A chromosome-scale genome assembly of Isatis indigotica,an important medicinal plant used in traditional Chinese medicine显示文摘Isatis indigotica(2n=14)is an important medicinal plant in China. Its dried leaves and roots(called Isatidis Folium and Isatidis Radix, respectively)are broadly used in traditional Chinese medicine for curing diseases caused by bacteria and viruses such as influenza and viral pneumonia.Various classes of compounds isolated from this species have been identified as effective ingredients.Previous studies based on transcriptomes revealed only a few candidate genes for the biosynthesis of these active compounds in this medicinal plant. Here,we report a high-quality chromosome-scale genome assembly of I.indigotica with a total size of 293.88 Mb and scaffold N50=36.16 Mb using single-molecule real-time long reads and high-throughput chromosome conformation capture techniques. We annotated 30,323 highconfidence protein-coding genes.Based on homolog searching and functional annotations,we identified many candidate genes involved in the biosynthesis of main active components such as indoles, terpenoids, and phenylpropanoids. In addition,we found that some key enzyme-coding gene families related to the biosynthesis of these components were expanded due to tandem duplications, which likely drove the production of these major active compounds and explained why I.indigotica has excellent antibacterial and antiviral activities. Our results highlighted the importance of genome sequencing in identifying candidate genes for metabolite synthesis in medicinal plants. | Minghui Kang Haolin Wu Qiao Yang Li Huang Quanjun Hu Tao Ma Zaiyun Li Jianquan Liu | 2020 | Horticulture Research2020,7,1: | 9

Hybrid speciation via inheritance of alternate alleles of parental isolating genes显示文摘It is increasingly realized that homoploid hybrid speciation(HHS),which involves no change in chromosome number,is an important mechanism of speciation.HHS will likely increase in frequency as ecological and geographical barriers between species are continuing to be disrupted by human activities.HHS requires the establishment of reproductive isolation between a hybrid and its parents, but the underlying genes and genetic mechanisms remain largely unknown. In this study,we reveal by integrated approaches that reproductive isolation originates in one homoploid hybrid plant species through the inheritance of altemate alleles at genes that determine parental premating isolation. The parent species of this hybrid species are reproductively isolated by differences in flowering time and survivorship on soils containing high concentrations of iron.We found that the hybrid species inherits alleles of parental isolating major genes related to flowering time from one parent and alleles of major genes related to iron tolerance from the other parent. In this way,it became reproductively isolated from one parent by the difference in flowering time and from the other by habitat adaptation(iron tolerance). These findings and further modeling results suggest that HHS may occur relatively easily via the inheritance of alternate parental premating isolating genes and barriers. | Zefu Wang Yuanzhong Jiang Hao Bi Zhiqiang Lu Yazhen Ma Xiaoyue Yang Ningning Chen Bin Tian Bingbing Liu Xingxing Mao Tao Ma Stephen P.DiFazio Quanjun Hu Richard J.Abbott Jianquan Liu | 2021 | Molecular Plant2021,14,2: | 4
Recent advances in crystalline carbon nitride for photocatalysis显示文摘Semiconductor photocatalytic technology has become one of the most important means to solve the current energy shortage and environmental pollution. Compared with the traditional photocatalysts, graphite carbon nitride has a wider range of light-harvesting and more stable physical and chemical properties. However, traditional thermally induced polymerization of nitrogen-containing precursors produces the melon-based carbon nitride solids with an amorphous or semi-crystalline structure, resulting in low conductivity and moderate photocatalytic activity. Recently, crystalline carbon nitride has attracted more and more attention in improving photocatalytic performance. Some significant progress regarding crystalline carbon nitride for the preparation of solar-fuel and environmental purification has also been made. This review describes the recent advances in the design and synthesis of crystalline carbon nitride photocatalysts. A brief description of the unique physical and chemical properties of crystalline carbon nitride was given. Later, the synthetic and modification strategies are being introduced. Then, the photocatalytic application of crystalline carbon nitride was discussed, mainly including photocatalytic H2 production, photocatalytic CO2 reduction, and photocatalytic degradation of pollutants. Finally, the challenges and future directions of crystalline carbon nitride photocatalysts are briefly introduced. | Jingjun Liu Wei Fu Yulong Liao Jiajie Fan Quanjun Xiang | 2021 | Journal of Materials Science & Technology2021,,32: | 2
