1斑马鱼nos2a基因的生长调控作用显示文摘为了在斑马鱼中探索诱导型一氧化氮合酶(Nos2)在生长调控中的作用,通过CRISPR/Cas9基因编辑技术,首次构建了可稳定遗传的nos2a缺失型(nos2a^(-/-))斑马鱼家系。通过监测其生长过程发现,nos2a^(-/-)斑马鱼的整体生长趋势较nos2a正常型(nos2a^(+/+))斑马鱼偏低,且nos2a^(-/-)成鱼的体型显著小于同期的nos2a^(+/+)斑马鱼(P<0.01):nos2a^(+/+)成鱼体长(29.53±0.60)mm,nos2a^(-/-)成鱼体长(26.34±0.69)mm;nos2a^(+/+)成鱼体质量(512.80±30.75)mg,nos2a^(-/-)成鱼体质量(394.40±16.91)mg;而在肌纤维密度(P=0.718)及躯椎骨的横截面积上(P=0.548)两者没有显著差异。qPCR检测发现,出膜8 d时,nos2a^(-/-)斑马鱼的生长激素基因gh1的mRNA水平(P<0.01)及类胰岛素生长因子基因igf1的mRNA水平(P<0.001)均显著低于nos2a^(+/+)斑马鱼;出膜68 d时,nos2a^(-/-)斑马鱼的gh1(P<0.0001)和igf1(P<0.0001)的mRNA水平同样显著低于nos2a^(+/+)斑马鱼。以上结果表明,斑马鱼中nos2a的功能缺失导致鱼体生长受到抑制,而gh1及igf1 mRNA的水平下调是其潜在原因。高磊 黄亚娟 PENGLEE Rachit 易欣鑫 鲍宝龙 2021上海海洋大学学报2021,30,4:4

6Human epidermal growth factor receptor-2 in oesophageal cancers:An observational study显示文摘AIM:To determine the incidence of human epidermal growth factor receptor 2(HER2) over expression in oesophageal cancers.METHODS:A retrospective study,of one hundred consecutive cases of endoscopic histological samples of oesophageal cancers from a single British cancer network were included.Cancer cases were diagnosed between April 2007 and June 2010.HER2 over expression was assessed using immunohistochemistry,those that scored '0' and '+1' were considered 'negative' for HER2;those that scored '+3' were considered 'Positive'.Cases that were scored '+2' on immunohistochemistry further went on to have HER2 gene analysis using the Ventana HER brightfield dual-colourin situ hybridisations(HER B DISH) assay and either came back to be positive or negative for HER2 over expression.Overall survival was measured from date of histological diagnosis until date of death.93% of the cases were followed up till five years or death,and all were followed up till two years.Cases of gastro-oesophageal junctional tumours were excluded.RESULTS:The median age of our sample was 66 years(range:38-91 years).Eighty one were male and 19 female.Ninety-one of the cases were adenocarcinoma of the oesophagus and the rest were cases of squamous cell carcinoma.The anatomical distribution of the tumours was;upper oesophagus 2,middle oesophagus 11,and 87 were in the lower oesophagus.Operative resection was completed in 15 cases;seven cases had attempted surgical resections,i.e.,open and close,33 patients received definitive chemo-radiation and 52 had palliative treatment.Twenty-five of the cancers showed evidence of HER2 over expression,all were adenocarcinomas.Of the 25 cases that showed evidence of HER2 over expression,21(84%) were located in the lower third of the oesophagus.On staging,24 out of the 25 HER2 positive cases were at stage 3 or more(13 at stage 3 and 11 at stage 4),For HER2 negative cases 37 were at stage 3 and 32 were staged as stage 4.Seventeen out of twenty five cases(68%) with HER2 over expression received palliative therapy,in comparison to thirty five out of seventy five(46.7%) in tumours not expressing HER2.No significant difference in overall survival was demonstrated between patients whose cancers showed evidence of HER2 over expression and those who did not;median overall survival for HER2 positive tumours was 15 mo(95%CI,11-19 mo) compared to 13 mo(95%CI,9-17 mo) for HER2 negative ones.Two years cumulative survival for cases with HER2 over expression was 33.7% compared to 31.6% in cases without HER2 over expression(P = 0.576).Only cancer's stage significantly affected overall survival on both univariant and multivariable analysis(P = 0.034 and P = 0.009 respectively).None of the patients included in this study received Trastuzumab.CONCLUSION:Twenty-seven point five percent of oesophageal adenocarcinomas showed evidence of HER2 over expression.Routine testing for human HER2 in oesophageal adenocarcinomas can have significant implication on treatments offered to patients that may potentially affect their prognosis.Hazem Al-Momani Rachel Barnes Ahmed El-Hadi Rachit Shah Wyn G Lewis Paul Edwards 2012World Journal of Gastroenterology2012,18,44:1

