1A Chromosome-Scale Genome Assembly of Paper Mulberry(Bmussonetia papyrifera)Provides New Insights into Its Forage and Papermaking Usage显示文摘Paper mulberry(Broussonetia papyrifera)is a well-known woody tree historically used for Cai Lun papermaking,one of the four great inventions of ancient China.More recently,Paper mulberry has also been used as forage to address the shortage of feedstuff because of its digestible crude fiber and high protein contents.In this study,we obtained a chromosome-scale genome assembly for Paper mulberry using integrated approaches,including Illumina and PacBio sequencing platform as well as Hi-C,optical,and genetic maps.The assembled Paper mulberry genome consists of 386.83 Mb,which is close to the estimated size,and 99.25%(383.93 Mb)of the assembly was assigned to 13 pseudochromosomes.Comparative genomic analysis revealed the expansion and contraction in the flavonoid and lignin biosynthetic gene families,respectively,accounting for the enhanced flavonoid and decreased lignin biosynthesis in Paper mulberry.Moreover,the increased ratio of syringyl-lignin to guaiacyl-lignin in Paper mulberry underscores its suitability for use in medicine,forage,papermaking,and barkcloth making.We also identified the rootassociated microbiota of Paper mulberry and found that Pseudomonas and Rhizobia were enriched in its roots and may provide the source of nitrogen for its stems and leaves via symbiotic nitrogen fixation.Collectively,these results suggest that Paper mulberry might have undergone adaptive evolution and recruited nitrogen-fixing microbes to promote growth by enhancing flavonoid production and altering lignin monomer composition.Our study provides significant insights into genetic basis of the usefulness of Paper mulberry in papermaking and barkcloth making,and as forage.These insights will facilitate further domestication and selection as well as industrial utilization of Paper mulberry worldwide.Xianjun Peng Hui Liu Peilin Chen Feng Tang Yanmin Hu Fenfen Wang Zhi Pi Meiling Zhao Naizhi Chen Hui Chen Xiaokang Zhang Xueqing Yan Min Liu Xiaojun Fu Guofeng Zhao Pu Yao Lili Wang He Dai Xuming Li Wei Xiong Wencai Xu Hongkun Zheng Haiyan Yu Shihua Shen 2019Molecular Plant2019,12,5:18
2Patterns of DNA cytosine methylation between haploids and corresponding diploids in rice显示文摘Eighteen pairs of diploid-haploid twin-seedlings were identified and screened out fromspecial rice SARII-628 population. Five pairs of themwere selected and randomly designated as A, B, C, Dand E. Simple sequence repeats (SSR) analysisshowed that there was no difference among 310 siteswhich indicated that there was no base mutation onDNA primary structure. DNA methylation plays animportant role in gene expression regulation duringgrowth and development stages in eukaryotes. Amodified AFLP technique (methylation-sensitiveAFLP, MSAP) was employed to detect the DNA me-thylation patterns in the 5′-CCGG sites of the fivepairs of twin-seedlings. Although no methylationmutation was detected among the five diploids,forty-three methylation mutation sites were foundfrom the corresponding haploids. The MSAP ratios,which were the ratios of MSAP sites to the total am-plified sites, in five haploids were 18.79%, 19.35%,18.49%, 18.45% and 18.75%, respectively. And cor-responding full methylation levels (5′-CmCGG indouble strands) of those haploids were 10.58%,11.3%, 10.11%, 10.09% and 10.34%, respectively.Both MSAP and full methylation levels in the fivehaploids were higher than that of their correspondingdiploids, which suggested that hypermethylation oc-curred in some 5′-CCGG sites. Five types of MASPpatterns among the five pairs of twin-seedlings weredetected as follows: (1) no changes, methylation lev-els were the same in both haploids and diploids; (2) demethylation, diploid was methylated but no me-thylation in the same site in haploid; (3) hypermethy-lation, the methylation level in haploid was higher than those in diploid; (4) hypomethylation, methyla-tion in haploid was lower than those in diploid; (5) undecided pattern, change trend of methylation lev-els in haploids was not decided. The bands of 18 sites were reclaimed, then sequenced and searched on website to determine the sites of those sequences on rice chromosomes. The result showed that the methylation mutation involved the whole rice genome and 12 pairs of chromosomes. The mutation was site-related and there were different mutation sites for different haploids. 