1NLRP3-dependent pyroptosis is required for HIV-1 gp120- induced neuropathology显示文摘The human immunodeficiency virus-1(HIV-1)envelope protein gp120 is the major contributor to the pathogenesis of HIVassociated neurocognitive disorder(HAND).Neuroinflammation plays a pivotal role in gp120-induced neuropathology,but how gp120 triggers neuroinflammatory processes and subsequent neuronal death remains unknown.Here,we provide evidence that NLRP3 is required for gp120-induced neuroinflammation and neuropathy.Our results showed that gp120-induced NLRP3-dependent pyroptosis and IL-1βproduction in microglia.Inhibition of microglial NLRP3 inflammasome activation alleviated gp120-mediated neuroinflammatory factor release and neuronal injury.Importantly,we showed that chronic administration of MCC950,a novel selective NLRP3 inhibitor,to gp120 transgenic mice not only attenuated neuroinflammation and neuronal death but also promoted neuronal regeneration and restored the impaired neurocognitive function.In conclusion,our data revealed that the NLRP3 inflammasome is important for gp120-induced neuroinflammation and neuropathology and suggest that NLRP3 is a potential novel target for the treatment of HAND.Xiaolong He Weijun Yang Zhijie Zeng Yi Wei Jie Gao Bao Zhang Li Li Liqun Liu Yu Wan Qing Zeng Zelong Gong Liting Liu Hanyun Zhang Yubin Li Shaojie Yang Tongtong Hu Lixian Wu Eliezer Masliah Shenghe Huang Hong Cao 2020Cellular & Molecular Immunology2020,17,3:11
2AFAP1-AS1: a rising star among oncogenic long non-coding RNAs显示文摘Long non-coding RNAs(lncRNAs)have become a hotspot in biomedical research.This interest reflects their extensive involvement in the regulation of the expression of other genes,and their influence on the occurrence and development of a variety of human diseases.Actin filament associated protein 1-Antisense RNA 1(AFAP1-AS1)is a recently discovered oncogenic lncRNA.It is highly expressed in a variety of solid tumors,and regulates the expression of downstream genes and signaling pathways through adsorption and competing microRNAs,or by the direct binding to other proteins.Ultimately,AFAP1-AS1 promotes proliferation,chemotherapy resistance,and resistance to apoptosis,maintains stemness,and enhances invasion and migration of tumor cells.This paper summarizes the research concerning AFAP1-AS1 in malignant tumors,including the clinical application prospects of AFAP1-AS1 as a potential molecular marker and therapeutic target of malignant tumors.We also discuss the limitations in the knowledge of AFAP1-AS1 and directions of further research.AFAP1-AS1 is expected to provide an example for studies of other lncRNA molecules.Fang Xiong Kunjie Zhu Su Deng Hongbin Huang Liting Yang Zhaojian Gong Lei Shi Yi He Yanyan Tang Qianjin Liao Jianjun Yu Xiaoling Li Yong Li Guiyuan Li Zhaoyang Zeng Wei Xiong Shanshan Zhang Can Guo 2021Science China(Life Sciences)2021,64,10:4
3Liquid metal as reconnection agent for peripheral nerve injury显示文摘This study demonstrated an unconventional way to cure peripheral nerve injury(PNI) with liquid metal gallium employed as the reconnection agent. In vivo experiments were performed, in which transected sciatic nerve of mouse was reconnected by liquid metal gallium.The nerve signals detected was found to be almost the same as those from the complete nerve, where the negative bursting firing caused by PNI was absent on the neural discharge curve after nerve-reconnection surgery. Meanwhile the atrophy tendency of gastrocnemius muscle was distinctly procrastinated according to the results of pathological examinations, which showed fibrillation potentials emerged immediately for mice with PNI but did not emerge until the third month for those received nerve-reconnection surgery. Furthermore, physical properties of gallium were studied, showing that its impedance was slightly influenced by the frequency of transmitted signal and the temperature, which confirmed the stability of gallium in future clinical usage. 4Synthesis of Chitosan-Hydroxyapatite Composites and Its Effect on the Properties of Bioglass Bone Cement显示文摘Chitosan-hydroxyapatite(CS-HA) composite powders were synthesized via in situ co-precipitation method,through the reaction of Ca(NO3)2 and H3PO4 in the simulated body fluid(SBF) containing appropriate amount of chitosan.The thermal evolution,microstructure and morphology were studied by TG-DTA(thermogravimetry-differential thermal analysis),XRD(X-ray diffraction),FTIR(Fourier transform infrared spectroscopy) and TEM(transmission electron microscopy).The in vitro bioactivity test showed that the obtained CS-HA composites had higher capability of inducing calcium ions deposition.Effects of CS-HA composites on the bioactivity and compressive strength of bioglass bone cement were investigated.The results indicated that the bioactivity of bioglass bone cement could be improved further when CS-HA composite powders were added into the cement,and appropriate amount of CS-HA additive was favorable for compressive strength improvement of bioglass bone cement.Jingxiao Liu Fei Shi Ling Yu Liting Niu Shanshan Gao 2009Journal of Materials Science & Technology2009,25,4:3