9Identifying malicious accounts in blockchains using domain names and associated temporal properties显示文摘The rise in the adoption of blockchain technology has led to increased illegal activities by cybercriminals costing billions of dollars.Many machine learning algorithms are applied to detect such illegal behavior.These algorithms are often trained on the transaction behavior and,in some cases,trained on the vulnerabilities that exist in the system.In our approach,we study the feasibility of using the Domain Name(DN)associated with the account in the blockchain and identify whether an account should be tagged malicious or not.Here,we leverage the temporal aspects attached to the DN.Our approach achieves 89.53%balanced-accuracy in detecting malicious blockchain DNs.While our results identify 73769 blockchain DNs that show malicious behavior at least once,out of these,34171 blockchain DNs show persistent malicious behavior,resulting in 2479 malicious blockchain DNs over time.Nonetheless,none of these identified malicious DNs were reported in new officially tagged malicious blockchain DNs.Rohit Kumar Sachan Rachit Agarwal Sandeep Kumar Shukla 2023Blockchain(Research and Applications)2023,4,3:0
10The role of nitric oxide and neuronal nitric oxide synthase in zebrafish(Danio rerio)shoaling显示文摘Nitric oxide(NO)-the product of arginine metabolism catalyzed by nitric oxide synthases(NOS)-is a well-known neurotransmitter which plays an important role in metabolism and amino acid transportation in the nervous system.In particular,it can inhibit monoamine neurotransmitter transportation which affects animal behavior,especially social behavior.Shoaling-is a one kind of social behavior.It is a behavior that individual fish choose to join with their group within two factors;food and predation risk.Shoaling fish has quickly responded to predator and increased the change in feeding competition.In addition,shoaling also effect to stress response on stock density of aquaculture system.The effect of NO molecular signaling on the dopamine pathway was investigated using zebrafish(Danio rerio)as a model organism.Our aim was to understand the role of NOS and NO in shoaling behavior,which is typical of zebrafish.The concentration of NO in the zebrafish brain was modulated using a knockout for the neuronal NOS gene,and NO production was induced through treatment with L-arginine.The existence of NO in the zebrafish brain was confirmed by using a fluorescent probe.Dopamine concentration in the brain was measured by UPLC tandem mass spectrometer.We measured shoaling cohesion of all individual fish of D.rerio,using average distance between all pairs of fish(nearest neighbor distance)and analyzed tracking by Zebralab ViewPoint software.Collectively,our results suggest that a lower level of NO was associated with a higher level of dopamine,which in turn leads to the shoaling behavior.Rachit Penglee Lei Gao Yajuan Huang Liping Liu Sukkrit Nimitkul Baolong Bao 2021Aquaculture and Fisheries2021,6,6:0
