1First dark matter search results from the PandaX-Ⅰ experiment显示文摘We report on the first dark-matter(DM)search results from PandaX-I,a low threshold dual-phase xenon experiment operating at the China JinPing Underground Laboratory.In the 37-kg liquid xenon target with 17.4 live-days of exposure,no DM particle candidate event was found.This result sets a stringent limit for low-mass DM particles and disfavors the interpretation of previously-reported positive experimental results.The minimum upper limit,3.7×10-44cm2,for the spin-independent isoscalar DM-particle-nucleon scattering cross section is obtained at a DM-particle mass of 49 GeV/c2at 90%confidence level.XIAO MengJiao XIAO Xiang ZHAO Li CAO XiGuang CHEN Xun CHEN YunHua CUI XiangYi FANG DeQing FU ChangBo GIBONI Karl L. 3Differential Characteristics of the Upper Ordovician-Lower Silurian Wufeng-Longmaxi Shale Reservoir and its Implications for Exploration and Development of Shale Gas in/around the Sichuan Basin显示文摘The Upper Ordovician Wufeng-Lower Silurian Longmaxi shale is widely distributed in the Sichuan Basin and its periphery,which is the key stratum for marine shale gas exploration and development(E&D)in China.Based on sedimentary environment,material basis,storage space,fracability and reservoir evolution data,the reservoir characteristics of the Wufeng-Longmaxi shale and their significance for shale gas E&D are systematically compared and analyzed in this paper.The results show that(1)the depocenter of the Wufeng(WF)-Longmaxi(LM)shale gradually migrates from east to west.The high-quality shale reservoirs in the eastern Sichuan Basin are mainly siliceous shales,which are primarily distributed in the graptolite shale interval of WF2-LM5.The high-quality reservoirs in the southern Sichuan Basin are mainly calcareous-siliceous and organic-rich argillaceous shales,which are distributed in the graptolite shale interval of WF2-LM7.(2)Deep shale gas(the burial depth>3500 m)in the Sichuan Basin has high-ultrahigh pressure and superior physical properties.The organic-rich siliceous,calcareous-siliceous and organic-rich argillaceous shales have suitable reservoir properties.The marginal area of the Sichuan Basin has a higher degree of pressure relief,which leads to the argillaceous and silty shales evolving into direct cap rocks with poor reservoir/good sealing capacity.(3)Combining shale gas exploration practices and impacts of lithofacies,depth,pressure coefficient and brittle-ductile transition on the reservoir properties,it is concluded that the favorable depth interval of the Wufeng-Longmaxi shale gas is 2200~4000 m under current technical conditions.(4)Aiming at the differential reservoir properties of the Wufeng-Longmaxi shale in the Sichuan Basin and its periphery,several suggestions for future research directions and E&D of shale gas are formulated.WANG Ruyue HU Zongquan LONG Shengxiang LIU Guangxiang ZHAO Jianhua DONG Li DU Wei WANG Pengwei YIN Shuai 2019Acta Geologica Sinica(English Edition)2019,93,3:8
4The Influence of Extractable Organic Matter on Pore Development in the Late Triassic Chang 7 Lacustrine Shales, Yanchang Formation, Ordos Basin, China显示文摘To investigate the influence of extractable organic matter(EOM) on pore evolution of lacustrine shales, Soxhlet extraction, using dichloromethane, was performed on a series of Chang 7 shale samples(Ordos Basin, China) with vitrinite reflectance of 0.64% to 1.34%. Low-pressure gas adsorption experiments were conducted on the samples before and after extraction. The pore structure parameters were calculated from the gas adsorption data. The results show complex changes to the pore volumes and surface areas after extraction. The pore development of both the initial and extracted samples is strongly controlled by total organic carbon(TOC) content. Micropores developed mainly in organic matter(OM), while mesopores and macropores predominantly developed in fractions other than OM. The influence of EOM on micropores is stronger than on mesopores and macropores. 5Neomycin biosynthesis is regulated positively by AfsA-g and NeoR in Streptomyces fradiae CGMCC 4.7387显示文摘Neomycins are a group of aminoglycoside antibiotics with both clinical and agricultural applications.To elucidate the regulatory mechanism of neomycin biosynthesis,we completed draft genome sequencing of a neomycin producer Streptomyces fradiae CGMCC 4.7387 from marine sediments,and the neomycin biosynthesis gene cluster was identified.Inactivation of the afsA-g gene encoding a γ-butyrolactone(GBL) synthase in S.fradiae CGMCC 4.7387 resulted in a significant decrease of neomycin production.Quantitative RT-PCR analysis revealed that the transcriptional level of neoR and the aphA-neoGH operon were reduced in the afsA-g::aac(3)Ⅳ mutant.Interestingly,a conserved binding site of AdpA,a