| 1 | Microvascular pathological features and changes in related injury factors in a rat acute blood stasis model显示文摘OBJECTIVE: To examine the microvascular pathological characteristics and changes in related injury factors in a rat model of acute blood stasis.METHODS: A total of 75 Sprague-Dawley rats were divided randomly and equally into a control group and four experimental groups assessed at different times after the induction of stasis(0, 1, 3 or 6 h after stasis)(n = 15). The acute blood stasis model was established through rat tail-vein injection of high-molecular-weight dextran. After Electrocardiograph(ECG) detection at predetermined times(0,1, 3 and 6 h after induction of stasis), the rats were sacrificed and blood and cardiac samples were harvested for analysis. Hematoxylin-eosin(HE) staining and transmission electron microscopy were used for histopathological detection; an enzyme linked immunosorbent assay(ELISA) was used to detect thromboxane B2(TXB2) and 6-keto-prostaglandin F1α(6-Keto-PGF1α) concentrations; a real-time polymerase chain reaction(PCR) reaction system was used to detect intercellular adhesion molecule1(ICAM-1) and vascular cell adhesion molecule1(VCAM-1) m RNA expression; western blotting was used to detect vascular endothelial cadherin(VE-cadherin) protein expression.RESULTS: The ST segment in the ECG showed gradual elevation after induction of stasis and continued elevation at a high level at 3 and 6 h. The HE staining showed changes in myocardial cell necrosis and tissue dissociation after the induction of stasis, along with inflammatory infiltration. Results of transmission electron microscopy showed immediate changes in blood stasis and lumen occlusion in the microvasculature, along with endothelial cell swelling. After the induction of stasis, TXB2 concentrations gradually increased while 6-Keto-PGF1αconcentrations were immediately significantly reduced. The TXB2/6-Keto-PGF1αratio was maintained at a high level. ICAM-1 m RNA expression showed an unstable elevation while VCAM-1 m RNA expression was significantly reduced after the induction of stasis. Compared with the control group, VE-cadherin protein expression increased at 0 and 3 h after the induction of stasis, while no change occurred at 1 and 6 h.CONCLUSION: The pathological manifestations of acute blood stasis are microvascular blood retention, lumen stenosis and even occlusion. The condition is also called 'blood coagulation and weep' in Traditional Chinese Medicine. | 2 | Changes in nitric oxide,angiotensinⅡ,angiopoietin-like protein 4 mRNA,neuregulin 1 mRNA,and platelet endothelial cell adhesion molecule-1 in rats with acute blood stasis induced by high-molecular-weight dextran显示文摘OBJECTIVE: To investigate the influence of acute blood stasis on nitric oxide(NO), angiotensin Ⅱ(Ang Ⅱ), angiopoietin-like protein 4(ANGPTL4)m RNA, neuregulin 1(NRG-1) m RNA, and platelet endothelial cell adhesion molecule-1(PECAM-1) in rats with stasis induced by high-molecular-weight dextran(HMWD).METHODS: Seventy-five Sprague Dawley rats were divided randomly into five groups(n = 15 in each group): control group, immediate group, 1 h group,3 h group, and 6 h group. A model of acute blood stasis was established via injection of HMWD into the tail vein. After performing electrocardiogram at the predetermined times according to the grouping, we collected blood and cardiac samples for hematoxylin-eosin(HE) staining and histopathological examination via transmission electron microscopy. Enzyme-linked immunosorbent assay was used to detect plasma levels of NO, AngⅡ, and fibrinogen. Real-time polymerase chain reaction was used to detect the expression of ANGPTL4 m RNA and NRG-1 m RNA. Immunohistochemical methods were used to detect PECAM-1 protein expression.RESULTS: The rat model of blood stasis showed blood retention in the capillary lumens. The ST segment showed gradual elevation, and was still elevated at 3 and 6 h after induction of blood stasis.HE staining showed myocardial cell necrosis and dissolution after modeling, along with basement membrane rupture and mitochondrial structural damage. Transmission electron microscopy showed endothelial cell swelling and an increase in absorption vesicles immediately after modeling. Endothelial cell apoptosis was increased at 3 and 6 h after modeling. Cardiac muscle fibers were disordered and intercalated discs were blurred immediately after modeling. There were massive numbers of dissolved cardiac muscle fibers, ruptured basement membranes, and mitochondrial structural damage at 3 and 6 h after modeling. NO plasma concentration was significantly reduced immediately and 1 h after modeling, while it was