1Cadmium accumulation and tolerance of two castor cultivars in relation to antioxidant systems显示文摘To investigate the effects of Cd on tolerance and antioxidant activities of castor, two different castor(Ricinus communis L.) cultivars(Zibo No. 5 and Zibo No. 8) were used for a hydroponic experiment(0, 1 and 2 mg/L Cd) and a pot experiment using Cd-contaminated soil(34 mg/kg) with the addition of ethylenedinitrilotetraacetic acid(EDTA). The results indicated that there were significant differences between the two cultivars with respect to Cd uptake in shoots(113–248 mg/kg for Zibo No. 5 and 130–288 mg/kg Zibo No. 8), biomass tolerance indexes(64.9%–74.6% for Zibo No. 5 and 80.1%–90.9% for Zibo No. 8) in the hydroponic experiment and survival rates(0% for Zibo No. 5 and 100% for Zibo No. 8)determined by the addition of EDTA in the pot experiment, suggesting that Zibo No. 8 has higher tolerance than Zibo No. 5. Moreover, the castor cultivars have low bioconcentration factors(4.80% for Zibo No. 5 and 5.43% for Zibo No. 8) and low translocation factors(<1%).Consequently, Zibo No. 8 can participate in Cd phytostabilization in highly Cd-polluted areas. The results indicated that glutathione(GSH) as a non-enzymatic antioxidant, and antioxidant enzymes including superoxide dismutase(SOD), catalase(CAT) and guaiacol peroxidase(GPX), were cultivar- and dose-dependent. 2Hepatitis C Virus Infection Downregulates the Ligands of the Activating Receptor NKG2D显示文摘Natural killer (NK) cells are a major component of the host innate immune defense against various pathogens. Several viruses, including hepatitis C virus (HCV), have developed strategies to evade the NK-cell response. In our study, we found HCV infection could trigger DNA damage response by both ataxia telangiectasia mutated (ATM) and ATM- and Rad3-related (ATR) pathways. Recent reports had revealed that NKG2D ligands (NK cell-activating ligands) were upregulated when a major DNA damage checkpoint pathway was activated. However, here we found that DNA damage response was activated but NKG2D ligands were downregulated upon HCV infection. Further studies showed that the protease NS3/4A of HCV which had been shown relation with immune invasion contributed to the reduced expression of NKG2D ligands. 3A new Cu^(2+)-induced color reaction of a rhodamine derivative N-(3-carboxy)acryloyl rhodamine B hydrazide显示文摘A new rhodamine derivative,N-(3-carboxy)acryloyl rhodamine B hydrazide(CARB),has been synthesized,and its unusual spectroscopic reaction with Cu2+ has been investigated.The derivative exhibits a rapid and reversible non-fluorescent absorption upon coordination to Cu2+,which is a rather unusual phenomenon for rhodamine B derivatives.Stoichiometric measurements using the Job's method and the molar ratio method reveal that one CARB molecule combines two Cu2+ ions,and the two Cu2+ ions play different roles:one opens the spirocyclic structure and the other quenches the fluorescence of the xanthene moiety.This reaction mechanism is supported by a comparative study on the model compound N-acryloyl rhodamine B hydrazide as well as by the density functional theory calculations.Furthermore,the absorption response of CARB is highly selective for Cu2+ over other common ions,which implies that CARB may be used as a colorimetric probe for the rapid visual detection of Cu2+.SUN ChengDong CHEN JianMing MA HuiMin LIU Yang ZHANG JingHua LIU QingJun 2011Science China Chemistry2011,54,7:3
4Microbial diversity in the sediments of the southern Mariana Trench显示文摘Microbial diversity in the abyssal sediments beneath the seafloor of 30,94,and 151cm near the southern end of the Mariana Trench was analyzed in the Illumina HiSeq 2500 platform.Results show that the microbial populations were dominated by bacteria but merely no archaea were identifi ed at the three depths.In the bacterial community,Proteobacteria and Firmicutes dominated the total taxon tags,followed by Bacteroidetes,Actinobacteria,Planctomycetes,Cyanobacteria,and Chloroflexi,which together account for over 99%of the total population.Similar to that in the seawater in the trench,the operational taxonomic units(OTUs)belonging to Gammaproteobacteria from the sediment samples showed high abundance.However,common bacterial OTUs in the water of the trench including Nitrospirae and Marinimicrobia were hardly found in the sediments from the southern Mariana Trench or the hadal region.Therefore,this study documented for the first time the compositions of microbial diversity in the trench sediments,revealed the difference in microbial diversity in water and sediment of the trench and will enrich the knowledge on the microbial diversity in the abyssal areas.LI Yingjie CAO Wenrui WANG Yan MA Qingjun 2019Journal of Oceanology and Limnology2019,37,3:3

