5Durability of neutralizing antibodies and T-cell response post SARS-CoV-2 infection显示文摘The ongoing pandemic of coronavirus disease 19(COVID-19)is caused by a newly discoveredβcoronavirus named severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).How long the adaptive immunity triggered by SARS-CoV-2 can last is of critical clinical relevance in assessing the probability of second infection and efficacy of vaccination.Here we examined,using ELISA,the IgG antibodies in serum specimens collected from 17 COVID-19 patients at 6–7 months after diagnosis and the results were compared to those from cases investigated 2 weeks to 2 months post-infection.All samples were positive for IgGs against the S-and N-proteins of SARS-CoV-2.Notably,14 samples available at 6–7 months post-infection all showed significant neutralizing activities in a pseudovirus assay,with no difference in blocking the cell-entry of the 614D and 614G variants of SARS-CoV-2.Furthermore,in 10 blood samples from cases at 6–7 months post-infection used for memory T-cell tests,we found that interferonγ-producing CD4+and CD8+cells were increased upon SARS-CoV-2 antigen stimulation.Together,these results indicate that durable anti-SARS-CoV-2 immunity is common in convalescent population,and vaccines developed from 614D variant may offer protection from the currently predominant 614D variant of SARS-CoV-2.Yun Tan Feng Liu Xiaoguang Xu Yun Ling Weijin Huang Zhaoqin Zhu Mingquan Guo Yixiao Lin Ziyu Fu Dongguo Liang Tengfei Zhang Jian Fan Miao Xu Hongzhou Lu Saijuan Chen 2020Frontiers of Medicine2020,14,6:6
6Dent Resistance for Automobile Body Panels显示文摘Dent resistance of automobile body panels is an important property for automobile design and manufacture, but the study on this aspect is not still profound. This study is to summarize the testing methods and physical significations of static and dynamic dent resistance of automobile body panels combined with the author's study, and to analyze the dent behaviors in the round. Several influence factors on dent resistance are expatiated including the mechanical properties of materials, stress states after forming, bake hardening ability, modulus, methods of testing, and structure of specimens and so on. The automotive lightweight and application of high strength steel sheets and aluminum alloys sheets are analyzed, and the significance of testing of dent resistance, especially for dynamic dent resistance of auto-panels, and the finite element simulation analysis are emphasized. To explain the physical phenomenon of dent behaviors, the latest and concerned study results are also discussed. According to this study, a dent resistance test and evaluation standard of Society of Automotive Engineers of China for automotive body panel is presented and is being carried out, and an industry conference is hold to discuss the working-out of the standard, a primary schedule of this standard is confirmed now. 7Distinct immune signatures discriminate between asymptomatic and presymptomatic SARS-CoV-2^(pos)subjects显示文摘Increasing numbers of SARS-CoV-2-positive(SARS-CoV-2^(pos))subjects are detected at silent SARS-CoV-2 infection stage(SSIS).Yet,SSIS represents a poorly examined time-window wherein unknown immunity patterns may contribute to the fate determination towards persistently asymptomatic or overt disease.Here,we retrieved blood samples from 19 asymptomatic and 12 presymptomatic SARS-COV-2^(pos)subjects,47 age/gender-matched patients with mild or moderate COVID-19 and 27 normal subjects,and interrogated them with combined assays of 44-plex CyTOF,RNA-seq and Olink.Notably,both asymptomatic and presymptomatic subjects exhibited numerous readily detectable immunological alterations,while certain parameters including more severely decreased frequencies of CD107a^(low)classical monocytes,intermediate monocytes,non-classical monocytes and CD62L^(hi)CD8^(+)Tnaive cells,reduced plasma STC1 level but an increased frequency of CD4^(+)NKT cells combined to distinguish the latter.Intercorrelation analyses revealed a particular presymptomatic immunotype mainly manifesting as monocytic overactivation and differentiation blockage,a likely lymphocyte exhaustion and immunosuppression,yielding mechanistic insights into SSIS fate determination,which could potentially improve 5ARS-CoV-2 management.Shanhe Yu Caixia Di Shijun Chen Mingquan Guo Jiayang Yan Zhaoqin Zhu Li Liu Ruixue Feng Yinyin Xie Ruihong Zhang Juan Chen Mengxi Wang Dong Wei Hai Fang Tong Yin Jinyan Huang Saijuan Chen Hongzhou Lu Jiang Zhu Jieming Qu 2021Cell Research2021,31,11:4