3Neuroprotective effect of pretreatment with ganoderma lucidum in cerebral ischemia/reperfusion injury in rat hippocampus显示文摘Ganoderma lucidum is a traditional Chinese medicine,which has been shown to have both anti-oxidative and anti-inflammatory effects,and noticeably decreases both the infarct area and neuronal apoptosis of the ischemic cortex.This study aimed to investigate the protective effects and mechanisms of pretreatment with ganoderma lucidum(by intragastric administration)in cerebral ischemia/reperfusion injury in rats.Our results showed that pretreatment with ganoder-ma lucidum for 3 and 7 days reduced neuronal loss in the hippocampus,diminished the content of malondialdehyde in the hippocampus and serum,decreased the levels of tumor necrosis fac-tor-αand interleukin-8 in the hippocampus,and increased the activity of superoxide dismutase in the hippocampus and serum.These results suggest that pretreatment with ganoderma lucidum was protective against cerebral ischemia/reperfusion injury through its anti-oxidative and an-ti-inflammatory actions.Wangxin Zhang Qiuling Zhang Wen Deng Yalu Li Guoqing Xing Xianjun Shi Yifeng Du 2014Neural Regeneration Research2014,9,15:6
4Autotrophic and Heterotrophic Nitri?cation in a Highly Acidic Subtropical Pine Forest Soil显示文摘The occurrence of nitri?cation in some acidic forest soils is still a subject of debate.Identi?cation of main nitri?cation pathways in acidic forest soils is still largely unknown.Acidic yellow soil(Oxisol) samples were selected to test whether nitri?cation can occur or not in acidic subtropical pine forest ecosystems.Relative contributions of autotrophs and heterotrophs to nitri?cation were studied by adding selective nitri?cation inhibitor nitrapyrin.Soil NH^+_4-N concentrations decreased,but NO^-_3-N concentrations increased signi?cantly for the no-nitrapyrin control during the ?rst week of incubation,indicating that nitri?cation did occur in the acidic subtropical soil.The calculated net nitri?cation rate was 0.49 mg N kg^(-1)d^(-1)for the no-nitrapyrin control during the ?rst week of incubation.Nitrapyrin amendment resulted in a signi?cant reduction of NO^-_3-N concentration.Autotrophic nitri?cation rate averaged0.28 mg N kg^(-1)d^(-1)and the heterotrophic nitri?cation rate was 0.21 mg N kg^(-1)d^(-1)in the ?rst week.Ammonia-oxidizing bacteria(AOB) abundance increased slightly during incubation,but nitrapyrin amendment signi?cantly decreased AOB amo A gene copy numbers by about 80%.However,the ammonia-oxidizing archaea(AOA) abundance showed signi?cant increases only in the last 2weeks of incubation and it was also decreased by nitrapyrin amendment.Our results indicated that nitri?cation did occur in the present acidic subtropical pine forest soil,and autotrophic nitri?cation was the main nitri?cation pathway.Both AOA and AOB were the active biotic agents responsible for autotrophic nitri?cation in the acidic subtropical pine forest soil.FAEFLEN J.Sarwee LI Shiwei XIN Xiaoping Alan L.WRIGHT JIANG Xianjun 2016Pedosphere2016,26,6:6

