| 3 | Landscape Ecology:Coupling of Pattern,Process,and Scale显示文摘Landscape ecology provides new theoretical frameworks and methodologies for understanding complex ecological phenomena at multiple scales.Studies of landscape ecology focus on understanding the dynamics of eco-logical patterns and processes,and highlight the integration of multiple disciplines.In this paper,we discussed the problems and challenges that landscape ecology is currently facing,emphasizing the limitations of current methods used to describe dynamic landscape patterns and processes.We suggested that the focus should be on the integration of ground-based observation,mobile monitoring,transect survey,and remote-sensing monitoring,as well as improved coupling of experimental and model simulations.In addition,we outlined the research frontiers in landscape ecology,including scaling,integrated pattern and process modeling,and regional synthesis.Lastly,a brief review of pat-tern-process-scale coupling studies in China was provided.We concluded by pointing out that pattern-process-scale interactions,correlations between natural,economic,and social processes,and the coupling of human and natural systems will be major research areas in landscape ecology in the future. | FU Bojie LIANG Di LU Nan | 2011 | Chinese Geographical Science2011,21,4: | 24 |
| 4 | Antimicrobial activities of essential oil from Artemisiae argyi leaves显示文摘A study was conducted to determine the antimicrobial activities of essential oil from Artemisiae argyi leaves. The sample of the essential oil was analyzed by GC–MS. From 18 compounds representing the oils, Eucalyptole (18.42%), Spathulenol (14.32), 4-Methyl-1-(1-methylethyl)-3-cyclohexen-1-ol (3.10%), 3-Carene (2.64%) appeared as the main components. The screening of antimicrobial activity of the essential oil was evaluated using agar diffusion and broth microdilution methods. Gram-positive bacterial were more sensitive than gram-negative bacterial of the 8 microorganisms, and Staphylococcus aureus ATCC 6538 showed the lowest MIC (0.3125%) and MBC (0.625%). In the disc diffusion assay, Staphylococcus epidermidis ATCC 49134 and Bacillus subtilis ATCC 6633 showed obvious inhibitory activity. Survival curve showed that, 2MIC of Artemisiae argyi essential oil had a lethal effect on Candida albicans within the first 1 h. | 5 | Cardamonin from a medicinal herb protects against LPS-induced septic shock by suppressing NLRP3 inflammasome显示文摘Aberrant activation of NLRP3 inflammasome has been implicated in the pathogenesis of diverse inflammation-related diseases, and pharmacological molecules targeting NLRP3 inflammasome are of considerable value to identifying potential therapeutic interventions. Cardamonin(CDN), the major active ingredient of the traditional Chinese medicinal herb Alpinia katsumadai, has exerted an excellent anti-inflammatory activity, but the mechanism underlying this role is not fully understood. Here, we show that CDN blocks canonical and noncanonical NLRP3 inflammasome activation triggered by multiple stimuli. Moreover, the suppression of CDN on inflammasome activation is specific to NLRP3, not to NLRC4 or AIM2 inflammasome. Besides, the inhibitory effect is not dependent on the expression of NF-κB-mediated inflammasome precursor proteins. We also demonstrate that CDN suppresses the NLRP3 inflammasome through blocking ASC oligomerization and speckle formation in a dose-dependent manner.Importantly, CDN improves the survival of mice suffering from lethal septic shock and attenuates IL-1βproduction induced by LPS in vivo, which is shown to be NLRP3 dependent. | 6 | Land Use Change and Anthropogenic Driving Forces:A Case Study in Yanhe River Basin显示文摘Human activities alter land use patterns and affect landscape sustainability. It is therefore very important to investigate the relationship between land use change and human activities. This study focuses on the detection of changing land use patterns in the Yanhe River Basin in northern Loess Plateau of China between 1995 and 2008. Landscape metrics were used to analyze the changing land use patterns and to explore the related anthropogenic driving forces. Results show that:1) Totally, 186 590 ha of croplands were converted into alternate land-use types (equivalent to 61.7% of the original cropland area). The majority of cropland areas were found to be converted into grassland and woodland areas (accounting for 55.9% and 4.9% respectively of the original cropland areas). 