Pervasive hybridization during evolutionary radiation of Rhododendron subgenus Hymenanthes in mountains of southwest China显示文摘Radiations are especially important for generating species biodiversity in mountainous ecosystems. The contribution of hybridization to such radiations has rarely been examined. Here,we use extensive genomic data to test whether hybridization was involved in evolutionary radiation within Rhododendron subgenus Hymenanthes, whose members show strong geographic isolation in the mountains of southwest China. We sequenced genomes for 143 species of this subgenus and 93 species of four other subgenera, and found that Hymenanthes was monophyletic and radiated during the late Oligocene to middle Miocene.Widespread hybridization events were inferred within and between the identified clades and subclades. This suggests that hybridization occurred both early and late during diversification of subgenus Hymenan thes, although the extent to which hybridization, speciation through mixing-isolation-mixing or hybrid speciation, accelerated the diversification needs further exploration.Cycles of isolation and contact in such and other montane ecosystems may have together promoted species radiation through hybridization between diverging populations and species.Similar radiation processes may apply to other montane floras in this region and elsewhere. | Yazhen Ma Xingxing Mao Ji Wang Lei Zhang Yuanzhong Jiang Yuying Geng Tao Ma Liming Cai Shuangquan Huang Pete Hollingsworth Kangshan Mao Minghui Kang Yiling Li Wenlu Yang Haolin Wu Yang Chen Charles C.Davis Nawal Shrestha Richard H.Ree Zhenxiang Xi Quanjun Hu Richard I.Milne Jianquan Liu | 2022 | National Science Review2022,9,12: | 1
Phylotranscriptomics of Swertiinae(Gentianaceae)reveals that key floral traits are not phylogenetically correlated显示文摘Establishing how lineages with similar traits are phylogenetically related remains critical for understanding the origin of biodiversity on Earth.Floral traits in plants are widely used to explore phylogenetic relationships and to delineate taxonomic groups. The subtribe Swertiinae(Gentianaceae)comprises more than 350 species with high floral diversity ranging from rotate to tubular corollas and possessing diverse nectaries. Here we performed phylogenetic analysis of 60 species from all 15 genera of the subtribe Swertiinae sensu Ho and Liu,representing the range of floral diversity, using data from the nuclear and plastid genomes.Extensive topological conflicts were present between the nuclear and plastome trees.Three of the 15 genera represented by multiple species are polyphyletic in both trees.Key floral traits including corolla type, absence or presence of lobe scales, nectary type, nectary position, and stigma type are randomly distributed in the nuclear and plastome trees without phylogenetic correlation. We also revealed the likely ancient hybrid origin of one large clade comprising 10 genera with diverse floral traits. These results highlight the complex evolutionary history of this subtribe. The phylogenies constructed here provide a basic framework for further exploring the ecological and genetic mechanisms underlying both species diversification and floral diversity. | Chunlin Chen Brad R.Ruhfel Jialiang Li Zefu Wang Lushui Zhang Lei Zhang Xingxing Mao Ji Wang Dashan He Yue Luo Quanjun Hu Yuanwen Duan Xiaoting Xu Zhenxiang Xi Jianquan Liu | 2023 | Journal of Integrative Plant Biology2023,65,6: | 1