key activator in the GBL regulatory cascade,was discovered upstream of neoR,a putative regulatory gene encoding a protein with an ATPase domain and a tetratricopeptide repeat domain.When neoR was inactivated,the neomycin production was reduced about 40%in comparison with the WT strain.Quantitative RT-PCR analysis revealed that the transcriptional levels of genes in the aphA-neoGH operon were reduced clearly in the neoR::aac(3)Ⅳ mutant.Finally,the titers of neomycin were improved considerably by overexpression of qfsA-gand neoR in S.fradiae CGMCC 4.7387.Xiangxi Meng Wenzhao Wang Zhoujie Xie Pengwei Li Yue Li Zhengyan Guo Yingjian Lu Jie Yang Kaile Guan Zhaoxin Lu Huarong Tan Yihua Chen 2017Science China(Life Sciences)2017,60,9:7
6Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 7Synthesis,biological activities and 3D-QSAR studies of(R)-2-phenyl-4,5-dihydrothiazole-4-carboxamide derivatives containing a sulfur ether moiety显示文摘A series of(R)-2-phenyl-4,5-dihydrothiazole-4-carboxamide derivatives containing a sulfur ether moiety were synthesized and characterized on the basis of NMR and elemental analysis(EA). The crystal structure of(R)-N-(2-methyl-1-(methylthio)propan-2-yl)-2-(4-nitrophenyl)-4,5-dihydrothiazole-4-carboxamide(13 d) was determined to show R configuration. The bioasssy results indicated that most title compounds displayed good and broad spectrum antifungal activities against several phytopathogenic fungi. The structure activity relationships were discussed. Based on the antifungal activity of title compounds against Phytophthora capsici, a CoMSIA calculation was performed to establish a 3 D-QSAR model, which revealed that electrostatic and hydrophobic fields were the two most significant factors for antifungal activity. 8Assessment of the individual and combined effects of Rht8 and Ppd-D1a on plant height, time to heading and yield traits in common wheat显示文摘Grain yield in cereal crops is a complex trait controlled by multiple genes and influenced by developmental processes and environment. Here we report the effects of alleles Rht8 and Ppd-D1 a on plant height, time to heading, and grain yield and its component traits. Association analysis and quantitative trait locus mapping using phenotypic data from 15 environments led to the following conclusions. First, both Rht8 and Ppd-D1 a reduce plant height. However, Ppd-D1 a but not Rht8 causes earlier heading.Second, both Rht8 and Ppd-D1 a promote grain yield and affect component traits. Their combined effects are substantially larger than those conferred by either allele alone.Third, promotion of grain yield by Rht8 and Ppd-D1 a is through increasing fertile spikelet number. We speculate that Rht8 and Ppd-D1 a act independently and additively in control of plant height, grain yield and yield component. 9The RECEPTOR-LIKE PROTEIN53 immune complex associates with LLG1 to positively regulate plant immunity显示文摘Pattern recognition receptors(PRRs)sense ligands in pattern-triggered immunity(PTI).Plant PRRs include numerous receptor-like proteins(RLPs),but many RLPs remain functionally uncharacterized.Here,we examine an Arabidopsis thaliana RLP,RLP53,which positively regulates immune signaling.Our forward genetic screen for suppressors of enhanced disease resistance1(edr1)identified a point mutation in RLP53 that fully suppresses disease resistance and mildewinduced cell death in edr1 mutants.The rlp53 mutants showed enhanced susceptibility to virulent pathogens,including fungi,oomycetes,and bacteria,indicating that RLP53 is important for plant immunity.The ectodomain of RLP53 contains leucine-rich repeat(LRR)motifs.RLP53 constitutively associates with the LRR receptorlike kinase SUPPRESSOR OF BRASSINOSTEROIDINSENSITIVE1-ASSOCIATEDKINASE(BAK1)-INTERACTINGRECEPTORKINASE1(SOBIR1)and interacts with the co-receptor BAK1 in a pathogen-induced manner.The double mutation sobir1-12 bak1-5 suppresses edr1-mediated disease resistance,suggesting that EDR1 negatively regulates PTI modulated by the RLP53–SOBIR1–BAK1 complex.Moreover,the glycosylphosphatidylinositol(GPI)-anchored protein LORELEI-LIKE GPI-ANCHORED PROTEIN1(LLG1)interacts with RLP53 and mediates RLP53 accumulation in the plasma membrane.We thus uncovered the role of a novel RLP and its associated immune complex in plant defense responses and revealed a potential new mechanism underlying regulation of RLP immune function by a GPI-anchored protein.Renjie Chen Pengwei Sun Guitao Zhong Wei Wang Dingzhong Tang 2022Journal of Integrative Plant Biology2022,64,9:4
