| 1 | Porphyromonas gingivalis exacerbates ulcerative colitis via Porphyromonas gingivalis peptidylarginine deiminase显示文摘Abstract:Ulcerative Colitis(UC)has been reported to be related to Porphyromonas gingivalis(P.gingivalis).Porphyromonas gingivalis peptidylarginine deiminase(PPAD),a virulence factor released by P.gingivalis,is known to induce inflammatory responses.To explore the pathological relationships between PPAD and UC,we used homologous recombination technology to construct a P.gingivalis strain in which the PPAD gene was deleted(Δppad)and aΔppad strain in which the PPAD gene was restored(comΔppad).C57 BL/6 mice were orally gavaged with saline,P.gingivalis,Δppad,or comΔppad twice a week for the entire 40 days(days 0-40),and then,UC was induced by dextran sodium sulfate(DSS)solution for 10 days(days 31-40).P.gingivalis and comΔppad exacerbated DDS-induced colitis,which was determined by assessing the parameters of colon length,disease activity index,and histological activity index,butΔppad failed to exacerbate DDS-induced colitis.Flow cytometry and ELISA revealed that compared withΔppad,P.gingivalis,and comΔppad increased T helper 17(Th17)cell numbers and interleukin(IL)-17 production but decreased regulatory T cells(Tregs)numbers and IL-10 production in the spleens of mice with UC.We also cocultured P.gingivalis,Δppad,or comΔppad with T lymphocytes in vitro and found that P.gingivalis and comΔppad significantly increased Th17 cell numbers and decreased Treg cell numbers.Immunofluorescence staining of colon tissue paraffin sections also confirmed these results.The results suggested that P.gingivalis exacerbated the severity of UC in part via PPAD. | Xida Zhao Jingbo Liu Chong Zhang Ning Yu Ze Lu Shuwei Zhang Yuchao Li Qian Li Junchao Liu Dongjuan Liu Yaping Pan | 2021 | International Journal of Oral Science2021,13,3: | 5 |
| 2 | CsbZIP1-CsMYB12 mediates the production of bitter-tasting flavonols in tea plants (Camellia sinensis) through a coordinated activator–repressor network显示文摘Under high light conditions or UV radiation,tea plant leaves produce more flavonols,which contribute to the bitter taste of tea;however,neither the flavonol biosynthesis pathways nor the regulation of their production are well understood.Intriguingly,tea leaf flavonols are enhanced by UV-B but reduced by shading treatment.CsFLS,CsUGT78A14,CsMYB12,and CsbZIP1 were upregulated by UV-B radiation and downregulated by shading.CsMYB12 and CsbZIP1 bound to the promoters of CsFLS and CsUGT78A14,respectively,and activated their expression individually.CsbZIP1 positively regulated CsMYB12 and interacted with CsMYB12,which specifically activated flavonol biosynthesis.Meanwhile,CsPIF3 and two MYB repressor genes,CsMYB4 and CsMYB7,displayed expression patterns opposite to that of CsMYB12.CsMYB4 and CsMYB7 bound to CsFLS and CsUGT78A14 and repressed their CsMYB12-activated expression.While CsbZIP1 and CsMYB12 regulated neither CsMYB4 nor CsMYB7,CsMYB12 interacted with CsbZIP1,CsMYB4,and CsMYB7,but CsbZIP1 did not physically interact with CsMYB4 or CsMYB7.Finally,CsPIF3 bound to and activated CsMYB7 under shading to repress flavonol biosynthesis.These combined results suggest that UV activation and shading repression of flavonol biosynthesis in tea leaves are coordinated through a complex network involving CsbZIP1 and CsPIF3 as positive MYB activators and negative MYB repressors,respectively.The study thus provides insight into the regulatory mechanism underlying the production of bitter-tasting flavonols in tea plants. | Xuecheng Zhao Xiangsheng Zeng Ning Lin Shuwei Yu Alisdair RFernie Jian Zhao | 2021 | Horticulture Research2021,8,1: | 4 |
| 3 | CsTCPs regulate shoot tip development and catechin biosynthesis in tea plant (Camellia sinensis)显示文摘The growth of leaves and biosynthesis of characteristic secondary metabolites are critically important for tea production and quality control.However,little is known about the coordinated regulation of leaf development and catechin biosynthesis in tea plants.Here,we reported that TCP TFs are involved in both catechin biosynthesis and leaf development.An integrated analysis of catechin profiling and CsTCP expression in different tissues of plants under various environmental conditions at different developmental stages indicated significant correlations between the transcript levels of CIN-type TCPs and catechin production.CIN-type CsTCP3 and CsTCP4 and PCF-type CsTCP14 interacted with the MYB-bHLH-WD40 repeat(MBW)complex by forming a CsTCP3-CsTT8 heterodimer and modulating the transactivation activity of the promoters of anthocyanin synthase(CsANSl)and anthocyanidin reductase(CsANRl).Four types of microRNA/target modules,miR319b/CsTCP3-4,miR164b/CsCUC,miR396/CsGRF-GIF,and miR165b/HD-ZIPIII ones,were also identified and characterized for their functions in the regulation of the development of teaplant shoot tips and leaf shape.The results of these modules were reflected by their different expression patterns in developing buds and leaves that had distinctly different morphologies in three different teaplant varieties.Their roles in the regulation of catechin biosynthesis were also further verified by manipulation of microRNA319b(miR319b),which targets the transcripts of CsTCP3 and CsTCP4.Thus,CsTCPs represent at least one of these important groups of TFs that can integrate tea plant leaf development together with secondary metabolite biosynthesis.Our study provides new insight into shoot tip development and catechin production in tea plants and lays a foundation for further mechanistic understanding of the regulation of tea plant leaf development and secondary metabolism. | Shuwei Yu Penghui Li Xuecheng Zhao Mangmang Tan Muhammad Zulfiqar Ahmad Yujie Xu Million Tadege Jian Zhao | 2021 | Horticulture Research2021,8,1: | 2 |
