1Orally administered extract from Prunella vulgaris attenuates spontaneous colitis in mdr1a^(-/-) mice显示文摘AIM: To investigate the ability of a Prunella vulgaris(P. vulgaris) ethanolic extract to attenuate spontaneous typhlocolitis in mdr1a-/- mice. METHODS: Vehicle(5% ethanol) or P. vulgaris ethanolic extract(2.4 mg/d) were administered daily by oral gavage to mdr1a-/- or wild type FVBWT mice from 6 wk of age up to 20 wk of age. Clinical signs of disease were noted by monitoring weight loss. Mice experiencingweight loss in excess of 15% were removed from the study. At the time mice were removed from the study, blood and colon tissue were collected for analyses that included histological evaluation of lesions, inflammatory cytokine levels, and myeloperoxidase activity. RESULTS: Administration of P. vulgaris extracts to mdr1a-/- mice delayed onset of colitis and reduced severity of mucosal inflammation when compared to vehicle-treated mdr1a-/- mice. Oral administration of the P. vulgaris extract resulted in reduced(P < 0.05) serum levels of IL-10(4.6 ± 2 vs 19.4 ± 4), CXCL9(1319.0 ± 277 vs 3901.0 ± 858), and TNFα(9.9 ± 3 vs 14.8 ± 1) as well as reduced gene expression by more than two-fold for Ccl2, Ccl20, Cxcl1, Cxcl9, IL-1 α, Mmp10, VCAM-1, ICAM, IL-2, and TNFα in the colonic mucosa of mdr1a-/- mice compared to vehicle-treated mdr1a-/-mice. Histologically, several microscopic parameters were reduced(P < 0.05) in P. vulgaris-treated mdr1a-/-mice, as was myeloperoxidase activity in the colon(2.49 ± 0.16 vs 3.36 ± 0.06, P < 0.05). The numbers of CD4+ T cells(2031.9 ± 412.1 vs 5054.5 ± 809.5) and germinal center B cells(2749.6 ± 473.7 vs 4934.0 ± 645.9) observed in the cecal tonsils of P. vulgaris-treated mdr1a-/- were significantly reduced(P < 0.05) from vehicle-treated mdr1a-/- mice. Vehicle-treated mdr1a-/- mice were found to produce serum antibodies to antigens derived from members of the intestinal microbiota, indicative of severe colitis and a loss of adaptive tolerance to the members of the microbiota. These serum antibodies were greatly reduced or absent in P. vulgaris-treated mdr1a-/- mice. 2Use of shear wave elastography for the diagnosis and follow-up of biliary atresia: A meta-analysis显示文摘BACKGROUND Timely differentiation of biliary atresia(BA)from other infantile cholestatic diseases can impact patient outcomes.Additionally,non-invasive staging of fibrosis after Kasai hepatoportoenterostomy has not been widely standardized.Shear wave elastography is an ultrasound modality that detects changes in tissue stiffness.The authors propose that the utility of elastography in BA can be elucidated through meta-analysis of existing studies.AIM To assess the utility of elastography in:(1)BA diagnosis,and(2)post-Kasai fibrosis surveillance.METHODS A literature search identified articles that evaluated elastography for BA diagnosis and for post-Kasai follow-up.Twenty studies met criteria for meta-analysis:Eleven for diagnosis and nine for follow-up post-Kasai.Estimated diagnostic odds ratio(DOR),sensitivity,and specificity of elastography were calculated through a random-effects model using Meta-DiSc software.RESULTS Mean liver stiffness in BA infants at diagnosis was significantly higher than in non-BA,with overall DOR 24.61,sensitivity 83%,and specificity 79%.Post-Kasai,mean liver stiffness was significantly higher in BA patients with varices than in patients without,with DOR 16.36,sensitivity 85%,and specificity 76%.Elastography differentiated stage F4 fibrosis from F0-F3 with DOR of 70.03,sensitivity 96%,and specificity 89%.Elastography also differentiated F3-F4 fibrosis from F0-F2 with DOR of 24.68,sensitivity 85%,and specificity 81%.CONCLUSION Elastography has potential as a non-invasive modality for BA diagnosis and surveillance post-Kasai.This paper’s limitations include inter-study method heterogeneity and small sample sizes.Future,standardized,multi-center studies are recommended.Ellen S Wagner Hussien Ahmed H Abdelgawad Meghan Landry Belal Asfour Mark B Slidell Ruba Azzam 2022World Journal of Gastroenterology2022,28,32:5

