1Antidiabetic Effects of Gegen Qinlian Decoction via the Gut Microbiota Are Attributable to Its Key Ingredient Berberine显示文摘Gegen Qinlian Decoction(GQD),a traditional Chinese medicine(TCM)formula,has long been used for the treatment of common metabolic diseases,including type 2 diabetes mellitus.However,the main limitation of its wider application is ingredient complexity of this formula.Thus,it is critically important to identify the major active ingredients of GQD and to illustrate mechanisms underlying its action.Here,we compared the effects of GQD and berberine,a hypothetical key active pharmaceutical ingredient of GQD,on a diabetic rat model by comprehensive analyses of gut microbiota,short-chain fatty acids,proinflammatory cytokines,and ileum transcriptomics.Our results show that berberine and GQD had similar effects on lowering blood glucose levels,modulating gut microbiota,inducing ileal gene expression,as well as relieving systemic and local inflammation.As expected,both berberine and GQD treatment significantly altered the overall gut microbiota structure and enriched many butyrate-producing bacteria,including Faecalibacterium and Roseburia,thereby attenuating intestinal inflammation and lowering glucose.Levels of short-chain fatty acids in rat feces were also significantly elevated after treatment with berberine or GQD.Moreover,concentration of serum proinflammatory cytokines and expression of immune-related genes,including Nfkb1,Stat1,and Ifnrg1,in pancreatic islets were significantly reduced after treatment.Our study demonstrates that the main effects of GQD can be attributed to berberine via modulating gut microbiota.The strategy employed would facilitate further standardization and widespread application of TCM in many diseases.Xizhan Xu Zezheng Gao Fuquan Yang Yingying Yang Liang Chen Lin Han Na Zhao Jiayue Xu Xinmiao Wang Yue Ma Lian Shu Xiaoxi Hu Na Lyu Yuanlong Pan Baoli Zhu Linhua Zhao Xiaolin Tong Jun Wang 2020Genomics, Proteomics & Bioinformatics2020,18,6:29
2Multifunctional roles of carbon-based hosts for Li-metal anodes:A review显示文摘With its high theoretical capacity,lithium(Li)metal is recognized as the most potential anode for realizing a high-performance energy storage system.A series of questions(severe safety hazard,low Coulombic efficiency,short lifetime,etc.)induced by uncontrollable dendrites growth,unstable solid electrolyte interface layer,and large volume change,make practical application of Li-metal anodes still a threshold.Due to their highly appealing properties,carbon-based materials as hosts to composite with Li metal have been passionately investigated for improving the performance of Li-metal batteries.This review displays an overview of the critical role of carbon-based hosts for improving the comprehensive performance of Li-metal anodes.Based on correlated mainstream models,the main failure mechanism of Li-metal anodes is introduced.The advantages and strategies of carbon-based hosts to address the corresponding challenges are generalized.The unique function,existing limitation,and recent research progress of key carbon-based host materials for Li-metal anodes are reviewed.Finally,a conclusion and an outlook for future research of carbon-based hosts are presented.This review is dedicated to summarizing the advances of carbon-based materials hosts in recent years and providing a reference for the further development of carbonbased hosts for advanced Li-metal anodes.Xiaolin Yan Liang Lin Qiulin Chen Qingshui Xie Baihua Qu Laisen Wang Dong-Liang Peng 2021Carbon Energy2021,3,2:8
