| 1 | A Resource for Inactivation of MicroRNAs Using Short Tandem Target Mimic Technology in Model and Crop Plants显示文摘microRNAs (miRNAs)are endogenous small non-coding RNAs that bind to mRNAs and target them for cleavage and/or translational repression,leading to gene silencing.We previously developed short tandem target mimic (STTM)technology to deactivate endogenous miRNAs in Arabidopsis.Here,we created hundreds of STTMs that target both conserved and species-specific miRNAs in Arabidopsis,tomato,rice,and maize,providing a resource for the functional interrogation of miRNAs.We not only revealed the functions of several miRNAs in plant development,but also demonstrated that tissue-specific inactivation of a few miRNAs in rice leads to an increase in grain size without adversely affecting overall plant growth and development.RNA-seq and small RNAseq analyses of STTM156/157 and STTM165/166 transgenic plants revealed the roles of these miRNAs in plant hormone biosynthesis and activation,secondary metabolism,and ion-channel activity-associated electrophysiology,demonstrating that STTM technology is an effective approach for studying miRNA functions.To facilitate the study and application of STTM transgenic plants and to provide a useful platform for storing and sharing of information about miRNA-regulated gene networks,we have established an online Genome Browser (http://gffzzc750bc0108fa4044sbwckuquwkfbk6n6k.ffgz.tsg.suse.edu.cn/designindex2.php) to display the transcriptomic and miRNAomic changes in STTMinduced miRNA knockdown plants. | Ting Peng Mengmeng Qiao Haiping Liu Sachin Teotia Zhanhui Zhang Yafan Zhao Bobo Wang Dongjie Zhao Lina Shi Cui Zhang Brandon Le Kestrel Rogers Chathura Gunasekara Haitang Duan Yiyou Gu Lei Tian Jinfu Nie Jian Qi Fanrong Meng Lan Huang Qinghui Chen Zhenlin Wang Jinshan Tang Xiaoqing Tang Ting Lan Xuemei Chen Hairong Wei Quanzhi Zhao Guiliang Tang | 2018 | Molecular Plant2018,11,11: | 7 |
| 2 | Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). | 12 | High-precision potassium isotope analysis using the Nu Sapphire collision cell(CC)-MC-ICP-MS显示文摘This study presents high-precision analyses of stable potassium(K)isotope ratio using the recently-developed,collision-cell multi-collector inductively coupled plasma mass spectrometry(CC-MC-ICP-MS,Nu Sapphire).The accuracy of our analyses is confirmed by measuring well-characterized geostandards(including rocks and seawater).Our results are consistent with literature values and a precision of 0.04‰(2SD)has been achieved based on multiple measurements of BCR-2 geostandard over a six-month period.We also evaluate factors that may lead to artificial isotope fractionations,including the mismatches in K concentration and acid molarity between samples and bracketing standards,as well as potential matrices.As the K adsorption capacity of AGW50-X8(200-400 mesh)is reduced with an increasing amount of matrix elements,less than 150µg K was loaded during the column chemistry.To evaluate the potential use of K isotopes as an archive of paleo seawater composition,δ^(41)K values of an international seawater standard(IAPSO),a Mn-nodule(NOD-P-1),and two iron formation standards(FeR-2 and FeR-4)are reported.The δ^(41)K value of IAPSO is consistent with other seawater samples reported previously,further substantiating a homogeneous K isotopic distribution in modern global oceans.The K isotopes in Mn-nodule(NOD-P-1:−0.121±0.013‰)and iron formation samples(FeR-2:−0.538±0.009‰;FeR-4:−0.401±0.008‰)seem to be an effective tracer of their formation genesis and compositional changes of ancient seawater.Our results suggest that high-precision measurements of stable K isotopes can be routinely obtained and open up a large variety of geological applications,such as continental weathering,hydrothermal circulation and alteration of oceanic crust. | Wenjun LI Mengmeng CUI Qiqi PAN Jing WANG Bingyu GAO Shanke LIU Meng YUAN Benxun SU Ye ZHAO Fang-Zhen TENG Guilin HAN | 2022 | Science China Earth Sciences2022,65,8: | 1 |
