| 1 | Evaluation of Glycosaminoglycan in the Lumbar Disc Using Chemical Exchange Saturation Transfer MR at 3.0 Tesla:Reproducibility and Correlation with Disc Degeneration显示文摘Objective This study aims to explore the clinical applicability and relevance of glycosaminoglycan Chemical Exchange Saturation Transfer(gag CEST) for intervertebral disc. Methods 25 subjects ranging in age from 24 yrs to 74 yrs were enrolled. gag CEST was acquired using a single-slice TSE sequence on a 3T. Saturation used a continuous rectangular RF pulse with B1=0.8 μT and a fixed duration time =1100 ms. Sagittal image was obtained firstly without saturation pulse, and then saturated images were acquired at 52 offsets ranging from ±0.125 to ±7 parts per million(ppm). MR T2 relaxivity map was acquired at the identical location. Six subjects were scanned twice to assess scan-rescan reproducibility. Results Gag CEST intraclass correlation coefficient(ICC) of six subjects was 0.759 for nucleus pulposus(NP) and 0.508 for annulus fibrosus(AF). Bland-Altman plots showed NP had a mean difference of 0.10%(95% limits of agreement:-3.02% to 3.22%); while that of AF was 0.34%(95% limits of agreement:-2.28% to 2.95%). For the 25 subjects, gag CEST in NP decreased as disc degeneration increased, with a similar trend to T2 relaxivity. Gag CEST of AF showed a better correlation with disc degeneration than T2 relaxivity. | 3 | Handheld high-throughput plasmonic biosensor using computational on-chip imaging显示文摘We demonstrate a handheld on-chip biosensing technology that employs plasmonic microarrays coupled with a lens-free computational imaging system towards multiplexed and high-throughput screening of biomolecular interactions for point-of-care applications and resource-limited settings.This lightweight and field-portable biosensing device,weighing 60 g and 7.5 cm tall,utilizes a compact optoelectronic sensor array to record the diffraction patterns of plasmonic nanostructures under uniform illumination by a single-light emitting diode tuned to the plasmonic mode of the nanoapertures.Employing a sensitive plasmonic array design that is combined with lens-free computational imaging,we demonstrate label-free and quantitative detection of biomolecules with a protein layer thickness down to 3 nm.Integrating large-scale plasmonic microarrays,our on-chip imaging platform enables simultaneous detection of protein mono-and bilayers on the same platform over a wide range of biomolecule concentrations.In this handheld device,we also employ an iterative phase retrieval-based image reconstruction method,which offers the ability to digitally image a highly multiplexed array of sensors on the same plasmonic chip,making this approach especially suitable for high-throughput diagnostic applications in field settings. | Arif E Cetin Ahmet F Coskun Betty C Galarreta Min Huang David Herman Aydogan Ozcan Hatice Altug | 2014 | Light(Science & Applications)2014，3，1： | 6 |

| 5 | Contribution of NMDA receptors to dorsolateral prefrontal cortical networks in primates显示文摘Cognitive disorders such as schizophrenia and Alzheimer's disease are associated with dysfunction of the highly evolved dorsolateral prefrontal cortex(dl PFC), and with changes in glutamatergic N-methyl-D-aspartate receptors(NMDARs). Recent research on the primate dl PFC discovered that the pyramidal cell circuits that generate the persistent firing underlying spatial working memory communicate through synapses on spines containing NMDARs with NR2 B subunits(Glu N2B) in the post-synaptic density. This contrasts with synapses in the hippocampus and primary visual cortex, where Glu N2 B receptors are both synaptic and extrasynaptic. Blockade of Glu N2 B in the dl PFC markedly reduces the persistent firing of the Delay cells needed for neuronal representations of visual space. Cholinergic stimulation of nicotinic α7 receptors within the glutamate synapse is necessary for NMDAR actions. In contrast, α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors have only subtle effects on the persistent firing of Delay cells, but contribute substantially to the firing of Cue and Response cells. Systemic administration of the NMDAR antagonist ketamine reduces the persistent firing of Delay cells, but increases the firing of some Response cells. The reduction in persistent firing produced by ketamine may explain why this drug can mimic or worsen the cognitive symptoms of schizophrenia.
