1Randomized controlled trial of uncut Roux-en-Y vs Billroth Ⅱ reconstruction after distal gastrectomy for gastric cancer: Which technique is better for avoiding biliary reflux and gastritis?显示文摘AIM To identify which technique is better for avoiding biliary reflux and gastritis between uncut Roux-en-Y and Billroth Ⅱ reconstruction. METHODS A total of 158 patients who underwent laparoscopyassisted distal gastrectomy for gastric cancer at the First Hospital of Jilin University(Changchun, China) between February 2015 and February 2016 were randomized into two groups: uncut Roux-en-Y(group U) and Billroth II group(group B). Postoperative complications and relevant clinical data were compared between the two groups. RESULTS According to the randomization table, each group included 79 patients. There was no significant difference in postoperative complications between groups U and B(7.6% vs 10.1%, P = 0.576). During the postoperative period, group U stomach p H values were lower than 7 and group B p H values were higher than 7. After 1 year of follow-up, group B presented a higher incidence of biliary reflux and alkaline gastritis. However, histopathology did not show a significant difference in gastritis diagnosis(P = 0.278), and the amount of residual food and gain of weight between the groups were also not significantly different. At 3 mo there was no evidence of partial recanalization of uncut staple line, but at 1 year the incidence was 13%. CONCLUSION Compared with Billroth II reconstruction, uncut Rouxen-Y reconstruction is secure and feasible, and can effectively reduce the incidence of alkaline reflux, residual gastritis, and heartburn. 2An Investigation of Oxidative DNA Damage in Pharmacy Technicians Exposed to Antineoplastic Drugs in Two Chinese Hospitals Using The Urinary 8-OHdG Assay显示文摘Objective To investigate oxidative DNA damage in pharmacy technicians preparing antineoplastic drugs at the PIVAS (Pharmacy Intravenous Admixture Service) in two Chinese hospitals.Methods Urinary 8-OHdG served as a biomarker.5-Fluorouracil (5-FU) concentrations in air,masks and gloves were determined.The spill exposure of each PIVAS technician to antineoplastic drugs was investigated.Eighty subjects were divided into exposed group I,II,and control group I,II.Results 5-FU concentration ratios for gloves and masks in exposed group I were significantly higher than those in exposed group II (P<0.05 or P<0.01).The average urinary 8-OHdG concentrations in exposed group I,control group I,exposed group II,and control group II were 14.69±0.93,10.68±1.07,10.57±0.55,and 11.96±0.73 ng/mg Cr,respectively.Urinary 8-OHdG concentration in exposed group I was significantly higher than that in control group I or that in exposed group II (P<0.01).There was a significant correlation between urinary 8-OHdG concentrations and spill frequencies per technician (P<0.01).Conclusion There was detectable oxidative DNA damage in PIVAS technicians exposed to antineoplastic drugs.This oxidative DNA damage may be associated with their spill exposure experience and contamination of their personal protective equipment.HUANG Yu Wen JIAN Le ZHANG Mei Bian ZHOU Quan YAN Xiao Feng HUA Xu Dong ZHOU Ying HE Ji Liang 2012Biomedical and Environmental Sciences2012,25,1:30

6Surgical approaches for stage Ⅰ and Ⅱ thymoma-associated myasthenia gravis:feasibility of complete video-assisted thoracoscopic surgery (VATS) thymectomy in comparison with trans-sternal resection显示文摘Complete resection could be achieved in virtually all myasthenic patients with Masaoka stage Ⅰ and Ⅱ thymoma using the trans-sternal technique.Whether this is appropriate for minimally invasive approach is not yet clear.We evaluated the feasibility of complete video-assisted thoracoscopic surgery (VATS) thymectomy for the treatment of Masaoka stage Ⅰ and Ⅱ thymoma-associated myasthenia gravis,compared to conventional trans-sternal thymectomy.We summarized 33 patients with Masaoka stage Ⅰ and Ⅱ thymoma-associated myasthenia gravis between April 2006 and September 2011.Of these,15 patients underwent right-sided complete VATS (the VATS group) by using adjuvant pneuomomediastinum,comparing with 18 patients using the trans-sternal approach (the T3b group).No intraoperative death was found and no VATS case required conversion