| 1 | Osteoporosis:A Silent Disease with Complex Genetic Contribution显示文摘Osteoporosis is the most common multifactorial metabolic bone disorder worldwide with a strong genetic component. In this review, the evidence for a genetic contribution to osteoporosis and related phenotypes is summarized alongside with methods used to identify osteoporosis susceptibility genes. The key biological pathways involved in the skeleton and bone development are discussed with a particular focus on master genes clustered in these pathways and their mode of action. Furthermore, the most studied single nucleotide polymorphisms(SNPs) analyzed for their importance as genetic markers of the disease are presented. New data generated by nextgeneration sequencing in conjunction with extensive meta-analyses should contribute to a better understanding of the genetic basis of osteoporosis and related phenotype variability. | 2 | Effects of maternal diabetes on trophoblast cells显示文摘Diabetes mellitus(DM)is a health condition characterized by hyperglycemia over a prolonged period.There are three main types of DM:DM type 1(DM1),DM2 and gestational DM(GDM).Maternal diabetes,which includes the occurrence of DM1 and DM2 during pregnancy or GDM,increases the occurrence of gesttional complications and adverse fetal outcomes.The hyperglycemic intrauterine environment affects not only the fetus but also the placental development and function in humans and experimental rodents.The underlying mechanisms are still unclear,but some evidence indicates alterations in trophoblast proliferation,apoptosis and cell cycle control in diabetes.A proper coordination of trophoblast proliferation,differentiation and invasion is required for placental development.Initially,increased expression of proliferative markers in junctional and labyrinth zones of rat placentas and villous cytotrophoblast,syncytiotrophoblast,stromal cells and fetal endothelial cells in human placentas is reported among diabetics.Moreover,reduced apoptotic index and expression of some apoptotic genes are described in placentas of GDM women.In addition,cell cycle regulators including cyclins and cyclin-dependent kinase inhibitors seem to be affected by the hyperglycemic environment.More studies are necessary to check the balance between proliferation,apoptosis and differentiation in trophoblast cells during maternal diabetes. | Marlúcia Bastos Aires Anne Carolline Veríssimo dos Santos | 2015 | World Journal of Diabetes2015,6,2: | 9 |
| 3 | MicroRNAs as emerging biomarkers and therapeutic targets for pancreatic cancer显示文摘Despite tremendous efforts from scientists and clinicians worldwide, pancreatic adenocarcinoma(PDAC) remains a deadly disease due to the lack of early diagnostic tools and reliable therapeutic approaches. Consequently, a majority of patients(80%) display an advanced disease that results in a low resection rate leading to an overall median survival of less than 6 months. Accordingly, robust markers for the early diagnosis and prognosis of pancreatic cancer, or markers indicative of survival and/or metastatic disease are des-perately needed to help alleviate the dismal prognosis of this cancer. In addition, the discovery of new therapeutic targets is mandatory to design effective treatments. | 4 | Changing human-environment interactions in medium mountains:the Apuseni Mts(Romania)as a case study显示文摘The study of human-environment relationships in mountain areas is important for both theoretical and practical reasons, as many mountain areas suffer similar problems, such as depopulation, unemployment and natural hazards. Medium mountains constitute a special case within mountains, because they are more populated but less attractive as tourist destinations than high mountains. In this context, the Apuseni Mts(Romania) are considered as a case study. In this paper, we apply GIS-based, quantitative methods to characterize the strength and dynamics of human-environment interactions, taking into consideration some environmental factors(elevation, relative height, slope, river distance, lithology, land cover, natural attractions) as well as historical population and recent tourism data. We found that population density has strong(r2>0.8) relationships with all relief factors(elevation, relative height, slope, river distance), and that best-fit functions are nonlinear. We outlined the varying demographic scenarios by elevation zones and interpreted the historically switching sign of population change versus elevation relationship. We demonstrated that lithology also has an impact on the spatial distribution of population, although it is notindependent from the relief effect. The land cover of the mainly cultural landscape is very strongly correlated with relief parameters(especially slope), which suggests good adaptation. We pointed out the dominance of karst objects in the natural tourism potential of the Apuseni Mts and also explored further components of real tourism(spas, heritage, towns). | 5 | Immunotoxic potential of aeration lagoon effluents for the treatment of domestic and hospital wastewaters in the freshwater mussel Elliptio complanata显示文摘Municipal wastewaters are major sources of pollution for the aquatic biota. The purpose of this study was to determine the levels of some pharmaceutical products and the immunotoxic potential of a municipal wastewater aeration lagoon for the treatment of the domestic wastewaters of a small town with wastewater inputs from a 400-bed hospital complex. Endemic mussels were collected, caged and placed in the final aeration lagoon and at sites 1 km upstream and 1 km downstream of the effluent outfall in the receiving river for a period of 14 days. The results showed that the final aeration lagoon contained high levels of total coliforms, conductivity and low dissolved oxygen (2.9 mg/L) as well as detectable amounts of trimethoprim, carbamazepine, gemfibrozil, and norfloxacin at concentrations exceeding 50 ng/L. The lagoon effluent was indeed toxic to the mussel specimens, as evidenced by the appearance of mortality after 14 days (10% mortality), decreased mussel weight-to-shell-length ratio and loss of hemocyte viability. The number of adhering hemocytes, phagocytic activity, total nitrite levels and arachidonic cyclooxygenase activity were significantly higher in mussels placed in the final aeration lagoon. A multivariate analysis also revealed that water pH, conductivity, total coliforms and dissolved oxygen were the endpoints most closely linked with phagocytic activity, the amount of adhering hemocytes and loss of hemocyte viability.
