| 1 | MiR-122 in hepatic function and liver diseases显示文摘As the most abundant liver-specific microRNA,mi-croRNA-122(miR-122)is involved in various physio-logical processes in hepatic function as well as in liver pathology.There is now compelling evidence that miR-122,as a regulator of gene networks and pathways in hepatocytes,plays a central role in diverse aspects of hepatic function and in the progress of liver diseases.This liver-enriched transcription factors-regulated miRNA promotes differentiation of hepatocytes and regulates lipid metabolism.With regard to liver diseases,miR-122 was shown to stimulate hepatitis C virus(HCV)replication through a unique and unusual interaction with two binding sites in the 5′-UTR of HCV genome to mediate the stability of the viral RNA,whereas inhibit the expression and replication of hepatitis B virus(HBV)by a miR-122-cylin G1/p53-HBV enhancer regulatory pathway.In addition,miR-122 acts as a suppressor of cell prolif-eration and malignant transformation of hepatocytes with remarkable tumor inhibition activity.Notably,a clinical trial targeting miR-122 with the anti-miR-122 oli-gonucleotides miravirsen,the first miRNA targeted drug,has been initiated for treatment of HCV infection.With further understanding of the comprehensive roles of miR-122 in hepatic functions and the mechanisms in-volved in miR-122 down-regulation in chronic hepatitis or hepatocellular carcinoma,miR-122 appears to be a promising candidate for effective therapeutic ap-proaches against tumor and infectious diseases. | Jun Hu Yaxing Xu Junli Hao Saifeng Wang Changfei Li Songdong Meng | 2012 | Protein & Cell2012,3,5: | 22 |

| 3 | A new tripodal rhodamine B derivative as a highly selective and sensitive fluorescence chemosensor for copper(Ⅱ)显示文摘A new tripodal rhodamine B derivative 2 was designed and synthesized by tripodal trialdehyde and rhodamine B hydrazide for the first time. This derivative could be used as a fluorescent chemosensor for the selective and sensitive determination of copper(II) in Tris-HCl buffer and ethanol aqueous mixed media. Under the optimum conditions described herein,fluorescence enhancement at 557/577 nm was linearly related to the concentration of copper(II) in the range of 0.10 to 10.00×10-5 mol·L-1,with a corre-lation coefficient of R2=0.9964 (n=15) and a detection limit of 1.129×10-7 mol·L-1 (the relative standard deviation for five repeated measurements at 4.00×10-5 mol·L-1 Cu(II) was 2.2%). | 6 | Tunable nonlinear refractive index of two-dimensional MoS2, WS2, and MoSe2 nanosheet dispersions [Invited]显示文摘Liquid-phase-exfoliation technology was utilized to prepare layered MoS2, WS2, and MoSe2 nanosheets in cyclohexylpyrrolidone. The nonlinear optical response of these nanosheets in dispersions was investigated by observing spatial self-phase modulation(SSPM) using a 488 nm continuous wave laser beam. The diffraction ring patterns of SSPM were found to be distorted along the vertical direction right after the laser traversing the nanosheet dispersions. The nonlinear refractive index of the three transition metal dichalcogenides dispersions n2 was measured to be 10-7cm2W-1, and the third-order nonlinear susceptibility χ(3)10-9 esu. The relative change of effective nonlinear refractive index Δn2e∕n(2e) of the MoS2, WS2, and MoSe2 dispersions can be modulated 0.012–0.240, 0.029–0.154, and 0.091–0.304, respectively, by changing the incident intensities. | 7 | Bioinformatics analysis for structure and function of CPR of Plasmodium falciparum显示文摘Objective:To analyse the structure and function of NADPH-cytochrome p450 reductase(CYPOR or CPR) from Plasmodium falciparum(Pf),and to predict its’ drug target and vaccine target. Methods:The structure,function,drug target and vaccine target of CPR from Plasmodium falciparum were analyzed and predicted by bioinformatics methods.Results:PfCPR,which was older CPR,had close relationship with the CPR from other Plasmodium species,but it was distant from its hosts,such as Homo sapiens and Anopheles.PfCPR was located in the cellular nucleus of Plasmodium falciparum.335aa-352aa and 591aa - 608aa were inserted the interior side of the nuclear membrane,while 151aa-265aa was located in the nucleolus organizer regions.PfCPR had 40 function sites and 44 protein-protein binding sites in amino acid sequence.The teriary structure of laa-700aa was forcep-shaped with wings.15 segments of PfCPR had no homology with Homo sapien CPR and most were exposed on the surface of the protein.These segments had 25 protein-protein binding sites.While 