| 2 | Protective effects of human umbilical cord mesenchymal stem cell vein transplantation against spinal cord ischemia/reperfusion injury in rats显示文摘BACKGROUND: The majority of studies addressing spinal cord ischemia/reperfusion injury (SCIRI) have focused on drugs, proteins, cytokines, and various surgical techniques. A recent study reports that human umbilical cord mesenchymal stem cell (hUCMSC) transplantation achieves good therapeutic effects, but the mechanisms underlying nerve protection remain poorly understood. OBJECTIVE: To observe survival of transplanted hUCMSCs in SCIRI rat models and the influence on motor function in the hind limbs, to determine interleukin-8 expression and cellular apoptosis in spinal cord tissues, and to verify the hypothesis that hUCMSC transplantation exhibits protective effects on SCIRI. DESIGN, TIME AND SETTING: A randomized, controlled, animal experiment was performed at the Laboratory of the Department of Orthopedics in the First Affiliated Hospital of Soochow University, China between January 2007 and December 2008. MATERIALS: hUCMSCs were harvested from umbilical cord blood of healthy pregnant women after parturition in the Obstetrical Department of the First Affiliated Hospital of Soochow University, China. Rabbit anti-human BrdU monoclonal antibody was provided by DAKO, USA. Terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) Kit and enzyme-linked immunosorbent assay (ELISA) Kit were purchased by Wuhan Boster, China. METHODS: A total of 72 healthy, Wistar, adult rats were randomly assigned to three groups: sham-surgery, model, and transplantation, with 24 rats in each group. SCIRI was induced in the model and transplantation groups via the abdominal aorta block method. The infrarenal abdominal aorta was not blocked in the sham-surgery group. Prior to abdominal aorta occlusion, 0.2-0.3 mL bromodeoxyuridine (BrdU)-labeled hUCMSCs suspension (cell concentration 5 × 103/μL) was injected through the great saphenous vein of the hind limb, and an equal volume of physiological saline was administered to the model and sham-surgery groups. MAIN OUTCOME MEASURES: Pathological observation of rat spinal cord tissues was performed by hematoxylin-eosin staining at 6, 24, and 48 hours post-surgery. Immunohistochemistry was applied to determine hUCMSCs survival in the spinal cord. The amount of cellular apoptosis and interleukin-8 expression in spinal cord tissues was assayed utilizing the TUNEL and ELISA methods, respectively. Motor function in the hind limbs was evaluated according to Jacob's score. RESULTS: Numerous BrdU-positive cells were observed in spinal cord tissues from the transplantation group. The number of apoptotic cells and interleukin-8 levels significantly decreased in the transplantation group (P < 0.05), pathological injury was significantly ameliorated, and motor function scores significantly increased (P < 0.05) compared with the model group. CONCLUSION: Via vein transplantation, hUCMSCs were shown to reach and survive in the injury area. Results suggested that the transplanted hUCMSCs contributed to significantly improved pathological changes in the injured spinal cord, as well as motor function, following SCIRI. | 3 | Carbonaceous and nitrogenous disinfection byproduct precursor variation during the reversed anaerobic–anoxic–oxic process of a sewage treatment plant显示文摘Disinfection byproduct(DBP)precursors in wastewater during the reversed anaerobic–anoxic–oxic(A^2/O)process,as well as their molecular weight(MW)and polarity-based fractions,were characterized with UV scanning,fluorescence excitation emission matrix,Fourier transform infrared and nuclear magnetic resonance spectroscopy.Their DBP formation potentials(DBPFPs)after chlorination were further tested.Results indicated that the reversed A^2/O process could not only effectively remove the dissolved organic carbon(DOC)and dissolved total nitrogen in the wastewater,but also affect the MW distribution and hydrophilic–hydrophobic properties of dissolved organic matter(DOM).The accumulation of low MW and hydrophobic(HPO)DOM was possibly due to the formation of soluble microbial product-like(SMP-like)matters in the reversed A^2/O treatment,especially in the anoxic and aerobic processes.Moreover,DOM in the wastewater displayed a high carbonaceous disinfection byproduct formation potential(C-DBPFP)in the fractions of MW>100 k Da and MW<5 k Da,and revealed an increasing tendency of nitrogenous disinfection byproduct formation potential(N-DBPFP)with decrease