3Endostatin enhances antitumor effect of tumor antigen-pulsed dendritic cell therapy in mouse xenograft model of lung carcinoma显示文摘Objective: To investigate the antitumor effect of endostatin combined with tumor antigen-pulsed dendritic cell(DC)-T cell therapy on lung cancer.Methods: Transplanted Lewis lung cancer(LLC) models of C57BL/6 mice were established by subcutaneous injection of LLC cells in left extremity axillary. Tumor antigen-pulsed DC-T cells from spleen cells and bone of mice were cultured in vitro. Tumor-bearing mice were randomly divided into three groups, including DCT+endostatin group, DC-T group, and phosphate-buffered saline(PBS) control group. Microvessel density(MVD) of tumor tissue in tumor-bearing mice was determined by immunohistochemistry(IHC). The expressions of vascular endothelial growth factor(VEGF) and hypoxia-inducible factor-1α(HIF-1α) were determined by Western blotting and IHC staining. The proportions of CD8+ T cells, mature dendritic cells(m DC), tumor-associated macrophages [TAM(M1/M2)], and myeloid-derived suppressor cells(MDSC) in suspended cells of tumor tissue were determined by flow cytometry. The expressions of interleukin(IL)-6, IL-10, IL-17, transforming growth factor-β(TGF-β) and interferon-γ(IFN-γ) in suspended cells of tumor tissue were detected by enzyme-linked immune sorbent assay(ELISA).Results: DC-T cells combined with endostatin remarkably suppressed tumor growth. MVD of mice in DCT+endostatin group was significantly lower than that of the control group and DC-T monotherapy group. The expressions of VEGF, IL-6 and IL-17 in tumors were markedly decreased, but IFN-γ and HIF-1α increased after treating with DC-T cells combined with endostatin, compared to control group and DC-T group. In the DCT+endostatin group, the proportions of MDSC and TAM(M2 type) were significantly decreased, m DC and TAM(M1 type) were up-regulated, and CD8+ T cells were recruited to infiltrate tumors, in contrast to PBS control and DC-T monotherapy. 4Insights into triterpene synthesis and unsaturated fatty-acid accumulation provided by chromosomallevel genome analysis of Akebia trifoliata subsp . australis显示文摘Akebia trifoliata subsp.australis is a well-known medicinal and potential woody oil plant in China.The limited genetic information available for A.trifoliata subsp.australis has hindered its exploitation.Here,a high-quality chromosomelevel genome sequence of A.trifoliata subsp.australis is reported.The de novo genome assembly of 682.14 Mb was generated with a scaffold N50 of 43.11 Mb.The genome includes 25,598 protein-coding genes,and 71.18%(485.55 Mb)of the assembled sequences were identi fi ed as repetitive sequences.An ongoing massive burst of long terminal repeat(LTR)insertions,which occurred~1.0 million years ago,has contributed a large proportion of LTRs in the genome of A.trifoliata subsp.australis.Phylogenetic analysis shows that A.trifoliata subsp.australis is closely related to Aquilegia coerulea and forms a clade with Papaver somniferum and Nelumbo nucifera,which supports the well-established hypothesis of a close relationship between basal eudicot species.The expansion of UDP-glucoronosyl and UDP-glucosyl transferase gene families and fi-amyrin synthase-like genes and the exclusive contraction of terpene synthase gene families may be responsible for the abundant oleanane-type triterpenoids in A.trifoliata subsp.australis.Furthermore,the acyl-ACP desaturase gene family,including 12 stearoyl-acyl-carrier protein desaturase(SAD)genes,has expanded exclusively.A combined transcriptome and fatty-acid analysis of seeds at fi ve developmental stages revealed that homologs of SADs,acyl-lipid desaturase omega fatty acid desaturases(FADs),and oleosins were highly expressed,consistent with the rapid increase in the content of fatty acids,especially unsaturated fatty acids.The genomic sequences of A.trifoliata subsp.australis will be a valuable resource for comparative genomic analyses and molecular breeding.Hui Huang Juan Liang Qi Tan Linfeng Ou Xiaolin Li Caihong Zhong Huilin Huang Ian Max Moller Xianjin Wu Songquan Song 2021Horticulture Research2021,8,1:5
5NLOS Identification and Machine Learning Methods for Predicting the Outcome of 60GHz Ranging System显示文摘Millimeter-wave(MMW) signals in 60 GHz band have shown immense potential for accurate range estimation with precise time and multipath resolution. Nonline of sight(NLOS) propagation is a primary factor affecting the range precision. To improve range estimation,an Energy detector(ED) based normalized threshold algorithm which employs a Neural network(NN) is developed on the basis of NLOS identification. The maximum curl and standard deviation of the received energy block values are used to determine NLOS environment and the normalized thresholds for different Signal-to-noise ratios(SNRs). 