| 2 | Simultaneous incorporation of PTH(1-34) and nano-hydroxyapatite into Chitosan/Alginate Hydrogels for efficient bone regeneration显示文摘Tissue regeneration based on the utilization of artificial soft materials is considered a promising treatment for bone-related diseases.Here,we report cranial bone regeneration promoted by hydrogels that contain parathyroid hormone(PTH)peptide PTH(1-34)and nano-hydroxyapatite(nHAP).A combination of the positively charged natural polymer chitosan(CS)and negatively charged sodium alginate led to the formation of hydrogels with porous structures,as shown by scanning electron microscopy.Rheological characterizations revealed that the mechanical properties of the hydrogels were almost maintained upon the addition of nHAP and PTH(1-34).In vitro experiments showed that the hydrogel containing nHAP and PTH(1-34)exhibited strong biocompatibility and facilitated osteogenic differentiation of rat bone marrow mesenchymal stem cells(rBMSCs)via the Notch signaling pathway,as shown by the upregulated expression of osteogenic-related proteins.We found that increasing the content of PTH(1-34)in the hydrogels resulted in enhanced osteogenic differentiation of BMSCs.Implantation of the complex hydrogel into a rat cranial defect model led to efficient bone regeneration compared to the rats treated with the hydrogel alone or with nHAP,indicating the simultaneous therapeutic effect of nHAP and PTH during the treatment process.Both the in vitro and in vivo results demonstrated that simultaneously incorporating nHAP and PTH into hydrogels shows promise for bone regeneration,suggesting a new strategy for tissue engineering and regeneration in the future. | Zhiyuan Zou Le Wang Zhifei Zhou Qing Sun Delong Liu Yan Chen Hao Hu Yu Cai Sixiong Lin Zhengran Yu Bizhi Tan Wei Guo Zemin Ling Xuenong Zou | 2021 | Bioactive Materials2021,6,6: | 7 |
| 3 | Differential expression of immune-related genes between healthy volunteers and type 2 diabetic patients with spleen-deficiency pattern显示文摘OBJECTIVE: To investigate the clinical differentiation of spleen-deficiency pattern(SDP), a group of symptoms and signs defined in terms of Traditional Chinese Medicine for its clinical practice.METHODS: Peripheral venous blood(> 3 m L) was collected from each of six type 2 diabetes mellitus(T2DM)-SDP patients and six healthy volunteers. After the isolation of peripheral white blood cells(PWBCs), total RNA was extracted, and quality control was performed on all RNA samples. Microarray experiments were conducted using the Agilent human whole genome gene chip, and genes demonstrating differential expression were screened. Bioinformatics analysis was conducted on these genes using several online databases.RESULTS: We screened a total of 175 differentially expressed genes(DEGs), of which 111(63%) were down-regulated and 64(37%) were up-regulated in T2DM-SDP patients compared with healthy controls. Among the 175 genes, 158 had biological function annotations: 46(29%) were directly related to an individual's immune regulation or response, 25(16%) were associated with substance and energy metabolism of PWBCs which could also indirectly influence immunity, and the remaining87(55%) were involved in a variety of PWBC biological processes that might eventually influence the immune function. Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis revealed that the DEGs were predominantly enriched in seven immune-related pathways. Hierarchical cluster analysis identified gene expression patterns that were distinguishable between the two study groups.CONCLUSION: Our results suggest that T2DM-SDP patients experience significant hypoimmunity and/or immune dysfunctions, and possess a specific gene expression profile. | 14 | Influences of AMY1 gene copy number and protein expression on salivary alpha-amylase activity before and after citric acid stimulation in splenic asthenia children显示文摘OBjECTIVE:To compare the correlations between salivary alpha-amylase(sAA) activity and amylase,alpha 1(salivary) gene(AMY1) copy number or its gene expression between splenic asthenia and healthy children,and investigate the reasons of attenuated sAA activity ratio before and after citric acid stimulation in splenic asthenia children.METHODS:Saliva samples from 20 splenic asthenia children and 29 healthy children were collected before and after citric acid stimulation.AMY1 copy number,sAA activity,and total sAA and glycosylated sAA contents were determined,and their correlations were analyzed.RESULTS:Although splenic asthenia and healthy children had no differences in AMY1 copy number,splenic