6Adjuvant heparanase inhibitor PI-88 therapy for hepatocellular carcinoma recurrence显示文摘AIM: To demonstrate that administering heparanase inhibitor PI-88 at 160 mg/d is safe and promising in reducing hepatocellular carcinoma(HCC) recurrence for up to 3 year following curative resection. METHODS: A total of 143 patients(83.1% of the 172 participants in the phase Ⅱ study) participated in the follow-up study. Of these patients, 50 had received no treatment, 48 had received 160 mg/d PI-88, and 45 had received 250 mg/d PI-88 during the phase Ⅱ trial. Safety parameters and the following efficacy endpoints were investigated:(1) time to recurrence;(2) diseasefree survival; and(3) overall survival. RESULTS: PI-88 at 160 mg/d delayed the onset and frequency of HCC recurrence, and provided a clinically significant survival advantage for up to 3 years after treatment compared with those of the control group:(1) the recurrence-free rate increased from 50% to 63%, and(2) time to recurrence at the 36 th percentile was postponed by 78%. The efficacy of administering PI-88 at 250 mg/d was confounded by a high dropout rate(11 out of 54 patients). Additionally, subgroup analyses of patients with(1) multiple tumors or a single tumor ≥ 2 cm; and(2) hepatitis B or C revealed that administering PI-88 at 160 mg/d conferred the most significant survival advantage(56.8% improvement in disease-free survival, P = 0.045) for patients with both risk factors for recurrence. 9A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. 10PRGL:A cell wall proline-rich protein containning GASA domain in Gerbera hybrida显示文摘PRPs (proline-rich proteins) are a group of cell wall proteins characterized by their proline and hy- droproline-rich repetitive peptides. The expression of PRPs in plants is stimulated by wounding and environmental stress. GASA (gibberellic acid stimulated in Arabidopsis) proteins are small peptides sharing a 60 amino acid conserved C-terminal domain containing twelve invariant cysteine residues. Most of GASAs reported are localized to apoplasm or cell wall and their expression was regulated by gibberellins (GAs). It has been reported that, in French bean, these two proteins encoding by two distinct genes formed a two-component chitin-receptor involved in plant-pathogen interactions when plant was infected. We cloned a full-length cDNA of PRGL (proline-rich GASA-like) gene which encodes a protein containing both PRP and GASA-like domains. 13The carbon nanotubes-based materials and their applications for organic pollutant removal: A critical review显示文摘The carbon nanotubes(CNTs) as the emerging materials for organic pollutant removal have gradually become a burgeoning research field.Herein,a mini-review of CNTs-based materials curre ntly studies for organic pollutant elimination is presented.This review summarizes the preparation methods of CNTsbased materials.CNTs-based materials can be used as adsorbents to remove organic pollutants in wastewater.The adsorption mechanisms mainly include surface diffusio n,pore diffusion and adsorption reaction.Most importantly,an in-depth overview of CNTs-based materials currently available in advanced oxidation processes(AOPs) applications for wastewater treatment is proposed.CNTs-based materials can catalyze different oxidants(e.g.,hydrogen peroxide(H2 O2),persulfates(PMS/PDS),ozone(O3) and ferrate/permanganate(Fe(Ⅵ)/Mn(Ⅶ)) to generate more reactive oxygen species(ROS) for organic pollutant elimination.Moreover,the possible reaction mechanisms of removing organic pollutants by CNTs-based materials are summarized systematically and discussed in detail.Finally,application potential and future research directions of CNTs-based materials in the environmental remediation field are proposed.Jiali Peng Yongli He Chenying Zhou Shijun Su Bo Lai 2021Chinese Chemical Letters2021,32,5:7