7Development of a novel RNAi therapy:Engineered miR-31 exosomes promoted the healing of diabetic wounds显示文摘Rationale:Chronic wounds associated with diabetes exact a heavy burden on individuals and society and do not have a specific treatment.Exosome therapy is an extension of stem cell therapy,and RNA interference(RNAi)-based therapy is a type of advanced precision therapy.Based on the discovery of chronic wound-related genes in diabetes,we combined exosome therapy and RNAi therapy through an engineering approach for the treatment of diabetic chronic wounds.Methods:We combined exosome therapy and RNAi therapy to establish a precision therapy for diabetes-associated wounds via an engineered exosome approach.Results:First,chronic diabetic wounds express low levels of miR-31-5p compared with nondiabetic wounds,and an miR-31-5p mimic was shown to be effective in promoting the proliferation and migration of three wound-related cell types in vitro.Second,bioinformatics analysis,luciferase reporter assays and western blotting suggested that miR-31-5p promoted angiogenesis,fibrogenesis and reepithelization by inhibiting factor-inhibiting HIF-1(HIF1AN,also named FIH)and epithelial membrane protein-1(EMP-1).Third,engineered miR-31 exosomes were generated as a miR-31-5p RNAi therapeutic agent.In vivo,the engineered miR-31 exosomes promoted diabetic wound healing by enhancing angiogenesis,fibrogenesis and reepithelization.Conclusion:Engineered miR-31 exosomes are an ideal disease pathophysiology-initiated RNAi therapeutic agent for diabetic wounds.Jinghuan Huang Muyu Yu Wenjing Yin Bo Liang Ang Li Jingfeng Li Xiaolin Li Shichang Zhao Fang Liu 2021Bioactive Materials2021,6,9:3
8Freestanding polypyrrole nanotube/reduced graphene oxide hybrid film as flexible scaffold for dendrite-free lithium metal anodes显示文摘Lithium metal anode is the most potential anode material for the next generation high-energy rechargeable batteries owing to its highest specific capacity and lowest redox potential.Unfortunately,the uneven deposition of Li during plating/stripping and the formation of uncontrolled Li dendrites,which might cause poor battery performance and serious safety problems,are demonstrating to be a huge challenge for its practical application.Here,we show that a flexible and free-standing film hybriding with polypyrrole(PPy) nanotubes and reduced graphene oxide(rGO) can significantly regulate the Li nucleation and deposition,and further prohibit the formation of Li dendrites,owing to the large specific surface area,rich of nitrogen functional groups and porous structures.Finally,the high Coulombic efficiency and stable Li plating/stripping cycling performance with 98% for 230 cycles at 0.5 mA cm^(-2) and more than 900 hours stable lifespan are achieved.No Li dendrites form even at a Li deposition capacity as high as4.0 mA h cm^(-2).Besides,the designed PPy/rGO hybrid anode scaffold can also drive a superior battery performance in the lithium-metal full cell applications.Gan Luo Xiaolin Hu Wei Liu Guanjie Lu Qiannan Zhao Jie Wen Jian Liang Guangsheng Huang Bin Jiang Chaohe Xu Fusheng Pan 2021Journal of Energy Chemistry2021,30,7:3
9Space qualification mechanical tests of HTS filters for satellite application显示文摘High performance high-temperature superconducting (HTS) filters have been designed and constructed for satellite application. The filters are actually a superconducting integration of an 8-pole band-pass filter with an adjustable band-stop filter onto a single piece of LaAlO3 substrate (with dimension of 0.5×45×20 mm3). Typical results of the filters, i.e., Filters A (made by YBCO) & B (made by TBCCO) will be reported. The measured responses of Filter A showed excellent specifications, e.g., an insertion loss less than 0.1 dB, a return loss better than -22.5 dB in pass-band, band-edge steepness greater than 12 dB/MHz and out-of-band rejection at a certain band deeper than -110 dB. To satisfy the requirement of rocket launch and space operation, three filters of the above design have undergone mechanical environmental simulation tests for space qualification. Detailed analysis of the response curves of Filter B measured before and after the tests showed that no noticeable change in the per-formance can be found. 