1A Critical Time-Window for the Selective Induction of Hippocampal Memory Consolidation by a Brief Episode of Slow-Wave Sleep显示文摘Although extensively studied, the exact role of sleep in learning and memory is still not very clear. Sleep deprivation has been most frequently used to explore the effects of sleep on learning and memory, but the results from such studies are inevitably complicated by concurrent stress and distress. Furthermore, it is not clear whether there is a strict time-window between sleep and memory consolidation. In the present study we were able to induce time-locked slow-wave sleep(SWS) in mice by optogenetically stimulating GABAergic neurons in the parafacial zone(PZ), providing a direct approach to analyze the influences of SWS on learning and memory with precise time-windows. We found that SWS induced by light for 30 min immediately or 15 min after the training phase of the object-in-place task significantly prolonged the memory from 30 min to 6 h. However, induction of SWS 30 min after the training phase did not improve memory, suggesting a critical time-window between the induction of a brief episode of SWS and learning for memory consolidation.Application of a gentle touch to the mice during light stimulation to prevent SWS induction also failed to improve memory, indicating the specific role of SWS,but not the activation of PZ GABAergic neurons itself, in memory consolidation. Similar influences of light-induced SWS on memory consolidation also occurred for Y-maze spatial memory and contextual fear memory, but not for cued fear memory. SWS induction immediately before the test phase had no effect on memory performance, indicating that SWS does not affect memory retrieval. 2EXPONENTIAL STABILITY OF LINEAR DISTRIBUTED PARAMETER SWITCHED SYSTEMS WITH TIME-DELAY显示文摘In this paper, the exponential stability analysis for ODE switched systems with time delay is extended to distributed parameter switched systems(DPSS) in Hilbert space. For a given family of exponential stable subsystems, this paper focuses on finding conditions to guarantee the overall DPSS' exponential stability. Based on semigroup theory, by applying piecewise Lyapunov-Krasovskii functionals method incorporated average dwell time approach, sufficient conditions for exponential stability are derived. These conditions are given in the form of linear operator inequalities(LOIs)where the decision variables are operators in Hilbert space, and the stability properties depend on switching rule. Being applied to heat switched propagation equations, these LOIs are reduced to standard Linear Matrix Inequalities(LMIs). 3Risk assessment of Giardia in rivers of southern China based on continuous monitoring显示文摘The occurrence and risks of Giardia in China have been unclear to date,which has made it difficult to properly manage source water as well as to create reasonable drinking water standards.The levels of Giardia in river networks of several cities in Zhejiang Province,China were found to be in the range of 0-5 oocysts/10 L in the rainy season in 2008.The mortality due to Giardia infection for people in this region was calculated to be from 0 to 1.95 × 10?8 persons using a conditional probability equation.Based on multiple unboiled water intake routes,the disability-adjusted life years(DALYs) due to Giardia infection for people who consumed conventionally treated water was 0.625(95% CI:0.137-2.05) per 105 persons,with the symptom of hospitalization making the highest contribution to total DALYs(0.56 per 105 persons;95% CI:0.122-1.84).The DALYs decreased to 0.425(95% CI:0.137-2.05) per 105 persons per year for those consuming water treated with advanced technology.These values were lower than the acceptable risk(1.97 × 10?5 DALYs per year).This study revealed the risk of Giardia infection to the people in river networks of Zhejiang Province for the first time,and provides a method to evaluate the risk of Giardia infection.The results are useful for the modification of drinking water quality standards based on cost-benefit analysis.Wei An Dongqing Zhang Shumin Xiao Jianwei Yu Min Yang 2012Journal of Environmental Sciences2012,24,2:8