8Discovery process, clinical characteristics, and treatment of patients infected with avian influenza virus (H7N9) in Shanghai显示文摘During the spring of 2013,a novel avian-origin influenza A (H7N9) virus emerged and spread in Shanghai.On May 9,2013,33 lab-confirmed cases (2 children and 31 adults) have been reported,among whom 13 died.Among the 31 adult cases of H7N9 avian influenza A virus infections in humans,80.6% (25/31) were older men (average age 65 years).Clear history of poultry exposure was found in eight patients (8/33,24.3%) and the remaining 21 cases had been suspected poultry exposure.Clustered cases were found in two families (two cases from a father and his son,and the other two cases from a wife and her husband).However,no evidence of human-to-human transmission had been identified.Avian influenza is one of the most dangerous contagions in poultry worldwide,and avian influenza A virus is the major pathogen responsible.Transmission from poultry to humans has now been proven.Some gene segments of the strain have mutated,and further mutations might result in human-to-human transmission.Sun Yang Shen Yinzhong Lu Hongzhou 2014Chinese Medical Journal2014,,1:4
9Morphology observation for surface orange peel and fracture in tension sample of aluminum-alloy sheet and characterization of nano hardness显示文摘The tension property of aluminum-alloy sheet with different microstructures is measured,and the surface and tension fracture morphology of tension sample with and without orange peel are observed by using scanning electron microscope(SEM).Surface roughness and nano hardness of tension sample are measured.The results show that the average elongation of the samples with orange peel is lower than that without orange peel;especially the r value of perpendicular to the rolling direction is much lower than that without orange peel.The tension surface of the orange peel samples is very rough;various parameters of surface roughness are higher.Under the observation of SEM,a wider sliding band with a micro crack on the surface of orange peel sample can be found.The various parameters of surface roughness without orange peel sample are near to zero,the sliding band is narrow and without micro cracks.The dimple width in tensile fracture of orange peel sample is larger than that without orange peel sample,but shear lip is narrower.The nano hardness testing results show that samples with orange peel behave high elastic modulus,high hardness,and high maximum load,but low plastic deformation depth.These mentioned features can completely describe surface and fracture morphology of tension samples with orange peel.Ma Mingtu Mei Huasheng Lu Hongzhou Yang Hongya Wu Emei Zhou Mingbo 2012Engineering Sciences2012,10,6:4

12Evaluation of Epstein-Barr Virus Salivary Shedding in HIV/AIDS Patients and HAART Use: A Retrospective Cohort Study显示文摘Little data is available on the evaluation of the occurrence rates of Epstein-Barr virus(EBV) in saliva and relationship with highly active antiretroviral therapy(HAART) use in HIV/AIDS patients in China. We conducted a retrospective cohort study of EBV serological tests for HIV/AIDS patients who were treated in the hospitals for infectious diseases in Wuxi and Shanghai, China from May 2016 to April 2017. The EBV-seropositive samples were identified by ELISA. EBV-specific primers and probes were used for the quantitative detection of viral DNA from saliva via quantitative real-time polymerase chain reaction. CD4 cell counts of the HIV/AIDS patients were detected by a flow cytometry. A total of 372 HIV/AIDS patients were ultimately selected and categorized for this retrospective cohort study. For EBV IgG and IgM, the HIV/AIDS HAART use(H) and non-HAART use(NH) groups had significantly higher seropositive rates than the HIV-negative control group. The HIV/AIDS(NH) group had the highest seropositive rate(IgG, 94.27%; IgM, 68.98%) and the highest incidence of EBV reactivation or infection. For salivary EBV DNA-positive rates and quantities, the HIV/AIDS(H)(73.69%) and the HIV/AIDS(NH)(100%) groups showed significantly higher values than the HIV-negative control group(35.79%,[ twofold). Further, the salivary EBV DNA-negative population had significantly higher CD4 cell counts than the EBV DNA-positive population in the HIV/AIDS(H) group and the HIV/AIDS(NH) groups. 