10Antibody Cocktail Exhibits Broad Neutralization Activity Against SARS-CoV-2 and SARS-CoV-2 Variants显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)has precipitated multiple variants resistant to therapeutic antibodies.In this study,12 high-affinity antibodies were generated from convalescent donors in early outbreaks using immune antibody phage display libraries.Of them,two RBD-binding antibodies(F61 and H121)showed high-affinity neutralization against SARS-Co V-2,whereas three S2-target antibodies failed to neutralize SARS-Co V-2.Following structure analysis,F61 identified a linear epitope located in residues G446–S494,which overlapped with angiotensinconverting enzyme 2(ACE2)binding sites,while H121 recognized a conformational epitope located on the side face of RBD,outside from ACE2 binding domain.Hence the cocktail of the two antibodies achieved better performance of neutralization to SARS-Co V-2.Importantly,these two antibodies also showed efficient neutralizing activities to the variants including B.1.1.7 and B.1.351,and reacted with mutations of N501 Y,E484 K,and L452 R,indicated that it may also neutralize the recent India endemic strain B.1.617.The unchanged binding activity of F61 and H121 to RBD with multiple mutations revealed a broad neutralizing activity against variants,which mitigated the risk of viral escape.Our findings revealed the therapeutic basis of cocktail antibodies against constantly emerging SARS-Co V-2 variants and provided promising candidate antibodies to clinical treatment of COVID-19 patients infected with broad SARS-Co V-2 variants.Yuanyuan Qu Xueyan Zhang Meiyu Wang Lina Sun Yongzhong Jiang Cheng Li Wei Wu Zhen Chen Qiangling Yin Xiaolin Jiang Yang Liu Chuan Li Jiandong Li Tianlei Ying Dexin Li Faxian Zhan Youchun Wang Wuxiang Guan Shiwen Wang Mifang Liang 2021Virologica Sinica2021,36,5:3
11Molecular basis of CONSTANS oligomerization in FLOWERING LOCUS T activation显示文摘The transcription factor CONSTANS(CO)integrates day-length information to induce the expression of florigen FLOWERING LOCUS T(FT)in Arabidopsis.We recently reported that the C-terminal CCT domain of CO forms a complex with NUCLEAR FACTOR-YB/YC to recognize multiple cis-elements in the FT promoter,and the N-terminal tandem B-box domains form a homomultimeric assembly.However,the mechanism and biological function of CO multimerization remained unclear.Here,we report that CO takes on a head-to-tail oligomeric configuration via its B-boxes to mediate FT activation in long days.The crystal structure of B-boxesCOreveals a closely connected tandem B-box fold forming a continuous head-to-tail assembly through unique CDHH zinc fingers.Mutating the key residues involved in CO oligomerization resulted in a non-functional CO,as evidenced by the inability to rescue co mutants.By contrast,a transgene encoding a human p53-derived tetrameric peptide in place of the B-boxesCOrescued co mutant,emphasizing the essential role of BboxesCO-mediated oligomerization.Furthermore,we found that the four TGTG-bearing cis-elements in FT proximal promoter are required for FT activation in long days.Our results suggest that CO forms a multimer to bind to the four TGTG motifs in the FT promoter to mediate FT activation.Xiaolin Zeng Xinchen Lv Rui Liu Hang He Shiqi Liang Lixian Chen Fan Zhang Liu Chen Yuehui He Jiamu Du 2022Journal of Integrative Plant Biology2022,64,3:2

14Crystalline InGaZnO quaternary nanowires with superlattice structure for high-performance thin-film transistors显示文摘Amorphous indium-gallium-zinc oxide (a-IGZO) materials have been widely explored for various thin-film transistor (TFT) applications;however, their device performance is still restricted by the intrinsic material issues especially due to their non-crystalline nature. In this study, highly crystalline superlattice-structured IGZO nanowires (NWs) with different Ga concentration are successfully fabricated by enhanced ambient-pressure chemical vapor deposition (CVD). The unique superlattice structure together with the optimal Ga concentration (i.e., 31 at.