4Characteristics of Caragana korshinskii and Hippophae rhamnoides stemflow and their significance in soil moisture enhancement in Loess Plateau, China显示文摘Stemflow of xerophytic shrubs represents a significant component of water replenishment to the soil-root system and influences water utilization of plant roots at the stand scale,especially in water-scarce semi-arid ecosystems.The stemflow of two semi-arid shrubs(Caragana korshinskii and Hippophae rhamnoides)and its effect on soil moisture enhancement were evaluated during the growing season of 2011 in the semi-arid loess region of China.The results indicated that stemflow averaged 12.3%and 8.4%of the bulk precipitation for C.korshinskii and H.rhamnoides,respectively.Individual stemflow increased in a linear function with increasing rainfall depth.The relationship between funneling ratios and rainfall suggested that there existed a rainfall depth threshold of 11 mm for both C.korshinskii and H.rhamnoides.Averaged funneling ratios were 156.6±57.1 and49.5±30.8 for C.korshinskii and H.rhamnoides,respectively,indicating that the canopy architecture of the two shrubs was an effective funnel to channel stemflow to the root area,and C.korshinskii showed a greater potential to use stemflow water in the semi-arid conditions.For individual rainfall events,the wetting front depths were approximately 2 times deeper in the rooting zone around the stems than in the bare area outside canopy for both C.korshinskii and H.rhamnoides.Correspondingly,soil water content was also significantly higher in the root area around the shrub stem than in the area outside the shrub canopy.This confirms that shrub stemflow conserved in the deep soil layers may be an available moisture source for plant growth under semi-arid conditions.ShengQi JIAN ChuanYan ZHAO ShuMin FANG Kai YU 2014Journal of Arid Land2014,6,1:6

6Differentiation of neuron-like cells from mouse parthenogenetic embryonic stem cells显示文摘Parthenogenetic embryonic stem cells have pluripotent differentiation potentials,akin to fertilized embryo-derived embryonic stem cells.The aim of this study was to compare the neuronal differentiation potential of parthenogenetic and fertilized embryo-derived embryonic stem cells.Before differentiation,karyotype analysis was performed,with normal karyotypes detected in both parthenogenetic and fertilized embryo-derived embryonic stem cells.Sex chromosomes were identified as XX.Immunocytochemistry and quantitative real-time PCR detected high expression of the pluripotent gene,Oct4,at both the mRNA and protein levels,indicating pluripotent differentiation potential of the two embryonic stem cell subtypes.Embryonic stem cells were induced with retinoic acid to form embryoid bodies,and then dispersed into single cells.Single cells were differentiated in N2 differentiation medium for 9 days.Immunocytochemistry showed parthenogenetic and fertilized embryo-derived embryonic stem cells both express the neuronal cell markers nestin,βIII-tubulin and myelin basic protein.Quantitative real-time PCR found expression of neurogenesis related genes(Sox-1,Nestin,GABA,Pax6,Zic5 and Pitx1) in both types of embryonic stem cells,and Oct4 expression was significantly decreased.Nestin and Pax6 expression in parthenogenetic embryonic stem cells was significantly higher than that in fertilized embryo-derived embryonic stem cells.Thus,our experimental findings indicate that parthenogenetic embryonic stem cells have stronger neuronal differentiation potential than fertilized embryo-derived embryonic stem cells.Xingrong Yan Yanhong Yang Wei Liu Wenxin Geng Huichong Du Jihong Cui Xin Xie Jinlian Hua Shumin Yu Liwen Li Fulin Chen 2013Neural Regeneration Research2013,8,4:4
7Risk assessment of Giardia from a full scale MBR sewage treatment plant caused by membrane integrity failure显示文摘Membrane bioreactors(MBR) are highly efficient at intercepting particles and microbes and have become an important technology for wastewater reclamation. However, many pathogens can accumulate in activated sludge due to the long residence time usually adopted in MBR, and thus may pose health risks when membrane integrity problems occur.This study presents data from a survey on the occurrence of water-borne Giardia pathogens in reclaimed water from a full-scale wastewater treatment plant with MBR experiencing membrane integrity failure, and assessed the associated risk for green space irrigation. Due to membrane integrity failure, the MBR effluent turbidity varied between 0.23 and 1.90 NTU over a period of eight months. Though this turbidity level still met reclaimed water quality standards(≤ 5 NTU), Giardia were detected at concentrations of 0.3 to 95 cysts/10 L, with a close correlation between effluent turbidity and Giardia concentration. All β-giardin gene sequences of Giardia in the WWTP influents were genotyped as Assemblages A and B, both of which are known to infect humans. An exponential dose–response model was applied to assess the risk of infection by Giardia. The risk in the MBR effluent with chlorination was9.83 × 10-3, higher than the acceptable annual risk of 1.0 × 10-4. 