7RNA-seq analysis of unintended effects in transgenic wheat overexpressing the transcription factor GmDREB1显示文摘The engineering of plants with enhanced tolerance to abiotic stresses typically involves complex multigene networks and may therefore have a greater potential to introduce unintended effects than the genetic modification for simple monogenic traits. For this reason, it is essential to study the unintended effects in transgenic plants engineered for stress tolerance. We selected drought-and salt-tolerant transgenic wheat overexpressing the transcription factor, GmDREB1, to investigate unintended pleiotropic effects using RNA-seq analysis. We compared the transcriptome alteration of transgenic plants with that of wild-type plants subjected to salt stress as a control. We found that GmDREB1 overexpression had a minimal impact on gene expression under normal conditions.GmDREB1 overexpression resulted in transcriptional reprogramming of the salt response,but many of the genes with differential expression are known to mitigate salt stress and contribute incrementally to the enhanced stress tolerance of transgenic wheat. GmDREB1 overexpression did not activate unintended gene networks with respect to gene expression in the roots of transgenic wheat. 9Myelin Deficits Caused by Olig2 Deficiency Lead to Cognitive Dysfunction and Increase Vulnerability to Social Withdrawal in Adult Mice显示文摘Oligodendrocyte (OL) and myelin development are crucial for network integration and are associated with higher brain functions. Accumulating evidence has demonstrated structural and functional impairment of OLs and myelin in serious mental illnesses. However, whether these deficits contribute to the brain dysfunction or pathogenesis of such diseases still lacks direct evidence. In this study, we conditionally deleted Olig2 in oligodendroglial lineage cells (Olig2 cKO) and screened the behavioral changes in adult mice. We found that Olig2 ablation impaired myelin development, which further resulted in severe hypomyelination in the anterior cingulate cortex. Strikingly, Olig2 cKO mice exhibited an anxious phenotype, aberrant responses to stress, and cognitive deficits. Moreover, Olig2 cKO mice showed increased vulnerability to social avoidance under the mild stress of social isolation. 10High valence metals engineering strategies of Fe/Co/Ni-based catalysts for boosted OER electrocatalysis显示文摘Electrocatalysis for the oxygen evolution reactions(OER)has attracted much attention due to its important role in water splitting and rechargeable metal-air batteries.Therefore,designing highly efficient and low-cost catalysts for OER process is essential as the conventional catalysts still rely on precious metals.Transition metal-based compounds have been widely investigated as active OER catalysts,and renewed interest in the high valence metals engineered compounds has been achieved for superior catalytic activity and stability.However,an in-depth understanding of the construction strategies and induced effects for the high valence metals engineered catalysts is still lacking and desired.In this review,we have summarized the construction strategies of high valence metals as dopants or formed heterostructures with the iron/cobalt/nickel(Fe/Co/Ni)-based catalysts.Then the induced effects on Fe/Co/Ni-based catalysts by incorporating high valence metals,e.g.,accelerating the surface reconstruction,forming amorphous structure,generating vacancies/defects,and acting as stabilizers,are highlighted.The impacts of high valence metals on OER performance are elucidated based on different elements,including molybdenum(Mo),tungsten(W),cerium(Ce),vanadium(V),chromium(Cr),manganese(Mn),niobium(Nb),zirconium(Zr).The correlations of construction strategies,induced effects,catalytic activity and OER reaction pathways are elaborated.Finally,the remaining challenges for further enhancements of OER performance induced by high valence metals are presented.Lu Li Xianjun Cao Juanjuan Huo Junpeng Qu Weihua Chen Chuntai Liu Yufei Zhao Hao Liu Guoxiu Wang 2023Journal of Energy Chemistry2023,,1:3
