1PTEN and Ki67 expression is associated with clinicopathologic features of non-small cell lung cancer显示文摘Phosphatase and tensin homolog deleted on chromosome 10(PTEN) and the proliferating antigen Ki67 have been widely studied in several tumors.However,their role as indicator in non-small cell lung cancer(NSCLC)remains unknown.Here,we investigated the expression of PTEN and Ki67 in NSCLC tissues and paired normal lung tissues to identify whether these proteins are associated with lung cancer development and survival.Immunohistochemistry for PTEN and Ki67 was performed on 67 lung cancer tissues and 41 paired adjacent normal lung tissues to detect the expression of these two proteins.The expression of PTEN in NSCLC tissues(32.8%) was significantly lower than that in normal tissues(82.9%,P < 0.05).In contrast,the expression of Ki67 in NSCLC tissues(76.1%) was significantly higher than that in normal tissues(27.3%,P < 0.05).Expression of both PTEN and Ki67 were strongly associated with tumor histology,clinical stage,lymph node metastasis,differentiation and4-year postoperative survival rate(P < 0.05).However,PTEN expression was negatively correlated with Ki67 expression(r =-0.279,P < 0.05).In conclusion,low PTEN expression and Ki67 overexpression are associated with malignant invasion and lymph node metastasis of NSCLC.These proteins may serve as diagnostic and prognostic biomarkers of NSCLC.Yong Ji Mingfeng Zheng Shugao Ye Jingyu Chen Yijiang Chen 2014The Journal of Biomedical Research2014,28,6:16
2RNA Interference-based Suppression of Phosphoenolpyruvate Carboxylase Results in Susceptibility of Rapeseed to Osmotic Stress显示文摘The diverse functions of phosphoenolpyruvate carboxylase(PEPCase;EC 4.1.1.31) in C3 plants are not as well understood as in C4 plants.To investigate the functions of PEPCase in C3 plants,rapeseed(Brassica napus L.) PEPCase gene(referred to as BNPE15) was silenced by the RNA interference(RNAi) technique.Under normal growth conditions,no significant difference in lipid content and fatty acid composition were found between wild-type(WT) and transgenic rapeseed plants.However,when these plants were subjected to osmotic stress induced by osmoticum polyethylene glycol(PEG-6000),membrane permeability and membrane lipid peroxidization in roots and leaves of transgenic plants were higher than those of WT plants.It suggested that transgenic plants are more susceptible to osmotic stress than WT plants.Taken together,the results showed that the suppression of PEPCase by RNAi leads to susceptibility to osmotic stress in rapeseed,and PEPCase is involved in the response of C3 plants to environmental stress.Mei Chen Yunlai Tang Jingmei Zhang Mingfeng Yang Yinong Xu 2010Journal of Integrative Plant Biology2010,52,6:4
3Thioetherification of isoprene and butanethiol on transition metal phosphides显示文摘Thioetherification between mercaptan and diolefin is an efficient process to remove mercaptans in FCC gasoline at mild condition, during which the selective hydrogenation of diolefin to monoolefin is also expected. Here, Si O2 supported transition metal(Fe, Co, Ni, Mo and W) phosphides were tested for the thioetherification of isoprene and butanethiol on a fixed-bed reactor at 120 °C and 1.5 MPa H2, and their structure before and after reaction was characterized by means of XRD, HRTEM, N2 sorption, CO chemisorption, NH3-TPD, XPS and TG. It was found that, among different metal phosphides, Mo P/Si O2 showed the best performance, and the optimal nominal Mo P loading was 25%. Apart from the nature of metal, the density of metal and acid sites determined the catalyst performance. Metal site was mainly responsible for hydrogenation of isoprene, while acid site dominantly contributed to the thioetherification and the polymerization of olefins. Moreover, a balance between metallic and acidic functions is required to arrive at a desired performance. Excessive metal sites or acid sites led to the over-hydrogenation of isoprene or the severe polymerization of olefins, respectively. 