2Incidence trends and age distribution of colorectal cancer by subsite in Guangzhou,2000-2011显示文摘Introduction:Colorectal cancer(CRC) is the third most common cancer in China.The incidence of CRC has been increasing in recent years.The aim of this study was to explore the incidence trends and the age distribution of CRC by subsite in Guangzhou between 2000 and 2011.Methods:A total of 22,432 incident cases of CRC between 2000 and 2011 from Guangzhou Cancer Registry were identified.Crude incidence and age-standardized rates(ASRs),using the Segi’s world standard population,were calculated for CRC and CRC subsites.The incidence trend was analyzed and the annual percentage change(APC) in incidence was calculated by using JoinPoint software.Results:The crude incidence increased significantly from 23.4/10~5 in 2000 to 37.4/10~5 in 2011 for males and from20.9/10~5 to 30.5/10~5 for females.The ASRs of CRC incidence stabilized during the period of 2000-2011 for both males and females.The ages at the onset of CRC for both males and females during 2010-2011 were significantly higher compared with those during 2000-2002(males:f = 1.95,P = 0.05;females:f = 6.03,P < 0.01).For males aged50-64 years,the CRC incidence increased by 8.50%annually(P = 0.04) during 2000-2004 and by 1.68%annually(P = 0.03) during 2005-2011.For females aged 65 years and older,the CRC incidence increased by 5.77%annually(P = 0.03) during 2000-2004.There were no significant changes for the CRC incidences in males aged 49 and younger and 65 years and older and females aged 64 years and younger during 2000-2004,or for those in all females as well as males aged 49 years and younger and 65 years and older during 2005-2011.The percentage of colon cancer in all CRCs increased significantly for both males and females between the periods of 2000-2002 and 2010-2011.The ASRs of descending colon and sigmoid colon cancer incidences increased significantly for females during 2005-2011(APC,5.51%and 1.08%,respectively,both P < 0.05).Conclusions:The crude incidence of CRC increased significantly between 2000 and 2011 because of the aging,whereas the ASRs kept stable.The percentage of colon cancer in all CRCs increased significantly.Further surveillance,research,and intervention are needed to identify the causes of these changes and to reduce the incidence and mortality of CRC.Qin Zhou Ke Li Guo-Zhen Lin Ji-Chuan Shen Hang Dong Yu-Ting Gu Hua-Zhang Liu 2015Chinese Journal of Cancer2015,34,8:17
3Selective Ferroptosis Inhibitor Liproxstatin-1 Attenuates Neurological Deficits and Neuroinflammation After Subarachnoid Hemorrhage显示文摘Ferroptosis is a form of iron-dependent regulated cell death.Evidence of its existence and the effects of its inhibitors on subarachnoid hemorrhage(SAH)is still lacking.In the present study,we found that liproxstatin-1 protected HT22 cells against hemin-induced injury by protecting mitochondrial functions and ameliorating lipid peroxidation.In in vivo experiments,we demonstrated the presence of characteristic shrunken mitochondria in ipsilateral cortical neurons after SAH.Moreover,liproxstatin-1 attenuated the neurological deficits and brain edema,reduced neuronal cell death,and restored the redox equilibrium after SAH.The inhibition of ferroptosis by liproxstatin-1 was associated with the preservation of glutathione peroxidase 4 and the downregulation of acylCoA synthetase long-chain family member 4 as well as cyclooxygenase 2.In addition,liproxstatin-1 decreased the activation of microglia and the release of IL-6,IL-1 b,and TNF-a.These data enhance our understanding of cell death after SAH and shed light on future preclinical studies.Yang Cao Yin Li Chao He Feng Yan Jian-Ru Li Hang-Zhe Xu Jian-Feng Zhuang Hang Zhou Yu-Cong Peng Xiong-Jie Fu Xiao-Yang Lu Yuan Yao Yu-Yu Wei Yun Tong Yi-Fu Zhou Lin Wang 2021Neuroscience Bulletin2021,37,4:16
