4Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 6Tomato lncRNA23468 functions as a competing endogenous RNA to modulate NBS-LRR genes by decoying miR482b in the tomato-Phytophthora infestans interaction显示文摘Our previous studies indicated that tomato miR482b could negatively regulate the resistance of tomato to Phytophthora infestans and the expression of miR482b was decreased after inoculation with P.infestans.However,the mechanism by which the accumulation of miR482b is suppressed remains unclear.In this study,we wrote a program to identify 89 long noncoding RNA(lncRNA)-originated endogenous target mimics(eTMs)for 46 miRNAs from our RNA-Seq data.Three tomato lncRNAs,lncRNA23468,lncRNA01308 and lncRNA13262,contained conserved eTM sites for miR482b.When lncRNA23468 was overexpressed in tomato,miR482b expression was significantly decreased,and the expression of the target genes,NBS-LRRs,was significantly increased,resulting in enhanced resistance to P.infestans.Silencing lncRNA23468 in tomato led to the increased accumulation of miR482b and decreased accumulation of NBS-LRRs,as well as reduced resistance to P.infestans.In addition,the accumulation of both miR482b and NBS-LRRs was not significantly changed in tomato plants that overexpressed lncRNA23468 with a mutated eTM site.Based on the VIGS system,a target gene of miR482b,Solyc02g036270.2,was silenced.The disease symptoms of the VIGS-Solyc02g036270.2 tomato plants were in accordance with those of tomato plants in which lncRNA23468 was silenced after inoculation with P.infestans.More severe disease symptoms were found in the modified plants than in the control plants.Our results demonstrate that lncRNAs functioning as eTMs may modulate the effects of miRNAs in tomato and provide insight into how the lncRNA23468-miR482b-NBS-LRR module regulates tomato resistance to P.infestans.Ning Jiang Jun Cui Yunsheng Shi Guanglei Yang Xiaoxu Zhou Xinxin Hou Jun Meng Yushi Luan 2019Horticulture Research2019,6,1:12

9Puerarin ameliorated pressure overload-induced cardiac hypertrophy in ovariectomized rats through activation of the PPARα/PGC-1 pathway显示文摘Estrogen deficiency induces cardiac dysfunction and increases the risk of cardiovascular disease in postmenopausal women and in those who underwent bilateral oophorectomy.Previous evidence suggests that puerarin,a phytoestrogen,exerts beneficial effects on cardiac function in patients with cardiac hypertrophy.In this study,we investigated whether puerarin could prevent cardiac hypertrophy and remodeling in ovariectomized,aortic-banded rats.Female SD rats subjected to bilateral ovariectomy(OVX)plus abdominal aortic constriction(AAC).The rats were treated with puerarin(50 mg·kg^(−1)·d^(−1),ip)for 8 weeks.Then echocardiography was assessed,and the rats were sacrificed,their heart tissues were extracted and allocated for further experiments.We showed that puerarin administration significantly attenuated cardiac hypertrophy and remodeling in AAC-treated OVX rats,which could be attributed to activation of PPARα/PPARγcoactivator-1(PGC-1)pathway.Puerarin administration significantly increased the expression of estrogen-related receptorα,nuclear respiratory factor 1,and mitochondrial transcription factor A in hearts.Moreover,puerarin administration regulated the expression of metabolic genes in AAC-treated OVX rats.Hypertrophic changes could be induced in neonatal rat cardiomyocytes(NRCM)in vitro by treatment with angiotensin Ⅱ(Ang Ⅱ,1μM),which was attenuated by co-treatemnt with puerarin(100μM).We further showed that puerarin decreased Ang Ⅱ-induced accumulation of non-esterified fatty acids(NEFAs)and deletion of ATP,attenuated the Ang II-induced dissipation of the mitochondrial membrane potential,and improved the mitochondrial dysfunction in NRCM.Furthermore,addition of PPARαantagonist GW6471(10μM)partially abolished the anti-hypertrophic effects and metabolic effects of puerarin in NRCM.In conclusion,puerarin prevents cardiac hypertrophy in AAC-treated OVX rats through activation of PPARα/PGC-1 pathway and regulation of energy metabolism remodeling.This may provide a new approach to prevent the development of heart failure in postmenopausal women.Ning Hou Yin Huang Shao-ai Cai Wen-chang Yuan Li-rong Li Xia-wen Liu Gan-jian Zhao Xiao-xia Qiu Ai-qun Li Chuan-fang Cheng Shi-ming Liu Xiao-hui Chen Dao-feng Cai Jing-xuan Xie Min-sheng Chen Cheng-feng Luo 2021Acta Pharmacologica Sinica2021,42,1:11