13Satb2 expression in Foxci-promoted osteogenic differentiation of MC3T3-E1 cells is negatively regulated by microRNA-103-3p显示文摘The forkhead tran scripti on factor C1 (Foxci) is a cell-fate-determining factor that controls cranial bone development and osteogenic differentiation. Previously, it was demonstrated that various microRNAs (miRNAs) play important roles in osteogenesis and regulate the complex process of osteogenic differentiation. However, it remains unclear how miRNA expression changes during Foxci-promoted osteogenic differentiation. In this study, we successfully overexpressed the Foxci gene in MC3T3-E1 cells and investigated the alterations in the miRNA expression profile on day 3 after osteogenic induction by using a miRNA microarray. Nine downregulated miRNAs and eight upregulated miRNAs were found to be differentially expressed. Among these miRNAs, miR-103-3p was consistently downregulated in the Foxc7-overexpressing MC3T3-E1 cells and was identified as a negative regulator of osteogenic differentiation by using a gain-and Iose-of-funotion assay. The special AT-rich sequence-binding protein 2 (Satb2), a pivotal osteogenic transcription factor, was identified as the miR-103-3p targeting gene and was verified by real-time polymerase chain reaction, western blot analysis, and luciferase assay. Overexpression of miR-103-3p markedly inhibited the expression of Satb2 and attenuated Foxci-promoted osteogenic differentiation. 15Hymecromone:a clinical prescription hyaluronan inhibitor for efficiently blocking COVID-19 progression显示文摘Currently,there is no effective drugs for treating clinically COVID-19 except dexamethasone.We previously revealed that human identical sequences of SARS-CoV-2 promote the COVID-19 progression by upregulating hyaluronic acid(HA).As the inhibitor of HA synthesis,hymecromone is an approved prescription drug used for treating biliary spasm.Here,we aimed to investigate the relation between HA and COVID-19,and evaluate the therapeutic effects of hymecromone on COVID-19.Firstly,HA was closely relevant to clinical parameters,including lymphocytes(n=158;r=−0.50;P<0.0001),C-reactive protein(n=156;r=0.55;P<0.0001),D-dimer(n=154;r=0.38;P<0.0001),and fibrinogen(n=152;r=0.37;P<0.0001),as well as the mass(n=78;r=0.43;P<0.0001)and volume(n=78;r=0.41;P=0.0002)of ground-glass opacity,the mass(n=78;r=0.48;P<0.0001)and volume(n=78;r=0.47;P<0.0001)of consolidation in patient with low level of hyaluronan(HA<48.43 ng/mL).Furthermore,hyaluronan could directly cause mouse pulmonary lesions.Besides,hymecromone remarkably reduced HA via downregulating HAS2/HAS3 expression.Moreover,89%patients with hymecromone treatment had pulmonary lesion absorption while only 42%patients in control group had pulmonary lesion absorption(P<0.0001).In addition,lymphocytes recovered more quickly in hymecromone-treated patients(n=8)than control group(n=5)(P<0.05).These findings suggest that hymecromone is a promising drug for COVID-19 and deserves our further efforts to determine its effect in a larger cohort.Shuai Yang Yun Ling Fang Zhao Wei Li Zhigang Song Lu Wang Qiuting Li Mengxing Liu Ying Tong Lu Chen Daoping Ru Tongsheng Zhang Kaicheng Zhou Baolong Zhang Peng Xu Zhicong Yang Wenxuan Li Yuanlin Song Jianqing Xu Tongyu Zhu Fei Shan Wenqiang Yu Hongzhou Lu 2022Signal Transduction and Targeted Therapy2022,7,4:2

18The 4Rs approach to COVID-19 emergency management during the post-pandemic period:What lessons can be learned from Shenzhen,China?显示文摘The Omicron variant of SARS-CoV-2(or Omicron)is extremely contagious and has swept the world in a short period.Shenzhen—a new international city in China with a permanent population of 17.56 million and covering nearly 2000 square kilometers—was attacked by Omicron in 2022.However,the pandemic was controlled in a very short time as a result of prompt government reaction that prevented the variant’s further spread.The total number of cases in this wave of the pandemic was more than 2600.Shenzhen’s successful experience in tackling the Omicron wave deserves in-depth discussion.Proposed by American scholar Robert Heath,the 4Rs model aims to reduce the harm from a crisis through the measures of reduction,readiness,response,and recovery.This article presents the successful experience of Shenzhen’s local government and the Third People’s Hospital of Shenzhen,the only hospital in Shenzhen designated to use the 4Rs emergency management model for the treatment of COVID-19 during the 2022 Omicron wave of the pandemic.Rongfeng Zhou Hongzhou Lu 2022Infectious Medicine2022,1,4:1
