| 1 | Effects of icariin on cGMP-specific PDE5 and cAMP-specific PDE4 activities显示文摘Aim:To clarify the mechanism of the therapeutic action of icariin on erectlile dysfunction(ED).Methods:PDE5 was isolated from the human platelet and PDE4 form the rat liver tissue using the FPLC system (Pharmacia,Milton Keynes,UK)and the Mono Q column.The inhibitory effects of icariin on PDE5 and PDE4 activities were investigated by the two-step radioisotope procedure with [^3H]-c GMP/[^3H]-cAMP.Papaverine served as the control drug.Results:Icariin and papaverine showed dose-dependent inhibitory effects on PDE5 and PDE4 activities.The IC50 of Icariin and papaverine on PDE5 were 0.432μ mol/L and 0.680μmol/L,respectively and those on PDE4,73.50μmol/L and 3.07μmol/L,respectively.The potencies of selectivity of icariin and papaverine on PDE5(PDE4/PDE5 of IC50)were 167.67 times and 4.54 times,respectively.Conclusion:Icariin is a cGMP-specific PDE5 inhibitor that may be developed into an oral effective agent for the treatment of ED. | ZC Xin EK Kim CS Lin WJ Liu L Tian YM Yuan J Fu | 2003 | Asian Journal of Andrology2003，5，1： | 38 |
| 2 | Water in the upper mantle and deep crust of eastern China: concentration, distribution and implications显示文摘Understanding the concentration and distribution of water in the Earth's mantle plays a substantial role in studying its chemical, physical and dynamic processes. After a decade of research, a comprehensive dataset of water content in upper-mantle samples has been built for eastern China, which is now the only place with water-content data from such diverse types of natural samples, and provides an integrated picture of the water content and its distribution in the upper mantle at a continental scale. The main findings include the following:(i) the temporal heterogeneity of the water content in the lithospheric mantle from early Cretaceous(~120 Ma) to Cenozoic(<40 Ma) was tightly connected with the stability of the North China Craton(from its destruction to its consolidation);(ii) the heterogeneous water content in the Cenozoic lithospheric mantle beneath different blocks of eastern China was not only inherited from tectonic settings from which they came, but was also affected later by geological processes they experienced;(iii) the distinct water content between the lowermost crust and lithospheric mantle of eastern China and its induced rheological contrast at the base of the crust indicate that the continental crust–mantle boundary could behave either in a coupled or decoupled manner beneath different areas and/or at different stages;(iv) the alkali basalts of eastern China demonstrate a heterogeneous distribution of water content in the mantle;local and regional comparisons of the water content between the lithospheric mantle and basalts' source indicate that the Cenozoic alkali basalts in eastern China were not sourced from the lithospheric mantle.Instead, the inferred high water contents in the mantle sources suggest that the Cenozoic eastern China basalts were likely sourced from the mantle transition zone(MTZ); and(v) both oceanic and continental crusts may carry a certain amount of water back into the deep mantle of eastern China by plate subduction.Such recycled crustal materials have not only created a local water-rich zone, but have also introduced crustal geochemical signatures into the mantle, both accounting for crustal geochemical imprints in the intra-plate magmatic rocks of eastern China. | Qun-Ke Xia Jia Liu István Kovács Yan-Tao Hao Pei Li Xiao-Zhi Yang Huan Chen Ying-Ming Sheng | 2019 | National Science Review2019，6，1： | 16 |

| 5 | Genomic Analysis of the Natural Population of Wheat dwarf virus in Wheat from China and Hungary显示文摘During the last decade, the leafhopper transmitted Wheat dwarf virus (WDV) has become a serious problem both in northwestern China and Hungary. In order to study the molecular diversity and population structure of WDV in these two countries, 39 Chinese isolates and 16 Hungarian isolates were collected from different regions of China and Hungary, and their genomes were sequenced. All isolates belonged to the wheat strain of WDV and showed limited genetic diversity. The highest and lowest nucleotide sequence identities among isolates from China and Hungary were 99.9 and 90%, respectively. In all isolates, the lowest nucleotide sequence identity was 89.5% between MO10-1 and KP10-5, which were collected from Martonvàsàr and Kompolt, Hungary. Phylogenetic analyses showed the genome sequences of 55 WDV isolates belong to two big clades, but no clear correlation to geographical location. Population difference analyses indicated that the Chinese and Hungarian WDV populations have no significant difference. | 6 | Discovery and validation of prognostic markers in gastric cancer by genome-wide expression profiling显示文摘AIM: To develop a prognostic gene set that can predict patient overall survival status based on the whole genome expression analysis. METHODS: Using Illumina HumanWG-6 BeadChip followed by semi-supervised analysis, we analyzed the expression of 47 296 transcripts in two batches of gastric cancer patients who underwent surgical resection. Thirty-nine samples in the first batch were used as the training set to discover candidate markers correlated to overall survival, and thirty-three samples in the second batch were used for validation. RESULTS: A panel of ten genes were identified as prognostic marker in the first batch samples and classified patients into a lowand a high-risk group with significantly different survival times (P = 0.000047). This prognostic marker was then verified in an independent validation sample batch (P = 0.0009). By comparing with the traditional Tumor-node-metastasis (TNM) staging system, this ten-gene prognostic marker showed consistent prognosis results. It was the only independent prognostic value by multivariate Cox regression analysis (P = 0.007). Interestingly, six of these ten genes are ribosomal proteins, suggesting a possible association between the deregulation of ribosome related gene expression and the poor prognosis.