4Oral berberine improves brain dopa/dopamine levels to ameliorate Parkinson’s disease by regulating gut microbiota显示文摘The phenylalanine-tyrosine-dopa-dopamine pathway provides dopamine to the brain.Iin this process,tyrosine hydroxylase(TH)isthe rate-limiting enzyme that hydroxylates tyrosine and generates levodopa(L-dopa)with tetranydrobiopterin(BH_(4))as a coenzyme.Here,we show that oral berberine(BBR)might supply H^(·) through dihydroberberine(reduced BBR produced by bacterial nitroreductase)and promote the production of BHl from dihydrobiopterin;the increased BH,enhances TH activity,which accelerates the production of L-dopa by the gut bacteria.Oral BBR acts in a way similar to vitamins.The L-dopa produced by theintestinal bacteria enters the brain through the circulation and is transformed to dopamine.To verify the gut-brain dialog activatedby BBR's effect,Enterococcus foecalis or Enterococcus faecium was transplanted into Parkinson's disease(PD)mice.The bacteriasignificantly increased brain dopamine and ameliorated PD manifestation in mice;additionally,combination of BBR with bacteriashowed better therapeutic effect than that with bacteria alone.Moreover,2,4,6-trimethy-pyranylium tetrafluoroborate(TMP-TFB)-derivatized matrix-assisted laser desorption mass spectrometry(MALDI-MS)imaging of dopamine identihed elevated striataldopamine levels in mouse brains with oral Enterococcus,and BBR strengthened the imaging intensity of brain dopamine.Theseresults demonstrated that BBR was an agonist of TH in Enterococcus and could lead to the production of L-dopa in the gut.Furthermore,a study of 28 patients with hyperlipidemia conhrmed that oral BBR increased bloodfecal L-dopa by the intestinalbacteria.Hence,BBR might improve the brain function by upregulating the biosynthesis of-dopa in the gut microbiota through avitamin-like effect.Yan Wang Qian Tong Shu-Rong Ma Zhen-Xiong Zhao Li-Bin Pan Lin Cong Pei Han Ran Peng Hang Yu Yuan Lin Tian-Le Gao Jia-Wen Shou Xiao-Yang Li Xian-Feng Zhang Zheng-Wei Zhang Jie Fu Bao-Ying Wen Jin-Bo Yu Xuetao Cao Jian-Dong Jiang 2021Signal Transduction and Targeted Therapy2021,6,3:16
5Allyl isothiocyanate ameliorates lipid accumulation and inflammation in nonalcoholic fatty liver disease via the Sirt1/AMPK and NF-κB signaling pathways显示文摘BACKGROUND Allyl isothiocyanate(AITC),a classic anti-inflammatory and antitumorigenic agent,was recently identified as a potential treatment for obesity and insulin resistance.However,little is known about its direct impact on the liver.AIM To investigate the effect and underlying mechanism of AITC in nonalcoholic fatty liver disease(commonly referred to as NAFLD).METHODS To establish a mouse and cellular model of NAFLD,C57BL/6 mice were fed a high fat diet(HFD)for 8 wk,and AML-12 cells were treated with 200μM palmitate acid for 24 h.For AITC treatment,mice were administered AITC(100 mg/kg/d)orally and AML-12 cells were treated with AITC(20μmol/L).RESULTS AITC significantly ameliorated HFD-induced weight gain,hepatic lipid accumulation and inflammation in vivo.Furthermore,serum alanine aminotransferase and aspartate aminotransferase levels were markedly reduced in AITC-treated mice.Mechanistically,AITC significantly downregulated the protein levels of sterol regulatory elementbinding protein 1(SREBP1)and its lipogenesis target genes and upregulated the levels of proteins involved in fatty acidβ-oxidation,as well as the upstream mediators Sirtuin 1(Sirt1)and AMPactivated protein kinaseα(AMPKα),in the livers of HFD-fed mice.AITC also attenuated the nuclear factor kappa B(NF-κB)signaling pathway.Consistently,AITC relieved palmitate acid-induced lipid accumulation and inflammation in AML-12 cells in vitro through the Sirt1/AMPK and NF-κB signaling pathways.Importantly,further studies showed that the curative effect of AITC on lipid accumulation was abolished by siRNA-mediated knockdown of either Sirt1 or AMPKαin AML-12 cells.CONCLUSION AITC significantly ameliorates hepatic steatosis and inflammation by activating the Sirt1/AMPK pathway and inhibiting the NF-κB pathway.Therefore,AITC is a potential therapeutic agent for NAFLD.Chun-Xiao Li Jian-Guo Gao Xing-Yong Wan Yi Chen Cheng-Fu Xu Ze-Min Feng Hang Zeng Yi-Ming Lin Han Ma Ping Xu Chao-Hui Yu You-Ming Li 2019World Journal of Gastroenterology2019,25,34:14
