| 3 | Antimicrobial peptides: new hope in the war against multidrug resistance显示文摘The discovery of antibiotics marked a golden age in the revolution of human medicine. However,decades later, bacterial infections remain a global healthcare threat, and a return to the pre-antibiotic era seems inevitable if stringent measures are not adopted to curb the rapid emergence and spread of multidrug resistance and the indiscriminate use of antibiotics. In hospital settings, multidrug resistant(MDR) pathogens, including carbapenem-resistant Pseudomonas aeruginosa, vancomycin-resistant enterococci(VRE), methicillin-resistant Staphylococcus aureus(MRSA), and extendedspectrum β-lactamases(ESBL) bearing Acinetobacter baumannii, Escherichia coli, and Klebsiella pneumoniae are amongst the most problematic due to the paucity of treatment options,increased hospital stay, and exorbitant medical costs. Antimicrobial peptides(AMPs) provide an excellent potential strategy for combating these threats. Compared to empirical antibiotics, they show low tendency to select for resistance, rapid killing action, broad-spectrum activity, and extraordinary clinical efficacy against several MDR strains. | 4 | Comprehensive assessment on dynamic roof instability under fractured rock mass conditions in the excavation disturbed zone显示文摘The damage process of fractured rock mass showed that the fracture in rocks induced roof collapse in Yangchangwan Coal Mine,China. The rock mass was particularly weak and fractured. There occurred 6 large-scale dynamical roof falls in the excavation disturbed zone(EDZ) with the collapsing volume of 216 m3. First,the field detailed geological environment,regional seismic dynamics,and dynamic instability of roadways were generally investigated. | 6 | Experimental small bowel preservation using Polysol: A new alternative to University of Wisconsin solution, Celsior and histidine-tryptophan-ketoglutarate solution?显示文摘AIM: To evaluate the potential of Polysol, a newly developed preservation solution, in cold storage of small bowel grafts, compared with the current standards, University of Wisconsin solution (UW), Celsior and histidine-tryptophan-ketoglutarate solution (HTK). METHODS: Male Wistar rats were used as donors. Small bowels were retrieved, flushed and then stored in the respective 4 solutions for 18 h at 4℃. Functional integrity of the grafts was evaluated by isolated reperfusion with oxygenated Krebs-Henseleit buffer at 37℃ for 30 min in all 4 groups. RESULTS: Polysol preservation exhibited the highest tissue ATP concentration and the lowest release of LDH. Malondialdehyde, an index for tissue lipid peroxidation, was also the lowest in Polysol. Tissue oxygen consumption was significantly higher in Polysol than in the others. Of interest, UW-storage promoted 10-fold higher apoptosis than in the others. Moreover, electron microscopy revealed that the mucosal villi/micro-villi formation and the cell organelles, including mitochondria, were both significantly better preserved in Polysol, while deleterious alterations were apparent in the others, most notably in UW. Although Celsior and HTK exhibited the better trend of results than UW in some parameters, but could not reach the over-all superiority to UW.CONCLUSION: Cold storage using Polysol resulted in significantly better integrity and function of small bowel grafts than UW. | 8 | Isolation and identification of symbiotic bacteria from the skin, mouth, and rectum of wild and captive tree shrews显示文摘Endosymbionts influence many aspects of their hosts’ health conditions, including physiology, development, immunity, metabolism, etc. Tree shrews(Tupaia belangeri chinensis) have attracted increasing attention in modeling human diseases and therapeutic responses due to their close relationship with primates. To clarify the situation of symbiotic bacteria from their body surface, oral cavity, and anus, 12 wild and 12 the third generation of captive tree shrews were examined. Based on morphological and cultural characteristics, physiological and biochemical tests, as well as the 16 S rDNA full sequence analysis, 12 bacteria strains were isolated and identified from the wild tree shrews: body surface: Bacillus subtilis(detection rate 42%), Pseudomonas aeruginosa(25%), Staphlococcus aureus(33%), S. Epidermidis(75%), Micrococcus luteus(25%), Kurthia gibsonii(17%); oral cavity: Neisseria mucosa(58%), Streptococcus pneumonia(17%); anus: Enterococcus faecalis(17%), Lactococus lactis(33%), Escherichia coli(92%), Salmonella typhosa(17%); whereas, four were indentified from the third generation captive tree