2) Both cropland and woodland demonstrated an increasing fragmentation tendency while grasslands showed a decreasing fragmentation tendency. 3) Multiple driving forces of land use change were thought to act together to changes in landscape metrics in the Yanhe River Basin. The anthropogenic driving forces were analyzed from four perspectives:ecological conservation policy, labor force transfer, industrial development, and rural settlement. The policy of the GfG (Grain for Green) project was the main driving factor which expedited the conversion from cropland to woodland and grassland. Industrial development was also found to affect land use change through the direct impact of economic activities such as oil exploration and agricultural production, or through indirect impacts such as the industrial structures readjustment. Labor force transfer from rural to urban areas was found to follow the industrial structure readjustment and further drove land use change from cropland to off-farm land use. | 10 | TaqMan Real-time RT-PCR Assay for Detecting and Differentiating Japanese Encephalitis Virus显示文摘Objective To detect Japanese encephalitis virus(JEV) rapidly and distinguish its genotypes, a TaqMan-based reverse transcriptase quantitative polymerase chain reaction(RT-PCR) detection system was developed.Methods By aligning the full-length sequences of JEV(G1-G5), six sets of highly specific TaqMan real-time RT-PCR primers and probes were designed based on the highly conserved NS1, NS2, and M genes of JEV, which included one set for non-specific JEV detection and five sets for the detection of specific JEV genotypes. Twenty batches of mosquito samples were used to evaluate our quantitative PCR assay.Results With the specific assay, no other flavivirus were detected. The lower limits of detection of the system were 1 pfu/mL for JEV titers and 100 RNA copies/μL. The coefficients of variation of this real-time RT-PCR were all < 2.8%. | 14 | Bavachin enhances NLRP3 inflammasome activation induced by ATP or nigericin and causes idiosyncratic hepatotoxicity显示文摘Psoraleae Fructus(PF)is a well-known traditional herbal medicine in China,and it is widely used for osteoporosis,vitiligo,and other diseases in clinical settings.However,liver injury caused by PF and its preparations has been frequently reported in recent years.Our previous studies have demonstrated that PF could cause idiosyncratic drug-induced liver injury(IDILI),but the mechanism underlying its hepatotoxicity remains unclear.This paper reports that bavachin isolated from PF enhances the specific stimuli-induced activation of the NLRP3 inflammasome and leads to hepatotoxicity.Bavachin boosts the secretion of IL-ip and caspase-1 caused by ATP or nigericin but not those induced by poly(I:C),monosodium urate crystal,or intracellular lipopolysaccharide.Bavachin does not affect AIM2 or NLRC4 inflammasome activation.Mechanistically,bavachin specifically increases the production of nigericin-induced mitochondrial reactive oxygen species among the most important upstream events in the activation of the NLRP3 inflammasome.Bavachin increases the levels of aspartate transaminase and alanine aminotransferase in serum and hepatocyte injury accompanied by the secretion of IL-ip via a mouse model of lipopolysaccharide-mediated susceptibility to IDILI.These results suggest that bavachin specifically enhances the ATP-or nigericin-induced activation of the NLRP3 inflammasome.Bavachin also potentially contributes to PF-induced idiosyncratic hepatotoxicity.Moreover,bavachin and PF should be evaded among patients with diseases linked to the ATP-or nigericin-mediated activation of the NLRP3 inflammasome,which may be a dangerous factor for liver injury. | Nan Qin Guang Xu Yan Wang Xiaoyan Zhan Yuan Gao Zhilei Wang Shubin Fu Wei Shi Xiaorong Hou Chunyu Wang Ruisheng Li Yan Liu Jiabo Wang Haiping Zhao Xiaohe Xiao Zhaofang Bai | 2021 | Frontiers of Medicine2021,15,4: | 9 |
