2Cytokines in adipose-derived mesenchymal stem cells promote the healing of liver disease显示文摘Adipose-derived mesenchymal stem cells(ADSCs) are a treatment cell source for patients with chronic liver injury. ADSCs are characterized by being harvested from the patient's own subcutaneous adipose tissue, a high cell yield(i.e., reduced immune rejection response), accumulation at a disease nidus, suppression of excessive immune response, production of various growth factors and cytokines, angiogenic effects, antiapoptotic effects, and control of immune cells via cellcell interaction. We previously showed that conditioned medium of ADSCs promoted hepatocyte proliferation and improved the liver function in a mouse model of acute liver failure. Furthermore, as found by many other groups, the administration of ADSCs improved liver tissue fibrosis in a mouse model of liver cirrhosis. A comprehensive protein expression analysis by liquid chromatography with tandem mass spectrometry showed that the various cytokines and chemokines produced by ADSCs promote the healing of liver disease. In this review, we examine the ability of expressed protein components of ADSCs to promote healing in cell therapy for liver disease. Previous studies demonstrated that ADSCs are a treatment cell source for patients with chronic liver injury. 3ANALYTICAL SOLUTIONS FOR THE SLOSHING LOADING ON CIRCULAR CYLINDRICAL LIQUID TANKS WITH INTERIOR SEMI-POROUS BARRIERS显示文摘The estimation of sloshing loading on liquid tanks is of fundamental importance due to the wide and important applications of liquid tanks in various engineering aspects. Analytical solutions for the sloshing loading on circular cylindrical liquid tanks with an interior concentric vertical thin semi porous barrier under external excitations are presented. And the extensions of the solutions to the elastic vibrations and to the high frequency limit to count for the earthquake response are also included. A clear view on the influence of semi porous barriers to the sloshing response of oscillating tanks is obtained by the analyses of systematic computational results. The present results may offer guidance to the corresponding design for liquid tanks of various applications and also a basis of comparison for the development of more sophisticated numerical methods for hydrodynamic and hydro elastic analyses.Miao, G.P. 5Distribution of late gadolinium enhancement in various types of cardiomyopathies:Significance in differential diagnosis, clinical features and prognosis显示文摘The recent development of cardiac magnetic resonance(CMR)techniques has allowed detailed analyses of cardiac function and tissue characterization with high spatial resolution.We review characteristic CMR features in ischemic and non-ischemic cardiomyopathies(ICM and NICM),especially in terms of the location and distribution of late gadolinium enhancement(LGE).CMR in ICM shows segmental wall motion abnormalities or wall thinning in a particular coronary arterial territory,and the subendocardial or transmural LGE.LGE in NICM generally does not correspond to any particular coronary artery distribution and is located mostly in the mid-wall to subepicardial layer.The analysis of LGE distribution is valuable to differentiate NICM with diffusely impaired systolic function,including dilated cardiomyopathy,end-stage hypertrophic cardiomyopathy(HCM),cardiac sarcoidosis,and myocarditis,and those with diffuse left ventricular(LV)hypertrophy including HCM,cardiac amyloidosis and Anderson-Fabry disease.A transient low signal intensity LGE in regions of severe LV dysfunction is a particular feature of stress cardiomyopathy.In arrhythmogenic right ventricular cardiomyopathy/dysplasia,an enhancement of right ventricular(RV)wall with functional and morphological changes of RV becomes apparent.Finally,the analyses of LGE distribution have potentials to predict cardiac outcomes and response to treatments.Hiroshi Satoh Makoto Sano Kenichiro Suwa Takeji Saitoh Mamoru Nobuhara Masao Saotome Tsuyoshi Urushida Hideki Katoh Hideharu Hayashi 2014World Journal of Cardiology2014,6,7:8

7Identification of advanced spin-driven thermoelectric materials via interpretable machine learning显示文摘Machine learning is becoming a valuable tool for scientific discovery.Particularly attractive is the application of machine learning methods to the field of materials development,which enables innovations by discovering new and better functional materials.To apply machine learning to actual materials development,close collaboration between scientists and machine learning tools is necessary.However,such collaboration has been so far impeded by the black box nature of many machine learning algorithms.It is often difficult for scientists to interpret the data-driven models from the viewpoint of material science and physics.Here,we demonstrate the development of spin-driven thermoelectric materials with anomalous Nernst effect by using an interpretable machine learning method called factorized asymptotic Bayesian inference hierarchical mixture of experts(FAB/HMEs).Based on prior knowledge of material science and physics,we were able to extract from the interpretable machine learning some surprising correlations and new knowledge about spin-driven thermoelectric materials.Guided by this,we carried out an actual material synthesis that led to the identification of a novel spin-driven thermoelectric material.This material shows the largest thermopower to date.Yuma Iwasaki Ryohto Sawada Valentin Stanev Masahiko Ishida Akihiro Kirihara Yasutomo Omori Hiroko Someya Ichiro Takeuchi Eiji Saitoh Shinichi Yorozu 2019npj Computational Materials2019,,1:7

