1Repeatability and Reproducibility of Quantitative Assessment of the Retinal Microvasculature Using Optical Coherence Tomography Angiography Based on Optical Microangiography显示文摘Objective The aim of this study was to determine the repeatability and reproducibility of optical coherence tomography angiography(OCTA) based on optical microangiography(OMAG) measurements of macular vessels in normal eyes. Methods In this prospective cohort study, 40 eyes of 40 healthy volunteers underwent repeated OCTA(Cirrus HD-OCT 5000 angiography system, Carl Zeiss Meditec, Inc.) scans on two separate visit days. On each visit day, the eyes were scanned three times. The following parameters were used to quantitatively describe the OCTA images of the superficial vascular network: vessel area density(VAD), vessel skeleton density(VSD), vessel diameter index(VDI), vessel perimeter index(VPI), vessel complexity index(VCI), flux, and foveal avascular zone(FAZ). Coefficient of variation(CV) and intraclass correlation coefficient(ICC) were calculated for evaluating intravisit and intervisit repeatability, as well as interobserver reproducibility. Results The measurements showed high repeatability [CVs ≤ 4.2%(intravisit) and ≤ 4.6%(intervisit)] and interobserver reproducibility(ICCs ≥ 0.923) for all parameters. 2基于光学微血流成像技术的相干光断层扫描血流成像对视网膜血流定量分析的可重复性评价显示文摘目的评价基于光学微血流成像技术(OMAG)的相干光断层扫描血流成像(OCTA)在正常眼视网膜血流定量分析的可重复性。设计诊断试验。研究对象 40名(40眼)健康志愿者,年龄22~55岁,平均(33.0±8.8)岁。方法使用Zeiss Cirrus HD-OCT 5000扫描,以基于OMAG技术的AngioPlex^(TM)软件对视网膜浅层3 mm×3 mm OCTA图像的血流长度密度(VLD)、灌注密度(PD)及黄斑中心凹无血流信号区(FAZ)面积、周长、圆形度进行自动定量分析。采用变异系数(CV)评价观察日内及观察日间重复性,组内相关系数(ICC)评价观察者间再现性。主要指标变异系数(CV)和组内相关系数(ICC)。结果各血流参数观察日内CV(1.2±1.0)%^(4.2±5.5)%,均≤4.2%,观察日间CV(1.4±0.8)%^(4.6±4.9)%,均≤4.6%,观察日间CV高于观察日内CV,而中心凹、FAZ各参数CV高于旁中心凹CV;各血流参数观察者间ICC(0.923~0.994),均≥0.923,中心凹、FAZ各参数ICC高于旁中心凹。结论基于OMAG技术的OCTA对正常眼视网膜浅层血流各参数测量均具有较好的重复性和再现性。赵琦 王霄娜 杨文利 魏文斌 WANG Ruikang K. 游启生 楚仲頔 辛晨 2018眼科2018,27,2:7

4应用光学相干层析成像对大鼠舌的在体显微检测研究显示文摘目的 应用自行装置的光学相干层析成像(OCT)设备对大鼠舌苔与舌苔下浅表组织进行在体的显微成像研究,为光学相干层析成像技术在中医舌诊的应用可能性与其价值进行初步的探讨.方法 应用OCT,以Lab VIEW编写软件操作平台,将大鼠舌上表面进行显微成像检测,并对舌苔的某些观测内容进行量化处理;同时,与大鼠舌的常规组织切片结果进行对比性分析.结果 大鼠舌表面显微成像显示:分辨率为10~20 μm,深度约2 mm,能够清楚地分辨出舌苔层、舌苔舌体连接层与舌体层结构,在舌苔苔质的某些观察内容可以实现量化;OCT与组织学检测大鼠舌显微结构结果显示具有一定的相似性.结论 光学相干层析成像技术在大鼠舌诊中能实现成像研究,能实现一定的量化研究,具有一定的实用性价值.曾常春 钟会清 刘汉平 郭周义 何永红 Ruikang K Wang 2007北京中医药大学学报2007,30,8:6
5Nano-hydroxyapatite accelerates vascular calcification via lysosome impairment and autophagy dysfunction in smooth muscle cells显示文摘Vascular calcification(VC)is a common characteristic of aging,diabetes,chronic renal failure,and atherosclerosis.The basic component of VC is hydroxyapatite(HAp).Nano-sized HAp(nHAp)has been identified to play an essential role in the development of pathological calcification of vasculature.However,whether nHAp can induce calcification in vivo and the mechanism of nHAp in the progression of VC remains unclear.We discovered that nHAp existed both in vascular smooth muscle cells(VSMCs)and their extracellular matrix(ECM)in the calcified arteries from patients.Synthetic nHAp had similar morphological and chemical properties as natural nHAp recovered from calcified artery.nHAp stimulated osteogenic differentiation and accelerated mineralization of VSMCs in vitro.Synthetic nHAp could also directly induce VC in vivo.Mechanistically,nHAp was internalized into lysosome,which impaired lysosome vacuolar H+-ATPase for its acidification,therefore blocked autophagic flux in VSMCs.Lysosomal re-acidification by cyclic-3′,5′-adenosine monophosphate(cAMP)significantly enhanced autophagic degradation and attenuated nHAp-induced calcification.The accumulated autophagosomes and autolysosomes were converted into calcium-containing exosomes which were secreted into ECM and accelerated vascular calcium deposit.Inhibition of exosome release in VSMCs decreased calcium deposition.Altogether,our results demonstrated a repressive effect of nHAp on lysosomal acidification,which inhibited autophagic degradation and promoted a conversion of the accumulated autophagic vacuoles into exosomes that were loaded with undissolved nHAp,Ca^(2+),Pi and ALP.These exosomes bud off the plasma membrane,deposit within ECM,and form calcium nodules.Vascular calcification was thus accelerated by nHAP through blockage of autophagic flux in VSMCs.Qi Liu Yi Luo Yun Zhao Pingping Xiang Jinyun Zhu Wangwei Jing Wenjing Jin Mingyao Chen Ruikang Tang Hong Yu 2022Bioactive Materials2022,7,2:5