increased at 3 and 6 h.AngⅡ plasma concentration was decreased immediately after modeling, but increased at 1, 3, and6 h. Fibrinogen plasma concentration was significantly increased at immediate, 1, 3, and 6 h after modeling. PECAM-1 protein expression was obviously increased immediately after modeling, at 1, 6 h was found mild augment. Expression of Ang PTL4 m RNA was increased at immediate, 1, 3, and 6 h after modeling, and was found further augment at 3,and 6 h. Expression of NRG-1 m RNA was increased at immediate, 1, 3, and 6 h after modeling, and the strongest expression was at 1 h.CONCLUSION: The pathological manifestation of acute blood stasis is characterized by microvascular blood retention. | 5 | Tea polyphenols inhibit the growth and angiogenesis of breast cancer xenografts in a mouse model显示文摘Objective:To investigate the anti-angiogenic effect of tea polyphenols(TPS)on breast cancer and normal tissues in a mouse model.Methods:Breast cancer was successfully implanted into 48 BALB/c mice,which were then randomly divided into a TP oral gavage group,a TP local injection group,a ginsenoside Rg3 group,and a model control group according to a random number table.The tumor inhibitory rates of each group were calculated,while microvessel density(MVD)and the expression of vascular endothelial growth factor(VEGF),basic fibroblast growth factor(bFGF),and tissue inhibitor of metalloproteinase(TIMP-2)were detected by immunohistochemistry.Results:TPs could inhibit the growth of breast cancer xenografts in the mouse model.The tumor inhibition rates of the TP oral gavage and TP local injection groups were 37.43%and 40.94%,respectively.Compared with the model control group,MVD and VEGF and bFGF expression was downregulated(all P<.05),whereas TIMP-2 expression was elevated in the TP oral gavage and TP local injection groups(P=.015 and P=.032).TPs showed no significant effect on MVD and VEGF and TIMP-2 expression in the heart,brain,and kidney of the mouse model.Conclusion:TPs can restrict the growth of breast cancer by specifically inhibiting the angiogenesis of breast tumor tissue while having little effect on the normal tissue of important organs including the heart,brain,and kidney. | Peng Lv Fengqin Shi Xinyi Chen Li Xu Chong Wang Shaodan Tian Heng Yang Li Hou | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,2: | 2 |

| 8 | Expert consensus statement on diagnosis and treatment of cancer-related depressed mood state based on Chinese medicine显示文摘This consensus statement is organized into six parts:1)Definitions:cancer-related depressed mood state is defined as a group of depressive symptoms,rather than major depressive disorder.Thus,“cancer-related depression”or“depressed mood state”is introduced as standard terminology and associated with the Chinese medicine concept of“yu zheng”(depression syndrome).2)Pathogenesis:factors including psychological stress,cancer pain,cancer fatigue,sleep disorders,surgery trauma,chemotherapy,and radiation therapy are strongly associated with cancer-related depressed mood state.Crucial elements of pathogenesis are cancer caused by depression,depression caused by cancer,and the concurrence of phlegm,dampness,and stasis from constrained liver-qi and spleen deficiency.3)Symptoms:these include core symptoms,psychological symptoms,and somatic symptoms.Depressed mood and loss of interest are the main criteria for diagnosis.4)Clinical evaluation:based on the Mini-International Neuropsychiatric Interview and a numeric rating scale,and taking mood changes during cancer diagnosis and treatment into consideration,a questionnaire can be drafted to distinguish between major depressive disorder and cancer-related depression.The aim is to assist oncology clinicians to identify,treat,and refer patients with cancer-related depression.5)Diagnosis:diagnosis should be based on the Chinese Classification for Mental Disorders(CCMD-3),taking patients’mood changes during diagnosis and treatment into consideration.6)Treatment:treatments for cancer-related depression must be performed concurrently with cancer treatment.For mild depression,non-pharmacologic comprehensive therapies,including psychological intervention,music therapy,patient education,physical activity,and acupuncture,are recommended;for moderate depression,classical Chinese herbal formulas based on syndrome pattern differentiation combined with antidepressants are suggested;for severe depressive symptoms that have progressed to major depressive disorder, patients should be referred to a psychiatric clinician for specialized care. | Shaodan Tian Mei Jia Li Hou Xinyi Chen Dongyun Li Tianwei Guo | 2015 | Journal of Traditional Chinese Medical Sciences2015,2,4: | 1 |