9Improving the performance of conventional base isolation systems by an external variable negative stiffness device under near-fault and long-period ground motions显示文摘Recent studies have shown that base-isolated objects with long fundamental natural periods are highly influenced by long-period earthquakes. These long-period waves result in large displacements for isolators, possibly leading to exceedance of the allowable displacement limits. Conventional isolation systems, in general, fail to resist such large displacements. This has prompted the need to modify conventional base isolation systems. The current work focuses on the development of an external device, comprising a unit of negative and positive springs, for improving the performance of conventional base isolation systems. This unit accelerates the change in the stiffness of the isolation system where the stiffness of the positive spring varies linearly in terms of the displacement response of the isolated objects. The target objects of the present study are small structures such as computer servers, sensitive instruments and machinery. Numerical studies show that the increase in the damping of the system and the slope of the linear function is effective in reducing the displacement response. 10Inferring the Tree of Life of the order Cypriniformes, the earth's most diverse clade of freshwater fishes: Implications of varied taxon and character sampling显示文摘The phylogenetic relationships of species are fundamental to any biological investigation, including all evolutionary studies. Accurate inferences of sister group relationships provide the researcher with an historical framework within which the attributes or geographic origin of species (or supraspecific groups) evolved. Taken out of this phylogenetic context, interpretations of evolutionary processes or origins, geographic distributions, or speciation rates and mechanisms, are subject to nothing less than a biological experiment without controls. Cypriniformes is the most diverse clade of freshwater fishes with estimates of diversity of nearly 3,500 species. These fishes display an amazing array of morphological, ecological, behavioral, and geographic diversity and offer a tremendous opportunity to enhance our understanding of the biotic and abiotic factors associated with diversification and adaptation to environments. Given the nearly global distribution of these fishes, they serve as an important model group for a plethora of biological investigations, including indicator species for future cli-matic changes. The occurrence of the zebrafish, Danio rerio, in this order makes this clade a critical component in understanding and predicting the relationship between mutagenesis and phenotypic expressions in vertebrates, including humans. With the tremendous diversity in Cypriniformes, our understanding of their phylogenetic relationships has not proceeded at an acceptable rate, despite a plethora of morphological and more recent mo-lecular studies. Most studies are pre-Hennigian in origin or include relatively small numbers of taxa. Given that analyses of small numbers of taxa for molecular characters can be compromised by peculiarities of long-branch attraction and nodal-density effect, it is critical that significant progress in our understanding of the relationships of these important fishes occurs with increasing sampling of species to mitigate these potential problems. The recent Cypriniformes Tree of Life initiative is an effort to achieve this goal with morphological and molecular (mitochondrial and nuclear) data. In this early synthesis of our understanding of the phylogenetic relationships of these fishes, all types of data have contributed historically to improving our understanding, but not all analyses are complementary in taxon sampling, thus precluding direct understanding of the impact of taxon sampling on achieving accurate phylogenetic inferences. However, recent molecular studies do provide some insight and in some instances taxon sampling can be implicated as a variable that can influence sister group relationships. Other instances may also exist but without inclusion of more taxa for both mitochondrial and nuclear genes, one cannot distinguish between inferences being dictated by taxon sampling or the origins of the molecular data.Richard L. 12Efficacy of long-term rifaximin treatment for hepatic encephalopathy in the Japanese显示文摘BACKGROUND Hepatic encephalopathy(HE)is a complication of liver cirrhosis and can result in neuropsychological and neuromuscular dysfunctions in patients.Rifaximin,an antibiotic,has been reported to decrease the occurrence of overt HE and also improve cognitive function in studies from Europe and the United States of America.There is not enough evidence of the relationship between