3PSA controls hepatic lipid metabolism by regulating the NRF2 signaling pathway显示文摘The activity of proteinase is reported to correlate with the development and progression of nonalcoholic fatty liver disease(NAFLD).Puromycin-sensitive aminopeptidase(PSA/NPEPPS)is an integral nontransmembrane enzyme that functions to catalyze the cleavage of amino acids near the N-terminus of polypeptides.A previous study suggested that this enzyme acts as a regulator of neuropeptide activity;however,the metabolic function of this enzyme in the liver has not been explored.Here,we identified the novel role of PSA in hepatic lipid metabolism.Specifically,PSA expression was lower in fatty livers from NAFLD patients and mice(HFD,ob/ob,and db/db).PSA knockdown in cultured hepatocytes exacerbated diet-induced triglyceride accumulation through enhanced lipogenesis and attenuated fatty acid β-oxidation.Moreover,PSA mediated activation of the master regulator of antioxidant response,nuclear factor erythroid 2-related factor 2(NRF2),by stabilizing NRF2 protein expression,which further induced downstream antioxidant enzymes to protect the liver from oxidative stress and lipid overload.Accordingly,liver-specific PSA overexpression attenuated hepatic lipid accumulation and steatosis in ob/ob mice.Furthermore,in human liver tissue samples,decreased PSA expression correlated with the progression of NAFLD.Overall,our findings suggest that PSA is a pivotal regulator of hepatic lipid metabolism and its antioxidant function occurs by suppressing NRF2 ubiquitination.Moreover,PSA may be a potential biomarker and therapeutic target for treating NAFLD.Bangliang Huang Xin Xiong Linlin Zhang Xiufei Liu Yuren Wang Xiaoli Gong Qian Sang Yongling Lu Hua Qu Hongting Zheng Yi Zheng 2021Journal of Molecular Cell Biology2021,13,7:4
4Sequential infection with H1N1 and SARS-CoV-2 aggravated COVID-19 pathogenesis in a mammalian model, and covaccination as an effective method of prevention of COVID-19 and influenza显示文摘Influenza A virus may circulate simultaneously with the SARS-CoV-2 virus,leading to more serious respiratory diseases during this winter.However,the influence of these viruses on disease outcome when both influenza A and SARS-CoV-2 are present in the host remains unclear.Using a mammalian model,sequential infection was performed in ferrets and in K18-MCE2 mice,with SARS-CoV-2 infection following H1N1.We found that co-infection with H1N1 and SARS-CoV-2 extended the duration of clinical manifestation of COVID-19,and enhanced pulmonary damage,but reduced viral shedding of throat swabs and viral loads in the lungs of ferrets.Moreover,mortality was increased in sequentially infected mice compared with single-infection mice.Compared with singlevaccine inoculation,co-inoculation of PiCoVacc(a SARS-CoV-2 vaccine)and the flu vaccine showed no significant differences in neutralizing antibody titers or virus-specific immune responses.Combined immunization effectively protected K18-MCE2 mice against both H1N1 and SARS-CoV-2 infection.Our findings indicated the development of systematic models of co-infection of H1N1 and SARS-CoV-2,which together notably enhanced pneumonia in ferrets and mice,as well as demonstrated that simultaneous vaccination against HINT and SARS-CoV-2 may be an effective prevention strategy for the coming winter.Linlin Bao Wei Deng Feifei Qi Qi Lv Zhiqi Song Jiangning Liu Hong Gao Qiang Wei Pin Yu Yanfeng Xu Yajin Qu Fengdi Li Jing Xue Shuran Gong Mingya Liu Guanpeng Wang Shunyi Wang Binbin Zhao Bin Cong Chuan Qin 2021Signal Transduction and Targeted Therapy2021,6,6:4

10Relationship between joint development in rock and coal seams in the southeastern margin of the Ordos basin显示文摘To predict joint development characteristics of coal seams, joint characteristics of rock seams from 88field stations were observed and comparisons were made between joint characteristics of coal and rock seams at 10 coal outcrops. Additionally, detailed joint measurements of underground coal seams were taken at two coal mines. This study investigated the effects of seam thickness, lithology, and structure on joint development and established the relationship between joint development of coal and rock seams, which allowed predictions of predominant joint densities for the No.5 coal seam in the southeastern margin of the Ordos basin. The results show that outcrop and underground coal seams exhibit the same joint systems as rock seams. The joints are mainly upright. Predominant joints strike 55° on average, followed by joints striking 320°. The joint density of the coal seam is 18.7–22.5 times that of the sandstone seam at the same thickness. The predominant joint density of the No.5 coal seam, controlled by the structure, is 4–20 joints per meter. Joint densities exhibit high values at intersecting areas of faults and folds and decrease values in structurally stable areas. 