| 2 | Activation of mitogen-activated protein kinases in satellite glial cells of the trigeminal ganglion contributes to substance P-mediated inflammatory pain显示文摘Inflammatory orofacial pain,in which substance P (SP) plays an important role,is closely related to the cross-talk between trigeminal ganglion (TG) neurons and satellite glial cells (SGCs).SGC activation is emerging as the key mechanism underlying inflammatory pain through different signalling mechanisms,including glial fibrillary acidic protein (GFAP) activation,phosphorylation of mitogen-activated protein kinase (MAPK) signalling pathways,and cytokine upregulation.However,in the TG,the mechanism underlying SP-mediated orofacial pain generated by SGCs is largely unknown.In this study,we investigated whether SP is involved in inflammatory orofacial pain by upregulating interleukin (IL)-1β and tumour necrosis factor (TNF)-α from SGCs,and we explored whether MAPK signalling pathways mediate the pain process.In the present study,complete Freund’s adjuvant (CFA) was injected into the whisker pad of rats to induce an inflammatory model in vivo.SP was administered to SGC cultures in vitro to confirm the effect of SP.Facial expression analysis showed that pre-injection of L703,606 (an NK-1 receptor antagonist),U0126 (an inhibitor of MAPK/extracellular signal-regulated kinase [ERK] kinase [MEK] 1/2),and SB203580 (an inhibitor of P38) into the TG to induce targeted prevention of the activation of the NK-1 receptor and the phosphorylation of MAPKs significantly suppressed CFA-induced inflammatory allodynia.In addition,SP promoted SGC activation,which was proven by increased GFAP,p-MAPKs,IL-1β and TNF-α in SGCs under inflammatory conditions.Moreover,the increase in IL-1β and TNF-α was suppressed by L703,606,U0126 and SB203580 in vivo and in vitro.These present findings suggested that SP,released from TG neurons,activated SGCs through the ERK1/2 and P38 pathways and promoted the production of IL-1β and TNF-α from SGCs,contributing to inflammatory orofacial pain associated with peripheral sensitization. | Yanyan Zhang Ning Song Fei Liu Jiu Lin Mengke Liu Chaolan Huang Daqing Liao Cheng Zhou Hang Wang Jiefei Shen | 2019 | International Journal of Oral Science2019,11,3: | 6 |
| 3 | Receptor tyrosine kinases positively regulate BACE activity and Amyloid-β production through enhancing BACE internalization显示文摘Amyloid-β (AP) peptide, the primary constituent of senile plaques in Alzheimer's disease (AD), is generated byβ-secretase- and γ-secretase-mediated sequential proteolysis of the amyloid precursor protein (APP). The aspartic pro-tease, β -site APP cleavage enzyme (BACE), has been identified as the main P-secretase in brain but the regulation of itsactivity is largely unclear. Here, we demonstrate that both BACE activity and subsequent Aβ production are enhancedafter stimulation of receptor tyrosine kinases (RTKs), such as the receptors for epidermal growth factor (EGF) and nervegrowth factor (NGF), in cultured cells as well as in mouse hippocampus. Furthermore, stimulation of RTKs also inducesBACE internalization into endosomes and Golgi apparatus. This enhancement of BACE activity and Aβ production uponRTK activation could be specifically inhibited by Src family kinase inhibitors and by depletion of endogenous c-Src withRNAi, and could be mimicked by over-expressed c-Src. Moreover, blockage of BACE internalization by a dominantnegative form of Rab5 also abolished the enhancement of BACE activity and Aβ production, indicating the requirementof BACE internalization for the enhanced activity. | 8 | Effect of amide type modified rapeseed oil as lubricating additive on friction and wear behavior of steel-steel and steel-aluminum alloy systems显示文摘A new type of environmentally friendly lube additive-amide type modified rapeseed oil was synthesized and characterized by infrared spectrum. Its effect on the friction and wear behavior of steel-steel and steel-aluminum alloy systems were investigated with a four-ball machine and an Optimol SRV friction and wear tester respectively.The morphographies of the worn surfaces were analyzed by means of scanning electron microscopy(SEM). The worn surfaces of the 2024Al alloy block were analyzed by means of X-ray photoelectron spectroscopy(XPS). The results show that the modified rapeseed oil as additives can obviously decrease the wear rate and friction coefficient of steel pair and steel-aluminum frictional pair. | 17 | Nutritional Risk Screening with NRS2002 among Hospitalized Cancer Patients in China显示文摘Objective The Nutritional Risk Screening 2002(NRS 2002)was used to assess the nutritional risk of hospitalized oncology patients in China.This study explored the factors affecting the risk of nutrition to provide a scientific basis for the assessment and treatment of malnutrition in oncology patients.Methods We used the NRS 2002 to evaluate the nutritional risk in 48,831 hospitalized cancer patients.Descriptive statistical methods were used to describe the general patient information.A Chi-squared test was applied to analyze the relationship between NRS 2002 scores and different demographic characteristics,and the NRS 2002 scores of cancer patients with different characteristics were compared by one-way ANOVA.Results Among 48,831 patients,43.3%were women and 57.7%were men,and 36.5%(17,802)of patients were at risk of nutrition(score≥3).The NRS 2002 score was the highest in leukemia patients(2.93±1.28).A one-way ANOVA revealed that the differences in NRS 2002 scores among patients of different gender,ages,TNM stages,education levels,occupations and areas of residence were statistically significant(P<0.001).Male patients had slightly higher NRS 2002 scores than females(2.33 vs.2.17).The lowest NRS 2002 scores were in patients aged 45-59(2.00±1.26)years and the highest scores were in patients aged≥70(2.76±1.43)years.The NRS 2002 score of patients receiving surgery was the highest(2.45±1.41),and patients receiving surgery plus radiotherapy/chemotherapy was the lowest(2.00±1.26).The risk of nutrition was highest in patients who were farmers(2.34±1.37 scores)and lowest in office staff(2.15±1.32 scores).Patients living in rural areas had the highest risk of nutrition(2.32±1.37 scores).There were significant differences in the NRS 2002 scores for different cancer sub-types for different ethnic groups(P<0.05),except for Zhuang individuals(P=0.124).The risk of nutrition was highest in Uyghur patients(3.35±1.33 scores)and lowest in Mongolians(2.04±1.37 scores).Conclusion More attention should be paid to people at high risk of nutrition,such as elderly patients,patients with a high TNM stage,patients receiving surgical treatment,and patients living in rural areas.Active nutritional interventions should be carried out to improve the nutritional status of malnourished patients. | Ming Ming Zhou Tian Tian Wu Yu Han Wang Yuan Lin Zou Ke Di Xu Jiu Wei Cui Zeng Ning Li Jun Qiang Chen Ming Liu Zeng Qing Guo Hong Xia Xu Han Ping Shi Chun Hua Song Investigation on Nutrition Status and Clinical Outcome of Common Cancers(INSCOC)Group | 2022 | Journal of Nutritional Oncology2022,7,2: | 0 |