6Activation of mTORC1 in subchondral bone preosteoblasts promotes osteoarthritis by stimulating bone sclerosis and secretion of CXCL12显示文摘Increasing evidences show that aberrant subchondral bone remodeling plays an important role in the development of osteoarthritis(OA).However,how subchondral bone formation is activated and the mechanism by which increased subchondral bone turnover promotes cartilage degeneration during OA remains unclear.Here,we show that the mechanistic target of rapamycin complex 1(mTORC1)pathway is activated in subchondral bone preosteoblasts(Osterix+)from OA patients and mice.Constitutive activation of mTORC1 in preosteoblasts by deletion of the mTORC1 upstream inhibitor,tuberous sclerosis 1,induced aberrant subchondral bone formation,and sclerosis with little-to-no effects on articular cartilage integrity,but accelerated posttraumatic OA development in mice.In contrast,inhibition of mTORC1 in preosteoblasts by disruption of Raptor(mTORC1-specific component)reduced subchondral bone formation and cartilage degeneration,and attenuated post-traumatic OA in mice.Mechanistically,mTORC1 activation promoted preosteoblast expansion and Cxcl12 secretion,which induced subchondral bone remodeling and cartilage degeneration during OA.A Cxcl12-neutralizing antibody reduced cartilage degeneration and alleviated OA in mice.Altogether,these findings demonstrate that mTORC1 activation in subchondral preosteoblasts is not sufficient to induce OA,but can induce aberrant subchondral bone formation and secrete of Cxcl12 to accelerate disease progression following surgical destabilization of the joint.Pharmaceutical inhibition of the pathway presents a promising therapeutic approach for OA treatment.Chuangxin Lin Liangliang Liu Chun Zeng Zhong-Kai Cui Yuhui Chen Pinling Lai Hong Wang Yan Shao Haiyan Zhang Rongkai Zhang Chang Zhao Hang Fang Daozhang Cai Xiaochun Bai 2019Bone Research2019,7,1:12

9Comorbidity Burden of Dementia:A Hospital-Based Retrospective Study from 2003 to 2012 in Seven Cities in China显示文摘Dementia is increasing dramatically and imposes a huge burden on society. To date, there is a lack of data on the health status of patients with dementia in China. In an attempt to investigate the comorbidity burden of dementia patients in China at the national level, we enrolled 2,938 patients with Alzheimer's disease(AD), vascular dementia(Va D), or other types of dementia, who were admitted to tertiary hospitals in seven regions of China from January2003 to December 2012. The Charlson Comorbidity Index(CCI) was used to evaluate the comorbidity burden of the patients with dementia. Among these patients, 53.4% had AD, 26.3% had Va D, and 20.3% had other types of dementia. The CCI was 3.0 ± 1.9 for all patients,3.4 ± 1.8 for those with Va D, and 3.0 ± 2.1 for those with AD. The CCI increased with age in all patients, andthe length of hospital stay and daily expenses rose with age and CCI. Males had a higher CCI and a longer stay than females. Moreover, patients admitted in the last 5 years of the study had a higher CCI than those admitted in the first 5 years. We found that the comorbidity burden of patients with dementia is heavy. 10Strength and failure characteristics of brittle jointed rock-like specimens under uniaxial compression:Digital speckle technology and a particle mechanics approach显示文摘The strength and failure characteristics of most natural rock mass are influenced by discontinues such as fissures, joints, and weak surfaces. In the present study, the strength and failure behavior of ubiquitousjoint rock-like specimens under uniaxial loading have been investigated by DIC(digital image correlation) and discrete element numerical method(PFC2 D). The results are obtained. Firstly, the UCSJof specimens with c = 15° or 30° shows similar tendency while a goes from 0° to 75°. With c = 45° or 60°, the UCSJof specimens increases when a goes from 0° to 30° and decreases after a goes beyond 30°. With c = 75°, the peak UCSJvalue is reached when a = 0°. The UCSJvalue shows an increasing trend when a goes from 60° to 75°. Secondly, the ubiquitous-joint specimens present different failure modes for various levels of a and c(b-a). 