| 1 | Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021，41，11： | 29 |

| 7 | Significance and expression of Bax, Survivin and p53 in gastric carcinoma and precancerous lesions using tissue microarray显示文摘Objective: To explore the relationship between expressions of apoptosis-related protein Bax, Survivin and p53 and the molecular mechanisms of carcinogenesis and progression of gastric carcinoma. Methods: Tissue microarray and immunohistochemistry were used in this study. Results: The positive rate of Bax protein in gastric cancer (17.7%,17/96) was significantly lower than those in adjacent normal mucosa (51%), intestinal metaplasia (69.2%) and dysplasia (75%), P < 0.01. The positive rate of Survivin expression in gastric cancer (80.6%, 89/98) was significantly higher than that in adjacent normal mucosa (3.9%), P < 0.01. The positive rates of Survivin expression in tumors with different organ metastases (in lymph node metastasis 86.2%, liver 100% and ovarian 100%) were statistically higher than in tumors without metastasis (64.3%), P < 0.05. Bax expression was correlated with Survivin but not with mp53 that was closely related to Survivin expression (P < 0.05) in gastric cancer. Conclusion: The abnormal expressions of Bax, Survivin and mp53 were correlated with the tumorigenesis and progression of gastric carcinoma. | 11 | Three-dimensional properties of mesoscale cyclonic warm-core and anticyclonic cold-core eddies in the South China Sea显示文摘In general,a mesoscale cyclonic(anticyclonic)eddy has a colder(warmer)core,and it is considered as a cold(warm)eddy.However,recently research found that there are a number of'abnormal'mesoscale cyclonic(anticyclonic)eddies associated with warm(cold)cores in the South China Sea(SCS).These'abnormal'eddies pose a challenge to previous works on eddy detection,characteristic analysis,eddy-induced heat and salt transports,and even on mesoscale eddy dynamics.Based on a 9-year(2000–2008)numerical modelling data,the cyclonic warm-core eddies(CWEs)and anticyclonic cold-core eddies(ACEs)in the SCS are analyzed.This study found that the highest incidence area of the'abnormal'eddies is the northwest of Luzon Strait.In terms of the eddy snapshot counting method,8620 CWEs and 9879 ACEs are detected,accounting for 14.6%and 15.8%of the total eddy number,respectively.The size of the'abnormal'eddies is usually smaller than that of the'normal'eddies,with the radius only around 50 km.In the generation time aspect,they usually appear within the 0.1–0.3 interval in the normalized eddy lifespan.The survival time of CWEs(ACEs)occupies 16.3%(17.1%)of the total eddy lifespan.Based on two case studies,the intrusion of Kuroshio warm water is considered as a key mechanism for the generation of these'abnormal'eddies near the northeastern SCS. | Wenjin Sun Yu Liu Gengxin Chen Wei Tan Xiayan Lin Yuping Guan Changming Dong | 2021 | Acta Oceanologica Sinica2021，40，10： | 3 |

| 13 | Integration of full-length transcriptomics and targeted metabolomics to identify benzylisoquinoline alkaloid biosynthetic genes in Corydalis yanhusuo显示文摘Corydalis yanhusuo W.T.Wang is a classic herb that is frequently used in traditional Chinese medicine and is efficacious in promoting blood circulation,enhancing energy,and relieving pain.Benzylisoquinoline alkaloids(BIAs)are the main bioactive ingredients in Corydalis yanhusuo.However,few studies have investigated the BIA biosynthetic pathway in C.yanhusuo,and the biosynthetic pathway of species-specific chemicals such as tetrahydropalmatine remains unclear.We performed full-length transcriptomic and metabolomic analyses to identify candidate genes that might be involved in BIA biosynthesis and identified a total of 101 full-length transcripts and 19 metabolites involved in the BIA biosynthetic pathway.Moreover,the contents of 19 representative BIAs in C.yanhusuo were quantified by classical targeted metabolomic approaches.Their accumulation in the tuber was consistent with the expression patterns of identified BIA biosynthetic genes in tubers and leaves,which reinforces the validity and reliability of the analyses.Full-length genes with similar expression or enrichment patterns were identified,and a complete BIA biosynthesis pathway in C.yanhusuo was constructed according to these findings.Phylogenetic analysis revealed a total of ten enzymes that may possess columbamine-O-methyltransferase activity,which is the final step for tetrahydropalmatine synthesis.Our results span the whole BIA biosynthetic pathway in C.yanhusuo.Our full-length transcriptomic data will enable further molecular cloning of enzymes and activity validation studies. | Dingqiao Xu Hanfeng Lin Yuping Tang Lu Huang Jian Xu Sihui Nian Yucheng Zhao | 2021 | Horticulture Research2021，8，1： | 2 |

| 15 | Carboniferous integrative stratigraphy and timescale of China显示文摘The Carboniferous period lasted about 60 Myr, from ~358.9 Ma to ~298.9 Ma. According to the International Commission on Stratigraphy, the Carboniferous System is subdivided into two subsystems, i.e., Mississippian and Pennsylvanian, including 6 series and 7 stages. The Global Stratotype Sections and Points(GSSPs) of three stages have been ratified, the Tournaisian, Visean, and Bashkirian stages. The GSSPs of the remaining four stages(i.e., the Serpukhovian, Moscovian,Kasimovian, and Gzhelian) have not been ratified so far. This paper outlines Carboniferous stratigraphic subdivision and correlation on the basis of detailed biostratigraphy mainly from South China, and summarizes the Carboniferous chronostratigraphic framework of China. High-resolution biostratigraphic study reveals 37 conodont zones, 24 foraminiferal(including fusulinid) zones, 13 ammonoid zones, 10 brachiopod zones, and 10 rugose coral zones in the Carboniferous of China. The biostratigraphic framework based on these biozones warrants the precise correlation of regional stratigraphy of China(including2 subsystems, 4 series, and 8 stages) to that of the other regions globally. Meanwhile, the Carboniferous chemo-, sequence-,cyclo-, and event-stratigraphy of China have been intensively studied and can also be correlated worldwide.