asthenia children had positive correlations between AMY1 copy number and sAA activity before or after citric acid stimulation.Splenic asthenia children had a higher sAA glycosylated proportion ratio and glycosylated sAA content ratio,while their total sAA content ratio and sAA activity ratio were lower compared with healthy children.The glycosylated sAA content ratio was higher than the total sAA content ratio in both groups.Splenic asthenia and healthy children had positive correlations between total sAA or glycosylated sAA content and sAA activity.However,the role played by glycosylated sAA content in sAA activity in healthy children increased after citric acid stimulation,while it decreased in splenic asthenia children.CONCLUSION:Genetic factors like AMY1 copy number variations,and more importantly,sAA glycosylation abnormalities leading to attenuated sAA activity after citric acid stimulation,which were the main reasons of the attenuated sAA activity ratio in splenic asthenia children compared with healthy children. | Yang Zemin Lin Jing Chen Longhui Zhang Min Yang Xiaorong Chen Weiwen | 2015 | Journal of Traditional Chinese Medicine2015,35,3: | 0 |

| 17 | High strength and high creep resistant ZrB2/Al nanocomposites fabricated by ultrasonic-chemical in-situ reaction显示文摘In this study, the ZrB2/Al nanocomposites were fabricated via in-situ reaction of the Al-K2ZrF6-KBF4 system, assisted with ultrasonic vibration and spiral electromagnetic stirring. Microstructure, tensile property and creep behavior of the fabricated nanocomposites were further investigated. Microstructure observation showed that the ultrasonic vibration could prevent the fast growth as well as break the clusters of in-situ synthesized nanoparticles in melt, resulted in smaller size (10-50 nm) and relatively more uniform distribution of the in-situ nanoparticles located on the boundary of and/or inside the aluminum matrix grains in the final composites. The fabricated nanocomposites exhibited an enhancement in both strength and ductility, due to the elevated work hardening ability, i.e., improved dislocation propagating ability and decreased dynamic recovery of the existing dislocations induced by the in-situ nanoparticles. Meanwhile, the nanocomposites exhibited excellent creep resistance ability, which was about 2-18 times higher than those of the corresponding aluminum matrix. The stress exponent of 5 was identified for the fabricated nanocomposites, which suggested that their creep behavior was related to dislocation climb mechanism. The enhanced creep resistance of the nanocomposites was attributed to the Orowan strengthening and grain boundary strengthening induced by the ZrB2 nanoparticles. | 19 | Estimation and Long-term Trend Analysis of Surface Solar Radiation in Antarctica: A Case Study of Zhongshan Station显示文摘Long-term,ground-based daily global solar radiation (DGSR) at Zhongshan Station in Antarctica can quantitatively reveal the basic characteristics of Earth’s surface radiation balance and validate satellite data for the Antarctic region.The fixed station was established in 1989,and conventional radiation observations started much later in 2008.In this study,a random forest (RF) model for estimating DGSR is developed using ground meteorological observation data,and a highprecision,long-term DGSR dataset is constructed.Then,the trend of DGSR from 1990 to 2019 at Zhongshan Station,Antarctica is analyzed.The RF model,which performs better than other models,shows a desirable performance of DGSR hindcast estimation with an R^2 of 0.984,root-mean-square error of 1.377 MJ m^(-2),and mean absolute error of 0.828 MJ m^(-2).The trend of DGSR annual anomalies increases during 1990–2004 and then begins to decrease after 2004.Note that the maximum value of annual anomalies occurs during approximately 2004/05 and is mainly related to the days with precipitation (especially those related to good weather during the polar day period) at this station.In addition to clouds and water vapor,bad weather conditions (such as snowfall,which can result in low visibility and then decreased sunshine duration and solar radiation) are the other major factors affecting solar radiation at this station.The high-precision,longterm estimated DGSR dataset enables further study and understanding of the role of Antarctica in global climate change and the interactions between snow,ice,and atmosphere. | Zhaoliang ZENG Zemin WANG Minghu DING Xiangdong ZHENG Xiaoyu SUN Wei ZHU Kongju ZHU Jiachun AN Lin ZANG Jianping GUO Baojun ZHANG | 2021 | Advances in Atmospheric Sciences2021,38,9: | 0 |