| 11 | Dynamic changes of beta-amyloid protein deposition in hippocampus of female ovariectomized rats显示文摘BACKGROUND: To evaluate and summarize the effects of cerebral perfusion and vascular reserve on the treatment of SICAS. Recently, research on β-amyloid protein has focused on the regulatory effects of es- trogen or phytoestrogen on its deposition. However, there have been only a few reports on dynamic changes of β-amyloid protein deposition in hippocampus of ovariectomized rats. OBJECTIVE: To measureβ-amyloid protein deposition in the hippocampal formation of ovariectomized rats by using immunohistochemistry; to observe time-dependent dynamic changes. DESIGN: Randomized controlled animal study. SETTING: Third Xiangya Hospital of Central South University. MATERIALS: The experiment was carried out in the Central Laboratory of the Third Xiangya Hospital of Central South University from November 2005 to December 2006. Fifty healthy female Sprague Dawley (SD) rats, weighing (293 ± 10) g, were provided by the Animal Laboratory of Xiangya Medical College, Central South University. All rats had neither a childbearing history nor hepatic or renal disease, or skeletal deformity. β-amyloid protein immunohistochemical kit was provided by Wuhan Boster Company. The ex- periment was in accordance with animal ethics standards. METHODS: All rats were randomly divided into five groups, including normal control group (n = 10), sham operation group (n = 10), and ovariectomized group (n = 30). After anesthesia in the ovariectomized group, the bilateral ovaries were separated and resected. The same volume of fat was resected in the sham operation group. Rats from the normal control group, however, did not receive any surgical treatments. Rats in the normal control group and sham operation group were sacrificed by anesthesia 7 weeks after surgery. Every ten rats from the ovariectomized group was respectively sacrificed at 7, 15, and 30 weeks after surgery. Immunohistochemistry was used to detectβ-amyloid protein deposition in hippocampal sections. Cell counting and gray value measurements served to record the dynamic changes in β-amyloid protein deposi- tion. MAIN OUTCOME MEASURES: ① Morphological changes. ② Positive cell counts from β-amyloid protein stainings and gray value measurements. RESULTS: All 50 rats were involved in the final analysis. ① Morphological changes. β-amyloid-positive cells were detected in the hippocampus of all rats. Biebrich scarlet stained neurites with a swollen cytoplasm. A few β-amyloid-positive cells were observed in all groups 7 weeks after surgery, and plasma and neurites were slightly stained. By 15 weeks after surgery, a number of β-amyloid-positive cells were observed in the ovariectomized group, and plasma and neurites were also slightly stained. By 30 weeks after surgery, how- ever, many β-amyloid-positive cells were observed in the ovariectomized group. These cells were partially aggregated and darkly stained. ② Positive cell counts and gray value of β-amyloid protein in hippocam- pus. At 7 weeks after surgery, cell counts and gray value measurements were not significantly different in the ovariectomized group compared to the sham operation group and normal control group (P > 0.05). Cell counts and gray value measurements were higher in the ovariectomized group by 15 weeks compared to those by 7 weeks in the normal control group, sham operation group and ovariectomized group (P < 0.05). At 30 weeks after surgery, cell counts and gray value measurements were higher in the ovariectomized group compared to the normal control group. In addition, there were significant differences between sham opera- tion group and ovariectomized group at 7 and 15 weeks after operation (P < 0.05-0.01). Cell counts and gray value measurements increased in all groups over time. | 12 | Target prediction and activity verification for the antidepressant action of Huangqin(Radix Scutellariae Baicalensis)显示文摘OBJECTIVE:To decipher the antidepressant targets and mechanisms of Huangqin(Radix Scutellariae Baicalensis)(RSB)by a novel computational system based on prediction and experimental verification.METHODS:The putative targets of RSB against depression were identified from Traditional Chinese Medicine Systems Pharmacology(TCMSP)and Drug Bank.Next,protein-protein interaction network of the anti-depression targets of RSB were identified,and differentially expressed genes(DEGs)of depression were mined from the NCBI database.Then,Kyoto Encyclopedia of Genes and Genomes and Gene Ontology were used to analysis the common targets.Finally,the selected pathways and functions were verified by experimentation.RESULTS:Thirty