Enhanced chemical trapping and catalytic conversion of polysulfides by diatomite/MXene hybrid interlayer for stable Li-S batteries显示文摘Lithium-Sulfur (Li-S) batteries with high theoretical energy density are promising energy storage systems in the next decades, while the lithium polysulfides (LiPSs) shuttling caused by the sluggish sulfur redox reaction severely lowers the practical performance. The use of interlayer between the cathode and separator has been widely investigated to physically or chemically block the LiPSs, while the introduction of catalytic materials is a more effective strategy to accelerate the conversion of LiPSs. MXene with rich surface chemistry has shown its potential for facilitating the catalytic conversion, however, the aggregation of MXene sheets usually leads to the loss of the catalytic active sites. Herein, we report a diatomite/MXene (DE/MX) hybrid material as the bifunctional interlayer for improving the adsorption/conversion of LiPSs in Li-S batteries. The diatomite with porous structure and rich silica-hydroxyl functional groups could trap LiPSs effectively, while prevent the aggregation of MXene. The DE/MX based interlayer showed bifunctions of enhancing the chemical adsorption and promoting the conversion of LiPSs. The Li-S batteries with the DE/MX interlayer delivered an improved cycling stability with a low capacity decay of 0.059% per cycle over 1000 cycles at 1.0 C. Moreover, stable 200 cycles can be realized with a high sulfur loading electrode up to 6.0 mg cm^(−2). This work provides an effective strategy to construct bifunctional interlayers for hindering the shuttling of LiPSs and boosting the practical application of Li-S batteries. | Zehui Fan Chen Zhang Wuxing Hua Huan Li Yan Jiao Jingyi Xia Chuan-Nan Geng Rongwei Meng Yingxin Liu Quanjun Tang Ziyang Lu Tongxin Shang Guowei Ling Quan-Hong Yang | 2021 | Journal of Energy Chemistry2021,30,11: | 1

Generation of gene-target dogs using CRISPR/Cas9 system显示文摘Dear Editor,Dogs(Canis familiaris)serve as human companions and are raised to herd livestock, aid hunters, guard homes, perform police and rescue work, and guide the blind.Dogs exhibit close similarities to humans in terms of metabolic, physiological, and anatomical characteristics, and thus are ideal genetic and clinical models to study human diseases(Tsai et al.,2007).Gene target technology is a powerful tool to create new strains of animals with favorable traits. However, thus far,gene-target dogs have not been developed due to their unique species-specific reproductive characteristics, which limits the applications of dogs especially in the field of biomedical research.Recently, clustered regularly interspaced short palindromic repeats(CRISPRs)/CRISPR-associated(Cas)9 system was applied to edit specific genes with a high efficiency(Cong et al.,2013;Mali et al.,2013).Here we attempt to explore the feasibility of producing gene knockout(KO)dogs by using this technology.Beagle dog,the most widely used breed in biomedical research, was used as our animal model.Myostatin(MSTN)was chosen as the first gene of interest. | Qingjian Zou Xiaomin Wang Yunzhong Liu Zhen Ouyang Haibin Long Shu Wei Jige Xin Bentian Zhao Sisi Lai Jun Shen Qingchun Ni Huaqiang Yang Huilin Zhong Li Li Minhua Hu Quanjun Zhang Zhidong Zhou Jiaxin He Quanmei Yan Nana Fan Yu Zhao Zhaoming Liu Lin Guo Jiao Huang Guanguan Zhang Jun Ying Liangxue Lai Xiang Gao | 2015 | Journal of Molecular Cell Biology2015,7,6: | 1

Targeted Micellar Phthalocyanine for Lymph Node Metastasis Homing and Photothermal Therapy in an Orthotopic Colorectal Tumor Model显示文摘Tumor lymph node(LN)metastasis seriously affects the treatment prognosis.Studies have shown that nanoparticles with size of sub-50 nm can directly penetrate into LN metastases after intravenous administration. Here,we speculate through introducing targeting capacity, the nanoparticle accumulation in LN metastases would be further enhanced for improved local treatment such as photothermal therapy.Trastuzumabtargeted micelles(<50 nm)were formulated using a unique surfactantstripping approach that yielded concentrated phthalocyanines with strong near-infrared absorption.Targeted micellar phthalocyanine(T-MP)was an effective photothermal transducer and ablated HT-29 cells in vitro.A HER2-expressing colorectal cancer cell line(HT-29)was used to establish an orthotopic mouse model that developed metastatic disease in mesenteric sentinel LN.T-MP accumulated more in the LN metastases compared to the micelles conjugated with control IgG.Following surgical resection of the primary tumor, minimally invasive photothermal treatment of the metastatic LN with T-MP,but not the control micelles, extended mouse survival. Our findings demonstrate for the first time that targeted small-sized nanoparticles have potential to enable superior paradigms for dealing with LN metastases. | Hai‑Yi Feng Yihang Yuan Yunpeng Zhang Hai‑Jun Liu Xiao Dong Si‑Cong Yang Xue‑Liang Liu Xing Lai Mao‑Hua Zhu Jue Wang Qin Lu Quanjun Lin Hong‑Zhuan Chen Jonathan F.Lovell Peng Sun Chao Fang | 2021 | Nano-Micro Letters2021,13,11: | 1