25%Mo P/Si O2 was tested for 37 h time on stream, and butanethiol conversion maintained at 100%; although isoprene conversion remarkably decreased, the selectivity to isopentenes exceeded 80% after reaction for 11 h. 4Deoxygenation of methyl laurate to hydrocarbons on silica-supported Ni-Mo phosphides: Effect of calcination temperatures of precursor显示文摘SiO2-supported Ni-Mo bimetallic phosphides were prepared by temperature-programmed reduction(TPR) method from the phosphate precursors calcined at different temperatures. Their properties were characterized by means of ultraviolet-visible diffuse reflectance spectroscopy(UV-Vis DRS), H2temperature-programmed reduction(H2-TPR), X-ray diffraction(XRD), transmission electron microscopy(TEM), CO chemisorption, H2 and NH3temperature-programmed desorptions(H2-TPD and NH3-TPD). Their catalytic performances for the deoxygenation of methyl laurate were tested in a fixed-bed reactor. When the precursors were calcined at 400 and 500?C, respectively, Ni Mo P2 phase could be formed apart from Ni2 P and Mo P phases in the prepared C400 and C500 catalysts. However, when the precursors were calcined at600, 700 and 800°C, respectively, only Ni2 P and Mo P phases could be detected in the prepared C600, C700 and C800 catalysts. Also, in C400, C500 and C600 catalysts, Mo atoms were found to be entered in the lattice of Ni2 P phase, but the entering extent became less with the increase of calcination temperature. As the calcination temperature of the precursor increased, the interaction between Ni and Mo in the prepared catalysts decreased, and the phosphide crystallite size tended to increase, subsequently leading to the decrease in the surface metal site density and the acid amount. C600 catalyst showed the highest activity among the tested ones for the deoxygenation of methyl laurate. As the calcination temperature of the precursor increased, the selectivity to C12 hydrocarbons decreased while the selectivity to C11 hydrocarbons tended to increase. This can be mainly attributed to the decreased Ni-Mo interaction and the increased phosphide particle size. 5Tocilizumab in patients with moderate or severe COVID-19: a randomized, controlled, open-label, multicenter trial显示文摘Tocilizumab has been reported to attenuate the“cytokine storm”in COVID-19 patients.We attempted to verify the effectiveness and safety of tocilizumab therapy in COVID-19 and identify patients most likely to benefit from this treatment.We conducted a randomized,controlled,open-label multicenter trial among COVID-19 patients.The patients were randomly assigned in a 1:1 ratio to receive either tocilizumab in addition to standard care or standard care alone.The cure rate,changes of oxygen saturation and interference,and inflammation biomarkers were observed.Thirty-three patients were randomized to the tocilizumab group,and 32 patients to the control group.The cure rate in the tocilizumab group was higher than that in the control group,but the difference was not statistically significant(94.12%vs.87.10%,rate difference 95%CI−7.19%–21.23%,P=0.4133).The improvement in hypoxia for the tocilizumab group was higher from day 4 onward and statistically significant from day 12(P=0.0359).In moderate disease patients with bilateral pulmonary lesions,the hypoxia ameliorated earlier after tocilizumab treatment,and less patients(1/12,8.33%)needed an increase of inhaled oxygen concentration compared with the controls(4/6,66.67%;rate difference 95%CI−99.17%to−17.50%,P=0.0217).No severe adverse events occurred.More mild temporary adverse events were recorded in tocilizumab recipients(20/34,58.82%)than the controls(4/31,12.90%).Tocilizumab can improve hypoxia without unacceptable side effect profile and significant influences on the time virus load becomes negative.For patients with bilateral pulmonary lesions and elevated IL-6 levels,tocilizumab could be recommended to improve outcome.Dongsheng Wang Binqing Fu Zhen Peng Dongliang Yang Mingfeng Han Min Li Yun Yang Tianjun Yang Liangye Sun Wei Li Wei Shi Xin Yao Yan Ma Fei Xu Xiaojing Wang Jun Chen Daqing Xia Yubei Sun Lin Dong Jumei Wang Xiaoyu Zhu Min Zhang Yonggang Zhou Aijun Pan Xiaowen Hu Xiaodong Mei Haiming Wei Xiaoling Xu 2021Frontiers of Medicine2021,15,3:2