active compounds in RSB were predicted with high confidence by TCMSP and DrugBank,and seventy-one DEGs were identified in the GEO database.Besides,eight core target proteins were screened out by descending order of degree value,including ACHE,IL6,SLC6A4,FOS,SLC6A3,MAOB,DPP4,and JUN.These target genes were further found to be associated with pathways involved in neuronal apoptosis,such as pathways in cancer,Toll-like receptor signaling pathway,and TNF signaling.The cell proliferation assay and wound-healing assay results showed that RSB does not affect PC12 cell proliferation and chemotaxis.Unexpectedly,RSB protected PC12 cells from oxidative stress induced by H2O2 via inhibiting autophagy and apoptosis.We revealed significant changes in mice treated with 400 mg/kg RSB compared with the lipopolysaccharide mice.The possible mechanism for the antidepressive action of RSB is by reducing the expression of LC3-B in CA1 neurons.CONCLUSIONS:Our research partially expounds the mechanism of the antidepressant effect of RSB by the combination of network pharmacology prediction and experimental verification.Furthermore,it is also conducive to the application of Traditional Chinese Medicine within modern medicine. | LI Ganggang LU Ye HE Pei ZHANG Shiyue CHENG Yating ZHANG Shaodan PEI Lin | 2021 | Journal of Traditional Chinese Medicine2021,41,6: | 0 |

| 14 | The Dynamics of Interorganizational Collaboration in Disaster Management:A Network Study Based on Flood Disasters in China显示文摘Interorganizational collaboration networks have become an important tool for disaster management.However,research on how different organizations can effectively collaborate throughout the entire disaster management process in centralized states such as China is scarce.This study begins to fill this lacuna by investigating interorganizational collaboration in different phases of disaster management and analyzing changes in the structure of the networks constructed during the preparedness and response phases of the 2020 flood disaster in Hubei Province,China.Building on the complex adaptive systems(CAS)theory,we argue that interorganizational collaboration changes dynamically according to its tasks and requirements.In the preparedness phase,interorganizational collaborations primarily follow established plans and choose horizontal selforganized collaboration mechanisms.However,when the urgent information and resource requirements increase in the response phase,many organizations choose vertical mandatory collaboration mechanisms.We found that organizations at the central and provincial levels in China were well positioned to coordinate information and resources and strengthen the interorganizational collaboration and communication that is crucial in disaster management.These findings contribute to the study of interorganizational collaboration networks in disaster management. | Zhichao Li Long Liu Shaodan Liu | 2023 | International Journal of Disaster Risk Science2023,14,6: | 0 |
| 15 | Application of UAV-Based Imaging and Deep Learning in Assessment of Rice Blast Resistance显示文摘Rice blast is regarded as one of the major diseases of rice.Screening rice genotypes with high resistance to rice blast is a key strategy for ensuring global food security.Unmanned aerial vehicles(UAV)-based imaging,coupled with deep learning,can acquire high-throughput imagery related to rice blast infection.In this study,we developed a segmented detection model(called RiceblastSegMask)for rice blast detection and resistance evaluation.The feasibility of different backbones and target detection models was further investigated.RiceblastSegMask is a two-stage instance segmentation model,comprising an image-denoising backbone network,a feature pyramid,a trinomial tree fine-grained feature extraction combination network,and an image pixel codec module.The results showed that the model combining the image-denoising and fine-grained feature extraction based on the Swin Transformer and the feature pixel matching feature labels with the trinomial tree recursive algorithm performed the best.The overall accuracy for instance segmentation of RiceblastSegMask reached 97.56%,and it demonstrated a satisfactory accuracy of 90.29%for grading unique resistance to rice blast.These results indicated that low-altitude remote sensing using UAV,in conjunction with the proposed RiceblastSegMask model,can efficiently calculate the extent of rice blast infection,offering a new phenotypic tool for evaluating rice blast resistance on a field scale in rice breeding programs. | LIN Shaodan YAO Yue LI Jiayi LI Xiaobin MA Jie WENG Haiyong CHENG Zuxin YE Dapeng | 2023 | Rice science2023,30,6: | 0 |