shrews: body surface: S. epidermidis(75%); oral cavity: N.mucosa(67%); anus: L. lactis(33%), E. coli(100%). These results indicate that S. epidermidis, N. mucosa, L. lactis and E. coli were major bacteria in tree shrews, whereas, S. aureus, M. luteus, K. gibsonii, E. faecalis and S. typhosa were species-specific flora. | 11 | RegIV expression showing specificity to gastrointestinal tract and its potential role in diagnosing digestive tract neuroendocrine tumor显示文摘Regenerating gene IV (RegIV),a member of the regenerating gene family discovered in 2001,has been found to be involved in malignancy in several different organs including the stomach,colorectum,pancreas and prostate,but the overall expression profile of RegIV has not been reported. To learn more about RegIV,we evaluated its distribution by immunohistochemistry (IHC) in a total of 360 samples including 24 types of normal tissue,40 benign and malignant lesions,and 18 neuroendocrine tumors. We found that in normal tissues,in addition to its relative specificity for the gastrointestinal tract,RegIV was detected in the adrenal gland and mammary gland. Among all the malignancies of various histological types under evaluation,RegIV was found mostly in adenocarcinomas. Studies on additional sets of colorectal tumor samples showed that RegIV expression was predominant in colorectal adenoma (87.5%) and peritumoral tissue (100%) but not in cancer tissue (30.8%). | 15 | Metallogenic Mechanism and Tectonic Setting of Tungsten Mineralization in the Yangbishan Deposit in Northeastern China显示文摘The Yangbishan iron–tungsten deposit in the Shuangyashan area of Heilongjiang Province is located in the center of the Jiamusi Massif in northeastern China. The rare earth element and trace element compositions of the scheelite show that it formed in a reducing environment and inherited the rare earth element features of the ore-forming fluid. The geochemical characteristics of the gneissic granite associated with the tungsten mineralization show that the magma formed in this reducing environment and originated from the partial melting of metamorphosed shale that contained organic carbon and was enriched with tungsten. In addition, in situ Hf isotopic analysis of zircons from the gneissic granite indicates that they probably originated from the partial melting of a predominantly Paleo–Mesoproterozoic crustal source. According to LA-ICP-MS zircon dating, the Yangbishan orerelated gneissic granite has an Early Paleozoic crystallization age of 520.6 ± 2.8 Ma. This study, together with previous data, indicates that the massifs of northeastern China, including Erguna, Xing'an, Songliao, Jiamusi, and Khanka massifs, belonged to an orogenic belt that existed along the southern margin of the Siberian Craton during the late Pan-African period. | 16 | Comparison of outcomes after human leukocyte antigen-matched and haploidentical hematopoietic stem-cell transplantation for multiple myeloma显示文摘Background:Allogeneic stem-cell transplantation (SCT) is a well-established immunotherapeutic strategy for multiple myeloma (MM) with a potent and often sustained graft-vs.-myeloma effect.This multicenter investigation aimed to analyze the complications and survival of haploidentical SCT in patients with MM,and compare the main outcomes with matched-related donors (MRDs).Methods:Haploidentical and MRD SCT was identified from a cohort of 97 patients with MM who received a myeloablative transplantation in 13 hospitals from May 2001 to December 2017.A matched-pair analysis was designed.For each haplo recipient,the recipients were randomly selected from the MRD group and were matched according to the following criteria:year of the hematopoietic SCT (±2 years),disease status at transplantation,and the length of follow-up.ults:Seventy cases received MRD and 27 received haploidendcal transplantation.The two groups showed no significant ifferences regarding age,gender,cytogenetic risk,and diagnostic stage.The cumulative incidences of non-relapse mortality (NRM) at1 and 3 years based on donor type were 20.5%(95% confidence interval [CI],10.90-30.10%) and 24.2%(95% CI,13.81-34.59%) for the MRD group and 16.80%(95% CI,1.71-31.89%) and 28.70%(95% CI,8.71-48.69%) for the haplo group,respectively.Cumulative incidence of NRM did not differ significantly between the two groups (x2 =0.031,P =0.861).The cumulative incidences of progression-free survival (PFS) and 1 year and 3 years by type of donors were 59.8 %(95 % CI,48.24-71.36 %) and 45.4 %(95 % CI,33.44-57.36%),and 65.6%(95% CI,47.18-84.02%) and 26.8%(95% CI,7.59-46.01%) for MRD and haploidentical donor,respectively.Cumulative incidence of PFS did not differ significantly between the two groups (x2 =0.182,P =0.670).In multivariate analyses,no statistically significant differences were observed between haploidentical and MRD for relapse,NRM,PFS,and overall survival.There were no statistically differences on main outcomes after haploidentical and MRD.Conclusion:Haploidentical SCT could be performed safely and feasibly for patients with MM in need. | Yao Chen Wei-Jun Fu Lan-Ping Xu Han-Yun Ren Yong-Rong Lai Dai-Hong Liu Lin Liu Zi-Min Sun Yuan-Bin Wu Xin Wang Ling-Hui Xia Ming Jiang Tong-Lin Hu Ding-Ming Wan Xiao-Jun Huang | 2019 | Chinese Medical Journal2019,,15: | 4 |
