| 3 | Notch signaling mediated by TGF-β/Smad pathway in concanavalin A-induced liver fibrosis in rats显示文摘AIM To explore the exact interaction between Notch and transforming growth factor(TGF)-β signaling in liver fibrosis. METHODS We established a rat model of liver fibrosis induced by concanavalin A. Peripheral blood mononuclear cells(PBMCs) were isolated from the modeled rats, and cultured with γ-secretase inhibitor DAPT and TGF-β inhibitor for 24 h. The m RNA levels of Notch and TGF-β signaling were detected by quantitative real-time polymerase chain reaction. Expression of Notch and TGF-β proteins was analyzed by western blotting.RESULTS Compared to control rats, Notch and TGF-β signaling was activated in PBMCs of model rats. Administration of DAPT and TGF-β inhibitor suppressed Notch and TGF-β signal transducer in PBMCs of model rats. DAPT reduced m RNA and protein expression of TGF-β signaling, such as TGF-β1 and Smad3. TGF-β inhibitor also downregulated Notch1, Hes1 and Hes5, and m RNA and protein expression of the Notch signaling pathway.CONCLUSION Notch and TGF-β signaling play a role in liver fibrosis. | 4 | Prevalence of asthenopia and its risk factors in Chinese college students显示文摘AIM:To determine the prevalence of asthenopia and identify any associated risk factors in the college students in Xi'an,China. METHODS:From April to September 2012,1 500 students from five universities in Xi'an were selected according to a multi-stage stratified cluster sampling method.Data on demographic features,lifestyle or dietary habits,health status,living environment conditions,sleep and mental status,and asthenopia symptoms were collected through a self-administered validated questionnaire.Univariate logistic regression and multivariate logistic regression analysis modified by the factor analysis were performed to evaluate risk factors for asthenopia.RESULTS:Fifty-seven percent of the college students complained of asthenopia.Statistically significant risk factors for asthenopia in the univariate analysis included 13 variables.Multivariate analysis revealed a significant relationship between the use of computer and asthenopia(OR 1.21,95% CI:1.09 to 1.35).Good sleep and mental status(OR 0.86,95% CI:0.76 to 0.97),good living environment conditions(OR 0.67,95% CI:0.60 to 0.76),and high intake of green leafy vegetables(OR 0.89,95% CI:0.80 to 0.98) were found to be strong predictors of decreasing the occurrence of asthenopia complaints.CONCLUSION:Asthenopia symptom appeares to be common among college students;and it is strongly associated with computer use,psychosocial state,environment conditions and dietary habits,suggesting that additional studies are warranted to verify these risk factors and establish prevention guidelines,especially for college students. | Cheng-Cheng Han Rong Liu Ru-Ru Liu Zhong-Hai Zhu Rong-Bin Yu Le Ma | 2013 | International Journal of Ophthalmology(English edition)2013,6,5: | 12 |
| 5 | A phase I study of different doses and frequencies of pegylated recombinant human granulocyte-colony stimulating factor(PEG rhG-CSF) in patients with standard-dose chemotherapy-induced neutropenia显示文摘Objective: The recommended dose of prophylactic pegylated recombinant human granulocyte-colony stimulating factor(PEG rhG-CSF) is 100 μg/kg once per cycle for patients receiving intense-dose chemotherapy.However, few data are available on the proper dose for patients receiving less-intense chemotherapy. The aim of this phase I study is to explore the proper dose and administration schedule of PEG rhG-CSF for patients receiving standard-dose chemotherapy.Methods:Eligible patients received 3-cycle chemotherapy every 3 weeks.No PEG rhG-CSF was given in the first cycle.Patients experienced grade 3 or 4 neutropenia would then enter the cycle 2 and 3.In cycle 2,patients received a single subcutaneous injection of prophylactic PEG rhG-CSF on d 3,and received half-dose subcutaneous injection in cycle 3 on d 3 and d 5,respectively.Escalating doses(30,60,100 and 200μg/kg)of PEG rhG-CSF were investigated.Results:A total of 26 patients were enrolled and received chemotherapy,in which 24 and 18 patients entered cycle 2 and cycle 3 treatment,respectively.In cycle 2,the incidence of grade 3 or 4 neutropenia for patients receiving single-dose PEG rhG-CSF of 30,60,100 and 200 μg/kg was 66.67%,33.33%,22.22% and 0,respectively,with a median duration less than 1(0–2)d.No grade 3 or higher neutropenia was noted in cycle 3 in all dose cohorts.Conclusions:The pharmacokinetic and pharmacodynamic profiles of PEG rhG-CSF used in cancer patients were similar to those reported,as well as the safety.Double half dose administration model showed better efficacy result than a single dose model in terms of grade 3 neutropenia and above.The single dose of 60 μg/kg,100 μg/kg and double half dose of 30 μg/kg were recommended to the phase Ⅱ study,hoping to find a preferable method for neutropenia treatment. | Yan Qin Xiaohong Han Lin Wang Ping Du Jiarui Yao Di Wu Yuanyuan Song Shuxiang Zhang Le Tang Yuankai Shi | 2017 | Chinese Journal of Cancer Research2017,29,5: | 11 |