11Standard pancreatoduodenectomy versus extended pancreatoduodenectomy with modified retroperitoneal nerve resection in patients with pancreatic head cancer:a multicenter randomized controlled trial显示文摘Background:The extent of pancreatoduodenectomy for pancreatic head cancer remains controversial,and more high-level clinical evidence is needed.This study aimed to evaluate the outcome of extended pancreatoduodenectomy(EPD)with retroperitoneal nerve resection in pancreatic head cancer.Methods:This multicenter randomized trial was performed at 6 Chinese highvolume hospitals that enrolled patients between October 3,2012,and September 21,2017.Four hundred patients with stage I or II pancreatic head cancer and without specific pancreatic cancer treatments(preoperative chemotherapy or chemoradiation)within three months were randomly assigned to undergo standard pancreatoduodenectomy(SPD)or EPD,with the latter followed by dissection of additional lymph nodes(LNs),nerves and soft tissues 270◦on the right side surrounding the superior mesenteric artery and celiac axis.The primary endpoint was overall survival(OS)by intention-to-treat(ITT).The secondary endpoints were disease-free survival(DFS),mortality,morbidity,and postoperative pain intensity.Results:TheR1 ratewas slightly lower with EPD(8.46%)thanwith SPD(12.56%).The morbidity and mortality rates were similar between the two groups.The median OS was similar in the EPD and SPD groups by ITT in the whole study cohort(23.0 vs.20.2 months,P=0.100),while the median DFS was superior in the EPD group(16.1 vs.13.2 months,P=0.031).Patients with preoperative CA19–9<200.0 U/mL had significantly improved OS and DFS with EPD(EPD vs.SPD,30.8 vs.20.9 months,P=0.009;23.4 vs.13.5 months,P<0.001).The EPD group exhibited significantly lower locoregional(16.48%vs.35.20%,P<0.001)andmesenteric LNrecurrence rates(3.98%vs.10.06%,P=0.022).The EPD group exhibited less back pain 6 months postoperation than the SPD group.Conclusions:EPD for pancreatic head cancer did not significantly improve OS,but patients with EPD treatment had significantly improved DFS.In the subgroup analysis,improvements in bothOS and DFS in the EPD armwere observed in patients with preoperative CA19–9<200.0 U/mL.EPD could be used as an effective surgical procedure for patients with pancreatic head cancer,especially those with preoperative CA19–9<200.0 U/mL.Qing Lin Shangyou Zheng Xianjun Yu Meifu Chen Yu Zhou Quanbo Zhou Chonghui Hu Jing Gu Zhongdong Xu LinWang Yimin Liu Qingyu Liu MinWang Guolin Li He Cheng Dongkai Zhou Guodong Liu Zhiqiang Fu Yu Long Yixiong Li Weilin Wang Renyi Qin Zhihua Li Rufu Chen 2023Cancer Communications2023,43,2:3

13The Dynamic Experiment on Treating Acid Mine Drainage with Iron Scrap and Sulfate Reducing Bacteria Using Biomass Materials as Carbon Source显示文摘The study is aimed at the problem of high content of Cr^(6+),Cr^(3+)and SO_(4)^(2-)is high and low pH value in acid mine drainage(AMD).Moreover,treatment of AMD by sulfate reducing bacteria(SRB)requires the addition of carbon source,while the treating effectiveness is not good enough on its own.The sugarcane slag,the corn cob and the sunflower straw were selected as the SRB carbon source cooperating with iron scrap to construct the dynamic columns 1,2 and 3.The mechanism of removing Cr^(6+),Cr^(3+),SO_(4)^(2-)and H+and the regularity of sustained release of carbon source and TFe release was studied in AMD.The removal efficiency of heavy metal ions,the ability of sustained release of carbon source,and the ability of adjusting acid by the three dynamic columns were compared.The result shows that the average removal rates of Cr^(6+),Cr^(3+)and SO_(4)^(2-)in effluent of dynamic column 1,filled by sugarcane slag,iron scrap and SRB,were 96.9%,67.1%and 54.3%.The average release of TFe and chemical oxygen demand(COD)were 4.4 and 287.3 mg/L.Its average pH was 6.98.Compared with the performance of dynamic columns 1,2 and 3,dynamic column 1 performed best in removing Cr^(6+),Cr^(3+)and SO_(4)^(2-)from AMD and controlling the release of COD and TFe,adjusting the pH of the solution.The study is of significance in treatment of AMD by taking for biomass materials as SRB carbon source in cooperation with iron scrap.Xianjun Wang Junzhen Di Yanrong Dong Yu Yang Bing Liang Fankang Meng Tingting Wang Wenbo An Zengxin Li Jianzhi Guo 2021Journal of Renewable Materials2021,9,1:2
