| 1 | Ganoderic acid alleviates chemotherapy-induced fatigue in mice bearing colon tumor显示文摘Chemotherapy-related fatigue(CRF)is increasingly being recognized as one of the severe symptoms in patients undergoing chemotherapy,which not only largely reduces the quality of life in patients,but also diminishes their physical and social function.At present,there is no effective drug for preventing and treating CRF.Ganoderic acid(GA),isolated from traditional Chinese medicine Ganoderma lucidum,has shown a variety of pharmacological activities such as anti-tumor,anti-inflammation,immunoregulation,etc.In this study,we investigated whether GA possessed anti-fatigue activity against CRF.CT26 tumor-bearing mice were treated with 5-fluorouracil(5-FU,30 mg/kg)and GA(50 mg/kg)alone or in combination for 18 days.Peripheral and central fatigue-related behaviors,energy metabolism and inflammatory factors were assessed.We demonstrated that co-administration of GA ameliorated 5-FU-induced peripheral muscle fatigue-like behavior via improving muscle quality and mitochondria function,increasing glycogen content and ATP production,reducing lactic acid content and LDH activity,and inhibiting p-AMPK,IL-6 and TNF-αexpression in skeletal muscle.Co-administration of GA also retarded the 5-FU-induced central fatigue-like behavior accompanied by down-regulating the expression of IL-6,iNOS and COX2 in the hippocampus through inhibiting TLR4/Myd88/NF-κB pathway.These results suggest that GA could attenuate 5-FU-induced peripheral and central fatigue in tumor-bearing mice,which provides evidence for GA as a potential drug for treatment of CRF in clinic. | Abudumijiti Abulizi Ling Hu Ang Ma Fang-yu Shao Hui-ze Zhu Si-mei Lin Guang-ying Shao Yue Xu Jian-hua Ran Jing Li Hong Zhou Dong-mei Lin Lian-fu Wang Min Li Bao-xue Yang | 2021 | Acta Pharmacologica Sinica2021,42,10: | 5 |
| 2 | Clinical significance of HBME-1,Galectin-3,and CK19 expression and the status of BRAF mutation in papillary thyroid carcinoma显示文摘Objective The aim of this study was to explore the clinical significance of the expression of proteins human bone marrow endothelial cell markers(HBME-1), Galectin-3, and cytokeratin19(CK19), as well as the status of v-raf murine sarcoma viral oncogene homolog B1(BRAF) mutation in papillary thyroid carcinoma(PTC). Methods Immunohistochemical staining was performed in 82 specimens each of PTC and papillary benign lesions to detect the expression of HBME-1, Galectin-3, and CK19. Polymerase chain reaction(PCR) and gene sequencing were performed on 60 specimens each of PTC and papillary benign lesions to detect the status of BRAF mutation. Results The positive expression ratios of HBME-1, Galectin-3, and CK19 in PTC were 98.8%, 97.6% and 100% respectively, which were significantly higher than the expressions in papillary benign lesions(P < 0.05). No significant relationship was observed between the expression of these makers and the clinicopathological features of PTC. The sensitivity of co-expression of HBME-1 and CK19 or HBME-1 and Galectin-3 as diagnostic criteria of PTC was 99.9%, with a specificity of 95.4%. BRAF mutation was detected in 40 of 60 PTC(66.7%) specimens. There was a statistical difference in BRAF mutations between PTC and papillary benign lesions(P < 0.05); there were no associations between BRAF mutation and the clinicopathological features of PTC. Conclusion Combined immunohistochemical staining of HBME-1, Galectin-3, and CK19 can further improve the sensitivity and specificity of differential diagnosis of PTC. | 20 | Submerged fermentation improves bioactivity of mulberry fruits and leaves显示文摘Objective:Mulberry(Morus spp.)fruits and leaves have been proven to possess nutraceutical properties.Due to its fast and easy growing characteristics,mulberry fruits(MF)and leaves(ML)potentially emerge as a great source of functional foods.This study aims to enhance bioactivities(antioxidant,antiinflammation,and hypoglycemic activity)of MF and ML via submerged fermentation using bacteria(Lactobacillus plantarum TAR 4),yeast(Baker’s yeast and red yeast)and fungi(Tempeh and Tapai starter).Methods:In this study,25%(mass to volume ratio)of MF and ML were fermented(48 h)with 1%(mass to volume ratio)of different microbial cultures,respectively.Effects of different fermentations on MF and ML were determined based on the changes of total phenolics(TPC),flavonoids(TFC),anthocyanins,total sugar,DPPH activity,ferric reducing antioxidant power(FRAP),albumin denaturation inhibition activity(ADI),anti-lipoxygenase activity and a-amylase inhibition activity(AI).Results:Generally,ML had higher AI than MF.However,MF exhibited higher DPPH,FRAP and antilipoxygenase activity than ML.After all forms of fermentation,DPPH and AI activity of MF and ML were increased significantly(P<0.05).However,the effects of fermentation on TPC,FRAP,ADI and antilipoxygenase activity of MF were in contrast with ML.TPC,FRAP and anti-lipoxygenase activity of ML were enhanced,but reduced in MF after fermentation.Although the effects exerted by different microorganisms in MF and ML fermentation were different,the bioactivities of MF and ML were generally improved after fermentation.Fermentation by Tempeh starter enhanced TPC(by 2-fold),FRAP(by 2.3-fold),AI(at 10%increment)and anti-lipoxygenase activity(by 5-fold)of ML,whereas Tapai fermentation effectively enhanced the DPPH(at 17%increment)and ADI(by 2-fold)activity of MF.Conclusion:Findings of this study provide an insight into the future process design of MF and ML processing into novel functional foods. | Hui Qian Chuah Pei Ling Tang Ni Jing Ang Hui Yin Tan | 2021 | Chinese Herbal Medicines2021,13,4: | 0 |