4Systematic review of periprosthetic tibia fracture after total knee arthroplasties显示文摘AIM: To investigate the known incidences, treatment options, and related outcomes of periprosthetic tibia fractures after total knee arthroplasty(TKA).METHODS: A literature search was done to identify studies that fit the inclusion criteria. The database search yielded 185 results, which were further reduced by the exclusion criteria to 13 papers, totaling 157 patients that met these criteria. Incidence rates of the different types of periprosthetic tibia fractures were determined and their treatments were subsequently analyzed based on the fracture's subclass, with patient outcomes being overall favorable.RESULTS: Of the 144 documented patients, 54(37.5%) had a subclass C fracture, which are frequently seen in revision arthroplasties or when using cement intraoperatively. The fractures of subclasses A and B occur postoperatively. There were 90 subclass A and B fractures with incidences of 18.75% and 43.75% respectively. When broken down by type, 62(55.36%) were type 1, 24(21.4%) were type 2, 24(21.4%) were type 3, and 2(1.8%) were type 4. Furthermore, from the studies that included origin of injury, the types were further classified as having non-traumatic or traumatic origins. Type 1 had 78%(40/51) non-traumatic origin and 22%(11/51) traumatic origin. Fifteen fractures were type 2, but 5 were falls and 1 through a motor vehicle accident, giving a trauma causation of 40%(6/15). Of the 24 type 3 fractures, 12 were falls and 2 vehicular accidents, leading to a trauma causation of 58%(14/24).CONCLUSION: Type 1 fractures were the most common. 6Highly Fluorescent Dye-Doped Silica Nanoparticles Increase Flow Cytometry Sensitivity for Cancer Cell Monitoring显示文摘Early and accurate diagnosis and treatment of cancer depend on rapid,sensitive,and selective detection of tumor cells.Current diagnosis of cancers,especially leukemia,relies on histology and fl ow cytometry using single dye-labeled antibodies.However,this combination may not lead to high signal output,which can hinder detection,especially when the probes have relatively weak affi nities or when the receptor is expressed in a low concentration on the target cell surface.To solve these problems,we have developed a novel method for sensitive and rapid detection of cancer cells using dye-doped silica nanoparticles(NPs)which increases detection sensitivity in fl ow cytometry analyses between 10-and 100-fold compared to standard methods.Our NPs are~60 nm in size and can encapsulate thousands of individual dye molecules within their matrix.We have extensively investigated surface modifi cation strategies in order to make the NPs suitable for selective detection of cancer cells using fl ow cytometry.The NPs are functionalized with polyethylene glycol(PEG)to prevent nonspecifi c interactions and with neutravidin to allow universal binding with biotinylated molecules.By virtue of their reliable and selective detection of target cancer cells,these NPs have demonstrated their promising usefulness in conventional fl ow cytometry.Moreover,they have shown low background signal,high signal enhancement,and effi cient functionalization,either with antibody-or aptamer-targeting moieties.M.-Carmen Estévez Meghan B.OʼDonoghue Xiaolan Chen Weihong Tan 2009Nano Research2009,2,6:3

15Humanized mice are susceptible to Salmonella typhi infection显示文摘Salmonella enterica serovar Typhi is a pathogen that only infects humans.Currently,there is no animal model for studying this pathogen.Recently,alymphoid RAG2^(-/-)/γc^(-/-) mice engrafted with human leukocytes,known as humanized mice,have been successfully utilized to develop experimental models for several human-specific viral infections,including HIV,human-like dengue fever and hepatitis C virus.Little is known about the usefulness and feasibility of the humanized mouse model for the study of human-specific bacterial pathogens,such as S.typhi.The aim of this study was to determine if Salmonella enterica serovar Typhi could establish productive infection in humanized mice.Here we report that intravenous inoculation of S.typhi into humanized mice,but not controls,established S.typhi infections.High bacterial loads were found in the liver,spleen,blood and bone marrow of mice reconstituted with human leukocytes,but not in the unreconstituted control mice.Importantly,S.typhi-infected humanized mice lost significant body weight,and some of the infected mice displayed neurological symptoms.Our data suggest,for the first time,that humanized mice are susceptible to S.typhi challenge and that this model can be utilized to study the pathogenesis of S.typhito develop novel therapeutic strategies.M Firoz Mian Elisabeth A Pek Meghan J Chenoweth Ali A Ashkar 2011Cellular & Molecular Immunology2011,8,1:2