8Rabdosia serra alleviates dextran sulfate sodium salt-induced colitis in mice through anti-inflammation,regulating Th17/Treg balance,maintaining intestinal barrier integrity,and modulating gut microbiota显示文摘Rabdosia serra(R.serra),an important component of Chinese herbal tea,has traditionally been used to treat hepatitis,jaundice,cholecystitis,and colitis.However,the chemical composition of R.serra and its effect against colitis remain unclear.In this study,the chemical composition of the water extract of R.serra was analyzed using ultra performance liquid chromatography coupled with a hybrid linear ion trap quadrupole-orbitrap mass spectrometer(UPLC-LTQ-Orbitrap-MS).A total of 46 compounds,comprising ent-kaurane diterpenoids,flavonoids,phenolic acids,and steroids,were identified in the water extract of R.serra,and the extract could significantly alleviate dextran sulfate sodium salt-induced colitis by improving colon length,upregulating anti-inflammatory factors,downregulating proinflammatory factors,and restoring the balance of T helper 17/T regulatory cells.R.serra also preserved intestinal barrier function by increasing the level of tight junction proteins(zonula occludens 1 and occludin)in mouse colonic tissue.In addition,R.serra modulated the gut microbiota composition by increasing bacterial richness and diversity,increasing the abundance of beneficial bacteria(Muribaculaceae,Bacteroides,Lactobacillus,and Prevotellaceae_UCG-001),and decreasing the abundance of pathogenic bacteria(Turicibacter,Eubacterium_fissicatena_group,and Eubacterium_xylanophilum_group).Gut microbiota depletion by antibiotics further confirmed that R.serra alleviated colitis in a microbiota-dependent manner.Overall,our findings provide chemical and biological evidence for the potential application of R.serra in the management of colitis.Hongyi Li Yi Wang Shumin Shao Hui Yu Deqin Wang Chuyuan Li Qin Yuan Wen Liu Jiliang Cao Xiaojuan Wang Haibiao Guo Xu Wu Shengpeng Wang 2022Journal of Pharmaceutical Analysis2022,12,6:4

10SRSF1 plays a critical role in invariant natural killer T cell development and function显示文摘Invariant natural killer T(iNKT)cells are highly conserved innate-like T lymphocytes that originate from CD4^(+)CD8^(+)double-positive(DP)thymocytes.Here,we report that serine/arginine splicing factor 1(SRSF1)intrinsically regulates iNKT cell development by directly targeting Myb and balancing the abundance of short and long isoforms.Conditional ablation of SRSF1 in DP cells led to a substantially diminished iNKT cell pool due to defects in proliferation,survival,and TCRαrearrangement.The transition from stage 0 to stage 1 of iNKT cells was substantially blocked,and the iNKT2 subset was notably diminished in SRSF1-deficient mice.SRSF1 deficiency resulted in aberrant expression of a series of regulators that are tightly correlated with iNKT cell development and iNKT2 differentiation,including Myb,PLZF,Gata3,ICOS,and CD5.In particular,we found that SRSF1 directly binds and regulates pre-mRNA alternative splicing of Myb and that the expression of the short isoform of Myb is substantially reduced in SRSF1-deficient DP and iNKT cells.Strikingly,ectopic expression of the Myb short isoform partially rectified the defects caused by ablation of SRSF1.Furthermore,we confirmed that the SRSF1-deficient mice exhibited resistance to acute liver injury uponα-GalCer and Con A induction.Our findings thus uncovered a previously unknown role of SRSF1 as an essential post-transcriptional regulator in iNKT cell development and functional differentiation,providing new clinical insights into iNKT-correlated disease.Jingjing Liu Menghao You Yingpeng Yao Ce Ji Zhao Wang Fang Wang Di Wang Zhihong Qi Guotao Yu Zhen Sun Wenhui Guo Juanjuan Liu Shumin Li Yipeng Jin Tianyan Zhao Hai-Hui Xue Yuanchao Xue Shuyang Yu 2021Cellular & Molecular Immunology2021,18,11:3
11Congestive heart failure in COX2 deficient rats显示文摘Non-steroidal anti-inflammatory drugs(NSAIDs)inhibit prostaglandin(PG)formation by targeting cyclooxygenase(COX)1 and 2.Long-term use of NSAIDs that selectively inhibit COX2 increases the risk for thrombotic events,cardiac failure,and hypertension.However,the underlying mechanisms remain unclear.In this study,COX1-and COX2-deficient rats were created via Cas9/RNA-mediated gene targeting.DNA genotyping and Western blot analysis confirmed successful generation of COX1^(-/-) and COX2^(-/-) rats.Adult COX1^(-/-) rats grew normally,while more than 70%of COX2^(-/-) rats after wean died within 2 months.Echocardiography showed markedly reduced left ventricular ejection fraction and fractional shortening in adult COX2^(-/-) rats compared to those in wildtype(WT)controls.Histological analysis revealed accumulation of inflammatory cells and severe interstitial and perivascular fibrosis in COX2^(-/-)cardiac tissues.Moreover,cardiac ATP and acetyl-Co A production was dramatically decreased in COX2^(-/-) rats.Consistently,the expression of genes related to mitochondrial oxidation,such as those that encode for subunits of pyruvate dehydrogenase complex and acyl Co A dehydrogenases,were downregulated,while glycolytic hexokinase 1(HK1)was upregulated in COX2^(-/-) heart tissues.These observations indicate that COX2-deficient rats developed spontaneously heart failure,likely as a result of dysregulated cardiac energy metabolism.Qiangyou Wan Deping Kong Qian Liu Shumin Guo Chenchen Wang Yan Zhao Zun-Ji Ke Ying Yu 2021Science China(Life Sciences)2021,64,7:3
