1FABP4 secreted by M1-polarized macrophages promotes synovitis and angiogenesis to exacerbate rheumatoid arthritis显示文摘Increasing evidence shows that adipokines play a vital role in the development of rheumatoid arthritis(RA).Fatty acid-binding protein 4(FABP4),a novel adipokine that regulates inflammation and angiogenesis,has been extensively studied in a variety of organs and diseases.However,the effect of FABP4 on RA remains unclear.Here,we found that FABP4 expression was upregulated in synovial M1-polarized macrophages in RA.The increase in FABP4 promoted synovitis,angiogenesis,and cartilage degradation to exacerbate RA progression in vivo and in vitro,whereas BMS309403(a FABP4 inhibitor)and anagliptin(dipeptidyl peptidase 4 inhibitor)inhibited FABP4 expression in serum and synovial M1-polarized macrophages in mice to alleviate RA progression.Further studies showed that constitutive activation of mammalian target of rapamycin complex 1(mTORC1)by TSC1 deletion specifically in the myeloid lineage regulated FABP4 expression in macrophages to exacerbate RA progression in mice.In contrast,inhibition of mTORC1 by ras homolog enriched in brain(Rheb1)disruption specifically in the myeloid lineage reduced FABP4 expression in macrophages to attenuate RA development in mice.Our findings established an essential role of FABP4 that is secreted by M1-polarized macrophages in synovitis,angiogenesis,and cartilage degradation in RA.BMS309403 and anagliptin inhibited FABP4 expression in synovial M1-polarized macrophages to alleviate RA development.Hence,FABP4 may represent a potential target for RA therapy.Dong Guo Chuangxin Lin Yuheng Lu Hong Guan Weizhong Qi Hongbo Zhang Yan Shao Chun Zeng Rongkai Zhang Haiyan Zhang Xiaochun Bai Daozhang Cai 2022Bone Research2022,10,4:5

7Cerium and carbon-sulfur codoped mesoporous TiO_(2)nanocomposites for boosting visible light photocatalytic activity显示文摘Ce and C-S codoped mesoporous TiO_(2)nanocomposites were synthesized via a sol-gel method integrated with an evaporation-induced self-assembly approach.The basic physicochemical characteristics of the synthetic samples were analyzed via a series of characterization techniques.The results reveal that C-S and Ce codoping on mesoporous TiO_(2)enhances the photocatalytic activity owing to the synergistic effect caused by narrowing the band gap,enhancing adsorption,trapping and transferring the excited e^(-)/h^(+)pairs and suppressing the recombination of e^(-)/h^(+)pairs.Furthermore,the obtained C,S-TiO_(2)/CeO_(2)materials exhibit large specific surface areas and numerous pores which not only effectively improve the adsorption-enrichment capability,but also supply multi-dimensional mass and electron transfer channels.The photodegradation efficiency of RhB by C,S-TiO_(2)/CeO_(2)within 40 min is nearly 100%,and its degradation efficiency is 6.63 times that of undoped TiO_(2).Recycling experiments show that mesoporous C,S-TiO_(2)/CeO_(2)shows excellent recoverability and stability.Furthermore,by trapping experiments,·O_(2)e^(-)/h^(+)and·OH are the predominant active species and a possible reaction mechanism is proposed.Fei Zheng Faqin Dong Lin Zhou Jieyu Yu Xijie Luo Xingyu Zhang Zhenzhen Lv Luman Jiang Yuheng Chen Mengqing Liu 2023Journal of Rare Earths2023,41,4:1

10Construction of Tobacco Bax Inhibitor-1 ihpRNA Gene Silencing Vector and Transformation into Agrobacterium tumefaciens EHA105显示文摘The plasmid pGSA1285 was first modified by substituting its GUS sequence with the Chalcone synthase intron fragment from vector pFGC5941 to get the plant silencing expression vector that contained Kanamycin resistance site and was named as pGSA2285. Using PCR-based amplification, two different restriction sites at both ends of tobacco Bax inhibitor-1 (NtBI-1) gene were created, respectively, which made the construction of ihpRNA gene silencing vector more efficiently. Then, NtBI-1 genes were inserted into Multiple Cloning Site (MCS) of pGSA2285 respectively to form Bax inhibitor-1 ihpRNA gene silencing vector, named as pGSA4285, containing sense and anti-sense BI-1 sequence which was spliced by chalcone synthase intron. 11Develop potential multi-target drugs by self-assembly of quercetin with amino acids and metal ion to achieve significant efficacy in anti-Alzheimer’s disease显示文摘Exploring multi-target drugs(MTDs)is a prevalent method against Alzheimer’s disease(AD).However,the current developed MTDs based on the“framework combination”technique or using flavonoids have not realized better healing effects owing to a complex metabolic process and low bioavailability.Here,we propose an alternative strategy to develop self-assembled nanoparticles(NPs)as MTDs using 9-fluorenylmethyloxycarbonyl-tryptophan(Fmoc-Trp),quercetin(Que),and Fe^(2+)as building blocks through coordination and electrostatic interaction to generate Fmoc-Trp-Fe^(2+)-Que(FTFQ)NPs.Whether in cell or in vivo,FTFQ NPs exhibited considerable biocompatibility attributing to the inherent biological affinity of assembly units.By combining the advantages of assemble approach and nanostructures,the obtained FTFQ NPs greatly enhanced the bioavailability of Que and displayed synergistic therapeutic effects through reducing Aβaggregation in direct/indirect means and eliminating the toxicity induced by ROS and Aβoligomer/fibrils.Furthermore,FTFQ NPs could obviously restore the AD zebrafish’s mobility and stimulus response ability.More importantly,developing self-assembly NPs as MTDs could be an efficient and general method to promote other novel MTDs by extending the assemble units to other drugs,peptides,and metal ions.Based on above benefits,these self-assembled NPs could be as potential MTDs and show great promising application in AD treatment.Chunling Zhu Yuheng Yang Xiaowen Li Xingyu Chen Xucong Lin Xiaoping Wu 2022Nano Research2022,15,6:0

