2An atlas of CNV maps in cattle, goat and sheep显示文摘Copy number variation(CNV)is the most prevalent type of genetic structural variation that has been recognized as an important source of phenotypic variation in humans,animals and plants.However,the mechanisms underlying the evolution of CNVs and their function in natural or artificial selection remain unknown.Here,we generated CNV region(CNVR)datasets which were diverged or shared among cattle,goat,and sheep,including 886 individuals from 171 diverse populations.Using 9 environmental factors for genome-wide association study(GWAS),we identified a series of candidate CNVRs,including genes relating to immunity,tick resistance,multi-drug resistance,and muscle development.The number of CNVRs shared between species is significantly higher than expected(P<0.00001),and these CNVRs may be more persist than the single nucleotide polymorphisms(SNPs)shared between species.We also identified genomic regions under long-term balancing selection and uncovered the potential diversity of the selected CNVRs close to the important functional genes.This study provides the evidence that balancing selection might be more common in mammals than previously considered,and might play an important role in the daily activities of these ruminant species.Yongzhen Huang Yunjia Li Xihong Wang Jiantao Yu Yudong Cai Zhuqing Zheng Ran Li Shunjin Zhang Ningbo Chen Hojjat Asadollahpour Nanaei Quratulain Hanif Qiuming Chen Weiwei Fu Chao Li Xiukai Cao Guangxian Zhou Shudong Liu Sangang He Wenrong Li Yulin Chen Hong Chen Chuzhao Lei Mingjun Liu Yu Jiang 2021Science China(Life Sciences)2021,64,10:2

4A Hu sheep genome with the first ovine Y chromosome reveal introgression history after sheep domestication显示文摘The Y chromosome plays key roles in male fertility and reflects the evolutionary history of paternal lineages.Here,we present a de novo genome assembly of the Hu sheep with the first draft assembly of ovine Y chromosome(o MSY),using nanopore sequencing and Hi-C technologies.The o MSY that we generated spans 10.6 Mb from which 775 Y-SNPs were identified by applying a large panel of whole genome sequences from worldwide sheep and wild Iranian mouflons.Three major paternal lineages(HY1a,HY1b and HY2)were defined across domestic sheep,of which HY2 was newly detected.Surprisingly,HY2 forms a monophyletic clade with the Iranian mouflons and is highly divergent from both HY1a and HY1b.Demographic analysis of Y chromosomes,mitochondrial and nuclear genomes confirmed that HY2 and the maternal counterpart of lineage C represented a distinct wild mouflon population in Iran that diverge from the direct ancestor of domestic sheep,the wild mouflons in Southeastern Anatolia.Our results suggest that wild Iranian mouflons had introgressed into domestic sheep and thereby introduced this Iranian mouflon specific lineage carrying HY2 to both East Asian and Africa sheep populations.Ran Li Peng Yang Ming Li Wenwen Fang Xiangpeng Yue Hojjat Asadollahpour Nanaei Shangquan Gan Duo Du Yudong Cai Xuelei Dai Qimeng Yang Chunna Cao Weidong Deng Sangang He Wenrong Li Runlin Ma Mingjun Liu Yu Jiang 2021Science China(Life Sciences)2021,64,7:1