12Anxiolytic Effect of Increased NREM Sleep after Acute Social Defeat Stress in Mice显示文摘Social defeat stress(SDS)plays a major role in the pathogenesis of psychiatric disorders like anxiety and depression.Sleep is generally considered to involve recovery of the brain from prior experience during wakefulness and is altered after acute SDS.However,the effect of acute SDS on sleep/wake behavior in mice varies between studies.In addition,whether sleep changes in response to stress contribute to anxiety is not well established.Here,we first investigated the effects of acute SDS on sleep/wake states in the active period in mice.Our results showed that total sleep time(time in rapid eyemovement[REM]and non-REM[NREM]sleep)increased in the active period after acute SDS.NREM sleep increased mainly during the first 3 h after SDS,while REM sleep increased at a later time.Then,we demonstrated that the increased NREM sleep had an anxiolytic benefit in acute SDS.Mice deprived of sleep for 1 h or 3 h after acute SDS remained in a highly anxious state,while in mice with ad libitum sleep the anxiety rapidly faded away.Altogether,our findings suggest an anxiolytic effect of NREM sleep,and indicate a potential therapeutic strategy for anxiety.Xiang Feng Hui-Ying Zhao Yu-Jin Shao Hui-Fang Lou Li-Ya Zhu Shumin Duan Yan-Qin Yu 2020Neuroscience Bulletin2020,36,10:2
13Modulation of Fano resonances in symmetry-broken gold-SiO_2-gold nanotube dimers显示文摘Fano resonances in the symmetry-broken gold-SiO2-gold(BGSG)nanotubes and the associated dimers have been investigated based on the finite element method.In the BGSG nanotube,the symmetry breaking induced the interactions of the inner gold core and outer gold nanoshell plasmons of all multipolar orders and hence the red-shifts of the plasmon resonance modes and the enhanced quadrupole mode peaks were observed.The interference of the quadrupole mode peak with the subradiant dipole mode caused a Fano-dip in the scattering spectrum.By increasing the core offset-value in the BGSG nanotube,the Fano dip with low energy showed a red-shift and became deeper.Unexpectedly the plasmon coupling between a GSG nanotube and a BGSG nanotube can lead to two strong Fano dips in the scattering spectra of the dimer.It was further noted that the thin side of the BGSG nanotube located at two sides of the dimer gap can lead to the strong near-field coupling between two BGSG nanotubes and hence a deeper and broader Fano dip.WU DaJian YU HaiQun JIANG ShuMin WU XueWei LIU XiaoJun 2014Science China(Physics,Mechanics & Astronomy)2014,57,6:2

15XPS Studies of Magnetic Multilayers显示文摘NiOx/N81Fe19 and Co/AlOx/Co magnetic multilayers were fabricated by reactive RF/DC magnetron sputtering on clean glass substrates and oxidized Si (100) substrates, respectively. The exchange biasing field (Hex) between NiOx, and Ni81Fe19 as a function of NiOx, oxidation states was studied by X-ray photoelectron spectroscopy (XPS). The oxidation states and the oxide thickness of Al layers in magnetic multilayer films consisting of Co/AlOx/Co were also analyzed. It is found that the Hex of NiOx/Ni81Fe19 films only depends on Ni2+ but not on Ni3+ or Ni. The bottom Co can be completely covered by depositing an Al layer thicker than 2.0 nm. 17Anti-tumor responses to hypofractionated radiation in mice grafted with triple negative breast cancer is associated with decorin induction in peritumoral muscles显示文摘Triple negative breast cancer (TNBC)is the most lethal one for all types of breast cancer.Though radiotherapy is an efficient treatment,long-term survival rate of TNBC patients is still suboptimal. Hyprofractionated radiotherapy,an improved radiotherapy,has made an inspiring result in clinic. However,the mechanism underlying TNBC treated with hyprofractionated radiotherapy is not clear.Decorin (DCN)is a small poteoglycan of matrix which has an inhibitory effect on the breast cancer and is secreted by muscle under certain conditions.In this study,we demonstrated that peritumoral muscles secrete more DCN at higher dose irradiation than that at conventional irradiation dose in TNBC tumor-bearing mice.Thus,it indicates that DCN secreted from peritumoral muscle may be one of the reasons why hyprofractionated radiotherapy could inhibit the growth of TNBC more effectively.Moreover,we also indicated that the up-regulated DCN attenuated lung metastasis of TNBC.In conclusion,we demonstrated that hypofractionated radiation promotes the secretion of DCN in peritumoral muscle,thus enhancing the inhibitory effect on TNBC,which might help to optimize the strategy of radiotherapy for TNBC patients in the future.Qi Yu Kedao Xin Yu Miao Zhaobin Li Shen Fu Shudong Hu Qing Zhang Shumin Zhou 2018Acta Biochimica et Biophysica Sinica2018,50,11:1