13Support technology of deep roadway under high stress and its application显示文摘Roadway instability has been a major concern in the fields of mining engineering. This paper aims to provide practical and efficient strategy to support the roadways under high in-situ stress. A case study on the stability of deep roadways was carried out in an underground mine in Gansu province, China. Currently,the surrounding rock strata is extremely fractured, which results in a series of engineering disasters, such as side wall collapse and severe floor heave in the past decades. Aiming to solve these problems, an improved support method was proposed, which includes optimal bolt parameters and arrangement, floor beam layout by grooving, and full length grouting. Based on the modeling results by FLAC3D, the new support method is much better than the current one in terms of preventing the large deformation of surrounding rock and restricting the development of plastic zones. For implementation and verification, field experiments, along with deformation monitoring, were conducted in the 958 level roadway of Mining II areas. The results show that the improved support can significantly reduce surrounding rock deformation, avoid frequent repair, and maintain the long-term stability of the roadway. 14Stress-induced CsPbBr3 nanocrystallization on glass surface:Unexpected mechanoluminescence and applications显示文摘In this work,we discovered an unexpected mechanoluminescence (ML) phenomena occurring when transforming amorphous into crystalline,due to the stress-induced precipitation of CsPbBr3 perovskite nanocrystals on glass surface.It is revealed that,unlike the conventional thermal-induced phase transformation mechanism,the breakage of bonding of glass network provides the energy for nucleation and growth,and the shear stress avoids the long-range migration of structural units for crystallization.Such unique ML phenomenon enables the visualization of dynamical force that is inaccessible by common strategy,and so,opens up some novel applications,such as the pressure-sensitive 'glassy pencil' to learn people's writing habits,and the pb^2+-detection with good sensitivity and selectivity.These findings not only demonstrate an effective route for the preparation of perovskite materials in a green,time-saving,low cost,and scalable way,enrich the knowledge of glass crystallization mechanism,but also exploit a useful avenue to quantitatively visualize the dynamical force.Xiaoqiang Xiang Hang Lin Renfu Li Yao Cheng Qingming Huang Ju Xu Congyong Wang Xueyuan Chen Yuansheng Wang 2019Nano Research2019,12,5:8
15Inactivation of SACE_3446, a TetR family transcriptional regulator, stimulates erythromycin production in Saccharopolyspora erythraea显示文摘Erythromycin A is a widely used antibiotic produced by Saccharopolyspora erythraea;however,its biosynthetic cluster lacks a regulatory gene,limiting the yield enhancement via regulation engineering of S.erythraea.Herein,six TetR family transcriptional regulators(TFRs)belonging to three genomic context types were individually inactivated in S.erythraea A226,and one of them,SACE_3446,was proved to play a negative role in regulating erythromycin biosynthesis.EMSA and qRT-PCR analysis revealed that SACE_3446 covering intact N-terminal DNA binding domain specifically bound to the promoter regions of erythromycin biosynthetic gene eryAI,the resistant gene ermE and the adjacent gene SACE_3447(encoding a longchain fatty-acid CoA ligase),and repressed their transcription.Furthermore,we explored the interaction relationships