of MW.For polarity-based fractions,the HPO fraction of wastewater showed significantly higher C-DBPFP and N-DBPFP than hydrophilic and transphilic fractions.Therefore,although the reversed A^2/O process could remove most DBP precursors by DOC reduction,it led to the enhancement of DBPFP with the formation and accumulation of low MW and HPO DOM.In addition,strong correlations between C-DBPFPs and SUVA,and between N-DBPFPs and DON/DOC,were observed in the wastewater,which might be helpful for DBPFP prediction in wastewater and reclaimed water chlorination. | Huihui Han Hengfeng Miao Yajing Zhang Minfeng Lu Zhenxing Huang Wenquan Ruan | 2018 | Journal of Environmental Sciences2018,30,3: | 3 |
| 4 | Mitochondrial malic enzyme 2 promotes breast cancer metastasis via stabilizing HIF-1α under hypoxia显示文摘Objective: α-ketoglutarate(α-KG) is the substrate to hydroxylate collagen and hypoxia-inducible factor-1α(HIF-1α), which are important for cancer metastasis. Previous studies have shown that the upregulation of collagen prolyl 4-hydroxylase in breast cancer cells stabilizes the expression of HIF-1α by depleting α-KG levels. We hypothesized that mitochondrial malic enzyme 2(ME2) might also affect HIF-1α expression via modulating α-KG levels in breast cancer cells.Methods: We evaluated ME2 protein expression in 100 breast cancer patients using immunohistochemistry and correlated with clinicopathological indicators. The effect of ME2 knockout on cancer metastasis was evaluated using an orthotopic breast cancer model. The effect of ME2 knockout or knockdown on the levels of α-KG and HIF-1α proteins in breast cancer cell lines was determined both in vitro and in vivo.Results: ME2 was found to be upregulated in the human breast cancerous tissues compared with the matched precancerous tissues(P<0.001). The elevated expression of ME2 was associated with a poor prognosis(P=0.019).ME2 upregulation was also related to lymph node metastasis(P=0.016), pathological staging(P=0.033), and vascular cancer embolus(P=0.014). Also, ME2 knockout significantly inhibited lung metastasis in vivo. In the tumors formed by ME2 knockout cells, the levels of α-KG were significantly increased and collagen hydroxylation level did not change significantly but HIF-1α protein expression was significantly decreased, compared to the control samples. In cell culture, cells with ME2 knockout or knockdown demonstrated significantly higher α-KG levels but significantly lower HIF-1α protein expression than control cells under hypoxia. Exogenous malate and α-KG exerted similar effect on HIF-1α in breast cancer cells to ME2 knockout or knockdown. Additionally,treatment with malate significantly decreased 4 T1 breast cancer lung metastasis. | 5 | Estimation of earthquake losses by using macroeconomic approach显示文摘Most earthquake loss studies use an inventory approach by which the predicted damages in various categories of structures and facilities in a concerned region are estimated and added together to obtain the total estimated. Such an approach requires a detailed inventory database of the structures and facilities in the region, which is not always readily available in many regions of the world. Therefore, an alternative means of estimating earthquake losses is used based on several macroeconomic indices such as the gross domestic product (GDP) and population. Based on the published earthquake loss data during 1980—1995, the relations between GDP and earthquake losses have been formulated empirically for several intensity ranges. The world’s land surface was divided into unit cells with 0.5°×0.5°in size, the GDP of each cell was apportioned based on its population and the GDP and population of the region to which it belongs. | 11 | Vertical and horizontal displacements of a reservoir slope due to slope aging effect,rainfall,and reservoir water显示文摘Landslides are common hazards in reservoir areas and significantly affect dam operation and human lives.For the prevention and management of landslides,accurate assessment of the factors influencing their generation is essential.This study evaluated the key external factors influencing horizontal and vertical displacements of Luobogang Reservoir Slope in Hanyuan County,China.Displacements had been monitored by a surface-displacement-monitoring system consisting of 118 GPS stations during 2012-2015.To identify the external driving factors,their influence zones,and slope responses,we analyzed 32 months of displacement measurements and other multi-source datasets using