6Estimation of Lightning-Generated NO_(x) in the Mainland of China Based on Cloud-to-Ground Lightning Location Data显示文摘Lightning-generated nitrogen oxides(LNO_(x))have a major influence on the atmosphere and global climate change.Therefore,it is of great importance to obtain a more accurate estimation of LNO_(x).The aim of this study is to provide a reference for the accurate estimation of the total LNO_(x) in the mainland of China based on cloud-to-ground lightning(CG)location data from 2014 to 2018.The energy of each CG flash was based on the number of return strokes per CG flash,the peak current of each return stroke,and the assumed CG breakdown voltage.The energy of intracloud lightning(IC)was based on the estimated frequencies of IC and the assumed energy of each IC flash.Combining the energy of lightning and the number of nitric oxide(NO)molecules produced by unit energy(ρno),the total LNO_(x) production in the mainland of China was determined.The LNO_(x) in the mainland of China estimated in this study is in the range(0.157-0.321)×10^(9) kg per year[Tg(N)yr-1],which is on the high end of other scholars’works.Negative cloud-to-ground lightning(NCG)flashes produce the most moles of NO_(x),while positive cloud-to-ground lightning(PCG)flashes produce the least total moles of NO_(x).The breakdown voltage of PCG is greater than that of IC or NCG,while the latter has a greater output of LNO_(x).Qi LI Fengxia GUO Xiaoyu JU Ze LIU Mingjun GAN Kun ZHANG Binbin CAI 2023Advances in Atmospheric Sciences2023,40,1:0
7Mechanistic Insights into Membrane Protein Clustering Revealed by Visualizing EGFR Secretion显示文摘Most plasmalemmal proteins are organized into clusters to modulate various cellular functions.However,the machineries that regulate protein clustering remain largely unclear.Here,with EGFR as an example,we directly and in detail visualized the entire process of EGFR from synthesis to secretion onto the plasma membrane(PM)using a high-speed,high-resolution spinning-disk confocal microscope.First,colocalization imaging revealed that EGFR secretory vesicles underwent transport from the ER to the Golgi to the PM,eventually forming different distribution forms on the apical and basal membranes;that is,most EGFR formed larger clusters on the apical membrane than the basal membrane.A dynamic tracking image and further siRNA interference experiment confirmed that fusion of secretory vesicles with the plasma membrane led to EGFR clusters,and we showed that EGFR PM clustering may be intimately related to EGFR signaling and cell proliferation.Finally,we found that the size and origin of the secretory vesicles themselves may determine the difference in the distribution patterns of EGFR on the PM.More importantly,we showed that actin influenced the EGFR distribution by controlling the fusion of secretory vesicles with the PM.Collectively,a comprehensive understanding of the EGFR secretion process helps us to unravel the EGFR clustering process and elucidate the key factors determining the differences in the spatial distribution of EGFR PM,highlighting the correlation between EGFR secretion and its PM distribution pattern.Haijiao Xu Jinrui Zhang Yijia Zhou Guanfang Zhao Mingjun Cai Jing Gao Lina Shao Yan Shi Hongru Li Hongbin Ji Yikai Zhao Hongda Wang 2023Research2023,,1:0
8Effect of laser-discrete-quenching on bonding properties of electroplated grinding wheel with AISI 1045 steel substrate and nickel bond显示文摘To improve the bonding strength between the nickel bond and the hub of the electroplated diamond grinding wheel,a hybrid technique was proposed to combine laser prequenching steel substrate and post-electroplating nickel.To validate the effectiveness of the proposed technique,AISI 1045 substrate was nickel-coated.The bonding properties between the electroplated nickel coating and substrate with or without laser-discrete-quenching were discussed comparatively by scratch,indentation,and thermal shock tests.The results show that the prequenching treatment leads to phase transformation of AISI 1045 microstructure from the mixed pearlite and ferrite phases into the martensitic phase.Since the martensitic phase is characterized as a high corrosion resistance,the interface of substrate/coating is smooth and flat in the prequenched zone,and the coating is bonded well with the steel substrate.In contrast to the steel substrate without pre-quenching treatment,the proposed technique significantly enhanced the bonding strengths of the electroplated nickel-coating.On one hand,the average hardness of electroplated nickel-coating on the laser pre-quenched zone is increased by 18.7%,and the scratch depth with the same load become narrower and shallower.On the other hand,the coefficient of friction(CoF)and the vibration amplitude are reduced,and the coating is bonded effectively with the substrate to inhibit the crack initialization at the interface.This prevents effectively the coating from peeling off and improves significantly the thermal shock resistance property.Cong MAO Peihao CAI Yongle HU Yujie ZHONG Jishuang WEI Zhuming BI Yifeng JIANG Linfeng XIAO Mingjun ZHANG 2021Chinese Journal of Aeronautics2021,34,6:0

10Cell membrane sample preparation method of combined AFM and dSTORM analysis显示文摘A major role of cell membranes is to provide an ideal environment for the constituent proteins to perform their biological functions.A deep understanding of the membrane proteins assembly process under physiological conditions is quite important to elucidate both the structure and the function of the cell membranes.Along these lines,in this work,a complete workflow of the cell membrane sample preparation and the correlated AFM and dSTORM imaging analysis methods are presented.A specially designed,angle-controlled sample preparation device was used to prepare the cell membrane samples.The correlated distributions of the specific membrane proteins with the topography of the cytoplasmic side of the cell membranes can be obtained by performing correlative AFM and dSTORM measurements.These methods are ideal for systematically studying the structure of the cell membranes.The proposed method of the sample characterization was not only limited to the measurement of the cell membrane but also can be applied for both biological tissue section analysis and detection.Mingjun Cai Huili Wang Guanfang Zhao Hongru Li Jing Gao Hongda Wang 2022Biophysics Reports2022,8,4:0