of SACE_3446 and previously identified TFRs(SACE_3986 and SACE_7301)associated with erythromycin production.Given demonstrated relatively independent regulation mode of SACE_3446 and SACE_3986 in erythromycin biosynthesis,we individually and concomitantly inactivated them in an industrial S.erythraea WB.Compared with WB,the WBΔ3446 and WBΔ3446Δ3986 mutants respectively displayed 36%and 65%yield enhancement of erythromycin A,following significantly elevated transcription of eryAI and ermE.When cultured in a 5 L fermentor,erythromycin A ofWBΔ3446 and WBΔ3446Δ3986 successively reached 4095 mg/L and 4670 mg/L with 23%and 41%production improvement relative to WB.The strategy reported here will be useful to improve antibiotics production in other industrial actinomycete.Hang Wu Yansheng Wang Li Yuan Yongrong Mao Weiwei Wang Lin Zhu Panpan Wu Chengzhang Fu Rolf Muller David T.Weaver Lixin Zhang Buchang Zhang 2016Synthetic and Systems Biotechnology2016,1,1:7
16The biological effect of recombinant humanized collagen on damaged skin induced by UV-photoaging:An in vivo study显示文摘The application of medical devices to repair skin damage is clinically accepted and natural polymer enjoys an important role in this field,such as collagen or hyaluronic acid,etc.However,the biosafety and efficacy of these implants are still challenged.In this study,a skin damage animal model was prepared by UV-photoaging and recombinant humanized type Ⅲ collagen(rhCol Ⅲ)was applied as a bioactive material to implant in vivo to study its biological effect,comparing with saline and uncrosslinked hyaluronic acid(HA).Animal skin conditions were non-invasively and dynamically monitored during the 8 weeks experiment.Histological observation,specific gene expression and other molecular biological methods were applied by the end of the animal experiment.The results indicated that rhCol Ⅲ could alleviate the skin photoaging caused by UV radiation,including reduce the thickening of epidermis and dermis,increase the secretion of Collagen Ⅰ(Col Ⅰ)and Collagen Ⅲ(Col Ⅲ)and remodel of extracellular matrix(ECM).Although the cell-material interaction and mechanism need more investigation,the effect of rhCol Ⅲ on damaged skin was discussed from influence on cells,reconstruction of ECM,and stimulus of small biological molecules based on current results.In conclusion,our findings provided rigorous biosafety information of rhCol Ⅲ and approved its potential in skin repair and regeneration.Although enormous efforts still need to be made to achieve successful translation from bench to clinic,the recombinant humanized collagen showed superiorities from both safety and efficacy aspects.Jing Wang He Qiu Yang Xu Yongli Gao Peijie Tan Rui Zhao Zhanhong Liu Yajun Tang Xiangdong Zhu Chongyun Bao Hang Wang Hai Lin Xingdong Zhang 2022Bioactive Materials2022,7,5:7
17Comparison of continuous homogenous azeotropic and pressure-swing distillation for a minimum azeotropic system ethyl acetate/nhexane separation显示文摘Continuous homogenous azeotropic distillation(CHAD) and pressure-swing distillation(PSD) are explored to separate a minimum-boiling azeotropic system of ethyl acetate and n-hexane. The CHAD process with acetone as the entrainer and the PSD process with the pressures of 0.1 MPa and 0.6 MPa in two columns are designed and simulated by Aspen Plus. The operating conditions of the two processes are optimized via a sequential modular approach to obtain the minimum total annual cost(TAC). The computational results show that the partially heat integrated pressure-swing distillation(HIPSD) has reduced in the energy cost and TAC by 40.79% and 35.94%, respectively, than the conventional PSD, and has more greatly reduced the energy cost and TAC by 62.61% and 49.26% respectively compared with the CHAD process. 