980s.But the polycentricity emerged in the Beijing metropolitan area is very different by nature from that observed in Western cities. The subcenters emerged are adherent to the development scheme planned for the city,so it can be referred to as the so called 'planned polycentricity'. | SUN Tieshan HAN Zhenhai WANG Lanlan LI Guoping | 2012 | Chinese Geographical Science2012,22,4: | 3
Path following control of underactuated ships based on nonswitch analytic model predictive control显示文摘A path following controller is developed for underactuated ships with only surge force and yaw moment available to follow a predefined path. The proposed controller is based on nonswitch analytic model predictive control. It is shown that the optimal control law for a nonlinear path following system with ill-defined relative degree is continuous and nonsingular. The problem of ill-defined relative degree is solved. The path-following ability of the nonlinear system is guaranteed.Numerical simulations are provided to demonstrate the effectiveness of the proposed control law. | Xiaofei WANG Zaojian ZOU Tieshan LI Weilin LUO | 2010 | 控制理论与应用(英文版)2010,8,4: | 3

The new sideway of CNTF sig-nal transduction pathway显示文摘The action of ciliary neurotrophic factor (CNTF) on intercellular free Ca2+ concentrations [Ca2+]1 induced by glutamate (Glu) in primary cultured hippocampal neurons were detected with Fura2/AM, a Ca2+-sensitive fluorophore. and the morphological influence of G-protein on it was ob-jected. Glu could induce rapid increase of [Ca2+]1 in hippo-campal neurons. CNTF had no significant action on [Ca2+]1 in resting hippocampal neurons. However, after incubation of CNTF for 5 min, the increase of [Ca2+]1 in hippocampal neurons rapidly induced by Glu was inhibited. Pretussis toxin (PTX)-sensitive G protein could block the action. These results indicate that a new non-genomic rapid sideway might exist in the upper stream of CNTF signal transduction pathway, which was related to Ca2+ signal transduction. | YAN Jin HE Cheng LU Changlin WANG Chenghai BAO Xuan TANG Tieshan HUANG Xiuying SUN Fangzhen | 2001 | Chinese Science Bulletin2001,46,6: | 1

The role of PARP1 in the DNA damage response and its application in tumor therapy显示文摘Single-strand break repair protein poly(ADP-ribose)polymerase 1(PARP1)catalyzes the poly(ADPribosyl)ation of many key proteins in vivo and thus plays important roles in multiple DNA damage response pathways, rendering it a promising target in cancer therapy. The tumor-suppressor effects of PARP inhibitors have attracted significant interest for development of novel cancer therapies. However, recent evidence indicated that the underlying mechanism of PARP inhibitors in tumor therapy is more complex than previously expected. The present review will focus on recent progress on the role of PARP1 in the DNA damage response and PARP inhibitors in cancer therapy. The emerging resistance of BRCA-deficient tumors to PARP inhibitors is also briefly discussed from the perspective of DNA damage and repair. These recent research advances will inform the selection of patient populations who can benefit from the PARP inhibitor treatment and development of effective drug combination strategies. | Zhifeng Wang Fengli Wang Tieshan Tang Caixia Guo | 2012 | Frontiers of Medicine2012,6,2: | 1

Pharmacological evaluation and mechanistic study of compound Xishu Granule in hepatocellular carcinoma显示文摘Objective:In this study,we used HepG2 human hepatocellular carcinoma cells to study the effects of Compound Xishu Granule(CXG)on cell proliferation, apoptosis, and the cell cycle in vitro. We also used a xenograft tumor model to study the anti-tumor effects of CXG and related mechanisms in vivo. Methods:The effect of CXG on cell viability was measured using Cell Counting Kit-8 and a colony formation assay. The effect of CXG on apoptosis and the cell cycle was analyzed using flow cytometry. The in vivo anti-tumor effect of CXG was assessed by measuring the volume change in xenograft tumors after drug administration. The CXG anti-tumor mechanism was studied using western blotting assay to detect cell cycle and apoptotic associated proteins. Results:CXG suppressed HepG2 cell proliferation in a time-and dose-dependent manner in vitro.Colony formation experiments showed that CXG administration for 24 h significantly reduced HepG2 cell formations(P<.01).Flow cytometric analysis showed that CXG treatment for 48 h promoted apoptosis and blocked HepG2 cells in the G2/M phase.Western blotting results showed that Bax was significantly upregulated and Bcl-2 was down-regulated in graft tumor tissues and HepG2 cells after CXG administration, which increased the Bax/Bcl-2 ratio.PLK1,CDC25 C,CDK1,and Cyclin B1 expression were upregulated.CXG had a good inhibitory effect on graft tumor growth in vivo. Conclusion:CXG has good anti-tumor effects in vitro and in vivo.In vitro,CXG promoted HepG2 cell apoptosis and induced G2/M phase arrest. In vivo,CXG significantly inhibited graft tumor growth. The CXG mechanism in treating hepatocellular carcinoma may be that CXG can induce abnormal apoptotic and cell cycle associated protein expression, leading to mitotic catastrophe and apoptosis. | Pin Li Yuanyuan Shi Baosheng Zhao Wenhui Xu Ziying Xu Jingxuan Zhang Zhaojuan Guo Yucong Bi Tieshan Wang Yu Qin Ting Wang | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,3: | 0
Preparation and Magnetic Properties of Cu-Ni Core-shell Nanowires in Ion-track Templates显示文摘Cu-Ni core-shell nanowires, with an inner Cu core diameter of about 60 nm and varying Ni shell thicknesses(10, 30, 50, 60, and 80 nm), were successfully fabricated in porous polycarbonate(PC) iontrack templates by a two-step etching and electrodeposition method. In our experiment, the thickness of Ni shell can be effectively tuned through the etching time of templates. The core-shell structure was confirmed by scanning electron microscopy(SEM) and transmission electron microscopy(TEM). The X-ray diffraction(XRD) pattern elucidates the co-existence of characteristic peaks for both Cu and Ni, indicating no other phases were formed during preparation. Magnetic hysteresis loops measured via vibrating sample magnetometry(VSM) revealed that Cu-Ni core-shell nanowires with thinner Ni shell exhibited obviously diamagnetic character and together with a weak ferromagnetic activity, whereas ferromagnetic behavior was primarily measured for the wires with thicker Ni shell. With increasing Ni shell thickness, the squareness and coercivity value became smaller due to the shape anisotropy and the formation of multi-domain structure. | 陈永辉 DUAN Jinglai YAO Huijun MO Dan WANG Tieshan SUN Youmei 刘杰 | 2015 | Journal of Wuhan University of Technology(Materials Science)2015,30,4: | 0