10Arabidopsis E3 ligase KEG associates with and ubiquitinates MKK4 and MKK5 to regulate plant immunity显示文摘Mitogen-activated protein kinase(MAPK) cascades are highly conserved signaling modules that regulate plant immune responses. The Arabidopsis thaliana Raf-like MAPK kinase kinase ENHANCED DISEASE RESISTANCE1(EDR1) is a key negative regulator of plant immunity that affects the protein levels of MKK4 and MKK5, two important MAPK cascade members, but the underlying mechanism is poorly understood. Here, genome-wide phosphorylation analysis demonstrated that the E3 ligase KEEP ON GOING(KEG) is phosphorylated in the edr1 mutant but not the wild type, suggesting that EDR1 negatively affects KEG phosphorylation. The identified phosphorylation sites in KEG appear to be important for its accumulation. The keg-4 mutant, a previously identified edr1 suppressor, enhances susceptibility to the powdery mildew pathogen Golovinomyces cichoracearum. In addition, MKK4 and MKK5 protein levels are reduced in the keg-4 mutant. Furthermore,we demonstrate that MKK4 and MKK5 associate with full-length KEG, but not with truncated KEG-RK or KEG-RKA, and that KEG ubiquitinates and mediates the degradation of MKK4 and MKK5. 12The genome and gene editing system of sea barleygrass provide a novel platform for cereal domestication and stress tolerance studies显示文摘The tribe Triticeae provides important staple cereal crops and contains elite wild species with wide geneticdiversity and high tolerance to abiotic stresses. Sea barleygrass (Hordeum marinum Huds.), a wildTriticeae species, thrives in saline marshlands and is well known for its high tolerance to salinity and waterlogging. Here, a 3.82-Gb high-quality reference genome of sea barleygrass is assembled de novo, with 3.69Gb (96.8%) of its sequences anchored onto seven chromosomes. In total, 41 045 high-confidence (HC)genes are annotated by homology, de novo prediction, and transcriptome analysis. Phylogenetics, nonsynonymous/synonymous mutation ratios (Ka/Ks), and transcriptomic and functional analyses provide genetic evidence for the divergence in morphology and salt tolerance among sea barleygrass, barley, andwheat. The large variation in post-domestication genes (e.g. IPA1 and MOC1) may cause interspecies differences in plant morphology. The extremely high salt tolerance of sea barleygrass is mainly attributed tolow Na+ uptake and root-to-shoot translocation, which are mainly controlled by SOS1, HKT, and NHX transporters. 13Uncertainty quantification and reduction in metal additive manufacturing显示文摘Uncertainty quantification(UQ)in metal additive manufacturing(AM)has attracted tremendous interest in order to dramatically improve product reliability.Model-based UQ,which relies on the validity of a computational model,has been widely explored as a potential substitute for the time-consuming and expensive UQ solely based on experiments.However,its adoption in the practical AM process requires overcoming two main challenges:(1)the inaccurate knowledge of uncertainty sources and(2)the intrinsic uncertainty associated with the computational model.Here,we propose a data-driven framework to tackle these two challenges by combining high throughput physical/surrogate model simulations and the AM-Bench experimental data from the National Institute of Standards and Technology(NIST).We first construct a surrogate model,based on high throughput physical simulations,for predicting the three-dimensional(3D)melt pool geometry and its uncertainty with respect to AM parameters and uncertainty sources.We then employ a sequential Bayesian calibration method to perform experimental parameter calibration and model correction to significantly improve the validity of the 3D melt pool surrogate model.The application of the calibrated melt pool model to UQ of the porosity level,an important quality factor,of AM parts,demonstrates its potential use in AM quality control.The proposed UQ framework can be generally applicable to different AM processes,representing a significant advance toward physicsbased quality control of AM products.Zhuo Wang Chen Jiang Pengwei Liu Wenhua Yang Ying Zhao Mark F.Horstemeyer Long-Qing Chen Zhen Hu Lei Chen 2020npj Computational Materials2020,,1:2
14Fabricated temperature sensitive photocatalyst of PNIPAM@ZnO/C for controllable photocatalytic activity显示文摘PNIPAM@ZnO/C composite photocatalyst was prepared by cross-linking polymerization technology with N-isopropylacrylamide used as functional monomer, N,N'-methylenebis(acrylamide) used as crosslinking agent, ammonium persulfate used as initiator, and 3-(trimethoxysilyl) propyl methacrylate used as surface modification reagent. The morphology, structure, electrochemical and photocatalytic properties of as-prepared samples were characterized via the serial tests. The temperature-response performances of PNIPAM@ZnO/C were evaluated by the photocatalytic degradation of tetracycline(TC)under different temperatures. The results show that the synthesized composite photocatalysts possess the excellent and switchable photocatalytic activity. 