14Visualization of Ferroelectric Domains in Thin Films of Molecular Materials Using Confocal Micro-Raman Spectroscopy显示文摘Ferroelectrics are an important class of functional materials.Among all their unique properties,the study of their ferroelectric domains and domain walls is of great interest due to their importance in ferroelectric applications.There are many methods to characterize ferroelectric domains,namely,scanning probe microscopy,optical microscopy,electron microscopy,etc.Currently,newly emerged molecular ferroelectrics are attracting much attention from chemists,physicists and researchers in material sciences due to their structural flexibility,light mass,simple fabrication,etc.However,for the characterization of molecular ferroelectric domains,most conventional methods require either a complicated preparation process or direct contact between physical probes and material surfaces,limiting the development of molecular ferroelectric materials.In this report,we have demonstrated that confocal micro-Raman spectroscopy,as a nondestructive and noncontact in-situ method,is very suitable for studying the ferroelectric polarization and structures of domains in molecular ferroelectrics.Taking recently reported molecular ferroelectric trimethylchloromethyl ammonium trichlorocadmium(II)(TMCM-CdCl_(3))as an example,the non-180°domains have been characterized and visualized at different temperatures.Such a simple and extendable method requires minimum sample preparation,which would further benefit the research of molecular ferroelectric domain engineering and promote the miniaturization and integration of molecular ferroelectric films.ZHOU Wenqin FENG Zijie XIONG Yuan DU Guowei LIN Xiumei SU Qidong LOU Yuheng AN Shili YOU Yumeng 2022Chemical Research in Chinese Universities2022,38,6:0

16Combining Multiple Factors of LightGBM and XGBoost Algorithms to Predict the Morbidity of Double-High Disease显示文摘In recent years, cardiovascular and cerebrovascular diseases have seriously jeopardized people’s health. Double-high (hyperlipidemia and hypertension) is one of the main causes of cardiovascular and cerebrovascular diseases in clinical practice. To diagnose cardiovascular and cerebrovascular diseases early, a reliable prediction system should be developed to assist doctors. In this paper, the different elements and evaluate the weight of these elements on double-high diseases are analyzed by machine learning method. The LightGBM algorithm and XGBoost algorithm were employed to construct the prediction models, respectively. Significantly, the proposed model was trained by real physical examination data and five meaningful and useful biochemical indicators were selected to encoding the raw physical examination data to numerical vector. The selected features are systolic blood pressure, diastolic blood pressure, serum triglyceride, serum high-density lipoprotein and serum low-density lipoprotein. The mean square error (MSE) after calculating the logarithm of the predicted value and the true value was introduced to assess the prediction model. 17Junction matters in hydraulic circuit bio-design of microfluidics显示文摘Microfluidic channels are at micrometer scales;thus,their fluid flows are laminar,resulting in the linear dependence of pressure drop on flow rate in the length of the channel.The ratio of the pressure drop to flow rate,referred to as resistance,depends on channel size and dynamic viscosity.Usually,a microfluidic chip is analogous to an electric circuit in design,but the design is adjusted to optimize channel size.However,whereas voltage loss is negligible at the nodes of an electric circuit,hydraulic pressure drops at the nodes of microfluidic chips by a magnitude are comparable to the pressure drops in the straight channels.Here,we prove by experiment that one must fully consider the pressure drops at nodes so as to accurately design a precise microfluidic chip.In the process,we numerically calculated the pressure drops at hydraulic nodes and list their resistances in the range of flows as concerned.We resorted to machine learning to fit the calculated results for complex junctions.Finally,we obtained a library of node resistances for common junctions and used them to design three established chips that work for single-cell analysis and for precision allocation of solutes(in gradient and averaging concentration microfluidic networks).Endothelial cells were stimulated by generating concentrations of adriamycin hydrochloride from the last two microfluidic networks,and we analyzed the response of endothelial cells.The results indicate that consideration of junction resistances in design calculation brings experimental results closer to the design values than usual.This approach may therefore contribute to providing a platform for the precise design of organ chips.Yao Lin Dongliang He Zerui Wu Yurou Yao Zhanhao Zhang Yuheng Qiu Shan Wei Guangzhu Shang Xingyue Lei Ping Wu Weiping Ding Liqun He 2023Bio-Design and Manufacturing2023,6,1:0
18Addressing the Chengjiang conundrum: A palaeoecological view on the rarity of hurdiid radiodonts in this most diverse early Cambrian Lagerstatte显示文摘Over one hundred arthropod fossil species have been described from the famous Chengjiang Lagerst?tte(South China, Cambrian Stage 3, ca. 518 Ma) including a diverse assemblage of radiodonts–a group containing Anomalocaris and its relatives. These iconic stem-group euarthropods include some of the largest animals of the time, and some are known from hundreds of specimens. A longstanding conundrum has been the rarity or absence of hurdiids from Cambrian Series 2 Lagerst?tten like Chengjiang. This is because radiodonts are generally common in such deposits and the oldest radiodont ever discovered is a hurdiid. Furthermore, this family displays the widest geographic and temporal ranges of all radiodont families, and the highest diversity. Here we document the first hurdiid frontal appendages from Chengjiang, which display unique features within the family and may provide insights for understanding the character evolution of hurdiid appendages. The palaeoenvironmental distribution of hurdiids suggests that the rarity of hurdiids in Chengjiang may be due to a preference for deeper water environments,and the later success of this family from the Wuliuan onwards may relate to their ability to tolerate cooler water temperatures than other radiodont families.