to median sternotomy.Significant differences between the two groups regarding duration of surgery and volume of intraoperative blood loss (P=0.001 and P < 0.001,respectively) were observed.Postoperative morbidities were 26.7% and 33.3% for the VATS and T3b groups,respectively.All 33 patients were followed up for 12 to 61 months in the study.The cumulative probabilities of reaching complete stable remission and effective rate were 26.7% (4/15) and 93.3% (14/15) in the VATS group,which had a significantly higher complete stable remission and effective rate than those in the T3b group (P=0.026 and P=0.000,respectively).We conclude that VATS thymectomy utilizing adjuvant pneuomomediastinum for the treatment of stage Ⅰ and Ⅱ thymoma-associated myasthenia gravis is technically feasible but deserves further investigation in a large series with long-term follow-up.Zhicheng He Quan Zhu Wei Wen Liang Chen Hai Xu Hai Li 2013The Journal of Biomedical Research2013,27,1:15
7Large-scale analysis of small RNAs derived from traditional Chinese herbs in human tissues显示文摘Plant-derived microRNAs have recently been reported to function in human blood and tissues. Controversy was immediately raised due to possible contamination and the lack of large sample sizes. Here, we report thousands of unique small RNAs derived from traditional Chinese medicine(TCM) herbs found in human blood cells and mouse lung tissues using a large-scale analysis.We extracted small RNAs from decoctions of 10 TCM plants(Ban Zhi Lian, Chai Hu, Chuan Xin Lian, Di Ding Zi Jin, Huang Qin, Jin Yin Hua, Lian Qiao, Pu Gong Ying, Xia Ku Cao, and Yu Xing Cao) and obtained millions of RNA sequences from each herb. We also obtained RNA-Seq data from the blood cells of humans who consumed herbal decoctions and from the lung tissues of mice administered RNAs from herbal decoctions via oral gavage. We identified thousands of unique small RNA sequences in human blood cells and mouse lung tissues. Some of these identified small RNAs from Chuan Xin Lian and Hong Jing Tian could be mapped to the genomes of the herbs, confirming their TCM plant origin. 8Orthogonal Test Design for Optimization of the Extraction of Flavonid from the Fructus Gardeniae显示文摘Objective It is imperative to provide some consistent experimental results for the extraction of flavonid from Fructus Gardeniae.Methods The key extraction parameters that influenced the yield of flavonid from Fructus Gardeniae were optimized by employing an orthogonal experiment [L9(3)4],including the ratio of buffer solution (Na2B4O7·10H2O) to raw material,concentration of Fructus Gardeniae in extracting solution,extraction time and pH of buffer solution.An UV/Vis detector was used to perform the qualitative and quantitative analyses of the extracted flavonid with the using of the standard sample.Results The maximum extraction yield of the crude extract was 5.0533 (mg/g) after 20 min when the mass ratio of Na2B4O7·10H2O to raw material was 0.4%,the concentration of Fructus Gardeniae in the extraction solution was 1/12 (g/mL),and pH of buffer solution was 4.5.The positive reactions to the Molish and HCl‐Mg tests suggested that the extracted compound was flavonoid,and FTIR measurements also identified the presence of flavonoid in the extracts.Conclusion This work is expected to provide a basis for further research,development,and utilization of Fructus gardenia in flavonid extraction.CAI Ding Jian SHU Qing XU Bao Quan PENG Li Mei HE Yan 2011Biomedical and Environmental Sciences2011,24,6:12
9Effect of High Suspension and Low Incision Surgery Based on Traditional Ligation of Chinese Medicine in Treatment of Mixed Haemorrhoids:A Multi-centre,Randomized,Single-Blind,Non-inferiority Clinical Trial显示文摘Objective:To observe the clinical effect of high suspension and low incision(HSLI)surgery on mixed haemorrhoids,compared with Milligan-Morgan haemorrhoidectomy.Methods:A multi-centre,randomized,singleblind,non-inferiority clinical trial was performed.Participants with mixed haemorrhoids from Xiyuan Hospital of China Academy of Chinese Medical Sciences,Beijing Rectum Hospital,Air Force Medical Center of People's Liberation Army of China,and Puyang Hospital of Traditional Chinese Medicine were enrolled from September 2016 to March 2018.By using a blocked