1Ca^(2+)-supplying black phosphorus-based scaffolds fabricated with microfluidic technology for osteogenesis显示文摘Effective osteogenesis remains a challenge in the treatment of bone defects.The emergence of artificial bone scaffolds provides an attractive solution.In this work,a new biomineralization strategy is proposed to facilitate osteogenesis through sustaining supply of nutrients including phosphorus(P),calcium(Ca),and silicon(Si).We developed black phosphorus(BP)-based,three-dimensional nanocomposite fibrous scaffolds via microfluidic technology to provide a wealth of essential ions for bone defect treatment.The fibrous scaffolds were fabricated from 3D poly(L-lactic acid)(PLLA)nanofibers(3D NFs),BP nanosheets,and hydroxyapatite(HA)-porous SiO2 nanoparticles.The 3D BP@HA NFs possess three advantages:i)stably connected pores allow the easy entrance of bone marrow-derived mesenchymal stem cells(BMSCs)into the interior of the 3D fibrous scaffolds for bone repair and osteogenesis;ii)plentiful nutrients in the NFs strongly improve osteogenic differentiation in the bone repair area;iii)the photothermal effect of fibrous scaffolds promotes the release of elements necessary for bone formation,thus achieving accelerated osteogenesis.Both in vitro and in vivo results demonstrated that the 3D BP@HA NFs,with the assistance of NIR laser,exhibited good performance in promoting bone regeneration.Furthermore,microfluidic technology makes it possible to obtain high-quality 3D BP@HA NFs with low costs,rapid processing,high throughput and mass production,greatly improving the prospects for clinical application.This is also the first BP-based bone scaffold platform that can self-supply Ca^(2+),which may be the blessedness for older patients with bone defects or patients with damaged bones as a result of calcium loss.Zhanrong Li Xingcai Zhang Jiang Ouyang Dandan Chu Fengqi Han Liuqi Shi Ruixing Liu Zhihua Guo Grace X.Gu Wei Tao Lin Jin Jingguo Li 2021Bioactive Materials2021,6,11:4
2Six sigma robust design optimization for thermal protection system of hypersonic vehicles based on successive response surface method显示文摘Lightweight design is important for the Thermal Protection System(TPS) of hypersonic vehicles in that it protects the inner structure from severe heating environment. However, due to the existence of uncertainties in material properties and geometry, it is imperative to incorporate uncertainty analysis into the design optimization to obtain reliable results. In this paper, a six sigma robust design optimization based on Successive Response Surface Method(SRSM) is established for the TPS to improve the reliability and robustness with considering the uncertainties. The uncertain parameters related to material properties and thicknesses of insulation layers are considered and characterized by random variables following normal distributions. By employing SRSM, the values of objective function and constraints are approximated by the response surfaces to reduce computational cost. The optimization is an iterative process with response surfaces updating to find the true optimal solution. 3Deficiency of two-pore segment channel 2 contributes to systemic lupus erythematosus via regulation of apoptosis and cell cycle显示文摘Background:Systemic lupus erythematosus(SLE)is a complex autoimmune disease,and the mechanism of SLE is yet to be fully elucidated.The aim of this study was to explore the role of two-pore segment channel 2(TPCN2)in SLE pathogenesis.Methods:Quantitative reverse transcription polymerase chain reaction(qRT-PCR)was used to detect the expression ofTPCN2 in SLE.We performed a loss-of-function assay by lentiviral construct in Jurkat and THP-1 cell.Knockdown ofTPCN2 were confirmed at the RNA level by qRT-PCR and protein level by Western blotting.Cell Count Kit-8 and flow cytometry were used to analyze the cell proliferation,apoptosis,and cell cycle ofTPCN2-deficient cells.In addition,gene expression profile ofTPCN2-deficient cells was analyzed by RNA sequencing(RNA-seq).Results:TPCN2 knockdown with short hairpin RNA(shRNA)-mediated lentiviruses inhibited cell proliferation,and induced apoptosis and cell-cycle arrest of G2/M phase in both Jurkat and THP-1 cells.We analyzed the transcriptome of knockdown-TPCN2-Jurkat cells,and screened the differential genes,which were enriched for the G2/M checkpoint,complement,and interleukin-6-Janus kinase-signal transducer and activator of transcription pathways,as well as changes in levels of forkhead box O,phosphatidylinositol 3-kinase/protein kinase B/mechanistic target of rapamycin,and T cell receptor pathways;moreover,TPCN2 significantly influenced cellular processes and biological regulation.Conclusion:TPCN2 might be a potential protective factor against SLE.Keke Li Jingkai Xu Ke Xue Ruixing Yu Chengxu Li Wenmin Fei Xiaoli Ning Yang Han Ziyi Wang Jun Shu Yong Cui 2022Chinese Medical Journal2022,,4:2
