18Altitudinal Variation in Digestive Tract Length in Yunnan Pond Frog (Pelophylax pleuraden)显示文摘The digestive tract plays an important role in digestion and the acquisition of food energy. Understanding the impact of abiotic environments on digestive tract morphology is especially important for evolution of digestive tract across different environments. Here, we investigated altitudinal variation in digestive tract length in the Yunnan Pond Frog(Pelophylax pleuraden) across five populations ranging from 1413 m to 1935 m a.s.l. in Ningnan County, Sichuan province in western China. Frogs were collected during the breeding season, from 1–5 June 2012. Our results revealed that females had longer digestive tract and relative digestive tract(i.e. digestive tract length / body size) lengths in comparison to males, on average, but the differences between them decreased with increasing altitude. 19The inhibitory effect of SEMA3C/3D mutations on Neuro-2a cells migration and axonal growth in patients with Hirschsprung′s disease显示文摘Objective:To explore the effect and mechanism of SEMA3C/3D mutations on axonal growth of neurons and cell migration in Hirschsprung′s disease(HSCR)patients.Methods:HEK293 Tcells were transfected with wild-type and mutant SEMA3C/3D plasmids.The supernatants that contained SEMA3C/3D-AP fusion proteins were collected and added into the Neuro-2acells.The changes in the cell morphology were observed by immunofluorescence staining.The expression and phosphorylation levels of cofilin,ERM and CRMP2 were determined by western blotting.The cell migration rate was measured by transwell assay.Results:Compared with wild-type SEMA3D,SEMA3D-P615T mutant suppressed cofilin phosphorylation in Neuro-2a cells(P <0.05).The neural cells treated by five mutant SEMA3C/3D-AP fusion proteins presented different levels of axon atrophy,growth cone collapse,and sometimes,loss of normal structure.SEMA3C-S329 G,SEMA3C-V337M and SEMA3D-P615T mutants cells exhibited a significantly reduced migration rate as compared with wild-type SEMA3C/3D treated cells(P <0.01).Conclusion:SEMA3C and SEMA3 D mutations in HSCR patients could lead to the inhibition of Neuro-2a cells′migration and axonal growth.The mechanism of SEMA3 DP615T mutant might be related to down-regulation of the expression of p-cofilin,which consequently lead to cytoskeleton structure collapse and migrating ability decrease.Our study might provide important clues for the pathogenesis of HSCR.Kaihui Yu Zhen Zhang Qi Li Ping Xiao Lin Su Long Li Shangling Pan Qian Jiang 2017广西医科大学学报2017,34,8:0
20Natural variation in the SVP contributes to the pleiotropic adaption of Arabidopsis thaliana across contrasted habitats显示文摘Coordinated plant adaptation involves the interplay of multiple traits driven by habitat-specific selection pressures. Pleiotropic effects, wherein genetic variants of a single gene control multiple traits, can expedite such adaptations. Until present, only a limited number of genes have been reported to exhibit pleiotropy. Here, we create a recombinant inbred line (RIL) population derived from two Arabidopsis thaliana (A. thaliana) ecotypes originating from divergent habitats. Using this RIL population, we identify an allelic variation in a MADS-box transcription factor, SHORT VEGETATIVE PHASE (SVP), which exerts a pleiotropic effect on leaf size and drought-versus-humidity tolerance. Further investigation reveals that a natural null variant of the SVP protein disrupts its normal regulatory interactions with target genes, including GRF3, CYP707A1/3, and AtBG1, leading to increased leaf size, enhanced tolerance to humid conditions, and changes in flowering time of humid conditions in A. thaliana. Remarkably, polymorphic variations in this gene have been traced back to early A. thaliana populations, providing a genetic foundation and plasticity for subsequent colonization of diverse habitats by influencing multiple traits.