randomization scheme,participants were assigned to two groups.The experimental group was treated with HSLI,while the control group was treated with Milligan-Morgan haemorrhoidectomy.The primary outcome was the clinical effect evaluated at 12 weeks after operation.The secondary outcomes included the number of haemorrhoids treated during the operation,pain scores,use of analgesics,postoperative oedema,wound healing,incidence of anal stenosis,anorectal manometry after operation,as well as surgical duration,length of stay and total hospitalization expenses.A safety evaluation was also conducted.Results:In total,246 eligible participants were enrolled,with 123 cases in each group.There was no significant difference in the clinical effect between the two groups(100.00% vs.99.19%,P>0.05).Compared with the control group,the number of external haemorrhoids treated during the operation and the pain scores after operation were significantly reduced in the experimental group(P<0.05 or P<0.01);the patient number with wound healing at 2 weeks after operation and the functional length of anal canal at 12 weeks after operation were significantly increased in the experimental group(P<0.05).There was no significant difference in the incidence of anal stenosis,the numbers of patients using analgesics and patients with postoperative oedema between the two groups after operation(P>0.05).The surgical duration and length of stay in the experimental group were significantly longer than those in the control group,and the total hospitalization expense was significantly higher than that in the control group(all P<0.05).No adverse events were reported in either group during the whole trial or follow-up period.Conclusion:HSLI had the advantages of preserving the skin of anal canal completely,alleviating postsurgical pain and promoting rapid recovery after operation.(Registration No.Chi CTR1900022883).JIA Xiao-qiang CAO Wei-wei QUAN Long-fang ZHAO Wei-bing CHENG Fang JIA Shan FENG Liu-quan WEI Xu-feng XIE Zhen-nian WANG Dong XU Chun-yan CUI Chun-hui CAI Xing-juan HE Lan-ye WANG Zhan-jun TIAN Ying SHI Shu-min SUN Si-miao SU Liang ZHAI Meng-fan 2021Chinese Journal of Integrative Medicine2021,27,9:11
10Fibroblast-derived CXCL12/SDF-1α promotes CXCL6 secretion and co-operatively enhances metastatic potential through the PI3K/Akt/m TOR pathway in colon cancer显示文摘AIM To investigate the underlying mechanism by which CXCL12 and CXCL6 influences the metastatic potential of colon cancer and internal relation of colon cancer and stromal cells. METHODS Western blotting was used to detect the expression of CXCL12 and CXCL6 in colon cancer cells and stromal cells. The co-operative effects of CXCL12 and CXCL6 on proliferation and invasion of colon cancer cells and human umbilical vein endothelial cells(HUVECs) were determined by enzyme-linked immunosorbent assay,and proliferation and invasion assays. The angiogenesis of HUVECs through interaction with cancer cells and stromal cells was examined by angiogenesis assay. We eventually investigated activation of PI3K/Akt/m TOR signaling by CXCL12 involved in the metastatic process of colon cancer.RESULTS CXCL12 was expressed in DLD-1 cancer cells and fibroblasts. The secretion level of CXCL6 by colon cancer cells and HUVECs were significantly promoted by fibroblasts derived from CXCL12. CXCL6 and CXCL2 could significantly enhance HUVEC proliferation and migration(P < 0.01). CXCL6 and CXCL2 enhanced angiogenesis by HUVECs when cultured with fibroblast cells and colon cancer cells(P < 0.01). CXCL12 also enhanced the invasion of colon cancer cells. Stromal cell-derived CXCL12 promoted the secretion level of CXCL6 and co-operatively promoted metastasis of colon carcinoma through activation of the PI3K/Akt/m TOR pathway.CONCLUSION Fibroblast-derived CXCL12 enhanced the CXCL6 secretion of colon cancer cells,and both CXCL12 and CXCL6 co-operatively regulated the metastasis via the PI3K/Akt/m TOR signaling pathway. 