1Knockout mouse models of insulin signaling: Relevance past and future显示文摘Insulin resistance is a hallmark of type 2 diabetes. In an effort to understand and treat this condition, re searchers have used genetic manipulation of mice to uncover insulin signaling pathways and determine the effects of their perturbation. After decades of research much has been learned, but the pathophysiology o insulin resistance in human diabetes remains contro versial, and treating insulin resistance remains a chal lenge. This review will discuss limitations of mouse models lacking select insulin signaling molecule genes In the most influential mouse models, glucose metabo lism differs from that of humans at the cellular, organ and whole-organism levels, and these differences limi the relevance and benefit of the mouse models both in terms of mechanistic investigations and therapeutic development. These differences are due partly to im mutable differences in mouse and human biology, and partly to the failure of genetic modifications to produce an accurate model of human diabetes. Several fac tors often limit the mechanistic insights gained from experimental mice to the particular species and strain including: developmental effects, unexpected meta bolic adjustments, genetic background effects, and technical issues. We conclude that the limitations and weaknesses of genetically modified mouse models of insulin resistance underscore the need for redirection of research efforts toward methods that are more directly relevant to human physiology.Anne E Bunner P Charukeshi Chandrasekera Neal D Barnard 2014World Journal of Diabetes2014,5,2:9

3Update in the treatment of intracranial atherosclerotic disease显示文摘This review highlights the recent evolution of the imaging,medical management,surgical options and endovascular therapies for symptomatic intracranial atherosclerotic disease(ICAD).Recent imaging developments including optical coherence tomography and other modalities to assess the intracranial arteries for symptomatic ICAD are reviewed,not only to diagnose ICAD but to determine if ICAD plaques have any high-risk features for treatment.Potential future developments in the treatment of ICAD are discussed,including the development of trackable drug-coated balloons for the cerebral circulation to treat primary or restenotic arteries,new iterations of self-expanding intracranial stents with easier delivery systems,and the re-examination of indirect surgical bypass techniques for revascularisation.In addition to these important technological developments,however,is the evolving evidence regarding the best treatment window for these techniques and additional factors in medical management which can improve patient outcomes in this devastating pathology.Zachary R Barnard Michael J Alexander 2020Stroke & Vascular Neurology2020,5,1:5
4Mosquito larvicidal properties of Orthosiphon thymiflorus(Roth)Sleesen.(Family:Labiatae)against mosquito vectors,Anopheles stephensi,Culex quinquefasciatus and Aedes aegypti(Diptera:Culicidae)显示文摘Objective:To determine the mosquito larvicidal activities of hexane,chloroform,ethyl acetate, acetone and methanol leaf extract of Orthosiphon thymiflorus(O.thymiflorus) against Anopheles stephensi(An.stephensi),Culex quinquefasciatus(Cx.quinquefasciatus) and Aedes aegypti (Ae.aegypti).Methods:The larvicidal activity was assayed against three mosquito species at various concentrations ranging from(50-450 ppm) under the laboratory conditions.The LC50 and LC90value of the O.thymiflorus leaf extract was determined by Probit analysis.Results:The LC50 values of hexane,chloroform,ethyl acetate,acetone and methanol extract of O.thymiflorus third instar larvae of An.stephensi were LC50= 201.39,178.76,158.06,139.22 and 118.74 ppm; Cx.quinquefasciatus were LC50=228.13,209.72.183.35,163.55 and 149.96 ppm and Ae.aegypti were LC50=215.65,197.91,175.05,154.80 and 137.26 ppm,respectively.Maximum larvicidal activity was observed in the methanolic extract followed by acetone,ethyl acetate chloroform and hexane extract.The larval mortality was observed after 24 h exposure.No mortality was observed in control.Conclusions:The present results suggest that the effective plant crude extracts have potential to be used as an ideal eco-friendly approach for the control of mosquito vectors.This study provides the first report on the larvicidal activity of this plant crude solvent extract of against An.stephensi,Cx.quinquefasciatus and Ae.aegypti mosquitoes.K Kovendan K Murugan S Vincent Donald R Barnard 2012Asian Pacific Journal of Tropical Medicine2012,5,4:2

6Simultaneous Measurements of Harmonic Waves at Fatigue-Cracked Interfaces显示文摘Nonlinear harmonic waves generated at cracked interfaces are investigated theoretically and experimentally.A compact tension specimen is fabricated and the amplitude of the transmitted wave is analyzed as a function of position along the fatigued crack surface.In order to measure as many nonlinear harmonic components as possible,broadband lithium niobate(LiNbO3)transducers are employed together with a calibration technique for making absolute amplitude measurements with fluid−coupled receiving transducers.Cracked interfaces are shown to generate high acoustic nonlinearities,which are manifested as harmonics in the power spectrum of the received signal.The first subharmonic f/2 and the second harmonic 2f waves are found to be dominant nonlinear components for an incident toneburst signal of frequency f.To explain the observed nonlinear behavior,a partially closed crack is modeled by planar half interfaces that can account for crack parameters,such as crack opening displacement and crack surface conditions.The simulation results show reasonable agreement with the experimental results.Hyunjo Jeong Dan Barnard 2011Chinese Physics Letters2011,28,8:2
7Inflatable rock bolt bond strength versus rock mass rating (RMR): A comparative analysis of pull-out testing data from underground mines in Nevada显示文摘The purpose of this paper is to establish confidence in anticipated minimum bond strength for inflatable rock bolts by comparing the bond strength to variable geotechnical conditions using the rock mass rating(RMR) system. To investigate a correlation between these parameters,the minimum bond strength of pull-out tested inflatable rock bolts was compared to the RMR of the rock in which these bolts were placed. Bond strength vs. RMR plots indicate that expected minimum bond strength is positively correlated with RMR; however,the correlation is not strong. Cumulative percent graphs indicate that 97% of pull-out tests result in a minimum bond strength of 3.3 and 1.7 ton/m in RMR P 45 and <45,respectively.Although lower bond strengths are more commonly encountered in low RMR ground,high bond strengths are possible as well,yielding higher variability in bond strengths in low RMR ground. Bond strength of friction bolts relies on contact between the rock bolt and drill hole. Experience in Nevada indicates that RMR is known to affect both the quality and consistency of drill holes which likely affects bond strength. Drilling and bolting in low RMR ground is more sensitive to drilling and bolting practices,and strategies for maximizing bond strength in these conditions are discussed.Barnard Chase Kallu Raj R.