Double knock-in pig models with elements of binary Tet-On and phiC31 integrase systems for controllable and switchable gene expression显示文摘Inducible expression systems are indispensable for precise regulation and in-depth analysis of biological process.Binary Tet-On system has been widely employed to regulate transgenic expression by doxycycline.Previous pig models with tetracycline regulatory elements were generated through random integration. This process often resulted in uncertain expression and unpredictable phenotypes, thus hindering their applications. Here,by precise knock-in of binary Tet-On 3G elements into Rosa26 and Hipp11 locus, respectively,a double knock-in reporter pig model was generated. We characterized excellent properties of this system for controllable transgenic expression both in vitro and in vivo. Two att P sites were arranged to flank the td Tomato to switch reporter gene.Single or multiple gene replacement was efficiently and faithfully achieved in fetal fibroblasts and nuclear transfer embryos. To display the flexible application of this system,we generated a pig strain with Dox-inducing h KRASexpression through phiC31 integrase-mediated cassette exchange. After eight months of Dox administration, squamous cell carcinoma developed in the nose, mouth, and scrotum, which indicated this pig strain could serve as an ideal large animal model to study tumorigenesis.Overall, the established pig models with controllable and switchable transgene expression system will provide a facilitating platform for transgenic and biomedical research. | Qin Jin Xiaoyu Yang Shixue Gou Xiaoyi Liu Zhenpeng Zhuang Yanhui Liang Hui Shi Jiayuan Huang Han Wu Yu Zhao Zhen Ouyang Quanjun Zhang Zhaoming Liu Fangbing Chen Weikai Ge Jingke Xie Nan Li Chengdan Lai Xiaozhu Zhao Jiaowei Wang Meng Lian Lei Li Longquan Quan Yinghua Ye Liangxue Lai Kepin Wang | 2022 | Science China(Life Sciences)2022,65,11: | 1
Detection and analysis system for hybridization images of lab-in-a-tube microarray显示文摘A lab-in-a-tube microarray system is developed for sample inspection and signal detection by fabricating a flat transparent window cap of the Eppendorf tube. The oli- gonucleotide microarray is immobilized on the inner surface of the cap. A small vessel is placed in an Eppendorf tube for storing hybridization solutions. With the microarray system, the full biochemical processes, including gene fragment am- plification, fluorescence labeling, hybridization, and fluores- cence detection, have been performed in the sealed tube without opening the cap. The images are obtained from a fluorescence microscope and captured by a CCD, and the data are transported to a computer through the universal serial bus (USB). After noise reduction, signal intensity is deter- mined from hybridization image and the presence of gene fragments is identified. The final data output includes sample information, process steps, and hybridization results. A lab-in- a-tube microarray system for detecting ten respiratory vi- ruses at a single detection is designed. High detection throug- hput and accuracy have been demonstrated with the system. | LIU Quanjun ZHOU Qin BAI Yunfei GE Qinyu LU Zuhong | 2005 | Chinese Science Bulletin2005,50,24: | 0

Gold nanorod translocation through a solid-state nanopore显示文摘Recently, nanopores have been used in an essential technique for detecting single molecule with high sensitivity. The initial application of nanopores to DNA and RNA sequencing has been expanded to sensing proteins and nanoparticles, including Bovine serum albumin, silica nanoparticles, polystyrene beads, and others. In our study, for the first time,a positively charged gold nanorod was investigated using a solid-state nanopore device.Various gold nanorods passed through the nanopore with different current blockages and duration times, providing a measurement of the nanorod diameter, length, and charge. Our findings indicate that nanopore sensing might be a new method for characterizing the size, shape, and charge of nanoparticles. | Liping Liu Jinglin Kong Xiao Xie Hongwen Wu Xiaofeng Ye Zhiliang Zhao Lei Wang Quanjun Liu | 2014 | Chinese Science Bulletin2014,59,7: | 0