18Berberine treats atherosclerosis via a vitamine-like effect downregulating Choline-TMA-TMAO production pathway in gut microbiota显示文摘Trimethylamine-N-oxide(TMAO)derived from the gut microbiota is an atherogenic metabolite.This study investigates whether or not berberine(BBR)could reduce TMAO production in the gut microbiota and treat atherosclerosis.Effects of BBR on TMAO production in the gut microbiota,as well as on plaque development in atherosclerosis were investigated in the culture of animal intestinal bacterial,HFD-fed animals and atherosclerotic patients,respectively.We found that oral BBR in animals lowers TMAO biosynthesis in intestine through interacting with the enzyme/co-enzyme of choline-trimethylamine lyase(CutC)and flavincontaining monooxygenase(FMO)in the gut microbiota.This action was performed by BBR’s metabolite dihydroberberine(a reductive BBR by nitroreductase in the gut microbiota),via a vitamine-like effect down-regulating Choline-TMA-TMAO production pathway.Oral BBR decreased TMAO production in animal intestine,lowered blood TMAO and interrupted plaque formation in blood vessels in the HFD-fed hamsters.Moreover,21 patients with atherosclerosis exhibited the average decrease of plaque score by 3.2%after oral BBR(0.5 g,bid)for 4 months(^(*)P<0.05,n=21);whereas the plaque score in patients treated with rosuvastatin plus aspirin,or clopidogrel sulfate or ticagrelor(4 months,n=12)increased by 1.9%.TMA and TMAO in patients decreased by 38 and 29%in faeces(^(*)P<0.05;^(*)P<0.05),and 37 and 35%in plasma(^(***)P<0.001;^(*)P<0.05),after 4 months on BBR.BBR might treat atherosclerotic plaque at least partially through decreasing TMAO in a mode of action similar to that of vitamins.Shu-Rong Ma Qian Tong Yuan Lin Li-Bin Pan Jie Fu Ran Peng Xian-Feng Zhang Zhen-Xiong Zhao Yang Li Jin-Bo Yu Lin Cong Pei Han Zheng-Wei Zhang Hang Yu Yan Wang Jian-Dong Jiang 2022Signal Transduction and Targeted Therapy2022,7,8:6
19Activation of mitogen-activated protein kinases in satellite glial cells of the trigeminal ganglion contributes to substance P-mediated inflammatory pain显示文摘Inflammatory orofacial pain,in which substance P (SP) plays an important role,is closely related to the cross-talk between trigeminal ganglion (TG) neurons and satellite glial cells (SGCs).SGC activation is emerging as the key mechanism underlying inflammatory pain through different signalling mechanisms,including glial fibrillary acidic protein (GFAP) activation,phosphorylation of mitogen-activated protein kinase (MAPK) signalling pathways,and cytokine upregulation.However,in the TG,the mechanism underlying SP-mediated orofacial pain generated by SGCs is largely unknown.In this study,we investigated whether SP is involved in inflammatory orofacial pain by upregulating interleukin (IL)-1β and tumour necrosis factor (TNF)-α from SGCs,and we explored whether MAPK signalling pathways mediate the pain process.In the present study,complete Freund’s adjuvant (CFA) was injected into the whisker pad of rats to induce an inflammatory model in vivo.SP was administered to SGC cultures in vitro to confirm the effect of SP.Facial expression analysis showed that pre-injection of L703,606 (an NK-1 receptor antagonist),U0126 (an inhibitor of MAPK/extracellular signal-regulated kinase [ERK] kinase [MEK] 1/2),and SB203580 (an inhibitor of P38) into the TG to induce targeted prevention of the activation of the NK-1 receptor and the phosphorylation of MAPKs significantly suppressed CFA-induced inflammatory allodynia.In addition,SP promoted SGC activation,which was proven by increased GFAP,p-MAPKs,IL-1β and TNF-α in SGCs under inflammatory conditions.Moreover,the increase in IL-1β and TNF-α was suppressed by L703,606,U0126 and SB203580 in vivo and in vitro.These present findings suggested that SP,released from TG neurons,activated SGCs through the ERK1/2 and P38 pathways and promoted the production of IL-1β and TNF-α from SGCs,contributing to inflammatory orofacial pain associated with peripheral sensitization.Yanyan Zhang Ning Song Fei Liu Jiu Lin Mengke Liu Chaolan Huang Daqing Liao Cheng Zhou Hang Wang Jiefei Shen 2019International Journal of Oral Science2019,11,3:6