14The ER membrane protein complex subunit Emc3 controls angiogenesis via the FZD4/WNT signaling axis显示文摘The endoplasmic reticulum(ER) membrane protein complex(EMC) regulates the synthesis and quality control of membrane proteins with multiple transmembrane domains. One of the membrane spanning subunits, EMC3, is a core member of the EMC complex that provides essential hydrophilic vestibule for substrate insertion. Here, we show that the EMC subunit Emc3 plays critical roles in the retinal vascular angiogenesis by regulating Norrin/Wnt signaling. Postnatal endothelial cell(EC)-specific deletion of Emc3 led to retarded retinal vascular development with a hyperpruned vascular network, the appearance of bluntended, aneurysm-like tip endothelial cells(ECs) with reduced numbers of filopodia and leakage of erythrocytes at the vascular front. Diminished tube formation and cell proliferation were also observed in EMC3 depleted human retinal endothelial cells(HRECs). We then discovered a critical role for EMC3 in expression of FZD4 receptor of β-catenin signaling using RNA sequencing, real-time quantitative PCR(RT-q PCR) and luciferase reporter assay. Moreover, augmentation of Wnt activity via lithium chloride(Li Cl) treatment remarkably enhanced β-catenin signaling and cell proliferation of HRECs. Additionally, Li Cl partially reversed the angiogenesis defects in Emc3-c KO mice. 16Blueberry malvidin-3-galactoside modulated gut microbial dysbiosis and microbial TCA cycle KEGG pathway disrupted in a liver cancer model induced by HepG2 cells显示文摘HCC(Hepatocellular Carcinoma)is a critical health issue worldwide.Our previous animal experiment has confirmed that blueberry malvidin-3-galactoside(M3G)can regulate the progression of HCC.In this study,feces samples from the same batch of mice were collected to explore the regulatory mechanism of M3G on intestinal microbiota and microbial TCA cycle metabolism KEGG pathway in HCC mice based on 16S rRNA sequencing and metagenomics.Our results showed that blueberry M3G increased the microbial diversity and regulated the structure of intestinal microbiota in mice,such as increasing the abundance of Clostridia(butyric acid-producing bacteria),Oscillospira and Ruminococcus,and reducing the abundance of pathogenic Erysipelotrichi.Compared with the group of liver cancer and 5-fluorouracil,blueberry M3G significantly regulated microbial TCA cycle KEGG pathway via improving the expression of key proteins(porA,DLAT,aceE,PC and OGDH).Additionally,we found which the abundance of Muribaculum intestinale increased by blueberry M3G may be an important factor affecting the microbial TCA cycle KEGG pathway via the pearson correlation(R)analysis of protein and microbiota.Taken together,these results demonstrate that the blueberry M3G has the potential to be an intestinal microbiota regulator and an adjuvant to HCC therapy.Zhen Cheng Jie Lin Ningxuan Gao Xiyun Sun Xianjun Meng Ruihai Liu Yunen Liu Weisheng Wang Bin Li Yuehua Wang 2020Food Science and Human Wellness2020,9,3:2
17Formation of Field Reversed Configuration (FRC) on the Yingguang-I device显示文摘As a hybrid approach to realizing fusion energy,Magnetized Target Fusion(MTF)based on the Field Reversed Configuration(FRC),which has the plasma density and confinement time in the range between magnetic and inertial confinement fusion,has been recently widely pursued around the world.To investigate the formation and confinement of the FRC plasma injector for MTF,the Yingguang-I,which is an FRC test device and contains a multi-bank program-discharged pulsed power sub-system,was constructed at the Institute of Fluid Physics(IFP),China.This paper presents the pulsed power components and their parameters of the device in detail,then