11Transcriptomic landscape regulated by the 14 types of bone morphogenetic proteins(BMPs)in lineage commitment and differentiation of mesenchymal stem cells(MSCs)显示文摘Mesenchymal stem cells(MSCs)are ubiquitously-existing multipotent progenitors that can self-renew and differentiate into multiple lineages including osteocytes,chondrocytes,adipocytes,tenocytes and myocytes.MSCs represent one of the most commonly-used adult progenitors and serve as excellent progenitor cell models for investigating lineagespecific differentiation regulated by various cellular signaling pathways,such as bone morphogenetic proteins(BMPs).As members of TGFb superfamily,BMPs play diverse and important roles in development and adult tissues.At least 14 BMPs have been identified in mammals.Different BMPs exert distinct but overlapping biological functions.Through a comprehensive analysis of 14 BMPs in MSCs,we demonstrated that BMP9 is one of the most potent BMPs in inducing osteogenic differentiation of MSCs.Nonetheless,a global mechanistic view of BMP signaling in regulating the proliferation and differentiation of MSCs remains to be fully elucidated.Here,we conducted a comprehensive transcriptomic profiling in the MSCs stimulated by 14 types of BMPs.Hierarchical clustering analysis classifies 14 BMPs into three subclusters:an osteo/chondrogenic/adipogenic cluster,a tenogenic cluster,and BMP3 cluster.We also demonstrate that six BMPs(e.g.,BMP2,BMP3,BMP4,BMP7,BMP8,and BMP9)can induce ISmads effectively,while BMP2,BMP3,BMP4,BMP7,and BMP11 up-regulate Smad-independent MAP kinase pathway.Furthermore,we show that many BMPs can upregulate the expression of the signal mediators of Wnt,Notch and PI3K/AKT/mTOR pathways.While the reported transcriptomic changes need to be further validated,our expression profiling represents the first-of-its-kind to interrogate a comprehensive transcriptomic landscape regulated by the 14 types of BMPs in MSCs.Linghuan Zhang Qing Luo Yi Shu Zongyue Zeng Bo Huang Yixiao Feng Bo Zhang Xi Wang Yan Lei Zhenyu Ye Ling Zhao Daigui Cao Lijuan Yang Xian Chen Bin Liu William Wagstaff Russell R*Reid Hue H*Luu Rex C*Haydon Michael J*Lee Jennifer Moriatis Wolf Zhou Fu Tong-Chuan He Quan Kang 2019Genes & Diseases2019,6,3:11
12Signaling pathways and targeted therapy for myocardial infarction显示文摘Although the treatment of myocardial infarction(MI)has improved considerably,it is still a worldwide disease with high morbidity and high mortality.Whilst there is still a long way to go for discovering ideal treatments,therapeutic strategies committed to cardioprotection and cardiac repair following cardiac ischemia are emerging.Evidence of pathological characteristics in MI illustrates cell signaling pathways that participate in the survival,proliferation,apoptosis,autophagy of cardiomyocytes,endothelial cells,fibroblasts,monocytes,and stem cells.These signaling pathways include the key players in inflammation response,e.g.,NLRP3/caspase-1 and TLR4/MyD88/NF-κB;the crucial mediators in oxidative stress and apoptosis,for instance,Notch,Hippo/YAP,RhoA/ROCK,Nrf2/HO-1,and Sonic hedgehog;the controller of myocardial fibrosis such as TGF-β/SMADs and Wnt/β-catenin;and the main regulator of angiogenesis,PI3K/Akt,MAPK,JAK/STAT,Sonic hedgehog,etc.Since signaling pathways play an important role in administering the process of MI,aiming at targeting these aberrant signaling pathways and improving the pathological manifestations in MI is indispensable and promising.Hence,drug therapy,gene therapy,protein therapy,cell therapy,and exosome therapy have been emerging and are known as novel therapies.In this review,we summarize the therapeutic strategies for MI by regulating these associated pathways,which contribute to inhibiting cardiomyocytes death,attenuating inflammation,enhancing angiogenesis,etc.so as to repair and re-functionalize damaged hearts.Qing Zhang Lu Wang Shiqi Wang Hongxin Cheng Lin Xu Gaiqin Pei Yang Wang Chenying Fu Yangfu Jiang Chengqi He Quan Wei 2022Signal Transduction and Targeted Therapy2022,7,4:11