9Shen Qi Wan attenuates renal interstitial fibrosis through upregulating AQP1显示文摘Renal interstitial fibrosis(RIF)is the crucial pathway in chronic kidney disease(CKD)leading to the end-stage renal failure.However,the underlying mechanism of Shen Qi Wan(SQW)on RIF is not fully understood.In the current study,we investigated the role of Aquaporin 1(AQP1)in SQW on tubular epithelial-to-mesenchymal transition(EMT).A RIF mouse model induced by adenine and a TGF-β1-stimulated HK-2 cell model were etablished to explore the involvement of AQP 1 in the protective effect of SQW on EMT in vitro and in vivo.Subsequently,the molecular mechanism of SQW on EMT was explored in HK-2 cells with AQP1 knockdown.The results indicated that SQW alleviated kidney injury and renal collagen deposition in the kidneys of mice induced by adenine,increased the protein expression of E-cadherin and AQP1 expression,and decreased the expression of vimentin andα-smooth muscle actin(α-SMA).Similarly,treatmement with SQW-containing serum significantly halted EMT process in TGF-β1 stimulated HK-2 cells.The expression of snail and slug was significantly upregulated in HK-2 cells after knockdown of AQP1.AQP1 knockdown also increased the mRNA expression of vimentin andα-SMA,and decreased the expression of E-cadherin.The protein expression of vimentin increased,while the expression of E-cadherin and CK-18 significantly decreased after AQP1 knockdown in HK-2 cells.These results revealed that AQP1 knockdown promoted EMT.Furthermore,AQP1 knockdown abolished the protective effect of SQW-containing serum on EMT in HK-2 cells.In sum,SQW attentuates EMT process in RIF through upregulation of the expression of AQP1.LIN Yiyou WEI Jiale ZHANG Yehui HUANG Junhao WANG Sichen LUO Qihan YU Hongxia JI Liting ZHOU Xiaojie LI Changyu 2023Chinese Journal of Natural Medicines2023,21,5:1

12Auricular vagus nerve acupressure for patients with emotional distress under the COVID-19 pandemic:A smartphone-based,randomized controlled trial显示文摘Objective: To confirm whether self-administered AVNA treatment is effective in improving emotional distressunder the COVID-19 pandemic.Methods: A smartphone-based online, randomized, controlled trial was designed from 26 February 2020 to 28April 2020 in four study sites, including Wuhan, Beijing, Shenyang, and Guangzhou of China. Local residentswho had considerable emotional distress with a score of the Hospital Anxiety and Depression Scale (HADS) ≥9 were recruited. Participants were randomly assigned to three times of AVNA (n = 191) per day, in morning,around noon, and in evening or usual care (UC, n = 215) once daily for 14 days. The primary outcome was theresponse rate, which was the proportion of participants whose Hospital Anxiety and Depression Scale (HADS)score reduced from baseline by ≥ 50%. The assessment was conducted at baseline, 3 days, and 14 days.Results: The AVNA group had a markedly higher response rate than the UC group at 3 days (35.6% vs. 24.9%,P = 0.02) and at 14 days (70.7% vs. 60.6%, P = 0.02). The AVNA group showed significantly greater reductionin scores of HADS at the two measurement points and BAI at 3 days (P ≤ 0.03), with average respective effectsize of 0.217 and 0.195. Participants with AVNA spent less time falling asleep and rated their sleep qualitybeing remarkably higher than those with UC at endpoint.Conclusion: During COVID-19 pandemic period, treatment with self-administrated AVNA was more effectivethan UC in reducing emotional distress of isolated populations. 