8Intrinsic physical relationships between rotor modal shapes and instantaneous vibrational energy flow transmission characteristics:Theoretical and numerical analysis and application显示文摘The modal vibration of the rotor is the main cause of excessive vibration of the aeroengine overall structure.To attenuate the vibration of the rotor under different modal shapes from the perspective of energy control,the intrinsic physical relationships between rotor modal shapes and instantaneous vibrational energy flow transmission characteristics is derived from the general equation of motion base on the structural intensity method.A dual-rotor-support-casing coupling model subjected to the rotor unbalanced forces is established by the finite element method in this paper.The transmission,conversion and balance relationships of the vibrational energy flow for the rotors in the first-order bending modal shape,the conical whirling modal shape and the translational modal shape are analyzed,respectively.The results show that the vibrational energy flow transmitted to the structure can be converted into the strain energy,the kinetic energy and the energy dissipated by the damping of the structure.The vibrational energy flow transmission characteristics of rotors with different modal shapes are quite different.Especially for the first-order bending modal shape,the vibrational energy flow and the strain energy are transmitted and converted to each other in the middle part of the rotor shaft,resulting in large deformation at this part.To attenuate this harmful vibration,the influences of grooving on the shaft on the first-order bending vibration are studied from the perspective of transmission control of vibrational energy flow.This study can provide theoretical references and guidance for the vibration attenuation of the rotors in different modal shapes from a more essential perspective.Yingqun MA Qingjun ZHAO Wei ZHAO Binbin LIU Long HAO 2020Chinese Journal of Aeronautics2020,33,12:1

12Structural analysis of a shrimp thymidylate synthase reveals species-specific interactions with dUMP and raltitrexed显示文摘Thymidylate synthase(TS)is a key enzyme in the de novo biosynthesis of thymidine monophosphate,serving as a well-known drug target in chemotherapy against cancers and infectious diseases.Additional to its clinical value,TS is supposed to be a promising drug target in aquatic-disease control.To facilitate designing pathogen-specific TS inhibitors for shrimp-disease control,we report the crystal structures of TS from Litopenaeus vannamei(LvTS)in the apo form,LvTS-dUMP complex and LvTS-dUMP-raltitrexed complex at 2.27Å,1.54Å,and 1.56Åresolution,respectively.LvTS shares a similar fold with known TSs,existing as a dimer in the crystal.The apo LvTS and LvTS-dUMP take an open conformation,and raltitrexed binding induces structural changes into a closed conformation in LvTS-dUMP-raltitrexed.Compared to those in other known TS-dUMP-raltitrexed complexes with the closed conformation,the C-terminal loop in LvTS-dUMP-raltitrexed shifts its position away from the bound raltitrexed;the distance between C6 of dUMP and Sγof the catalytic cysteine is obviously longer than that in the known TS structures with closed conformations,resembling that in the TS structures with open conformations.Other species-specific interactions with dUMP and raltitrexed are also observed.Therefore,LvTS-dUMP-raltitrexed adopts a loosely closed conformation with structural features intermediate between the closed and the open conformations that were reported in other TSs.Our study provides the first crustcean TS structure,and reveals species-specific interactions between TSs and the ligands,which would facilitate designing pathogen-specific TS inhibitors for shrimp-disease control.LIU Changshui ZANG Kun LI Shihao LI Fuhua MA Qingjun 2020Journal of Oceanology and Limnology2020,38,6:1