13Biomaterial-based strategies for maxillofacial tumour therapy and bone defect regeneration显示文摘Issues caused by maxillofacial tumours involve not only dealing with tumours but also repairing jaw bone defects.In traditional tumour therapy,the systemic toxicity of chemotherapeutic drugs,invasive surgical resection,intractable tumour recurrence,and metastasis are major threats to the patients’lives in the clinic.Fortunately,biomaterial-based intervention can improve the efficiency of tumour treatment and decrease the possibility of recurrence and metastasis,suggesting new promising antitumour therapies.In addition,maxillofacial bone tissue defects caused by tumours and their treatment can negatively affect the physiological and psychological health of patients,and investment in treatment can result in a multitude of burdens to society.Biomaterials are promising options because they have good biocompatibility and bioactive properties for stimulation of bone regeneration.More interestingly,an integrated material regimen that combines tumour therapy with bone repair is a promising treatment option.Herein,we summarized traditional and biomaterial-mediated maxillofacial tumour treatments and analysed biomaterials for bone defect repair.Furthermore,we proposed a promising and superior design of dual-functional biomaterials for simultaneous tumour therapy and bone regeneration to provide a new strategy for managing maxillofacial tumours and improve the quality of life of patients in the future.Bowen Tan Quan Tang Yongjin Zhong Yali Wei Linfeng He Yanting Wu Jiabao Wu Jinfeng Liao 2021International Journal of Oral Science2021,13,1:9
14Effects of dietary supplementation with essential oils and protease on growth performance,antioxidation,inflammation and intestinal function of weaned pigs显示文摘This experiment evaluated the impacts of essential oils(EO)and protease(PRO),independently or in combination,on growth performance,antioxidation,inflammation and intestinal function of weaned pigs.One hundred and sixty weaned pigs(21 d of age,BW of 6.74±0.20 kg)were randomly divided into 4 treatments with 8 replicate pens of 5 pigs per pen.Dietary treatments included the following:1)control diet(CON),2)CON with 300 mg/kg essential oils(EO),3)CON with 500 mg/kg protease(PRO),4)CON with 300 mg/kg essential oil and 500 mg/kg protease(EO + PRO).On d 8,one pig from each penwas selected for sampling.The remaining pigs were fed for an additional week and growth performance was monitored during this period.Dietary treatments had no marked effects(P>0.05)on the growth performance of pigs.However,pigs receiving EO diet had higher(P<0.05)serumglutathione peroxidase(GSH-Px)activity,and tended to decrease(P=0.063)serum concentration of tumor necrosis factor-a(TNF-a).In addition,pigs receiving EO diet had higher(P<0.05)abundances of phylum Actinobacteria,and genera Bifidobacterium,and lower(P<0.05)phylum Bacteroidetes and genera Alloprevotella in colonic digesta.Pigs receiving PRO diet decreased(P<0.05)the serum concentration of malondialdehyde(MDA)and diamine oxidase activity,increased(P<0.05)the villus height and the ratio of villus height to crypt depth in duodenum,increased sucrase activity in jejunal mucosa,and also increased the abundance of phylum Actinobacteria in colonic digesta.Furthermore,the synergistic effects of EO and PROwas observed(P<0.05)for pigs with decreasing serum TNF-a concentration and increasing serum GSH-Px activity.Collectively,the results indicated that dietary supplementation of EO and PRO had no significant effects on growth performance of weaned pigs.EO diet appeared to improve antioxidant activity and intestinal microbiota,while PRO diet improved intestinal morphology and digestive enzyme activity,and there was a synergistic effect of EO and PRO on reducing inflammatory parameters in weaned pigs.Xie Peng Qiang Zhou Cheng Wu Jian Zhao Quan Tan Ying He Liang Hu Zhengfeng Fang Yan Lin Shengyu Xu Bin Feng Jian Li Yong Zhuo Chris Van Ginneken Xuemei Jiang De Wu Lianqiang Che 2022Animal Nutrition2022,,2:8

17Comprehensive contour prediction model of work rolls in hot wide strip mill显示文摘The predictive calculation of comprehensive contour of work rolls in the on-line strip shape control model during hot rolling consists of two important parts of wear contour calculation and thermal contour calculation, which have a direct influence on the ac- curacy of shape control. A statistical wear model and a finite difference thermal contour model of work rolls were described. The comprehensive contour is the equivalence treatment of the sum of grinding, wear, and thermal contours. This comprehensive contour calculation model has been applied successfully in the real on-line strip shape control model. Its high precision has been proved through the large amounts of actual roll profile measurements and theoretical analyses.