6Distribution of Asbestos Enterprises and Asbestosis Cases--China, 1997−2019显示文摘What is already known on this topic?Asbestos is classified as a Class I Carcinogen by the International Agency for Research on Cancer(IARC)because exposure causes mesothelioma and lung cancer in addition to asbestosis and plaques.So far,asbestos has been banned in 67 countries,but chrysotile,a commonly encountered form of asbestos,is still widely used in China and most developing countries.Most asbestos-caused cancers are not reported,recorded,and compensated in many countries.What is added by this report?Enterprises manufacturing asbestos products have been migrating from economically developed Eastern China to relatively underdeveloped central and western regions between 2010 and 2019.Asbestosis cases reported in Tianjin,Beijing,Shandong,Xinjiang,Gansu,Qinghai,and Sichuan accounted for a large proportion of the total cases in China,which was inconsistent with the distribution of asbestos-related enterprises(AREs).The reported asbestosis cases versus total pneumoconiosis cases declined from 2.81%to 0.39%from 2006–2017,and this proportion reached 0.69%in 2018.What are the implications for public health practice?Robust occupational and environmental health assessments and reporting are needed to define the epidemiology of asbestos-related lung diseases,and management of using asbestos and existing products containing asbestos need strengthening and follow-up.Enterprises should be encouraged to use safer substitutes and gradually ban asbestos materials in China.Mingfeng Chen Huanqiang Wang Jianfang Zhang Chen Yu Weidong Liu Yunhao Xu 2020China CDC weekly2020,2,18:2

8Microstructural evolution of fine-grained ZA27 alloy during partial remelting显示文摘The microstructural evolution process of fined-grained ZA27 alloy during partial remelting has been investigated. The relationship between the as-cast and semi-solid microstructures has been discussed in particular. The results indicate that a semi-solid microstructure with small and spheroidal primary particles can be obtained when the ZA27 alloy is partially remelted. The microstructural evolution can be divided into four stages, the initial coarsening of the dendrites due to coalescence of dendrite arms, structural separation resulted from the melting of residual interdendritic eutectic, spheroidization due to the partial melting of solid particles and final coarsening attributed to the coalescence and Ostwald ripening. An equiaxed dendrite in the as-cast microstructure may evolve into one spheroidal particle in the semi-solid microsturucture after being partially remelted. The more equiaxed the dendrites in an as-cast microstructure are, the more spheroidal the solid particles in the semi-solid microstructure will be. Finer primary particles could be obtained if the alloy with finer as-cast microstructure was partially remelted. 13Engineered extracellular vesicles for concurrent Anti-PDL1 immunotherapy and chemotherapy显示文摘Immune checkpoint inhibitors(ICI)targeting PD-1/PD-L1 have been approved for the treatment of a variety of cancers.However,the efficacy of antibody-based ICIs could be further improved by mitigating anti-drug antibodies,proteolytic cleavage,and on-target off-tumor toxicity.One strategy for accomplishing this is through the use of extracellular vesicles(EVs),cell derived submicron vesicles with many unique properties.We constructed an engineered MDA-MB-231 cell line for harvesting EVs.This was accomplished by overexpressing a high-affinity variant human PD-1 protein(havPD-1),while simultaneously knocking out intrinsic PD-L1 and beta-2 microglobulin.The engineered havPD-1 EVs reduced PD-L1 overexpressing cancer cell proliferation and induced cellular apoptosis.Moreover,the EVs were shown to efficiently block PD-L1 mediated T cell suppression.Meanwhile antibody-dependent cellular cytotoxicity and complement-dependent