14Resveratrol induces AMPK and mTOR signaling inhibition-mediated autophagy and apoptosis in multiple myeloma cells显示文摘Resveratrol,a natural compound extracted from the skins of grapes,berries,or other fruits,has been shown to have anti-tumor effects against multiple myeloma(MM)via promoting apoptosis and inhibiting cell viability.In addition to apoptosis,autophagy also plays a significant role in anti-tumor effects.However,whether autophagy is involved in anti-MM activity of resveratrol remains unclear.In this study,human MM cell lines U266,RPMI-8226,and NCI-H929 were treated with resveratrol.Cell Counting Kit-8 assay and colony formation assay were used to measure cell viability.Western blot analysis was used to detect apoptosis-and autophagy-associated proteins.3-Methyladenine(3-MA)was applied to inhibit autophagy.Results showed that resveratrol inhibited cell viability and colony formation via promoting apoptosis and autophagy in MM cell lines U266,RPMI-8226,and NCI-H929.Resveratrol promoted apoptosis-related proteins,Caspase-3 activating poly-ADP-ribose polymerase and Caspase-3 cleavage,and decreased the protein level of Survivin in a dose-dependent manner.Additionally,resveratrol upregulated the levels of LC3 and Beclin1 in a dose-dependent way,indicating that autophagy might be implicated in anti-MM effect of resveratrol.Furthermore,3-MA relieved the cytotoxicity of resveratrol by blocking the autophagic flux.Resveratrol increased the phosphorylation of adenosine monophosphate(AMP)-activated protein kinase and decreased the phosphorylation of mammalian target of rapamycin(mTOR)and its downstream substrates p70S6K and 4EBP1 in a dose-dependent manner,leading to autophagy.Therefore,our results suggest that resveratrol exerts anti-MM effects through apoptosis and autophagy,which can be used as a new therapeutic strategy for MM in clinic.Ruye Ma Dandan Yu Yu Peng Hongfei Yi Yingcong Wang Taofang Cheng Bingqing Shi Guang Yang Weiming Lai Xiaosong Wu Ye Lu Jumei Shi 2021Acta Biochimica et Biophysica Sinica2021,53,6:7

16Atomic Fe-Zn dual-metal sites for high-efficiency pH-universal oxygen reduction catalysis显示文摘An effective electrocatalyst being highly active in all pH range for oxygen reduction reaction(ORR)is crucial for energy conversion and storage devices.However,most of the high-efficiency ORR catalysis was reported in alkaline conditions.Herein,we demonstrated the preparation of atomically dispersed Fe-Zn pairs anchored on porous N-doped carbon frameworks(Fe-Zn-SA/NC),which works efficiently as ORR catalyst in the whole pH range.It achieves high half-wave potentials of 0.78,0.85 and 0.72 V in 0.1 M HClO4,0.1 M KOH and 0.1 M phosphate buffer saline(PBS)solutions,respectively,as well as respectable stability.The performances are even comparable to Pt/C.Furthermore,when assembled into a Zn-air battery,the high power density of 167.2 mWcm−2 and 120 h durability reveal the feasibility of Fe-Zn-SA/NC in real energy-related devices.Theoretical calculations demonstrate that the superior catalytic activity of Fe-Zn-SA/NC can be contributed to the lower energy barriers of ORR at the Fe-Zn-N6 centers.This work demonstrates the potential of Fe-Zn pairs as alternatives to the Pt catalysts for efficient catalytic ORR and provides new insights of dual-atom catalysts for other energy conversion related catalytic reactions.Jie Xu Shuhua Lai Defeng Qi Min Hu Xianyun Peng Yifan Liu Wei Liu Guangzhi Hu Heng Xu Fan Li Chao Li Jia He Longchao Zhuo Jiaqiang Sun Yuan Qiu Shusheng Zhang Jun Luo Xijun Liu 2021Nano Research2021,14,5:6

19Synthesis and biological evaluation of nitric oxide-releasing sixalkoxyl biphenyl derivatives as anticancer agents显示文摘A series of novel nitric oxide-donating sixalkoxyl biphenyl derivatives (14a-l) were synthesized by coupling furoxan with alkoxyl biphenyl skeleton using amino acids as the spacers, and their cytotoxicity against HepG2 cells in vitro were evaluated by MTT method. It was found that 14c, 14d, 14f, 14i, 14j and 14k showed more potent cytotoxic activities than control 5-?uorouracil.
