| 2 | Void and cloud patterns in a dusty plasma显示文摘Voids and complex cloud patterns have been observed in a radio frequency (rf) discharge dusty plasma system. The structures and behaviors of these patterns were investigated in two- and three-dimensions by shifting an il- lumination laser sheet beam up and down. Two-dimensional (2D) circular voids, thin ring voids, and their coexistent voids were observed at the early stage of particle growth. The cor- responding three-dimensional (3D) structures were dome, shell, and their coexistent voids, respectively. The complex fluid-like cloud patterns occurred above the 3D dome voids. | 6 | Numerical analysis of time-varying wear with elastic deformation in line contact显示文摘Wear is an important factor for failures of mechanical components. Current research on wear is mainly focused on experiments while the numerical simulation of wear is hardly used owing to the complexities of the wear process. Explaining the effect of friction on the wear process is important, as it will lead to a deeper understanding of the evolution of wear. This study proposed a numerical method to expound the wear process in the contact between an elastic cylinder and a half-space simulating the ring-block tester. There are two difficulties during the calculation; one is that the contact shapes vary with time, causing the pressure distribution to change simultaneously and the other is the integral equation for calculating the contact pressure under different worn shapes. In the present study, the wear rate was computed using Archard's law and the wear process was calculated step by step until the specified total sliding distance was achieved. During each step of the calculation, the contact topography was updated. The simulation intuitively reproduced the contact state of change from line to surface contact throughout the wear process. | 8 | BRD4 interacts with PML/RARα in acute promyelocytic leukemia显示文摘Bromodomain-containing 4(BRD4)has been considered as an important requirement for disease maintenance and an attractive therapeutic target for cancer therapy.This protein can be targeted by JQ1,a selective small-molecule inhibitor.However,few studies have investigated whether BRD4influenced acute promyelocytic leukemia (APL),and whether BRD4had interaction with promyelocytic leukemia-retinoic acid receptor a (PML/RARa)fusion protein to some extent.Results from cell viability assay,cell cycle analysis,and Annexin-V/PI analysis indicated that JQ1inhibited the growth of NB4cells,an APL-derived cell line,and induced NB4cell cycle arrest at G1and apoptosis.Then,we used co-immunoprecipitation (co-IP)assay and immunoblot to demonstrate the endogenous interaction of BRD4and PML/RARa in NB4cells.Moreover,downregulation of PML/RARa at the mRNA and protein levels was observed upon JQ1treatment.Furthermore,results from the RT-qPCR,ChIP-qPCR,and re-ChIP-qPCR assays showed that BRD4and PML/RARu co-existed on the same regulatory regions of their target genes.Hence,we showed a new discovery of the interaction of BRD4and PML/ RARa,as well as the decline of PML/RARa expression,under JQ1treatment. | Qun Luo Wanglong Deng Haiwei Wang Huiyong Fan Ji Zhang | 2018 | Frontiers of Medicine2018,12,6: | 1 |
| 9 | Implicit discontinuous Galerkin method on agglomerated high-order grids for 3D simulations显示文摘High quality of geometry representation is regarded essential for high-order methods to maintain their high-order accuracy. An agglomerated high-order mesh generating method is investigated in combination with discontinuous Galerkin(DG) method for solving the 3D compressible Euler and Navier-Stokes equations. In this method, a fine linear mesh is first generated by standard commercial mesh generation tools. By taking advantage of an agglomeration method, a quadratic high-order mesh is quickly obtained, which is coarse but provides a high-quality geometry representation, thus very suitable for high-order computations. High-order discretizations are performed on the obtained grids with DG method and the discretized system is treated fully implicitly to obtain steady state solutions. Numerical experiments on several flow problems indicate that the agglomerated high-order mesh works well with DG method in dealing with flow problems of curved geometries. | 12 | Gut microbiome,cognitive function and brain structure:a multi-omics integration analysis显示文摘Background:Microbiome-gut-brain axis may be involved in the progression of age-related cognitive impairment and relevant brain structure changes,but evidence from large human cohorts is lacking.This study was aimed to investigate the associations of gut microbiome with cognitive impairment and brain structure based on multi-omics from three independent populations.Methods:We included 1430 participants from the Guangzhou Nutrition and Health Study(GNHS)with both gut microbiome and cognitive assessment data available as a discovery cohort,of whom 272 individuals provided fecal samples twice before cognitive assessment.We selected 208 individuals with baseline microbiome data for brain magnetic resonance imaging during the follow-up visit.Fecal 16S rRNA and shotgun metagenomic sequencing,tar-geted serum metabolomics,and cytokine measurements were performed in the GNHS.The validation analyses were conducted in an Alzheimer’s disease case-control study(replication study 1,n=90)and another community-based cohort(replication study 2,n=1300)with cross-sectional dataset.Results:We found protective associations of specific gut microbial genera(Odoribacter,Butyricimonas,and Bac-teroides)with cognitive impairment in both the discovery cohort and the replication study 1.Result of Bacteroides was further validated in the replication study 2.Odoribacter was positively associated with hippocampal volume(β,0.16;95%CI 0.06-0.26,P=0.002),which might be mediated by acetic acids.Increased intra-individual alterations in gut microbial composition were found in participants with cognitive impairment.We also identified several serum metabolites and inflammation-associated metagenomic species and pathways linked to impaired cognition.Conclusions:Our findings reveal that specific gut microbial features are closely associated with cognitive impair-ment and decreased hippocampal volume,which may play an important role in dementia development. | Xinxiu Liang Yuanqing Fu Wen-ting Cao Zhihong Wang Ke Zhang Zengliang Jiang Xiaofang Jia Chun-ying Liu Hong-rou Lin Haili Zhong Zelei Miao Wanglong Gou Menglei Shuai Yujing Huang Shengdi Chen Bing Zhang Yu-ming Chen Ju-Sheng Zheng | 2022 | Translational Neurodegeneration2022,11,1: | 1 |