cytotoxicity were not observed.The havPD-1 EVs treatment resulted in robust anti-tumor activity in both preventative co-implantation and therapeutic xenograft tumor models reconstituted with human T cells.The efficacy of the havPD-1 EVs was shown to be comparable to clinical anti-PD1 monoclonal antibodies.Additionally,loading the havPD-1 EVs with a potent PARP inhibitor was shown to further augment treatment efficacy.In brief,the engineered universal EVs harboring havPD-1 proteins can be used for cancer concurrent immunotherapy and chemotherapy.Yundi Chen Lixue Wang Mingfeng Zheng Chuandong Zhu Guosheng Wang Yiqiu Xia Ethan J.Blumenthal Wenjun Mao Yuan Wan 2022Bioactive Materials2022,7,3:1
14Efficient activation of peroxymonosulfate by hollow cobalt hydroxide for degradation of ibuprofen and theoretical study显示文摘Hollow microsphere structure cobalt hydroxide(h-Co(OH)2) was synthesized via an optimized solvothermal-hydrothermal process and applied to activate peroxymonosulfate(PMS) for degradation of a typical pharmaceutically active compound,ibuprofen(IBP).The material characterizations confirmed the presence of the microscale hollow spheres with thin nanosheets shell in h-Co(OH)2,and the crystalline phase was assigned to a-Co(OH)2.h-Co(OH)2 could efficiently activate PMS for radicals production,and 98.6% of IBP was degraded at 10 min.The activation of PMS by h-Co(OH)2 was a pHindependent process,and pH 7 was the optimum condition for the activation-degradation system.Scavenger quenching test indicated that the sulfate radical(SO4^·-) was the primary reactive oxygen species for IBP degradation,which contributed to 75.7%.Fukui index(f^-) based on density functional theory(DFT) calculation predicted the active sites of IBP molecule for SO4^·- attack,and then IBP degradation pathway was proposed by means of intermediates identification and theoretical calculation.The developed hollow Co(OH)2 used to efficiently activate PMS is promising and innovative alternative for organic contaminants removal from water and wastewater.Mingfeng Ma Long Chen Jingzhu Zhao Wen Liu Haodong Ji 2019Chinese Chemical Letters2019,30,12:1

19Solar term peak of onset and death in 1 597 patients with acute ischemic stroke Circular statistical analysis显示文摘BACKGROUND: Previously, time data were analyzed by using constituent ratio or relative ratio; however, circular statistical analysis could exactly provide average peak phase of diseases. OBJECTIVE: To investigate the correlation of solar term peak with onset and death of acute ischemic stroke. DESIGN: Retrospective case analysis. SETTINGS: Emergency Department of Foshan Municipal Hospital of Traditional Chinese Medicine; Department of Science and Education, the Second People's Hospital of Foshan. PARTICIPANTS: A total of 1 597 patients with acute ischemic stroke were selected from Emergency Room, Department of Neurology, Foshan Municipal Hospital of Traditional Chinese Medicine from 1994 to 2002. There were 875 males and 722 females, and their ages ranged from 33 to 97 years. All cases met the diagnostic criteria of acute cerebral infarction modified by the Second National Cerebrovascular Disease Academic Meeting; meanwhile, they were diagnosed with CT/MRI test. Patients and their relatives provided the confirmed consent. METHODS: Solar term of onset was retrospectively analyzed in 1 597 patients with acute ischemic stroke; among them, solar term of death in 90 cases were analyzed by using circular statistical analysis to calculate peak phase of onset and death of acute ischemic stroke and investigate the correlation of solar term with onset and death of acute ischemic stroke. MAIN OUTCOME MEASURES: Onset and death time of patients with acute ischemic stroke. RESULTS: Solar term of onset in 1 597 patients, especially solar term of death in 90 patients, was not concentrated (P > 0.05), and specific peak phase was not found out. Acute ischemic stroke low attacked from vernal equinox to summer begins, but death caused by acute ischemic stroke high attacked from grain buds to autumn begins.
