1Cloned pigs derived from somatic cell nuclear transferembryos cultured in vitro atlow oxygen tension显示文摘Pig cloning has great potential to human xenotransplantation. The present study was designed to establish a more efficient system for producing cloned pigs by somatic cell nuclear transfer (SCNT). Our approach was as follows: SCNT embryos were reconstructed by using fetal fibroblasts of Chinese miniature pig as donors and in vitro matured oocytes of prepubertal gilts as recipients. Reconstructed em- bryos were induced by electrical fusion/activation and cultured in BSA-containing North Carolina State University 23 medium (NCSU-23) or Porcine Zygote Medium (PZM-3) at the gas condition of 5% CO2, 7% O2, 88% N2. A total of 230 cloned embryos were transferred to three surrogate sows, producing three piglets. One of them is apparently healthy. The clonal provenance of the piglet was indicated by its coat color and confirmed by DNA microsatellite analysis. 2Experimental investigation on the spread of aqueous foam over ethanol surface显示文摘The presence of ethanol has an adverse effect on foam spreading,and ethanol fire is difficult to extinguish with aqueous fire-fighting foams.Thus,it is necessary to explore the foam formulation suitable for ethanol fuels and study the spreading behavior of foam over ethanol surface.In the current work,stable foams based on hydrocarbon surfactant(SDS),fluorocarbon surfactant(FC1157),and polymers(XG)were prepared by using the compressed-air foam system.The spreading behaviors of foam on polar ethanol and non-polar heptane surface were observed and compared.Furthermore,the effects of stabilizer concentrations,foam flow rates and expansion ratios on foam spreading performance were investigated,respectively.The results indicate that aqueous SDS foam can spread on the heptane layer continuously,but it is difficult to cover the ethanol surface.The addition of XG and FC1157 can synergistically improve the spreading performance of aqueous foam over ethanol.Depending on stabilizer concentrations,there are remarkable differences in foam spreading behaviors.Besides,different foam application parameters including expansion ratios and foam flow rates significantly affect the foam spreading rate,despite the same foam formulation.The research methods and results guide the optimal design of foam formulations as well as the practical application of aqueous foam for ethanol fire extinguishment.Xiaoyang Yu Fan Li Xuyang Miao Ning Jiang Ruowen Zong Shouxiang Lu Changhai Li State 2020Chinese Journal of Chemical Engineering2020,28,11:2

4Theoretical expressions for soil particle detachment rate due to saltation bombardment in wind erosion显示文摘Saltation bombardment is a dominate dust emission mechanism in wind erosion.For loose surfaces,splash entrainment has been well understood theoretically.However,the mass loss predictions of cohesive soils are generally empirical in most wind erosion models.In this study,the soil particle detachment of a bare,smooth,dry,and uncrusted soil surface caused by saltation bombardment is modeled by means of classical mechanics.It is shown that detachment rate can be analytically expressed in terms of the kinetic energy or mass flux of saltating grains and several common mechanical parameters of soils,including Poisson's ratio,Young's modulus,cohesion and friction angle.The novel expressions can describe dust emission rate from cohesive surfaces and are helpful to quantify the anti-erodibility of soil.It is proposed that the mechanical properties of soils should be appropriately included in physically-based wind erosion models.XuYang Liu WenXiao Ning ZhenTing Wang 2020Research in Cold and Arid Regions2020,12,4:1

7Seasonal influence on the development of parthenotes and cloned embryos in pigs显示文摘Seasonal influence on the developmental ability of porcine embryos was studied by comparisons of the number of cumulus-oocytes-complexes (COCs) per ovary pair and meiotic maturation ability of oocytes in different seasons; and by the observations on developmental competence of the parthenogenetic oocytes and cloned embryos collected from different seasons. We found that the number of COCs per ovary pairs was significantly higher in spring compared to the other seasons (P<0.05). However, no significant difference was found in the rate of oocytes progressing to MⅡ in four seasons (P<0.05). In addition, the rate of blastocyst formation of parthenogenetic embryos in summer was obviously declined compared to the other seasons (P<0.05), and an increased blastocyst rate of cloned porcine embryos was found in spring compared to autumn and winter (P<0.05). 9Exosomes from young healthy human plasma promote functional recovery from intracerebral hemorrhage via counteracting ferroptotic injury显示文摘Intracerebral hemorrhage (ICH), as a type of life-threatening and highly disabled disease, has limited therapeutic approaches. Here, we show that exosomes derived from young healthy human plasma exhibiting typical exosomes features could facilitate functional recovery of ICH mice. When these exosomes are intraventricularly delivered into the brain after ICH, they mainly distribute around the hematoma and could be internalized by neuronal cells. Strikingly, exosomes administration markedly enhanced the behavioral recovery of ICH mice through reducing brain injury and cell ferroptosis. MiRNA sequencing revealed that microRNA-25-3p (miR-25- 3p) was differentially expressed miRNA in the exosomes from young healthy human plasma, compared with exosomes from the old control. Importantly, miR-25-3p mimicked the treatment effect of exosomes on behavioral improvement, and mediated the neuroprotective effect of exosomes against ferroptosis in ICH. Furthermore, luciferase assay and western blotting data illustrated that P53 as assumed the role of a downstream effector of miR-25-3p, thereby regulating SLC7A11/GPX4 pathway to counteract ferroptosis. Taken together, these findings firstly reveal that exosomes from young healthy human plasma improve functional recovery through counteracting ferroptotic injury by regulating P53/SLC7A11/GPX4 axis after ICH.