13 other segments all possessed function sites. | 8 | Direct observation of interlayer coherent acoustic phonon dynamics in bilayer and few-layer PtSe2显示文摘This work reports the real-time observation of the interlayer lattice vibrations in bilayer and few-layer PtSe_2 by means of the coherent phonon method. The layer-breathing mode and standing wave mode of the interlayer vibrations are found to coexist in such a kind of group-10 transition metal dichalcogenides(TMDCs). The interlayer breathing force constant standing for perpendicular coupling(per effective atom) is derived as 7.5 N/m,2.5 times larger than that of graphene. The interlayer shearing force constant is comparable to the interlayer breathing force constant, which indicates that PtSe2 has nearly isotropic interlayer coupling. The low-frequency Raman spectroscopy elucidates the polarization behavior of the layer-breathing mode that is assigned to have A1 g symmetry. The standing wave mode shows redshift with the increasing number of layers, which successfully determines the out-of-plane sound velocity of PtSe2 experimentally. Our results manifest that the coherent phonon method is a good tool to uncover the interlayer lattice vibrations, beyond the conventional Raman spectroscopy limit. In these inclusion complexes,combined cavity and portal binding in TMeQ[6] were observed,and the length of the bridged alkylene was found to play an important role not only in balancing the overall hydrophilic/hydrophobic interaction between the host and the guest,but also in defining the structure of the resulting inclusion complexes. For the guest 1,2-ethylenedipyridine (Edpy),TMeQ[6] includes a positively charged pyridine ring of Edpy to form an unsymmetrical inclusion complex; for the guest 1,4-butylenedipyridine (Bdpy),TMeQ[6] includes a positively charged pyridine ring of Bdpy,but the different competitive interactions in and between the related inclusion complexes could lead to a fast exchange between the hosts and guests. For the guests with longer bridge chains,such as 1,6-hexamethylenedipyridine (Hdpy) or 1,8-octylenedipyridine (Odpy),a stable pseudorotaxane inclusion complex is formed by combining the hydrophobic cavity and the outer portal dipoleion interactions. | 18 | Effects of mesencephalic astrocyte-derived neurotrophic factor on sepsis-associated acute kidney injury显示文摘BACKGROUND:To determine the protective role of mesencephalic astrocyte-derived neurotrophic factor(MANF) in regulating sepsis-associated acute kidney injury(S-AKI).METHODS:A total of 96 mice were randomly divided into the control group,control+MANF group,S-AKI group,and S-AKI+MANF group.The S-AKI model was established by injecting lipopolysaccharide(LPS) at 10 mg/kg intraperitoneally.MANF(200 μg/kg) was administered to the control+MANF and S-AKI+MANF groups.An equal dose of normal saline was administered daily intraperitoneally in the control and S-AKI groups.Serum and kidney tissue samples were obtained for biochemical analysis.Western blotting was used to detect the protein expression of MANF in the kidney,and enzyme-linked immunosorbent assay(ELISA) was used to determine expression of MANF in the serum,pro-inflammatory cytokines(tumor necrosis factor-α [TNF-α] and interleukin-6 [IL-6]).Serum creatinine(SCr),and blood urea nitrogen(BUN)were examined using an automatic biochemical analyzer.In addition,the kidney tissue was observed for pathological changes by hematoxylin-eosin staining.The comparison between two groups was performed by unpaired Student’s t-test,and statistics among multiple groups were carried out using Tukey’s post hoc test following one-way analysis of variance(ANOVA).A P-value <0.05 was considered statistically significant.RESULTS:At the early stage of S-AKI,MANF in the kidney tissue was up-regulated,but with the development of the disease,it was down-regulated.Renal function was worsened in the S-AKI group,and TNF-α and IL-6 were elevated.The administration of MANF significantly alleviated the elevated levels of SCr and BUN and inhibited the expression of TNF-α and IL-6 in the kidney.The pathological changes were more extensive in the S-AKI group than in the S-AKI+MANF group.CONCLUSION:MANF treatment may significantly alleviate renal injury,reduce the inflammatory response,and alleviate or reverse kidney tissue damage.MANF may have a protective effect on S-AKI,suggesting a potential treatment for S-AKI. | Saifeng Chen Xuewei Hao Guo Chen Guorong Liu Xiaoyan Yuan Peiling Shen Dongfeng Guo | 2023 | World Journal of Emergency Medicine2023,14,5: | 0 |