the long-term use of rifaximin and its clinical effects in the Japanese.AIM To determine the clinical effects of long-term rifaximin therapy in decompensated liver cirrhosis patients,with overt HE or hyperammonemia.METHODS In this single-center retrospective observational cohort study,we reviewed the data of 38 patients who had taken rifaximin at the dose of 1200 mg/d for more than 24 wk.The primary outcome measured was the efficacy of long-term rifaximin use,and secondary outcome measured was the safety of its long-term use as determined by its influence on portosystemic shunts as well as Escherichia coli-related infections.Moreover,we compared the prognosis between the rifaximin group and control cases,matched for hepatic elasticity assessed by magnetic resonance ela-stography,age,and Child-Pugh classification.RESULTS Of the 38 patients included in the study,12(31.6%)had overt HE,27(71.1%)had complications of esophageal varices,and 9(23.7%)had hepatocellular carcinoma(HCC).The control group was matched for age,Child-Pugh classification,liver stiffness,and presence of HCC.The median of serum ammonia level before treatment was 104μg/dL(59-297),and 2 wk after treatment,it significantly decreased to 85μg/dL(34-153)(P=0.002).A significantly low value of 80.5μg/dL(44-150)was maintained 24 wk after treatment.The long-term use of rifaximin did not cause a decline in liver function.Diarrhea occurred in 2 patients,who improved with the administration of probiotics,and there were no cases of aborted rifaximin therapy owing to adverse events.In patients with Child C,the survival was short,but there was no significant difference compared with that of the control group.CONCLUSION Rifaximin therapy improves overt HE.The long-term use of rifaximin in the Japanese is effective and safe.Shinya Nishida Koichi Hamada Noriyuki Nishino Daizo Fukushima Ryota Koyanagi Yoshinori Horikawa Yoshiki Shiwa Satoshi Saitoh 2019World Journal of Hepatology2019,11,6:3
13Activation of protein kinase A alters subnuclear distribution pattern of human steroidogenic factor 1 in living cells显示文摘Background The aim of this study was to identify the subnuclear distribution pattern of human orphan nuclear receptor steroidogenic factor 1 (SF-1) in living cells with and without the activation of protein kinase A (PKA) signal pathway, and thus try to explain the unknown mechanism by which PKA potentiates SF-1 transactivation. Methods Full-length cDNAs of wild type and a naturally occurring mutant (G35E) human SF-1 were cloned and fused with green fluorescent protein (GFP). Subcellular distribution pattern of human SF-1 in living cells, whose PKA signaling was either activated or not, was studied by laser confocal microscopy after the validity of the gene sequence was confirmed.Results The transactivation ability of the GFP-SF-1 chimeric protein was highly conserved. Wild type human SF-1 diffused homogeneously within the nuclei of cells when PKA was not active, and converged to clear foci when PKA was activated. Mutant SF-1 diffused within the nuclei even in the presence of PKA activation, surprisingly aggregating as fluorescent dots inside the nucleoli, a phenomenon not altered by PKA.Conclusions Activation of PKA causes wild type, but not mutant SF-1 to alter its subnuclear distribution pattern to a transactivationally active form (foci formation). 20PiggyBac transposon-mediated gene delivery efficiently generates stable transfectants derived from cultured primary human deciduous tooth dental pulp cells(HDDPCs) and HDDPC-derived iPS cells显示文摘The ability of human deciduous tooth dental pulp cells(HDDPCs) to differentiate into odontoblasts that generate mineralized tissue holds immense potential for therapeutic use in the field of tooth regenerative medicine. Realization of this potential depends on efficient and optimized protocols for the genetic manipulation of HDDPCs. In this study, we demonstrate the use of a Piggy Bac(PB)-based gene transfer system as a method for introducing nonviral transposon DNA into HDDPCs and HDDPC-derived inducible pluripotent stem cells. The transfection efficiency of the PB-based system was significantly greater than previously reported for electroporation-based transfection of plasmid DNA. Using the neomycin resistance gene as a selection marker, HDDPCs were stably transfected at a rate nearly 40-fold higher than that achieved using conventional methods. Using this system, it was also possible to introduce two constructs simultaneously into a single cell. The resulting stable transfectants, expressing td Tomato and enhanced green fluorescent protein, exhibited both red and green fluorescence. The established cell line did not lose the acquired phenotype over three months of culture. Based on our results, we concluded that PB is superior to currently available methods for introducing plasmid DNA into HDDPCs. There may be significant challenges in the direct clinical application of this method for human dental tissue engineering due to safety risks and ethical concerns. However, the high level of transfection achieved with PB may have significant advantages in basic scientific research for dental tissue engineering applications, such as functional studies of genes and proteins.
