1The Arabidopsis catalase triple mutant reveals important roles of catalases and peroxisome-derived signaling in plant development显示文摘Hydrogen peroxide(H_2O_2)is generated in many metabolic processes.As a signaling molecule,H_2O_2plays important roles in plant growth and development,as well as environmental stress response.In Arabidopsis,there are three catalase genes,CAT1,CAT2,and CAT3.The encoded catalases are predominately peroxisomal proteins and are critical for scavenging H_2O_2.Since CAT1 and CAT3 are linked on chromosome 1,it has been almost impossible to generate cat1/3 and cat1/2/3 mutants by traditional genetic tools.In this study,we constructed cat1/3 double mutants and cat1/2/3 triple mutants by CRISPR/Cas9 to investigate the role of catalases.The cat1/2/3 triple mutants displayed severe redox disturbance and growth defects under physiological conditions compared with wild-type and the cat2/3 double mutants.Transcriptome analysis showed a more profound transcriptional response in the cat1/2/3 triple mutants compared to the cat2/3 mutants.These differentially expressed genes are involved in plant growth regulation as well as abiotic and biotic stress responses.In addition,expression of OXI1(OXIDATIVE SIGNAL INDUCIBLE 1)and several MAPK cascade genes were changed dramatically in the catalase triple mutant,suggesting that H_2O_2produced in peroxisomes could serve as a peroxisomal retrograde signal.Tong Su Pingping Wang Huijuan Li Yiwu Zhao Yao Lu Peng Dai Tianqi Ren Xiaofeng Wang Xuezhi Li Qun Shao Dazhong Zhao Yanxiu Zhao Changle Ma 2018Journal of Integrative Plant Biology2018,60,7:3
2Clinical significance of extended high-risk human papillomavirus genotyping and viral load in cervical cancer and precancerous lesions显示文摘Persistent infections with specific high-risk human papillomavirus(HR-HPV)strains are the leading cause of cervical cancer and precancerous lesions.HPV-16 and HPV-18 are associated with more than 70%of cervical cancer.However,with recent widespread vaccination efforts against cervical cancer,the infection rates of HPV-16 and HPV-18 have decreased across all age groups,while the infection rates of other HR-HPV strains have increased.The non-16/18 HR-HPV strains play an important role in cervical lesions.These strains can be identified with extended genotyping,and the 2019 American Society for Colposcopy and Cervical Pathology(ASCCP)guidelines recommended an HPV-based testing to assess the risk of cervical disease in patients.We reviewed and analyzed the clinical benefits of applying extended HR-HPV genotyping,which was published by the International Agency for Research on Cancer(HPV-16,18,31,33,35,39,45,51,52,56,58,59,66,and 68),to cervical cancer screening.This review concluded that cervical cancer screening needs to include extended HR-HPV genotyping.The examination of extended HR-HPV genotyping in cervical intraepithelial lesions and cervical cancers can help guide clinical practices.Pingping Su Jincheng Ma Lirui Yu Shuting Tang Pengming Sun 2023Gynecology and Obstetrics Clinical Medicine2023,3,1:2

8Crosstalk between IL-15Rα^(+)tumor-associated macrophages and breast cancer cells reduces CD8+T cell recruitment显示文摘Background:Interleukin-15(IL-15)is a promising immunotherapeutic agent owing to its powerful immune-activating effects.However,the clinical benefits of these treatments are limited.Crosstalk between tumor cells and immune cells plays an important role in immune escape and immunotherapy drug resistance.Herein,this study aimed to obtain in-depth understanding of crosstalk in the tumor microenvironment for providing potential therapeutic strategies to prevent tumor progression.Methods:T-cell killing assays and co-culture models were developed to determine the role of crosstalk between macrophages and tumor cells in breast cancer resistant to IL-15.Western blotting,histological analysis,CRISPR-Cas9 knockout,multi-parameter flow cytometry,and tumor cell-macrophage co-injection mouse models were developed to examine the mechanism by which IL-15Rα^(+)tumor-associated macrophages(TAMs)regulate breast cancer cell resistance to IL-15.Results:We found thatmacrophages contributed to the resistance of tumor cells to IL-15,and tumor cells induced macrophages to express high levels of theαsubunit of the IL-15 receptor(IL-15Rα).Further investigation showed that IL-15Rα^(+)TAMs reduced the protein levels of chemokine CX3C chemokine ligand 1(CX3CL1)in tumor cells to inhibit the recruitment of CD8^(+)T cells by releasing the IL-15/IL-15Rαcomplex(IL-15Rc).Administration of an IL-15Rc blocking peptide markedly suppressed breast tumor growth and overcame the resistance of cancer cells to anti-programmed cell death protein 1(PD-1)antibody immunotherapy.Interestingly,Granulocyte-macrophage colony-stimulating