| 4 | miR-7/TGF-β2 axis sustains acidic tumor microenvironment-induced lung cancer metastasis显示文摘Acidosis,regardless of hypoxia involvement,is recognized as a chronic and harsh tumor microenvironment(TME)that educates malignant cells to thrive and metastasize.Although overwhelming evidence supports an acidic environment as a driver or ubiquitous hallmark of cancer progression,the unrevealed core mechanisms underlying the direct effect of acidification on tumorigenesis have hindered the discovery of novel therapeutic targets and clinical therapy.Here,chemical-induced and transgenic mouse models for colon,liver and lung cancer were established,respectively.miR-7 and TGF-β2 expressions were examined in clinical tissues(n=184).RNA-seq,miRNA-seq,proteomics,biosynthesis analyses and functional studies were performed to validate the mechanisms involved in the acidic TME-induced lung cancer metastasis.Our data show that lung cancer is sensitive to the increased acidification of TME,and acidic TME-induced lung cancer metastasis via inhibition of miR-7-5 p.TGF-β2 is a direct target of miR-7-5 p.The reduced expression of miR-7-5 p subsequently increases the expression of TGF-β2 which enhances the metastatic potential of the lung cancer.Indeed,overexpression of miR-7-5 p reduces the acidic p H-enhanced lung cancer metastasis.Furthermore,the human lung tumor samples also show a reduced miR-7-5 p expression but an elevated level of activated TGF-β2;the expressions of both miR-7-5 p and TGF-β2 are correlated with patients’survival.We are the first to identify the role of the miR-7/TGF-β2 axis in acidic p H-enhanced lung cancer metastasis.Our study not only delineates how acidification directly affects tumorigenesis,but also suggests miR-7 is a novel reliable biomarker for acidic TME and a novel therapeutic target for non-small cell lung cancer(NSCLC)treatment.Our study opens an avenue to explore the p H-sensitive subcellular components as novel therapeutic targets for cancer treatment. | Tao Su Suchao Huang Yanmin Zhang Yajuan Guo Shuwei Zhang Jiaji Guan Mingjing Meng Linxin Liu Caiyan Wang Dihua Yu Hiu-Yee Kwan Zhiying Huang Qiuju Huang Elaine Lai-Han Leung Ming Hu Ying Wang Zhongqiu Liu Linlin Lu | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 2 |
| 5 | β-delayed proton decays near the proton drip line显示文摘We briefly reviewed the experimental study on a-delayed proton decays near the proton drip line published by our group during the period of 1996―2004, namely the first observation of the a-delayed proton decays of 9 new nuclides in the rare-earth region and the new measurements of a-delayed proton decays of 5 nuclides in the mass ~90 region near the N = Z line with the aid of the “p-?” coincidence in combination with a He-jet tape transport system. In the meantime some important experimental technique details were supplemented. The experimental results, including the half -lives, spins, parities, deformations and production reaction cross sections for the 14 nuclei were summarized and compared with the current nuclear-model predictions, and then the following points were represented. (1) The experimental half-lives for 85Mo and 92Rh as well as the predicted “waiting point” nuclei 89Ru and 93Pd are 5―10 times longer than the theoretical predictions given by M?ller et al. using a macroscopic-microscopic model. It considerably influences the predictions of the abundances of the nuclides produced in the rp-process. (2) The current-model predictions are not consistent with the experimental assignments of the spins and parities for the proton drip-line nuclei 142Ho and 128Pm. However, the nuclear potential energy surface (PES) calculated by using a Woods- Saxon-Strutinsky method reproduced the experimental results. | 7 | Growth of SiC as Binder to Adhere Diamond Particle and Tribological Properties of Diamond Particles Coated SiC显示文摘The purpose of this work was to grow SiC as binder to adhere diamond particles to graphite substrate by low pressure chemical vapor deposition(LPCVD) at 1100 °C and 100 Pa using methyltrichlorosilane(MTS: CH3 SiCl3) as precursor. The composite coatings on graphite substrates were analyzed by various techniques. Results show that a dense SiC coating with a cloud-cluster shape was formed both on the diamond particles and the substrate after deposition. The thermal stress(290.6 MPa) strengthened the interfacial bonding between the diamond particle and the SiC coating, which is advantageous for the purpose of adhering diamond particles to graphite substrate. The applied load of sliding wear test was found to affect not only the friction coefficient, but also the wear surface morphology.
