| 1 | Roles of a sustained activation of NCED3 and the synergistic regulation of ABA biosynthesis and catabolism in ABA signal production in Arabidopsis显示文摘ABA, acting as a stress signal, plays crucial roles in plant resistance to water stress. Because ABA signal production is based on ABA biosynthesis, the regulation of NCED, a key enzyme in the ABA biosynthesis pathway, is normally thought of as the sole factor controlling ABA signal production. Here we demonstrate that ABA catabolism in combination with a synergistic regulation of ABA biosynthesis plays a crucial role in governing ABA signal production. Water stress induced a significant accumulation of ABA, which exhibited different patterns in detached and attached leaves. ABA catabolism followed a temporal trend of exponential decay for both basic and stress ABA, and there was little difference in the catabolic half-lives of basic ABA and stress ABA. Thus, the absolute rate of ABA catabolism, i.e. the amount of ABA catabolized per unit time, increases with increased ABA accumulation. From the dynamic processes of ABA biosynthesis and catabolism, it can be inferred that stress ABA accumulation may be governed by a synergistic regulation of all the steps in the ABA biosynthesis pathway. Moreover, to maintain an elevated level of stress ABA sustained activation of NCED3 should be required. This inference was supported by further findings that the genes encoding major enzymes in the ABA biosynthesis pathway, e.g. NCED3, AAO3 and ABA3 were all activated by water stress, and with ABA accumulation progressing, the expressions of NCED3, AAO3 and ABA3 remained activated. | 2 | Bioinformatics analysis for structure and function of CPR of Plasmodium falciparum显示文摘Objective:To analyse the structure and function of NADPH-cytochrome p450 reductase(CYPOR or CPR) from Plasmodium falciparum(Pf),and to predict its’ drug target and vaccine target. Methods:The structure,function,drug target and vaccine target of CPR from Plasmodium falciparum were analyzed and predicted by bioinformatics methods.Results:PfCPR,which was older CPR,had close relationship with the CPR from other Plasmodium species,but it was distant from its hosts,such as Homo sapiens and Anopheles.PfCPR was located in the cellular nucleus of Plasmodium falciparum.335aa-352aa and 591aa - 608aa were inserted the interior side of the nuclear membrane,while 151aa-265aa was located in the nucleolus organizer regions.PfCPR had 40 function sites and 44 protein-protein binding sites in amino acid sequence.The teriary structure of laa-700aa was forcep-shaped with wings.15 segments of PfCPR had no homology with Homo sapien CPR and most were exposed on the surface of the protein.These segments had 25 protein-protein binding sites.While 13 other segments all possessed function sites. Results:P6NOS were not available,but nicotinamide adenine dinucleotide 2’-phosphate reduced tetrasodium(NADPH)-cytochrome p450 reductase(CPR) were gained.PiCPR was in the nucleus of Plasmodium berghei,while 134aa-229aa domain was localize in nucleolar organizer. | 4 | On brain activity mapping: Insights and lessons from Brain Decoding Project to map memory patterns in the hippocampus显示文摘The BRAIN project recently announced by the president Obama is the reflection of unrelenting human quest for cracking the brain code, the patterns of neuronal activity that define who we are and what we are. While the Brain Activity Mapping proposal has rightly emphasized on the need to develop new technologies for measuring every spike from every neuron, it might be helpful to consider both the theoretical and experimental aspects that would accelerate our search for the organizing principles of the brain code. Here we share several insights and lessons from the similar proposal, namely, Brain Decoding Project that we initiated since 2007. We provide a specific example in our initial mapping of real-time memory traces from one part of the memory circuit, namely, the CA1 region of the mouse hippocampus. We show how innovative behavioral tasks and appropriate mathematical analyses of large datasets can play equally, if not more, important roles in uncovering the specific-to-general feature-coding cell assembly mechanism by which episodic memory, semantic knowledge, and imagination are generated and organized. Our own experiences suggest that the bottleneck of the Brain Project is not only at merely developing additional new technologies, but also the lack of efficient avenues to disseminate cutting edge platforms and decoding expertise to neuroscience community. Therefore, we propose that in order to harness unique insights and extensive knowledge from various investigators working in diverse neuroscience subfields, ranging from perception and emotion to memory and social behaviors, the BRAIN project should create a set of International and National Brain Decoding Centers at which cutting-edge recording technologies and expertise on analyzing large datasets analyses can be made readily available to the entire community of neuroscientists who can apply and schedule to perform cutting-edge research. | TSIEN Joe Z. | 6 | Inter-specific and Inter-generic Hybridization Compatibility of Eriobotrya Species (Loquat) and Related Genera显示文摘We studied the cross-compatibility among 91 inter-specific combinations and 21 inter-generic combinations in 7 Eriobotrya plants and 2 related genera(Raphiolepis indica Lindl. and Photinia serrulata Lindl.) using emasculation, bagging, and artificial pollination. Our results showed that28 of the 91 inter-specific combinations set no fruit, which means nearly 30% of the combinations were incompatible. In the remaining 63 combinations, most showed partial cross-compatibility, and a few showed complete cross compatibility. Eriobotrya plants were incompatible with plants from their related genera(R. indica Lindl. and P. serrulata Lindl.). Backcrossing produced 5 compatible combinations, which could set fruits and produce F1 progeny but only after embryo rescue. Fruit setting ratios varied among various species used as male or female parents.E. prinoides Rehd. & Wils., common loquat(E. japonica) and Eriobotrya × daduheensis, used as female parents resulted in an average fruitsetting ratio of 36.2%–58.2%. E. deflexa Nakai and its two forms, and E. elliptica Lindl. as female parents resulted in 2.9%–16.3% average fruitsetting ratio; however, the fruit set ratio was higher(22.4%–43.1%) if they were used as male parents. | 7 | Radiation-associated Angiosarcoma After Postoperative Adjuvant Radiotherapy for Cervical Cancer:A Case Report and Review of Published Reports显示文摘Objective Radiation-associated angiosarcoma(RAAS)is rare.We present an unusual case of RAAS presenting only 1.5 years after radiotherapy for cervical cancer and review relevant published reports.Case report A 59-year-old woman with stage I B2 cervical cancer underwent radical hysterectomy and pelvic external beam radiation therapy.Seventeen months later,violaceous lesions began to appear,quickly covering her entire lower abdominal wall,perineum,and upper thighs.These lesions were accompanied by small nodules and ulceration.Immunohistochemistry revealed positive expression of CD31 and CD34.Genetic testing revealed MYC amplification.The patient was finally diagnosed with RAAS and treated with albumin-bound paclitaxel and bevacizumab.However,her condition deteriorated quickly and she died 7 months later.Conclusion This case illustrates the importance of extensive biopsy of skin lesions suspected of denoting RAAS as early as possible.Pathological examination for CD31,CD34,and MYC assists early establishment of the diagnosis. | Yayu HUANG Jing SUN Genlai LIN Guifen MA | 2022 | Clinical Cancer Bulletin2022,1,2: | 0 |