| 13 | 5-HMF attenuates inflammation and demyelination in experimental autoimmune encephalomyelitis mice by inhibiting the MIF-CD74 interaction显示文摘The neuroprotective role of 5-hydroxymethyl-2-furfural(5-HMF)has been demonstrated in a variety of neurological diseases.The aim of this study is to investigate the effect of 5-HMF on multiple sclerosis(MS).IFN-γ-stimulated murine microglia(BV2 cells)are considered a cell model of MS.With 5-HMF treatment,microglial M1/2 polarization and cytokine levels are detected.The interaction of 5-HMF with migration inhibitory factor(MIF)is predicted using online databases.The experimental autoimmune encephalomyelitis(EAE)mouse model is established,followed by a 5-HMF injection.The results show that 5-HMF facilitates IFN-γ-stimulated microglial M2 polarization and attenuates the inflammatory response.According to the network pharmacology and molecular docking results,5-HMF has a binding site for MIF.Further results show that blocking MIF activity or silencing CD74 enhances microglial M2 polarization,reduces inflammatory activity,and prevents ERK1/2 phosphorylation.5-HMF inhibits the MIF-CD74 interaction by binding to MIF,thereby inhibiting microglial M1 polarization and enhancing the anti-inflammatory response.5-HMF ameliorates EAE,inflammation,and demyelination in vivo.In conclusion,our research indicates that 5-HMF promotes microglial M2 polarization by inhibiting the MIF-CD74 interaction,thereby attenuating inflammation and demyelination in EAE mice. | Dongsheng Guan Yingxia Li Yinglin Cui Huanghong Zhao Ning Dong Kun Wang Deqi Ren Tiantian Song Xiaojing Wang Shijie Jin Yinghe Gao Mengmeng Wang | 2023 | Acta Biochimica et Biophysica Sinica2023,55,8: | 0 |
| 14 | Mediating role of resilience in the relationship between COVID-19 related stigma and mental health among COVID-19 survivors:a cross-sectional study显示文摘Background The global coronavirus disease 2019(COVID-19)has caused many negative effects on physical and mental health of patients who have survived COVID-19.Apart from some long-lasting physical sequelae,those COVID-19 survivors are also suffering stigma and discrimination at different levels around the world.The current study aims to assess the role resilience played in stigma and mental disorders among COVID-19 survivors.Methods The cross-sectional study was carried out among former COVID-19 patients in Jianghan District(Wuhan,China)from June 10 to July 25,2021.The demographic questions,the Impact of Events Scale-Revised,the Generalized Anxiety Disorder Questionnaire,the Patient Health Questionnaire,the Resilience Style Questionnaire and the Short Version of COVID-19 Stigma Scale of 12 items were used to collect relevant information of the participants.Descriptive analyses,Pearson correlation analysis and Structural Equation Modeling were used to make data description and analysis.Results A total of 1541 out of 1601 COVID-19 survivors(887 females and 654 males)were included in the analysis.Perceived stigma of those COVID-19 survivors correlates significantly with anxiety(r=0.335,P<0.001),depression(r=0.325,P<0.001)and post-traumatic stress disorder(PTSD)(r=0.384,P<0.001).It has a direct effect on COVID-19 survivors’anxiety(β=0.326,P<0.001),depression(β=0.314,P<0.001),PTSD(β=0.385,P<0.001)and their resilience(β=−0.114,P<0.01).Resilience partially mediated the association between perceived stigma and anxiety(β=0.020,P<0.01),depression(β=0.020,P<0.01),and PTSD(β=0.014,P<0.01)among COVID-19 survivors.Conclusion Stigma has a significant negative impact on mental health,while resilience plays a mediator role in the relationship between stigma and mental health among COVID-19 survivors.Based on our study,we suggested that when designing psychological interventions for COVID-19 survivors,consideration should be taken into account to reduce stigma and improve resilience. | Weijun Xiao Xiaoyang Liu Hao Wang Yiman Huang Zhenwei Dai Mingyu Si Jiaqi Fu Xu Chen Mengmeng Jia Zhiwei Leng Dan Cui Winnie W.S.Mak Liming Dong Xiaoyou Su | 2023 | Infectious Diseases of Poverty2023,12,2: | 0 |