10Recent progress on the recycling technology of Li-ion batteries显示文摘Lithium-ion batteries(LIBs)have been widely applied in portable electronic devices and electric vehicles.With the booming of the respective markets,a huge quantity of spent LIBs that typically use either LiFePO_(4) or Li N_(x)Co_(y)Mn_(z)O_(2) cathode materials will be produced in the very near future,imposing significant pressure for the development of suitable disposal/recycling technologies,in terms of both environmental protection and resource reclaiming.In this review,we firstly do a comprehensive summary of the-state-of-art technologies to recycle Li N_(x)Co_(y)Mn_(z)O_(2) and LiFePO_(4)-based LIBs,in the aspects of pretreatment,hydrometallurgical recycling,and direct regeneration of the cathode materials.This closed-loop strategy for cycling cathode materials has been regarded as an ideal approach considering its economic benefit and environmental friendliness.Afterward,as for the exhausted anode materials,we focus on the utilization of exhausted anode materials to obtain other functional materials,such as graphene.Finally,the existing challenges in recycling the LiFePO_(4) and Li N_(x)Co_(y)Mn_(z)O_(2) cathodes and graphite anodes for industrial-scale application are discussed in detail;and the possible strategies for these issues are proposed.We expect this review can provide a roadmap towards better technologies for recycling LIBs,shed light on the future development of novel battery recycling technologies to promote the environmental benignity and economic viability of the battery industry and pave way for the large-scale application of LIBs in industrial fields in the near future.Yuqing Wang Ning An Lei Wen Lei Wang Xiaotong Jiang Feng Hou Yuxin Yin Ji Liang 2021Journal of Energy Chemistry2021,30,4:10
11Transition of human diets and agricultural economy in Shenmingpu Site, Henan, from the Warring States to Han Dynasties显示文摘Historical records and archaeological remains indicate that the Chinese agricultural economy changed significantly from the Warring States (475-221 BC) to Han Dynasties (206 BC-220 AD), i.e., from rice-millet based agriculture to rice-millet-wheat based agriculture. However, the variation of human diets and the inner relationship between human diets and the agricultural transition during this period remain poorly understood. In this paper, the C and N stable isotopes from human and animal bones at the Shenmingpu site (SMPS), Xichuan County, Henan Province were analyzed. If some outliers (M34, M36, M102) were excluded, the mean 13C value (( 16.7±0.8)‰, n=15) of humans in Han Dynasties was dramatically lower than that in the Warring States (( 12.7±0.8)‰, n=14), indicating that the cultivation of rice and wheat, especially wheat, had been more widely popularized in Han Dynasties. Meanwhile, the range of 15N values of humans (6.6‰-9.3‰) in Han Dynasties was narrower than that of the Warring States (6.2‰-10.4‰), suggesting that the animal protein resources in human foods during Han Dynasties were more concentrated. 