| 17 | Antimicrobial peptide LL-37 forms complex with bacterial DNA to facilitate blood translocation of bacterial DNA and aggravate ulcerative colitis显示文摘Bacterial DNA(bacDNA) is frequently found in serum of patient with ulcerative colitis(UC) and Crohn's disease, even blood bacterial culture is negative. How bacDNA evades immune elimination and is translocated into blood remain unclear. Here, we showed that bacDNA avoids elimination and disables bacteriakilling function of antimicrobial peptide LL-37(Cramp in mice) by forming complex with LL-37, which is inducible after culture with bacteria or bacterial products. Elevated LL-37-bacDNA complex was found in plasma and lesions of patients with UC. LL-37-bacDNA promoted inflammation by inducing Th1, Th2 and Th17 differentiation and activating toll-like receptor-9(TLR9). The complex also increased paracellular permeability, which possibly combines its inflammatory effects to promote local damage and bacDNA translocation into blood. Cramp-bacDNA aggravated mouse colitis severity while interference with the complex ameliorated the disease. The study identifies that inflammatogenic bacDNA utilizes LL-37 as a vehicle for blood translocation and to evade immune elimination. | 18 | 3’-Methoxydaidzein exerts analgesic activity by inhibiting voltage-gated sodium channels显示文摘Isoflavones are widely consumed by people around the world in the form of soy products, dietary supplements and drugs. Many isoflavones or related crude extracts have been reported to exert pain-relief activities, but the mechanism remains unclear. Voltage-gated sodium channels(VGSCs) play important roles in excitability of pain sensing neurons and many of them are important nociceptors. Here, we report that several isoflavones including 3’-methoxydaidzein(3 MOD), genistein(GEN) and daidzein(DAI) show abilities to block VGSCs and thus to attenuate chemicals and heat induced acute pain or chronic constriction injury(CCI) induced pain hypersensitivity in mice. Especially, 3 MOD shows strong analgesic potential without inducing addiction through inhibiting subtypes NaV1.7, NaV1.8 and NaV1.3 with the IC50 of 181 ± 14, 397 ± 26, and 505 ± 46 nmol·L–1, respectively, providing a promising compound or parent structure for the treatment of pain pathologies. Using principal component analysis,a comprehensive evaluation of socio-economic development level and ecological environment quality in Jinghe County is conducted by combining with Excel. Their coupling relationship is studied,and quantitative coordination degree between social economy and ecological environment in Jinghe County is obtained. The results show that in recent 50 years,socio-economic development level in Jinghe County is rising,but the ecological environment quality is falling. Their coupling degree C changes during [- 1. 260, + 0. 482],in other words,their coupling relationship is changing between ' reluctant coordination' and ' not coordination'. | 20 | Quantitative analysis of biotechnical reinforcement for a steep slope consisting of composite coal-gangue-soil medium adjacent to a mined-out area显示文摘The engineering and geological characteristics of a steep slope consisting of coal gangue, rock and soil medium in Hua- ting coal mine have been comprehensively investigated. Owing to humid weather, heavy rainfall, vegetation and porous characteris- tics of the soil and rock mass, the steep slope will be destabilized and induce mud-rock flow or derive hazard easily. Firstly, based on the classical slope reinforcement theory, some regularity between the shear and displacement in the destabilized zone of the slope with or without root strength contribution is presented. Then, based on the experimental and statistical analysis of root strength, hy- drological characteristics and stability status, etc., some possible biotechnical techniques for reinforcement of the steep slope have been suggested.
