| 1 | Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29 |
| 2 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 3 | Neural Mechanisms of Mental Fatigue Revisited: New Insights from the Brain Connectome显示文摘Maintaining sustained attention during a prolonged cognitive task often comes at a cost: high levels of mental fatigue. Heuristically, mental fatigue refers to a feeling of tiredness or exhaustion, and a disengagement from the task at hand;it manifests as impaired cognitive and behavioral performance. In order to effectively reduce the undesirable yet preventable consequences of mental fatigue in many real-world workspaces, a better understanding of the underlying neural mechanisms is needed, and continuous efforts have been devoted to this topic. In comparison with conventional univariate approaches, which are widely utilized in fatigue studies, convergent evidence has shown that multivariate functional connectivity analysis may lead to richer information about mental fatigue. In fact, mental fatigue is increasingly thought to be related to the deviated reorganization of functional connectivity among brain regions in recent studies. In addition, graph theoretical analysis has shed new light on quantitatively assessing the reorganization of the brain functional networks that are modulated by mental fatigue. This review article begins with a brief introduction to neuroimaging studies on mental fatigue and the brain connectome, followed by a thorough overview of connectome studies on mental fatigue. | 7 | Flexible and flame-retarding phosphorylated MXene/polypropylene composites for efficient electromagnetic interference shielding显示文摘Flame-retardant composites with high electromagnetic interference(EMI)shielding performance are desirable for electronic device packaging.Despite great potential of MXene for high EMI,it still remains a great challenge to develop high-performance flame-retardant polymer/MXene composites with excellent EMI shielding effectiveness because of the poor oxidative stability of MXene.Herein,phosphorylated MXene/polypropylene(PP)composites are prepared by coating phosphorylated MXene on PP fabric followed by spraying polyethylenimine(PEI)and hot-pressing.The phosphorylated MXene proves to be more durable against oxidation than pure MXene due to the protection effect of polyphosphates.Upon hot-pressing,melted PP fibers are fused together at their contact points and thus as-prepared composites are bi-continuous with two interpenetrating phases.The composites show significantly improved thermal stability and flame retardancy relative to pure PP,with a low total heat release(THR)of 3.7 kJ/g and a heat release rate(HRR)of 50.0 W/g,which are reduced by 78%and 87%,respectively.In addition,the composites exhibit a high electrical conductivity of~36,700 S/m and an EMI shielding performance of~90 d B over the whole frequency range of 8–12 GHz with a thickness of~400μm.The as-developed PP/MXene composites hold great promise for reliable protection of next-generation electronic devices working in complex environments. | Tingting Tang Shanchi Wang Yue Jiang Zhiguang Xu Yu Chen Tianshu Peng Fawad Khan Jiabing Feng Pingan Song Yan Zhao | 2022 | Journal of Materials Science & Technology2022,,16: | 1 |

| 11 | Carbon footprint of crop production in Heilongjiang land reclamation area,China显示文摘In the context of global warming,agriculture,as the second-largest source of greenhouse gas emissions after industry,had attracted widespread attention from all walks of life to reduce agricultural emissions.The carbon footprint of the planting production system of the Heilongjiang Land Reclamation Area(HLRA),an important commodity grain base in China,was evaluated and analyzed in this paper.On this basis,this paper sought feasible strategies to reduce carbon emissions from two aspects:agronomic practices and cropping structure adjustment,which were particularly crucial to promote the low-carbon and sustainable development of agriculture in HLRA.Therefore,using the accounting methods in IPCC and Low Carbon Development and Guidelines for the Preparation of Provincial Greenhouse Gas Inventories compiled by the Chinese government,relevant data were collected from 2000 to 2017 in HLRA and accounted for the carbon emissions of the planting production system in four aspects:carbon emissions from agricultural inputs,N_(2)O emissions from managed soils,CH_(4) emissions from rice cultivation and straw burning