12A novel degradable injectable HLC-HPA hydrogel with antiinflammatory activity for biomedical materials:Preparation,characterization,in vivo and in vitro evaluation显示文摘In situ injectable hydrogels,which have great potential in tissue engineering,are characterized by simple preparation,minimal invasiveness and adaptation to complex shapes.However,injectable hydrogels have higher requirements for biocompatibility and safety due to their use in vivo implantation.Therefore,in this study,a human-like collagen(HLC)-based in situ gel-forming injectable HLC-HPA hydrogel was synthesized by combining the amino group of HLC with the carboxyl group of HPA activated using 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide hydrochloride(EDC)and N-hydroxysuccinimide(NHS),followed by crosslinking by horseradish peroxidase(HRP)and H_2O_2,and used as scaffold material for tissue engineering.The hydrogel stiffness,gel time and biodegradation rate could be easily and independently adjusted by varying the H_2O_2 and HRP concentrations.Scanning electron microscope(SEM)clarified the homogeneous porous and interconnected internal structures of the hydrogel.In vitro cell viability and in vivo degradation experiments confirmed that the HLC-HPA hydrogel had good biodegradability and excellent biocompatibility.Interestingly,in cultured macrophages,the HLC-HPA hydrogel showed anti-inflammatory activity by reducing the amount of the pro-inflammatory cytokines tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)and increasing the secretion of the anti-inflammatory cytokine interleukin-10(IL-10)induced by lipopolysaccharide(LPS).Meanwhile,in animal experiments,HLC-HPA hydrogels exhibit excellent biocompatibility and have properties of hemostasis and reduction of inflammatory response.Therefore,the HLC-HPA hydrogel prepared in this study has great potential for development for use in the biomedical field.GAO Shang QU LinLin ZHU ChenHui OUYANG PingKai FAN DaiDi 2020Science China(Technological Sciences)2020,63,11:1
13Ginsenoside Rk3 is a novel PI3K/AKT-targeting therapeutics agent that regulates autophagy and apoptosis in hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC)is the third leading cause of cancer death worldwide.Ginsenoside Rk3,an important and rare saponin in heat-treated ginseng,is generated from Rg1 and has a smaller molecular weight.However,the anti-HCC efficacy and mechanisms of ginsenoside Rk3 have not yet been characterized.Here,we investigated the mechanism by which ginsenoside Rk3,a tetracyclic triterpenoid rare ginsenoside,inhibits the growth of HCC.We first explored the possible potential targets of Rk3 through network pharmacology.Both in vitro(HepG2 and HCC-LM3 cells)and in vivo(primary liver cancer mice and HCC-LM3 subcutaneous tumor-bearing mice)studies revealed that Rk3 significantly inhibits the proliferation of HCC.Meanwhile,Rk3 blocked the cell cycle in HCC at the G1 phase and induced autophagy and apoptosis in HCC.Further proteomics and siRNA experiments showed that Rk3 regulates the phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)pathway to inhibit HCC growth,which was validated by molecular docking and surface plasmon resonance.In conclusion,we report the discovery that ginsenoside Rk3 binds to PI3K/AKT and promotes autophagy and apoptosis in HCC.Our data strongly support the translation of ginsenoside Rk3 into novel PI3K/AKT-targeting therapeutics for HCC treatment with low toxic side effects.Linlin Qu Yannan Liu Jianjun Deng Xiaoxuan Ma Daidi Fan 2023Journal of Pharmaceutical Analysis2023,13,5:1

18Intravenous injection of AAV-PHP.eB across the blood-brain barrier in the adult mouse for central nervous system gene therapy显示文摘Objective To verify the neurotypicality of AAV-PHP.eB after tail vein injection in adult mice and its efficiency in crossing the blood-brain barrier(BBB).Methods The rAAV-SYN-GFP plasmid was constructed, and adult C57 BL mice were injected with AAV PHP.eB: SYN-GFP in the tail vein(300 nL, virus titer 3 × 10~9 vg) and in the prefrontal lobe(50 L, virus tite5 × 10^(11) vg). The green fluorescent protein(GFP) signal in the brain was observed at two weeks, while the GFP signal in the peripheral organs was observed at four weeks. Results Two weeks after tail vein injection, GFP expression was observed throughout the brain especially in the cortex, hippocampus, and geniculate nucleus. No GFP signal was observed or detected by western blotting in the peripheral organs after four weeks. GFP signal was observed mainly at the loca site after prefrontal lobe injection.Conclusion AAV-PHP.eB: SYN-GFP can effectively cross the BBB in adult mice. 20Phenotypic and Genetic Characteristics of a Bordetella hinzii Isolated from a Rex Rabbit显示文摘A Bordetella hinzii strain,isolated from the lung of a dead breast-feeding infant rex rabbit with a respiratory infection,was used as study object to investigate its phenotypic and genetic characteristics. The strain could grow on Mac Conkey agar and was gram-negative,oxidase-positive,catalase-positive,short rodshaped,and it can produce alkali from malonate. Its 6S rRNA sequence was partially identical with B. hinzii( AF177667). Animal experiments revealed nasal mucous hyperemia,tracheal congestion and bronchopneumonia in 28-day old rex rabbits.