11Study on Phase Change Material in Grooved Bricks for Energy Efficiency of the Buildings显示文摘Phase change materials(PCMs)are an interesting technology due to their high density and isothermal behavior during phase change.Phase change material plays a major role in the energy saving of the buildings,which is greatly aided by the incorporation of phase change material into building products such as bricks,cement,gypsum board,etc.In this study,an experiment has been conducted with three identical small chambers made up of normal,grooved and PCM-treated grooved bricks.Before the inclusion of PCM in grooved bricks,PCM material behavior has been studied by different techniques such as DSC,TG/DTA,SEM,and XRD.Thermal properties and thermal stability were investigated by differential scanning calorimeter(DSC)and thermogravimetric analyzer(TGA)respectively.Scanning electron microscopy(SEM)and X-ray diffraction(XRD)were used to determine the microstructure and crystalloid phase of the PCM before and after the accelerated thermal cycling test(0,60,120).These three identical model rooms built were exposed at a temperature just above 40°C with a heater.When the maximum outdoor temperature was 40-41°C,then the temperature of the PCM-treated grooved chamber was 32-33°C.The PCM-treated wall was tested and compared with a conventional and grooved wall.The difference between the PCM-treated grooved chamber and the untreated one was 8-9°C.PCM-treated bricks provided more efficient internal heat retention in summer when the outside temperature increased.Niharika Pawar Yasmeen Qureshi Rachit Agarwal Srinivasarao Naik B 2023Journal of Architectural Environment & Structural Engineering Research2023,6,2:0
12COVID19: Forecasting Air Quality Index and Particulate Matter (PM2.5)显示文摘Urbanization affects the quality of the air,which has drastically degraded in the past decades.Air quality level is determined by measures of several air pollutant concentrations.To create awareness among people,an automation system that forecasts the quality is needed.The COVID-19 pandemic and the restrictions it has imposed on anthropogenic activities have resulted in a drop in air pollution in various cities in India.The overall air quality index(AQI)at any particular time is given as the maximum band for any pollutant.PM2.5 is a fine particulate matter of a size less than 2.5 micrometers,the inhalation of which causes adverse effects in people suffering from acute respiratory syndrome and other cardiovascular diseases.PM2.5 is a crucial factor in deciding the overall AQI.The proposed forecasting model is designed to predict the annual PM2.5 and AQI.The forecasting models are designed using Seasonal Autoregressive Integrated Moving Average and Facebook’s Prophet Library through optimal hyperparameters for better prediction.An AQI category classification model is also presented using classical machine learning techniques.The experimental results confirm the substantial improvement in air quality and greater reduction in PM2.5 due to the lockdown imposed during the COVID-19 crisis.R.Mangayarkarasi C.Vanmathi Mohammad Zubair Khan Abdulfattah Noorwali Rachit Jain Priyansh Agarwal 2021Computers, Materials & Continua2021,,6:0
13aguA基因缺失对嗜水气单胞菌致病性的影响显示文摘嗜水气单胞菌(Aeromonas hydrophila)是水环境常见的病原体,具有较强致病性。在本研究中,首次敲除了嗜水气单胞菌aguA基因,通过转录组分析发现aguA基因敲除后细菌的黏附、致病、胺代谢等生物过程下降。通过qRT-PCR进一步验证发现10个黏附基因、9个发病基因和8个胺代谢基因的mRNA表达水平明显下降;此外,转录组水平分析结合qRT-PCR验证发现Ⅵ型分泌系统基因、Ⅱ型分泌系统基因、鞭毛合成相关基因、菌毛合成相关基因、溶血基因、Ⅰ型分泌系统基因、肠毒素基因、RTX基因等毒力基因的RNA表达水平也出现下调。与野生菌株(WT)相比,aguA敲除株(ΔaguA)感染金鱼的致病力降低了86.6%,且aguA敲除株生物膜形成能力降低了16.6%。以上结果显示,aguA基因与嗜水气单胞菌的致病性密切相关。赵东康 Rachit PENGLEE 车金远 易欣鑫 邱超达 鲍宝龙 2023上海海洋大学学报2023,32,3:0
14Neoadjuvant therapy for esophageal cancer显示文摘Esophageal cancer is increasing in incidence more than any other visceral malignancy in North America. Adenocarcinoma has become the most common cell type. Surgery remains the primary treatment modality for locoregional disease. Overall survival with surgery alone has been dismal, with metastatic disease the primary mode of treatment failure after an R0 surgical resection. Cure rates with chemotherapy or radiation therapy alone have been disappointing as well. For these reasons, over the last decade multi-modality treatment has gained increasing acceptance as the standard of care.