emissions.Then carbon uptake consisted of seeds and straws.Finally,with farmers'incomes were set as the objective function and carbon emissions per unit of gross production value was set as the constraint,this paper simulated and optimized the cropping structure in HLRA.The results showed that there was a“stable-growing-declining”trend in the total carbon emissions and carbon uptake of the planting production system in HLRA,with total carbon emissions of 2.84×10^(10) kg and total carbon uptake of 7.49×10^(10) kg in 2017.In the past 18 years,carbon emissions per unit area and carbon emissions per unit of gross production had both shown a decreasing trend.To achieve further efficiency gains and emission reductions in the planting production system,it was recommended that the local governments strengthen the comprehensive use of straw resources,optimize irrigation and fertilization techniques,and adjust the cropping structure,i.e.,increase the planting area of maize and soybeans and reduce the planting area of rice,and increase subsidies to protect the economic returns of planters. | Tianshu Chu Lu Yu Derui Wang Zengling Yang | 2022 | International Journal of Agricultural and Biological Engineering2022,15,1: | 0 |

| 17 | Landscape influences and management countermeasures of ginseng planting near Changbai mountain nature reserve显示文摘Ginseng planting in Northeast China brings economic benefits but affects forest landscape integrity and native ecological processes.In order to quantify the impacts of ginseng planting on the forest landscape,Fusong County in Jilin Province was selected as a study area.The number and distribution of ginseng fields over different time was quantified based on remote sensing and ground surveys.Grid analysis and multiple regression analysis were used to study the impacts of ginseng planting on the landscape.The results showed that altitudes and slopes of ginseng fields increased and became increasingly scattered and smaller closer to the Changbai Mountain Nature Reserve.Ginseng fields and abandoned fields increased total patches and total area of the local forested landscape,and shared edge lengths between ginseng fields and forests,resulting in continuous fragmentation of the landscape.Although the total area of existing and abandoned ginseng fields accounts for a small fraction of the total landscape,their negative impacts on ecosystem conservation is significant.The local government needs to rationally plan ginseng planting,scientifically implement the restoration of abandoned ginseng lands,and enhance awareness of ginseng farmers to environmental stewardship.Our study has important significance for maintaining the healthy and stable development of the local ginseng industry and for improving the quality of regional ecological environment. | Mingfang Tang Sha Qiu Lijie Liu Tao Li Shanlin Li Tianshu Yu | 2022 | Journal of Forestry Research2022,33,6: | 0 |
| 18 | Negative correlation between acetyl-CoA acyltransferase 2 and cetuximab resistance in colorectal cancer显示文摘The emergence of anti-EGFR therapy has revolutionized the treatment of colorectal cancer(CRC).However,not all patients respond consistently well.Therefore,it is imperative to conduct further research to identify the molecular mechanisms underlying the development of cetuximab resistance in CRC.In this study,we find that the expressions of many metabolism-related genes are downregulated in cetuximab-resistant CRC cells compared to their sensitive counterparts.Specifically,acetyl-CoA acyltransferase 2(ACAA2),a key enzyme in fatty acid metabolism,is downregulated during the development of cetuximab resistance.Silencing of ACAA2 promotes proliferation and increases cetuximab tolerance in CRC cells,while overexpression of ACAA2 exerts the opposite effect.RTK-Kras signaling might contribute to the downregulation of ACAA2 expression in CRC,and ACAA2 predicts CRC prognosis in patients with Kras mutations.Collectively,our data suggest that modulating ACAA2 expression contributes to secondary cetuximab resistance in Kras wild-type CRC patients.ACAA2 expression is related to Kras mutation and demonstrates a prognostic role in CRC patients with Kras mutation.Thus,ACAA2 is a potential target in CRC with Kras mutation. | Yitao Yuan Xun Sun Mengling Liu Suyao Li Yu Dong Keshu Hu Jiayu Zhang Bei Xu Sining Ma Hesheng Jiang Pengcong Hou Yufu Lin Lu Gan Tianshu Liu | 2023 | Acta Biochimica et Biophysica Sinica2023,55,9: | 0 |