6Polarization sensitive optical coherence tomography with single input for imaging depth-resolved collagen organizations显示文摘Collagen organization plays an important role in maintaining structural integrity and determining tissue function.Polarization-sensitive optical coherence tomography(PSOCT)is a promising noninvasive three-dimensional imaging tool for mapping collagen organization in vivo.While PSOCT systems with multiple polarization inputs have demonstrated the ability to visualize depth-resolved collagen organization,systems,which use a single input polarization state have not yet demonstrated sufficient reconstruction quality.Herein we describe a PSOCT based polarization state transmission model that reveals the depth-dependent polarization state evolution of light backscattered within a birefringent sample.Based on this model,we propose a polarization state tracing method that relies on a discrete differential geometric analysis of the evolution of the polarization state in depth along the Poincare sphere for depth-resolved birefringent imaging using only one single input polarization state.We demonstrate the ability of this method to visualize depth-resolved myocardial architecture in both healthy and infarcted rodent hearts(ex vivo)and collagen structures responsible for skin tension lines at various anatomical locations on the face of a healthy human volunteer(in vivo).Peijun Tang Mitchell A.Kirby Nhan Le Yuandong Li Nicole Zeinstra G.Nina Lu Charles E.Murry Ying Zheng Ruikang K.Wang 2021Light(Science & Applications)2021,10,12:2
7Influence of viscosity on the phase transformation of amorphous calcium carbonate in fluids:An understanding of the medium effect in biomimetic mineralization显示文摘Biological mineral generation via an amorphous precursor is a topic of great current interest. Various factors such as the temperature, solution composition and presence of organic molecules can influence this important inorganic process. Here we demonstrate that this mineral transformation can actually readily be regulated by solution viscosity, a fundamental but often overlooked property. In our experiment, amorphous calcium carbonate (ACC), a key model compound in biomimetic mineralization studies, is synthesized and dispersed into inert dispersants with different viscosities and the crystallization process is examined by using FT-IR spectroscopy and XRD. It is found that the inhibition of the transformation of ACC becomes more significant with increasing fluid viscosity. This phenomenon can be explained by the differences in ion diffusion in different media. Furthermore, the resulting crystals always have different morphologies and size distributions although they all have the calcite structure. 9HIV-1 Vpr protein activates the NF-κB pathway to promote G2/M cell cycle arrest显示文摘Viral protein R(Vpr) plays an important role in the replication and pathogenesis of Human immunodeficiency virus type 1(HIV-1). Some of the various functions attributed to Vpr, including the induction of G2/M cell cycle arrest, activating the NF-κB pathway, and promoting viral reverse transcription, might be interrelated. To test this hypothesis, a panel of Vpr mutants were investigated for their ability to induce G2/M arrest and to activate the NF-κB pathway. The results showed that the Vpr mutants that failed to activate NF-κB also lost the activity to induce G2/M arrest, which suggests that inducing G2/M arrest via Vpr depends at least partially on the activation of NF-κB. 14Inorganic ionic polymerization:From biomineralization to materials manufacturing显示文摘Biomineralization process regulates the growth of inorganic minerals by complex molecules,proteins,and cells,endowing bio-materials with marvels structures and excellent properties.The intricate structures and compositions found in biominerals have inspired scientists to design and synthesize numerous artificial biomimetic materials.The methodology for controlling the formation of inorganics plays a pivotal role in achieving biomimetic structures and compositions.However,the current approach predominantly relies on the classical nucleation theory,which hinders the precise preparation of inorganic materials by replicating the biomineralization strategy.Recently,the development of“inorganic ionic polymerization”strategy has enabled us to regulate the arrangement of inorganic ions from solution to solid phase,which establishes an artificial way to produce inorganic materials analogous to the biomineralization process.Based on inorganic ionic polymerization,a series of achievements have been realized for the biomimetic preparation,including moldable construction of inorganic materials,hard tissue regeneration,and high-performance biomimetic materials.Moreover,the utilization of inorganic ionic polymerization has also facilitated the production of numerous advanced materials,including novel structures that exceed the current knowledge of materials science.The inorganic ionic polymerization system provides new artificial strategies and methodologies for the controllable synthesis of inorganics,which mimics the biomineralization process,paving the way for the future development of more high-performance materials.Yanhua Sang Kexin Qin Ruikang Tang Zhaoming Liu 2024Nano Research2024,17,2:1