12The role of RIP3 in cardiomyocyte necrosis induced by mitochondrial damage of myocardial ischemia-reperfusion显示文摘Myocardial damage caused by myocardial ischemia-reperfusion injury (MIRI)is difficult to be alleviated because cardiomyocyte necrosis is an irreversible and unregulated death form.Recently, necroptosis,a necrosis form caused by tumor necrosis factor-α (TNF-α)and Fas ligand (FasL),was found to be regulated by receptor interacting protein 3 (RIP3)and RIP3-receptor interacting protein 1 (RIP1)-mixed lineage kinase domain like protein (MLKL)pathway.But it is unclear whether they also play a regulatory role in MIRI-induced necroptosis.Our previous results showed that in rat MIRI,RIP3 could translocate and express highly in mitochondria.Therefore,it is important to explore proteins that interact with RIP3 which was translocated to mitochondria.The aim of this study was to explore the role of RIP3 in cardiomyocyte necrosis induced by mitochondrial damage of hypoxia/reoxygenation (H/R).Our results showed that H/R could cause RIP3-depended mitochondrial fragmentation and necrosis-based death;and RIP3-promoted H/R-induced necroptosis in H9c2 cells through increasing lactate dehydrogenase release and inhibiting cell viability.This process did not require RIP1 or MLKL but dynamin-related protein 1 (Drp1),which was related to Drpl activation,reactive oxygen species elevation,and △Ψm decline.This study provides novel insights into the role of RIP3 in cardiomyocyte injury during H/R.RIP3 may serve as a potential target for the treatment of MIRI.Huifang Hou Yongling Wang Qiong Li Zaibing Li Yan Teng Jingyu Li Xiaoying Wang Junli Chen Ning Huang 2018Acta Biochimica et Biophysica Sinica2018,50,11:10
13Laboratory tests and numerical simulations of brittle marble and squeezing schist at Jinping II hydropower station,China显示文摘Four 16.7 km-long tunnels with diameters ranging from 12.4 to 14.6 m are now under construction at Jinping II hydropower station along the Yalong River.The tunnels pass through Triassic rocks below Jinping Mountain.The tunnels are characterized with high overburden,long alignment and complex geological conditions.Brittle failure in marble and squeezing in schist are the primary problems in tunnelling.This paper introduces the studies of laboratory tests on Jinping II marble as well as numerical prediction of excavation damaged zone (EDZ) of tunnel section in brittle marble and determination of reinforced concrete lining thickness for restraining time-dependent deformation in the schist tunnel section.Laboratory tests indicate that Jinping II marble presents a complex brittle-ductile-plastic transition behavior of post-peak response with increasing confining pressure.Such behavior can be described numerically with the Hoek-Brown model.The EDZ was calibrated and predicted using both fast Lagrangian analysis of continua (FLAC) and particle flow code (PFC).The predicted result of EDZ in sections with different qualities of rock mass under various overburden pressures is quite helpful in understanding EDZ characterization and support design.A power-law creep model was used to support the lining design,especially in determining the lining thickness.Field convergence measurement data over 19 months were used to calibrate the creep model properties,followed by a sensibility analysis of reinforced concrete lining thickness that was launched to present the maximum lining compressive stress.Chunsheng Zhang,Weijiang Chu,Ning Liu,Yongsheng Zhu,Jing Hou East China Investigation and Design Institute,China Hydropower Engineering Consulting Group Co.,Hangzhou,310014,China 2011Journal of Rock Mechanics and Geotechnical Engineering2011,3,1:10
14Characterization and allergic role of IL-33-induced neutrophil polarization显示文摘Neutrophils are involved in the pathogenesis of allergy.However,the contribution of the different functionally polarized neutrophils in allergy needs to be clarified.We sought to define the characteristics of interleukin(IL)-33-induced neutrophils and the involvement of this subset of polarized neutrophils in allergic pathogenesis.Freshly isolated neutrophils were treated with different cytokines and the cytokine expression levels were detected by realtime PCR.The gene expression profile of IL-33-induced neutrophils was determined by microarray assay.Adoptive transfer assay was