15Long non-coding RNA AFAP1-AS1 accelerates lung cancer cells migration and invasion by interacting with SNIP1 to upregulate c-Myc显示文摘Actin filament associated protein 1 antisense RNA 1(named AFAP1-AS1)is a long non-coding RNA and overexpressed in many cancers.This study aimed to identify the role and mechanism of AFAP1-AS1 in lung cancer.The AFAP1-AS1 expression was firstly assessed in 187 paraffin-embedded lung cancer and 36 normal lung epithelial tissues by in situ hybridization.The migration and invasion abilities of AFAP1-AS1 were investigated in lung cancer cells.To uncover the molecular mechanism about AFAP1-AS1 function in lung cancer,we screened proteins that interact with AFAP1-AS1 by RNA pull down and the mass spectrometry analyses.AFAP1-AS1 was highly expressed in lung cancer clinical tissues and its expression was positively correlated with lung cancer patients'poor prognosis.In vivo experiments confirmed that AFAP1-AS1 could promote lung cancer metastasis.AFAP1-AS1 promoted lung cancer cells migration and invasion through interacting with Smad nuclear interacting protein 1(named SNIP1),which inhibited ubiquitination and degradation of c-Myc protein.Upregulation of c-Myc molecule in turn promoted the expression of ZEB1,ZEB2,and SNAIL gene,which ultimately enhanced epithelial to mesenchymal transition(EMT)and lung cancer metastasis.Understanding the molecular mechanism by which AFAP1-AS1 promotes lung cancer's migration and invasion may provide novel therapeutic targets for lung cancer patients'early diagnosis and therapy.Yu Zhong Liting Yang Fang Xiong Yi He Yanyan Tang Lei Shi Songqing Fan Zheng Li Shanshan Zhang Zhaojian Gong Can Guo Qianjin Liao Yujuan Zhou Ming Zhou Bo Xiang Xiaoling Li Yong Li Zhaoyang Zeng Guiyuan Li Wei Xiong 2021Signal Transduction and Targeted Therapy2021,6,7:1
16Wear Characteristics of the Nuclear Control Rod Drive Mechanism(CRDM)Movable Latch Serviced in High Temperature Water显示文摘The current research of nuclear control rod drive mechanism(CRDM)movable latch only makes a simple measurement of wear mass.The wear volume and difference in various claw surfaces are ignored and the degradation mechanism of each claw surface is not clear.In this paper,a detailed degradation analysis was carried out on each claw surface of movable latch combined with wear result and worn morphology.Results indicate that the boundary of carbide is preferred for corrosion because carbide presents a nobler Volta potential compared to the metal matrix or boundary region.Due to the oscillation of drive shaft between the claw surfaces of movable latch,the dominant wear mechanism on the upper surface of claw(USC)and lower surface of claw(LSC)is plastic deformation caused by impact wear.Mechanical impact wear will cause the fragmentation of carbides because of the high hardness and low ductility of carbides.Corrosion promotes the broken carbides to fall off from the metal matrix.The generated fine carbides(abrasive particles)cause extra abrasive wear on USC when the movable brings the drive shaft upward or downward.As a result,USC has a higher wear volume than LSC.This research proposes a method to evaluate the wear on the whole movable latches using a 3D full-size scanner.Tianda Yu Guozhong Fu Yanqing Yu Liting Zhu Maofu Liu Wei Li Qiang Deng Zhenbing Cai 2022Chinese Journal of Mechanical Engineering2022,35,2:1

20Assembling ionic liquid into porous molecular filler of mixed matrix membrane to trigger high gas permeability,selectivity,and stability for CO_(2)/CH_(4) separation显示文摘As an emerging zero-dimensional nano crystalline porous material,porous organic cages(POCs)with soluble properties in organic solvents,are promising candidates as molecular fillers in mixed matrix membranes(MMMs).The pore structure of POCs should be adjusted to trigger efficient gas separation performance,and the interaction between filler and matrix should be optimized.In this work,ionic liquid(IL)was introduced into the molecular fillers of CC3,to construct the IL@CC3/PIM-1 membrane to effectively separate CO_(2) from CH_(4).The advantages of doping IL include:(1)narrowing the cavity size of POCs from 4.4 to 3.9Åto enhance the diffusion selectivity,(2)strengthening the CO_(2) solubility to heighten the gas permeability,and(3)improving the compatibility between filler and matrix to upgrade membrane stability.After the optimization of the membrane composite,the IL@CC3/PIM-1-10%membrane possesses the CO_(2) permeability of 7868 Barrer and the CO_(2)/CH_(4) selectivity of 73.4,which compared to the CC3/PIM-1-10%membrane,improved by 15.9%and 106.2%,respectively.Furthermore,the membrane has maintained a stable separation performance at varied temperatures and pressures during the long-term test.The proposed method offers an efficient way to improve the performance of POCs-based MMMs in gas separation.Liting Yu Liqin Hao Yang Feng Jia Pang Mengwei Guo Liangjun Li Weidong Fan Lili Fan Rongming Wang Zixi Kang Daofeng Sun 2024Nano Research2024,17,5:0