the empirical orthogonal function.Overall,the results show that slope aging effect,rainfall,and reservoir water levels are three main driving factors.For horizontal displacement,aging effect is the most critical factor and predominantly affects the edges of landslides,the gob cave,and the public building zones.The secondary factor is the reservoir water level,which mainly acts on the boundary between the slope and reservoir water surface.The closer the slope zone is to the reservoir water,the more significant the impact is.Regarding vertical displacement,the most important factor is rainfall.The vertical displacement caused by rainfall accounts for 56.76% of the total vertical displacements.However,rainfall induces elastic displacements that generally cause less damage to the slope.The secondary factor is aging effect,and the vertical displacement caused by aging effect accounts for 9.42%.However,seven individual zones are highly affected by slope aging effect,which is consistent with the distribution of public buildings. | Dongzhen Jia Ye Zhou Xiufeng He Nan Xu Zhixiang Yang Minfeng Song | 2021 | Geodesy and Geodynamics2021,12,4: | 1 |
| 12 | Ultrathin Zn-free anode based on Ti_(3)C_(2)T_(x) and nanocellulose enabling high-durability aqueous hybrid Zn-Na battery with Zn2+/Na+co-intercalation mechanism显示文摘With low cost and high safety,aqueous zinc-based batteries have received considerable interest.Nevertheless,the excess utilization of zinc metal in the anodes of these batteries reduces energy density and increases costs.Herein,an ultrathin electrode of approximately 6.2μm thick is constructed by coating Ti_(3)C_(2)T_(x)/nanocellulose hybrid onto a stainless steel foil.This electrode is used as the Zn-free anode for aqueous hybrid Zn-Na battery,in which,a concentrated electrolyte is used to improve electrochemical reversibility.The Ti_(3)C_(2)T_(x)/nanocellulose coating is found to improve the electrolyte wettability,facilitate desolvation process of hydrated Zn^(2+) ions,lower nucleation overpotential,improve zinc plating kinetics,guide horizontal zinc plating along the Zn(002)facet,and inhibit parasitic side reactions.It is also found that the Na_(3)V_(2)(PO_(4))_(3) cathode material adopts a highly reversible Zn^(2+)/Na^(+)co-intercalation charge storage mechanism in this system.Thanks to these benefits,the assembled hybrid Zn-Na battery exhibits excellent rate capability,superior cyclability,and good anti-freezing ability.This work provides a new concept of electrode design for electrochemical energy storage. | Hao Chen Weijun Zhou Minfeng Chen Qinghua Tian Xiang Han Jizhang Chen | 2023 | Nano Research2023,16,1: | 1 |

| 17 | Tumor-responsive copper-activated disulfiram for synergetic nanocatalytic tumor therapy显示文摘Exploring alternative biomedical use of traditional drugs in different disease models is highly important as it can reduce the cost of drug development and overcome several critical issues of traditional chemodrugs such as low chemotherapeutic efficiency,severe side effect,and drug resistance.Disulfiram(DSF),a clinically approved alcohol-aversion drug,was recently demonstrated tofeature tumor-growth suppression effect along with the co-administration of Cu^(2+)species,but direct Cu^(2+)administration mode might cause severe toxicity originating from low Cu^(2+)accumulation into the tumor and nonspecific Cu^(2+)distribution-induced cytotoxicity.Based on the intriguing drug-delivery performance of nanoscale metal-organic frameworks(MOFs),we herein construct HKUST nMOFs as the Cu^(2+)self-supplying nanocarriers for efficient delivery of the D SF drug.The mildly acidic condition of tumor microenvironment initially triggered the release of Cu ions from HKUST nMOFs,which further reacted with the encapsulated DSF toform toxic Cu(DDTC)2(activation)for tumor chemotherapy.Especially,during the Cu(DDTC)2 complexation,Cu^(+)species were formed concomitantly,triggering the intratumoral nanocatalytic therapy for the generation of reactive oxygen species to synergistically destroying the tumor cells/tissue.As a result,synergetic tumor-responsive chemotherapy and nanocatalytic therapy are enabled by DSF@HKU ST nanodrugs,as demonstrated by the dominant anticancer efficacy with satisfied biocompatibility both in vitro and in vivo.The present work offers a sophisticated strategy for tumor-responsive nontoxic-to-toxic therapeutic with high biocompatibility. | Hao Chen Xi Li Minfeng Huo Liying Wang Yu Chen Wei Chen Bailiang Wang | 2021 | Nano Research2021,14,1: | 1 |