used to investigate the function of IL-33-induced neutrophils in an ovalbumin(OVA)-induced allergic asthma model.IL-33-treated neutrophils selectively produced IL-4,IL-5,IL-9 and IL-13(referred as to N(IL-33)cells)and displayed a distinctive gene expression profile in sharp contrast to resting and lipopolysaccharide(LPS)-treated neutrophils.IL-33-induced neutrophils expressed high Levels of IL-1R2 on cell surface,whereas resting and LPS-treated neutrophils did not,indicating IL-1R2 might be used as a biomarker for N(IL-33)cells.Importantly,N(IL-33)neutrophils exist in the lungs of OVA-induced allergic asthma mice.Adoptive transfer of N(IL-33)neutrophils significantly promotes the severity of the lung pathogenesis in this model.IL-33 induces neutrophil polarization through c-Jun N-terminal kinase-and nuclear factor-κB-dependent pathways.A previously unappreciated neutrophil polarization driven by IL-33 with unique cell surface markers and cytokine/chemokineproducing gene profile was defined.The newly identified N(IL-33)subpopulation may have significant contribution to IL-33-related pathogenesis.Bo Sun Linnan Zhu Yaling Tao Hai-Xi Sun Yang Li Peng Wang Yuzhu Hou Yang Zhao Xiaodong Zhang Lianfeng Zhang Ning Na Yong Zhao 2018Cellular & Molecular Immunology2018,15,8:9

163-year Treatment of Tenofovir Alafenamide vs.Tenofovir Disoproxil Fumarate for Chronic HBV Infection in China显示文摘Background and Aims:Tenofovir alafenamide(TAF)has similar efficacy to tenofovir disoproxil fumarate(TDF)but with improved renal and bone safety in chronic hepatitis B patients studied outside of China.We report 3-year results from two phase 3 studies with TAF in China(Clinicaltrials.gov:NCT02836249 and NCT02836236).Methods:Chinese hepatitis B e antigen(HBeAg)-positive and-negative chronic hepatitis B patients with viremia and elevated alanine aminotransferase were randomized 2:1 to TAF or TDF treatment groups and treated in a double-blind fashion for 144 weeks(3 years).Efficacy responses were assessed by individual study while safety was assessed by a pooled analysis.Results:Of the 334 patients(180 HBeAg-positive and 154 HBeAg-negative)randomized and treated,baseline characteristics were similar between groups.The overall mean age was 38 years and 73%were male.The mean HBV DNA was 6.4 log10 IU/mL.The median alanine aminotransferase was 88 U/L,and 37%had a history of antiviral use.At week 144,the proportion with HBV DNA<29 IU/mL was similar among the two groups,with TAF at 83%vs.TDF at 79%,and TAF at 93%vs.TDF at 92%for the HBeAg-positive and-negative patients,respectively.In each study,higher proportions of TAF than TDF patients showed normalized alanine aminotransferase(via the American Association for the Study of Liver Diseases and the China criteria)and showed loss of HBsAg;meanwhile,the HBeAg seroconversion rates were similar.Treatment was well-tolerated among the TAF patients,who showed a smaller median decline in creatinine clearance(−0.4 vs.−3.2 mL/min;p=0.014)and less percentage change in bone mineral density vs.TDF at hip(−0.95%vs.−1.93%)and spine(+0.35%vs.−1.40%).Conclusions:In chronic hepatitis B patients from China,TAF treatment provided efficacy similar to TDF but with better renal and bone safety at 3 years.Jinlin Hou Qin Ning Zhongping Duan You Chen Qing Xie Fu-Sheng Wang Lunli Zhang Shanming Wu Hong Tang Jun Li Feng Lin Yongfeng Yang Guozhong Gong John FFlaherty Anuj Gaggar Shuyuan Mo Cong Cheng Gregory Camus Chengwei Chen Yan Huang Jidong Jia Mingxiang Zhang GS-US-320-0110 and GS-US-320-0108 China Investigators 2021Journal of Clinical and Translational Hepatology2021,9,3:7

A radar polarimetric coefficients matrix(RPCM) with a simpler expression is presented for the monostatic radar polarization scattering matrix(PSM) measurement. Using a rhombic dihedral corner reflector and a metallic sphere, the RPCM can be obtained by solving a set of equations, which can be used to find the true PSM for any target.
