1Mouse macrophage specific knockout of SIRT1 influences macrophage polarization and promotes angiotensin Ⅱ-induced abdominal aortic aneurysm formation显示文摘Abdominal aortic aneurysm(AAA) is a vascular degenerative disease. Macrophage polarization and the balance between classically activated macrophages(M1) and alternatively activated macrophages(M2)are crucial for AAA pathogenesis. The present study aims to investigate the roles of macrophage SIRT1 in AAA formation and macrophage polarization. We found that in mouse peritoneal macrophages, SIRT1 expression was decreased after M1 stimulation, but was enhanced after M2 stimulation. Results from SIRT1^(flox/flox) mice and macrophage specific SIRT1 knockout mice with treatment of angiotensin II(Ang II)for 4 weeks showed that macrophage specific deficiency of SIRT1 increased the incidence of AAA and exacerbated the severity, including more severe aneurysm types, enlarged diameter of the aneurysm and increased degradation of elastin. In mouse aortas, SIRT1 deficiency increased the proinflammatory M1 molecule inducible nitric oxide synthase(i NOS), and decreased M2 molecules such as arginase 1(Arg1) and mannose receptor(MR). Furthermore, in peritoneal macrophages, SIRT1 deficiency increased the expression of M1 inflammatory molecules, but decreased the expression of M2 molecules. Overexpression of SIRT1 had the opposite effects. 3Truncated glycoprotein E of varicella-zoster virus is an ideal immunogen for Escherichia coli-based vaccine design显示文摘Varicella-zoster virus(VZV)is a highly infectious agent responsible for both varicella and herpes zoster disease.Despite high efficacy,there remain safety and accessibility concerns with the licensed vaccines.Here,we sought to produce a VZV g E immunogen using an E.coli expression system.We found that the soluble expression and yield of g E protein could be enhanced via C-terminal truncations to the protein,thereby facilitating a robust and scalable purification process for the purpose of vaccine manufacturing.The lead truncated g E(aa 31–358),hereafter referred to as tg E,was a homogenous monomer in solution and showed excellent antigenicity.Finally,we assessed and compared the immunogenicity of tg E with commercial v Oka LAV and Shingrix vaccine.We found that aluminum-adjuvanted tg E was immunogenic as compared with v Oka LAV.When adjuvanted with AS01B,a two-dose immunization of tg E showed comparable or better potency in antibody responses and cell-mediated immunity with those of the Shingrix vaccine at the same dosage,especially in terms of the proportion of IFN-γ-expressing CD4^(+)T cells.In conclusion,this method of E.coli-mediate tg E expression offers a cost-effective and scalable strategy to generate an ideal VZV g E immunogen for the development of both varicella and zoster vaccines.Tingting Chen Jie Sun Sibo Zhang Tingting Li Liqin Liu Wenhui Xue Lizhi Zhou Siting Liang Zhili Yu Qingbing Zheng Hai Yu Tong Cheng Jun Zhang Ying Gu Shaowei Li Ningshao Xia 2023Science China(Life Sciences)2023,66,4:1

8Bone morphogenetic protein 4 (BMP4) promotes hepatic glycogen accumulation and reduces glucose level in hepatocytes through mTORC2 signaling pathway显示文摘Liver is an important organ for regulating glucose and lipid metabolism.Recent studies have shown that bone morphogenetic proteins(BMPs)may play important roles in regu-lating glucose and lipid metabolism.Inour previous studies,we demonstrated that BMP4 signif-icantly inhibits hepatic steatosis and lowers serum triglycerides,playing a protective role against the progression of non-alcoholic fatty liver disease(NAFLD).However,the direct impact of BMP4 on hepatic glucose metabolism is poorly understood.Here,we investigated the regulatory roles of BMP4 in hepatic glucose metabolism.Through a comprehensive analysis of the 14 types of BMPs,we found that BMP4 was one of the most potent BMPs in promoting hepatic glycogen accumulation,reducing the level of glucose in hepatocytes and effecting the expression of genes related to glucose metabolism.Mechanistically,we demonstrated that BMP4 reduced the hepatic glucose lewels through the activation of mTORC2 signaling pathway in vitro and in wivo.Collectively,our findings strongly suggest that BMP4 may play an essential role in regulating hepatic glucose metabolism.This knowledge should aid us to understand the molecular pathogenesis of NAFLD,and may lead to the development of novel therapeutics by exploiting the inhibitory effects of BMPs on hepatic glucose and lipid metabolism.Liqin An Qiong Shi Ying Zhu Hao Wang Qi Peng Jinghong Wu Yu Cheng Wei Zhang Yanyu Yi Zihao Bao Hui Zhang Yetao Luo Jiaming Fan 2021Genes & Diseases2021,8,4:1