17Raltitrexed as a synergistic hyperthermia chemotherapy drug screened in patient-derived colorectal cancer organoids显示文摘Objective:Organoids have recently been used as in vitro models to screen chemotherapy drugs in combination with hyperthermia treatment in colorectal cancer.Our research aimed to establish a library of patient-derived colorectal cancer organoids to evaluate synergism between chemotherapy drugs and hyperthermia;validate an index of the hyperthermia chemotherapy sensitization enhancement ratio(HCSER)to identify the chemotherapeutics most enhanced by hyperthermia;and recommend chemotherapy drugs for hyperthermic intraperitoneal treatment.Methods:Organoids were grown from cells extracted from colorectal cancer patient samples or colorectal cancer cell lines.Cells from both sources were encapsulated in 3 D Matrigel droplets,which were formulated in microfluidics and phase-transferred into identical cell-laden Matrigel microspheres.The microspheres were seeded in 96-well plates,with each well containing a single microsphere that developed into an organoid after 7 days.The organoids were used to evaluate the efficacy of chemotherapy drugs at both 37℃ as a control and 43℃ for 90 min to examine hyperthermia synergism.Cell viability was counted with 10%CCK8.Results:We successfully established a library of colorectal cancer organoids from 22 patient parental tumors.We examined the hyperthermia synergism of 7 commonly used hyperthermic intraperitoneal chemotherapy drugs.In 11 of the 22 patient organoids,raltitrexed had significant hyperthermia synergism,which was indexed as the highest HCSER score within each patient group.Conclusions:Our results primarily demonstrated the use of patient-derived colorectal cancer organoids as in vitro models to evaluate hyperthermia synergistic chemotherapeutics.We found that hyperthermia enhanced the effect of raltitrexed the most among the common anti-colorectal cancer drugs.Lisi Zeng Quanxing Liao Haoran Zhao Shengwei Jiang Xianzi Yang Hongsheng Tang Qingjun He Xiansheng Yang Shuxian Fang Jinfu He Weiwen Cui Laiqiang Huang Shaohua Ma Shuzhong Cui 2021Cancer Biology & Medicine2021,18,3:0
18Protection against malaria by immunization with plasmid DNA encoding cholera toxin B subunit and Plasmodium falciparum epitopes显示文摘Cholera toxin B subunit is a good carrier protein and an effective adjuvant which can boost both cellular and humoral immunity. DNA fragments encoding B cell, Th cell and CTL epitopes of P. falciparum CS, MSA-1, MSA-2 and RESA antigens were cloned down-stream of cholera toxin B subunit gene in the same reading frame. Another modification using IL2 as adjuvant was also made. High titer of anti-malaria epitopes antibodies and strong cellular immunogenicity were elicited after Balb/c mice were immunized three times with 100 μg recombinant plasmid DNA dissolved in 100 μL PBS. 200 vaccinees were challenged with mouse Plasmodium yoelli to investigate if cross protection existed. The protective efficacy was about 50%. And it is found that the protective efficacy is correlated with CTL activity which was considered to be the primary effects of anti-sporozoite protective immunity. 20Box-Behnken experimental design for optimizing process parameters in carbonate-promoted direct thiophene carboxylation reaction with carbon dioxide显示文摘A feasible synthesis route is developed for achieving the direct carboxylation of thiophene and CO_(2) in a relatively mild solvent-free carboxylate-assisted carbonate(semi)molten state.The effects of reaction factors on the carboxylate yield are investigated in the preliminary screening experiments,and the phase behavior analysis of the reaction medium is detected through the thermal characterization analysis of insitu high temperature X-ray diffraction measurement(in-situ XRD).The application of response surface methodology(RSM)based on the Box-Behnken design(BBD)is conducted to investigate the effect of the reaction parameters,such as reaction temperatu re,carbonate proportion,CO_(2) pressure and thiophene amount,on the product yield.The regressed second-order polynomial model equation well correlates all the independent variables.The analysis of variance(ANOVA)results reveal that the quadratic effect of reaction temperature is the most effective parameter in this carboxylation reaction owing to it’s the highest contribution to the sum of square(30.18%).The optimum reaction conditions for maximum product yield are the reaction temperature of 287℃,carbonate proportion of 32.20%,CO_(2) pressure of 1.0MPa and thiophene amount of 9.35 mmol.Operating under these selected experimental conditions,a high product yield(50.98%)can be achieved.Qingjun Zhang Youguang Ma Xigang Yuan Aiwu Zeng 2022Chinese Journal of Chemical Engineering2022,,10:0
