1使用cDNA微阵列和组织微阵列对三种上皮性卵巢肿瘤基因表达的分析显示文摘背景与目的卵巢癌是在妇科恶性肿瘤中死亡率最高的肿瘤,主要原因是由于缺乏有效的早期诊断方法。为了发现新的、特异性癌基因和进一步探索上皮性卵巢癌的临床意义,本文探索出一种新方法,通过结合cDNA微阵列和RNA原位杂交冰冻组织微阵列的方法寻找特异性卵巢癌基因。方法利用cDNA微阵列筛选在3种不同卵巢癌中(卵巢浆液性交界性肿瘤、卵巢浆液性腺癌和卵巢子宫内膜样腺癌)显示有意义表达的基因,并由RNA原位杂交冰冻组织微阵列证实其结果。结果40个基因和ESTs显示出在卵巢浆液性交界性肿瘤、浆液性和子宫内膜样卵巢癌3种类型之间基因表达有明显的差异;EPHB6、PTPRF、GFER、ERG25、PLRP1,FLJ22060和WISP2被进一步用于RNA原位杂交冰冻组织微阵列研究,其结果与cDNA微阵列研究结果相符合。结论cDNA微阵列和RNA原位杂交冰冻组织微阵列相结合是一种理想的寻找癌相关基因的方法,EPHB6,PTPRF,GFER,ERG25,PLRP1,FLJ22060和WISP2有可能成为新的上皮性卵巢癌候选基因。郑敏 Simon R Kononen J Sauter G Mihatsch MJ Moch H 2004癌症2004,23,7:8
2Changes in demand and supply of ecosystem services under scenarios of future land use in Vorarlberg, Austria显示文摘Ecosystem services(ES)are highly impacted by human-induced land-use change.Progressive urbanization and agricultural land abandonment in Western Europe necessitate assessments of future land-change impacts on ES to ensure sustainable service management.The present study aims at evaluating future demand and supply of three key services(flood protection,nearby recreation and biodiversity)in the mountainous region of Vorarlberg,Austria.We mapped the ES for the referenced time step 2016 and two scenarios for 2050,assuming the continuation of current land-change trends and pressure on landscape development.Results for the referenced landscape in 2016 show the highest ES supply for intermediate levels,while ES supply was low in the lowlands and valley bottoms and in high-elevation areas.We found a high positive correlation of ES with the distribution of forested areas.In contrast,service demand was highest in lowelevation areas and decreased with increasing elevation.This indicates that densely settled and intensively used agricultural areas currently suffer from ES undersupply.The projected future development of land use showed an increase in both supply and demand of the selected ES.The overall service supply increased more than the respective demand due to some reforestation of open land.As forests were found to be important synergistic areas for overall service provision,we expect decreasing demand on related services.Locally,demand was found to exceed the supply of ES,especially in the densely populated Rhine valley-requiring further policy interventions.Such ES-related information may contribute to regional policy making and ensure the long-term provision of ESs for future generations.SAUTER Isabel KIENAST Felix BOLLIGER Janine WINTER Benjamin PAZuR Robert 2019Journal of Mountain Science2019,16,12:5

6Reduced anoctamin 7(ANO7) expression is a strong and independent predictor of poor prognosis in prostate cancer显示文摘Objective:Anoctamin 7(ANO7)is a calcium2+-dependent chloride ion channel protein.Its expression is restricted to prostate epithelial cells.The exact function is unknown.This study aimed to analyze ANO7 expression and its clinical significance in prostate cancer(PCa).Methods:ANO7 expression was assessed by immunohistochemistry in 17,747 clinical PCa specimens.Results:ANO7 was strongly expressed in normal prostate glandular cells but often less abundant in cancer cells.ANO7 staining was interpretable in 13,594 cancer tissues and considered strong in 34.4%,moderate in 48.7%,weak in 9.3%,and negative in 7.6%.Reduced staining was tightly linked to adverse tumor features[high classical and quantitative Gleason grade,lymph node metastasis,advanced tumor stage,high Ki67 labeling index,positive surgical margin,and early biochemical recurrence(P<0.0001 each)].The univariate Cox hazard ratio for prostate-specific antigen(PSA)recurrence after prostatectomy in patients with negative vs.strong ANO7 expression was 2.98(95%confidence interval 2.61–3.38).The prognostic impact was independent of established pre-or postoperatively available parameters(P<0.0001).Analysis of annotated molecular data showed that low ANO7 expression was linked to TMPRSS2:ERG fusions(P<0.0001),elevated androgen receptor expression(P<0.0001),as well as presence of 9 of 11 chromosomal deletions(P<0.05 each).A particularly strong association of low ANO7 expression with phosphatase and tensin homolog(PTEN)deletion may indicate a functional relationship with the PTEN/AKT pathway.Conclusions:These data identify reduced ANO7 protein expression as a strong and independent predictor of poor prognosis in PCa.ANO7 measurement,either alone or in combination,might provide clinically useful prognostic information in PCa.Andreas Marx Lena Koopmann Doris Hoflmayer Franziska Büscheck Claudia Hube-Magg Stefan Steurer Till Eichenauer Till S.Clauditz Waldemar Wilczak Ronald Simon Guido Sauter Jakob R.Izbicki Hartwig Huland Hans Heinzer Markus Graefen Alexander Haese Thorsten Schlomm Christian Bernreuther Patrick Lebok Sarah Bonk 2021Cancer Biology & Medicine2021,18,1:3

13A guided tour:Pollen tube orientation in flowering plants显示文摘Sexual reproduction in flowering plants requires that two sperm cells are delivered to the embryo sac where double fertilization of an egg cell and of a central cell results in the formation of a diploid embryo and of the triploid nutritional endosperm tissue. The immobile male gametes are delivered to the immobile female gametophyte by a single cell, the pollen tube. The pollen tube must be able to germinate on a genetically appropriate stigma and it must be directed through the transmitting tract of the style from where it must target an ovule. Moreover, the pollen tube must enter the ovule at a defined opening, the micropyle, grow toward one of the two synergids and release the two sperm cells upon contact. This complex process requires recognition events with chemically based or physically supported cell-cell communication as well as directional cues for the growing pollen tube.