gives a brief description of progress in experiments of FRC formation.Experimental results of the pulsed power sub-system show that the peak current/magnetic field of 110 kA/0.3 T,10 kA/1.2 Tand 1.7 MA/3.4 Twere achieved in the bias,mirror and q-pinch circuits with quarter cycle of 80 ms,700 ms and 3.8 ms respectively.The induced electric field in the neutral gas was greater than 0.25 kV/cm when the ionization bank was charged to 70 kV.With H_(2) gas of 8 Pa,the plasma target of density 10^(16) cm^(-3),separatrix radius 4 cm,half-length 17 cm,equilibrium temperature 200 eV and lifetime 3 ms(approximately the half pulse width of the reversed field)have been obtained through the q-pinch method when the bias,mirror,ionization and θ-pinch banks were charged to 5 kV,5 kV,55 kV and ±45 kV respectively.The images from the high-speed end-on framing camera demonstrate the formation processes of FRC and some features agree well with the results with the two-dimension magneto hydrodynamics code(2D-MHD).Qizhi Sun Xianjun Yang Yuesong Jia Lulu Li Dongfan Fang Xiaoming Zhao Weidong Qin Zhengfen Liu Wei Liu Jun Li Yuan Chi Xiaoguang Wang 2017Matter and Radiation at Extremes2017,2,5:2
18Equivalency and unbiasedness of grey prediction models显示文摘In order to deeply research the structure discrepancy and modeling mechanism among different grey prediction models, the equivalence and unbiasedness of grey prediction models are analyzed and verified. The results show that all the grey prediction models that are strictly derived from x(0)(k) +az(1)(k) = b have the identical model structure and simulation precision. Moreover, the unbiased simulation for the homogeneous exponential sequence can be accomplished. However, the models derived from dx(1)/dt + ax(1)= b are only close to those derived from x(0)(k) + az(1)(k) = b provided that |a| has to satisfy|a| < 0.1; neither could the unbiased simulation for the homogeneous exponential sequence be achieved. 19Floralozone protects endothelial function in atherosclerosis by ameliorating NHE1显示文摘Endothelial dysfunction is the pathological basis of atherosclerosis.Incomplete understanding of endothelial dysfunction etiology has impeded drug development for this devastating disease despite the currently available therapies.Floralozone,an aroma flavor,specifically exists in rabbit ear grass.Recently,floralozone has been demonstrated to inhibit atherosclerosis,but the underlying mechanisms are undefined.The present study was undertaken to explore whether floralozone pharmacologically targets endothelial dysfunction and therefore exerts therapeutic effects on atherosclerosis.The Na+/H+exchanger 1(NHE1),a channel protein,plays a vital role in atherosclerosis.Whether NHE1 is involved in the therapeutic effects of floralozone on endothelial dysfunction has yet to be further answered.By performing oil red staining and hematoxylin–eosin staining,vascular functional study,and oxidative stress monitoring,we found that floralozone not only reduced the size of carotid atherosclerotic plaque but also prevented endothelial dysfunction in atherosclerotic rats.NHE1 expression was upregulated in the inner membrane of carotid arteries and H2O2-induced primary rat aortic endothelial cells.Inspiringly,floralozone prevented the upregulation of NHE1 in vivo and in vitro.Notably,the administration of NHE1 activator LiCl significantly weakened the protective effect of floralozone on endothelial dysfunction in vivo and in vitro.Our study demonstrated that floralozone exerted its protective effect on endothelial dysfunction in atherosclerosis by ameliorating NHE1.NHE1 maybe a drug target for the treatment of atherosclerosis,and floralozone may be an effective drug to meet the urgent needs of atherosclerosis patients by dampening NHE1.Ning Huang Yue Qiu Yanhua Liu Tianheng Liu Xianjun Xue Ping Song Jian Xu Yutian Fu Ruili Sun Yaling Yin Peng Li 2021Acta Biochimica et Biophysica Sinica2021,53,10:2