%) are found to effectively modulate the carrier concentration as well as efficiently suppress the oxygen vacancy formation for the superior NW device performance. In specific, the In1.8Ga1.8Zn2.4O7 NW field-effect transistor exhibit impressive device characteristics with the average electron mobility of ~ 110 cm^2-V^-1·s^-1 and on/off current ratio of ~ 10^6. Importantly, these NWs can also be integrated into NW parallel arrays for the construction of high-performance TFT devices, in which their performance is comparable to many state-of-the-art IGZO TFTs. 16The contribution of photoinduced charge-transfer enhancement to the SERS of uranyl(Ⅵ) in a uranyl-Ag_2O complex显示文摘Charge-transfer(CT) is an important enhancement mechanism in the field of surface-enhanced Raman scattering(SERS) that typically increases the Raman intensity of molecules by as much as 10–100 times.Herein, a low-cost Ag_2O aggregates substrate was prepared via a facile chemical precipitation method,and the calculated CT-based enhancement factor of the uranyl ions adsorbed on it reached as high as 10~5, a metal-comparable value. The efficient photoinduced CT process from the valence band of Ag_2O to the LUMO of uranyl ions under appropriate excitation sources resulted in the repulsion of the axial oxygen atoms of the O=U=O bond, which enhanced its polarizability, creating a more intense Raman mode. To the best of our knowledge, this study firstly reports such a strong photoinduced CT enhancement of uranyl ions, with concentrations of 10^(-8) mol L^(-1) or lower being detected using this Ag_2O substrate. Most importantly, this research has shown that the photoinduced CT enhancement also contributes to the SERS of uranyl ions on pure Ag substrates which have often been ascribed to the electromagnetic enhancement in previous studies. 18A 3D Geometry-Based Scattering Model for Vehicleto-Vehicle Wideband MIMO Relay-Based Cooperative Channels显示文摘In this paper, a three-dimensional(3D) geometry- based stochastic scattering model(GBSSM) for wideband multi-input multi-output(MIMO) vehicle-to-vehicle(V2V) relay-based cooperative fading channel based on geometrical three-cylinder is proposed. Non-line-of-sight(NLOS) propagation condition is assumed in amplify-and-forward(AF) cooperative networks from the source mobile station(S) to the destination mobile station(D) via the mobile relay station(R). We extend the proposed narrowband model to wideband and also introduce the carrier frequency and bandwidth into the model. To avoid complicated procedure in deriving the analytical expressions of the channel parameters and functions, the channel is realized first. 20Incomplete multi-view clustering via local and global co-regularization显示文摘The incompleteness of multi-view data is a phenomenon associated with real-world data mining applications,which brings a huge challenge for multi-view clustering.Although various types of clustering methods,which try to obtain a complete and consensus clustering result from a latent subspace,have been developed to overcome this problem,most methods excessively rely on views-public instances to bridge the connection with view-private instances.When lacking sufficient views-public instances,existing methods fail to transmit the information among incomplete views effectively.To overcome this limitation,we propose an incomplete multi-view clustering algorithm via local and global co-regularization(IMVC-LG).In this algorithm,we define a new objective function that is composed of two terms:local clustering from each view and global clustering from multiple views,which constrain each other to exploit the local clustering information from different incomplete views and determine a global consensus clustering result,respectively.Furthermore,an iterative optimization method is proposed to minimize the objective function.Finally,we compare the proposed algorithm with other state-of-the-art incomplete multi-view clustering methods on several benchmark datasets to illustrate its effectiveness.Jiye LIANG Xiaolin LIU Liang BAI Fuyuan CAO Dianhui WANG 2022Science China(Information Sciences)2022,65,5:1