10Biodegradation Behavior of Starch in Simulated White Water System of Old Corrugated Cardboard Pulping Process显示文摘Considering the serious barriers/issues induced by the accumulated starch generated in white water system of old corrugated cardboard(OCC)pulping process,large amounts of accumulated starch in white water would be decomposed by microorganisms and could not be utilized,thereby resulting in severe resource wastage and environmental pollution.This study mainly explored the effects of biodegradation/hydrolysis conditions of the two types of starch substrates(native starch and enzymatically(α-amylase)hydrolyzed starch),which were treated via microorganism degradation within the simulated white water from OCC pulping system and their biodegradation products on the key properties were characterized via X-ray diffraction(XRD),Fourier-transform infrared spectroscopy(FT-IR),and gel permeation chromatography(GPC)technologies.The effects of system temperature,pH value,starch concentration,and biodegradation time on starch biodegradation ratio and the characteristics of obtained biodegradated products from the two types of starches were studied.In addition,the effect ofα-amylase dosage on the biodegradation ratio of enzymatically hydrolyzed starch and its properties was investigated.It was found that the native starch presented a maximal degradation ratio at a system temperature of 55℃and pH value range of 5-7,respectively,the corresponding starch concentration within simulated white water system was 200 mg/L.Whereas the enzymatically hydrolyzed starch exhibited a highest degradation ratio at a system temperature of 50℃and pH value of 5.5,respectively,and the corresponding starch concentration within simulated white water system was 100 mg/L.It was verified that native starch is more readily bio-hydrolyzed and biodegradation-susceptive by microorganisms in simulated white water system of OCC pulping process,while the enzymatically hydrolyzed starch exhibits better biodegradation/hydrolysis resistance to the microbial degradation than that of native starch.This study provides a practical and interesting approach to investigate the starch hydrolysis or biodegradation behaviors in white water system of OCC pulping process,which would greatly contribute to the full recycling and valorized application of starch as a versatile additive during paperboard production.Jinhuan Guo Liqin Liu Xiaohong Zhang Zhengbai Cheng Shuangxi Nie Haibing Cao Na Li Md Manik Mian Xingye An Hongbin Liu 2023Paper And Biomaterials2023,8,1:0
11Design of B/N Co-doped micro/meso porous carbon electrodes from CNF/BNNS/ZIF-8 nanocomposites for advanced supercapacitors显示文摘Boron(B)and nitrogen(N)co-doped 3D hierarchical micro/meso porous carbon(BNPC)were successfully fabricated from cellulose nanofiber(CNF)/boron nitride nanosheets(BNNS)/zinc-methylimidazolate framework-8(ZIF-8)nanocomposites prepared by 2D BNNS,ZIF-8 nanoparticles,and wheat straw based CNFs.Herein,CNF/ZIF-8 acts as versatile skeleton and imparts partial N dopant into porous carbon structure,while the introduced BNNS can help strengthen the hierarchical porous superstructure and endow abundant B/N co-dopants within BNPC matrix.The obtained BNPC electrode possesses a high specific surface area of 505.4 m2/g,high B/N co-doping content,and desirable hydrophilicity.Supercapacitors assembled with BNPC-2(B/N co-doped porous carbon with a CNF/BNNS mass ratio of 1꞉2)electrodes exhibited exceptional electrochemical performance,demonstrating high capacitance stability even after 5000 charge-discharge cycles.The devices exhibited outstanding energy density and power density,as well as the highest specific capacitance of 433.4 F/g at 1.0 A/g,when compared with other similar reports.This study proposes a facile and sustainable strategy for efficiently fabrication of rich B/N co-doped hierarchical micro/meso porous carbon electrodes from agricultural waste biomass for advanced supercapacitor performance.Zhen Shang Xingye An Shuangxi Nie Na Li Haibing Cao Zhengbai Cheng Hongbin Liu Yonghao Ni Liqin Liu 2023Journal of Bioresources and Bioproducts2023,8,3:0

15Bug localization based on syntactical and semantic information of source code显示文摘The existing software bug localization models treat the source file as natural language, which leads to the loss of syntactical and structure information of the source file. A bug localization model based on syntactical and semantic information of source code is proposed. Firstly, abstract syntax tree(AST) is divided based on node category to obtain statement sequence. The statement tree is encoded into vectors to capture lexical and syntactical knowledge at the statement level.Secondly, the source code is transformed into vector representation by the sequence naturalness of the statement. Therefore,the problem of gradient vanishing and explosion caused by a large AST size is obviated when using AST to the represent source code. Finally, the correlation between bug reports and source files are comprehensively analyzed from three aspects of syntax, semantics and text to locate the buggy code. 16Bottlenecks and recent advancements in detecting Mycobacterium tuberculosis in patients with HIV显示文摘Tuberculosis(TB)remains a leading cause of deaths among patients with acquired immunodeficiency syndrome patients.Early diagnosis of TB is essential for administering timely anti‐TB therapy and improving health out-comes,particularly in the people living with HIV.However,conventional techniques used to detect Mycobacterium tuberculosis have significant draw-backs:for example,sputum smear microscopy has low sensitivity,and liquid culture is time‐consuming in patients with HIV‐TB co‐infection due to low sputum production.In addition,while immunological‐based methods involving tuberculin skin testing and interferon gamma release assays are commonly used for auxiliary TB diagnosis,they are often inaccurate in immunodeficient patients.Molecular techniques such as line probe assays,Xpert MTB,and lipoarabinomannan assay are recommended for early diagnosis by World Health Organization.However,no single technique is sufficicent for diagnosing HIV/TB co‐infection,suggesting that multiple diagnostic tests should be used to detect TB.Here,we summarize the drawbacks and advantages of existing TB‐diagnostic methods,as well as their applications to diagnosing HIV/TB co‐infection.We describe newly emerging technologies such as whole genome sequencing and mass spectrometry,with the aim of providing updated guidelines and alternative strategies for TB diagnosis,and particularly HIV/TB diagnosis.Zixun Lin Liqin Sun Cheng Wang Fuxiang Wang Jun Wang Qian Li Hongzhou Lu 2023iLABMED2023,1,1:0