16Geodynamics of oceanic plateau and plume head accretion and their role in Phanerozoic orogenic systems of China显示文摘We present three 3D numerical models of deep subduction where buoyant material from an oceanic plateau and a plume interact with the overriding plate to assess the influence on subduction dynamics,trench geometry,and mechanisms for plateau accretion and continental growth.Transient instabilities of the convergent margin are produced,resulting in:contorted trench geometry;trench migration parallel with the plate margin;folding of the subducting slab and orocline development at the convergent margin;and transfer of the plateau to the overriding plate.The presence of plume material beneath the oceanic plateau causes flat subduction above the plume,resulting in a 'bowed' shaped subducting slab.In plateau-only models,plateau accretion at the edge of the overriding plate results in trench migration around the edge of the plateau before subduction is re-established directly behind the trailing edge of the plateau.The plateau shortens and some plateau material subducts.The presence of buoyant plume material beneath the oceanic plateau has a profound influence on the behaviour of the convergent margin.In the plateau + plume model,plateau accretion causes rapid trench advance.Plate convergence is accommodated by shearing at the base of the plateau and shortening in the overriding plate.The trench migrates around the edge of the plateau and subduction is re-established well behind the trailing edge of the plateau,effectively embedding the plateau into the overriding plate.A slab window forms beneath the accreted plateau and plume material is transferred from the subducting plate to the overriding plate through the window.In all of the models,the subduction zone maintains a relatively stable configuration away from the buoyancy anomalies within the downgoing plate.The models provide a dynamic context for plateau and plume accretion in Phanerozoic accretionary orogenic systems such as the East China Orogen and the Central Asian Orogen(Altiads),which are characterised by accreted ophiolite complexes with diverse geochemical affinities,and a protracted evolution of accretion of exotic terranes including oceanic plateau and terranes with plume origins.Peter G.Betts Louis Moresi Meghan S.Miller David Willis 2015Geoscience Frontiers2015,6,1:2
17An assessment of interspecific competition between two introduced parasitoids of Diaphorina citri (Hemiptera: Liviidae)on caged citrus plants显示文摘Two parasitoids attacking nymphs of Asian citrus psyllid,Diaphorina citri Kuwayama (Hemiptera:Liviidae),Tamarixia radiata (Waterston)(Hymenoptera:Eulophi- dae)and Diaphoreneyrtus aligarhensis (Shafee,Alam &Agarwal)(Hymenoptera:Encyrtidae)are being released in California,USA in a classical biological control program. To evaluate the effect of multiple parasitoid species on D.citri mortality,we conducted mesocosm experiments under controlled conditions using a complete block design with 6 treatments (D.cirri nymphs exposed to:no parasitoids;D.aligarhensis or T.radiata alone;D.aligarhensis or T.radiata released first (by 48 h);and both species released simultaneously).Parasitism of D.citri nymphs by T.radiata exceeded 60%and was unchanged when D.aligarhensis were present.Parasitism by D.aligarhensis was greatest when T. radiata was absent (-28%)and was reduced in all treatments with T.radiata present (<3%).D.cirri mortality and parasitoid-related mortality olD.citri was consistent across parasitoid treatments.Laboratory results suggest that competition between D.aligarhensis and T.radiata is asymmetric and favors T.radiata.It may be difficult for D.aligarhensis to contribute significantly to D.cirri biological control where T.radiata is present.However, results reported here suggest that competition between T.radiata and D.aligarhensis is not likely to reduce parasitism by T.radiata or reduce parasitoid-induced mortality of D.citri.Meghan A.Vankosky Mark S.Hoddle 2019Insect Science2019,26,1:2