16Identifying factors that influence soil heavy metals by using categorical regression analysis:A case study in Beijing,China显示文摘Identifying the factors that influence the heavy metal contents of soil could reveal the sources of soil heavy metal pollution.In this study,a categorical regression was used to identify the factors that influence soil heavy metals.First,environmental factors were associated with soil heavy metal data,and then,the degree of influence of different factors on the soil heavy metal contents in Beijing was analyzed using a categorical regression.The results showed that the soil parent material,soil type,land use type,and industrial activity were the main influencing factors,which suggested that these four factors were important sources of soil heavy metals in Beijing.In addition,population density had a certain influence on the soil Pb and Zn contents.The distribution of soil As,Cd,Pb,and Zn was markedly influenced by interactions,such as traffic activity and land use type,industrial activity and population density.The spatial distribution of soil heavy metal hotspots corresponded well with the influencing factors,such as industrial activity,population density,and soil parent material.In this study,the main factors affecting soil heavy metals were identified,and the degree of their influence was ranked.A categorical regression represents a suitable method for identifying the factors that influence soil heavy metal contents and could be used to study the genetic process of regional soil heavy metal pollution.Jun Yang Jingyun Wang Pengwei Qiao Yuanming Zheng Junxing Yang Tongbin Chen Mei Lei Xiaoming Wan Xiaoyong Zhou 2020Frontiers of Environmental Science & Engineering2020,14,3:1

18Construction of cDNA library, nucleotide sequence and analysis of entire genome of classical swine fever virus strain Shimen显示文摘According to the previously published CSFV sequences, 18 pairs of primers have been designed and synthesized, which cover the entire genome of CSFV strain Shimen. Each cDNA fragment has been amplified by RT-PCR from the anticoagulant blood of strain Shimen infected pig. The PCR products have been cloned respectively and sequenced. Results show that the cDNA library of strain Shimen and its nucleotide sequence have been obtained. The genomic RNA of strain Shimen is 12 298 nucleotides in length, containing a 5’ and a 3’ noncoding region 373 and 231 nt long respectively. 19Restored intestinal integrity,nutrients transporters,energy metabolism,antioxidative capacity and decreased harmful microbiota were associated with IUGR piglet's catch-up growth before weanling显示文摘Background:Intrauterine growth restriction(IUGR)is a major inducer of higher morbidity and mortality in the pig industry and catch-up growth(CUG)before weanling could significantly restore this negative influence.But there was limited knowledge about the underlying mechanism of CUG occurrence.Methods:Eighty litters of newborn piglets were divided into normal birth weight(NBW)and IUGR groups according to birth weight.At 26 d,those piglets with IUGR but over average body weight of eighty litters of weaned piglets were considered as CUG,and the piglets with IUGR still below average body weight were considered as NCUG.This study was conducted to systemically compare the intestinal difference among NBW,CUG and NCUG weaned piglets considering the crucial role of the intestine for piglet growth.Results:The results indicated that the m RNA expression of nutrients(amino acids,glucose,and fatty acids)transporters,and mitochondrial electron transport chain(ETC)I were upregulated in CUG piglets'gut with improved morphology compared with those NCUG,as well as the ratio of P-AMPK/AMPK protein expression which is the indicator of energy metabolism.Meanwhile,CUG piglet's gut showed higher antioxidative capacity with increased SOD and GSHPx activity,decreased MDA levels,as well as higher m RNA expressions of Nrf2,Keap1,SOD,and GSH-Px.Furthermore,inflammatory parameters including TNF-α,IL-1β,IL-6,and IL-12 factors,and the activation of MAPK and NF-κB signaling pathways were significantly elevated in the NCUG intestine,while the protein expression of ZO-1,Occludin and Claudin-1 was reduced.The alpha diversity of fecal microbiota was higher in CUG piglets in contrast with NCUG piglets,and the increased beneficial bacteria and decreased pathogenic bacteria was also observed in CUG piglets.Conclusions:CUG piglet's intestine showed comprehensive restoration including higher nutrients transport,energy metabolism,antioxidant capacity,and intestinal physical barrier,while lower oxidative stress,inflammatory response,and pathogenic microbiota.Chang Cui Caichi Wu Jun Wang Ziwei Ma Xiaoyu Zheng Pengwei Zhu Nuan Wang Yuhua Zhu Wutai Guan Fang Chen 2023Journal of Animal Science and Biotechnology2023,14,1:1