factor(GMCSF)inducedγchain(γc)expression to promote tumor cell-macrophage crosstalk,which facilitated tumor resistance to IL-15.Additionally,we observed that the non-transcriptional regulatory function of hypoxia inducible factor-1alpha(HIF-1α)was essential for IL-15Rc to regulateCX3CL1 expression in tumor cells.Conclusions:The IL-15Rc-HIF-1α-CX3CL1 signaling pathway serves as a crosstalk between macrophages and tumor cells in the tumormicroenvironment of breast cancer.Targeting this pathway may provide a potential therapeutic strategy for enhancing the efficacy of cancer immunotherapy.Wenlong Zhang Qing Zhang Nanfei Yang Qian Shi Huifang Su Tingsheng Lin Zhonglei He Wenxin Wang Hongqian Guo Pingping Shen 2022Cancer Communications2022,42,6:0
9Isolation of Swine Corynebacteria and Analysis of 16S rDNA Gene显示文摘A strain of gram-positive bacillus was isolated from suppurative lung organs of nursery pigs in a pig farm,which was further characterized by morphological observation,cultivation test,biochemical test,drug sensitivity test,pathogenicity test and 16 S rDNA gene cloning and sequence analysis. The results showed that the isolate grew well in rabbit blood agar plate and horse serum tryptone soybean agar( TSA) plate under aerobic condition,which could lead to mortality of mice and were susceptible to cephalosporin antibiotics and fluoroquinolone antibiotics. 10Exploration of sea anemone-inspired high-performance biomaterials with enhanced antioxidant activity显示文摘Antioxidant biomaterials have attracted much attention in various biomedical fields because of their effective inhibition and elimination of reactive oxygen species(ROS)in pathological tissues.However,the difficulty in ensuring biocompatibility,biodegradability and bioavailability of antioxidant materials has limited their further development.Novel bioavailable antioxidant materials that are derived from natural resources are urgently needed.Here,an integrated multi-omics method was applied to fabricate antioxidant biomaterials.A key cysteine-rich thrombospondin-1 type Ⅰ repeat-like(TSRL)protein was efficiently discovered from among 1262 adhesive components and then used to create a recombinant protein with a yield of 500 mg L^(-1).The biocompatible TSRL protein was able to self-assemble into either a water-resistant coating through Ca^(2+)-mediated coordination or redox-responsive hydrogels with tunable physical properties.The TSRL-based hydrogels showed stronger 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging rates than glutathione(GSH)and ascorbic acid(Aa)and protected cells against external oxidative stress significantly more effectively.When topically applied to mice skin,TSRL alleviated epidermal hyperplasia and suppressed the degradation of collagen and elastic fibers caused by ultraviolet radiation B(UVB)irradiation,confirming that it enhanced antioxidant activity in vivo.This is the first study to successfully characterize natural antioxidant biomaterials created from marine invertebrate adhesives,and the findings indicate the excellent prospects of these biomaterials for great applications in tissue regeneration and cosmeceuticals.Lulu Wang Xiaokang Zhang Pingping Xu Jicheng Yan Yuzhong Zhang Hainan Su Chengjun Sun Qiang Lu Weizhi Liu 2022Bioactive Materials2022,7,4:0

14卵巢癌术前腹腔化疗的临床应用及其评价(英文)显示文摘Objective:To investigate the clinical efficacy of intraperitoneal chemotherapy before surgery for ovarian cancer.Methods:60 patients with stages II-IV of ovarian cancer were treated with intraperitoneal chemotherapy of CAP or TP regimen followed by a surgery treatment and another chemotherapy for 6 cycles.And then the efficiency of the therapy was evaluated by analyzing the changes of ascites, the serum CA-125 and CA-19-9 levels and the findings in the operation, and investigating the recurrence of cancer and the survival.Results:After 1-3 cycles of intraperitoneal chemotherapy, serum levels of CA-125 and CA-19-9 and carcinous ascites significantly reduced in all patients, ascites reduce was over 50% in 98.3% cases;all cases were successfully treated with cytoreduction and it was found during the operation that bulky tumor was reduced and looser so as to be easily isolated and removed;PFS of the patients was prolonged, while the toxicity and side-effects were not so serious as beyond the patient toleration.Conclusion:Intraperitoneal chemotherapy before surgery for ovarian cancer has an active efficacy in clinic, being able to improve conditions of surgery and increase the opportunity of maximal cytoreduction, and prolong survival of the patients, and should be a good selection for the treatment of advanced ovarian cancer.Dianyun Shi Mei Xiao Saiying Chen Bin Yang Qijiang Zhu Pingping Xu Chenfang Su 2009The Chinese-German Journal of Clinical Oncology2009,8,5:0