| 11 | Effects of potassium deficiency on photosynthesis,chloroplast ultrastructure,ROS,and antioxidant activities in maize(Zea mays L.)显示文摘Potassium(K)deficiency significantly decreases photosynthesis due to leaf chlorosis induced by accumulation of reactive oxygen species(ROS).But,the physiological mechanism for adjusting antioxidative defense system to protect leaf function in maize(Zea mays L.)is unknown.In the present study,four maize inbred lines(K-tolerant,90-21-3 and 099;K-sensitive,D937 and 835)were used to analyze leaf photosynthesis,anatomical structure,chloroplast ultrastructure,ROS,and antioxidant activities.The results showed that the chlorophyll content,net photosynthetic rate(P_n),stomatal conductance(G_s),photochemical quenching(q_P),and electron transport rate of PSII(ETR)in 90-21-3 and 099 were higher than those in D937 and 835 under K deficiency treatment.Parameters of leaf anatomical structure in D937 that were significantly changed under K deficiency treatment include smaller thickness of leaf,lower epidermis cells,and vascular bundle area,whereas the vascular bundle area,xylem vessel number,and area in 90-21-3 were significantly larger or higher.D937 also had seriously damaged chloroplasts and PSII reaction centers along with increased superoxide anion(O_2^-·)and hydrogen peroxide(H_2O_2).Activities of antioxidants,like superoxide dismutase(SOD),catalase(CAT),and ascorbate peroxidase(APX),were significantly stimulated in 90-21-3 resulting in lower levels of O_2^-·and H_2O_2.These results indicated that the K-tolerant maize promoted antioxidant enzyme activities to maintain ROS homeostasis and suffered less oxidative damage on the photosynthetic apparatus,thereby maintaining regular photosynthesis under K deficiency stress. | DU Qi ZHAO Xin-hua XIA Le JIANG Chun-ji WANG Xiao-guang HAN Yi WANG Jing YU Hai-qiu | 2019 | Journal of Integrative Agriculture2019,18,2: | 6 |
| 12 | Paeoniflorin-6′-0-benzene sulfonate alleviates collageninduced arthritis in mice by downregulating BAFF-TRAF2-NF-kB signaling: comparison with biological agents显示文摘Paeoniflorin-6′-0-benzene sulfonate (CP-25) is a new ester derivative of paeoniflorin with improved lipid solubility and oral bioavailability, as well as better anti-inflammatory activity than its parent compound. In this study we explored whether CP-25 exerted therapeutic effects in collagen-induced arthritis (CIA) mice through regulating B-cell activating factor (BAFF)-BAFF receptors-mediated signaling pathways. CIA mice were given CP-25 or injected with biological agents rituximab or etanercept for 40 days. In CIA mice, we found that T cells and B cells exhibited abnormal proliferation;the percentages of CD19^+ total B cells, CD19^+CD27^+-activated B cells, CD19^+BAFFR^+ and CD19^+TACI^+cells were significantly increased in PBMCs and spleen lymphocytes. CP-25 suppressed the indicators of arthritis, alleviated histopathology, accompanied by reduced BAFF and BAFF receptors expressions, inhibited serum immunoglobulin levels, decreased the Bell subsets percentages, and prevented the expressions of key molecules in NF-kB signaling. Furthermore, we showed that treatment with CP-25 reduced CD19+TRAF2^+ cell expressions stimulated by BAFF and decreased TRAF2 overexpression in HEK293 cells in vitro. Thus, CP-25 restored the abnormal T cells proliferation and B-cell percentages to the normal levels, and normalized the elevated levels of IgA, lgG2a and key proteins in NF-kB signaling. In comparison, rituximab and etanercept displayed stronger antiinflammatory activities than CP-25;they suppressed the elevated inflammatory indexes to below the normal levels in CIA mice. In summary, our results provide evidence that CP-25 alleviates CIA and regulates the functions of B cells through BAFF-TRAF2-NF-kB signaling. | 15 | Solar Energetic Particle Event of 2005 January 20:Release Times and Possible Sources显示文摘Based on cosmic ray data obtained by neutron monitors at the Earth's surface, and data on near-relativistic electrons measured by the WIND satellite, as well as on solar X-ray and radio burst data, the solar energetic particle (SEP) event of 2005 January 20 is studied. The results show that this event is a mixed event where the flare is dominant in the acceleration of the SEPs, the interplanetary shock accelerates mainly solar protons with energies below 130 MeV, while the relativistic protons are only accelerated by the solar flare. The interplanetary shock had an obvious acceleration effect on relativistic electrons with energies greater than 2 MeV. It was found that the solar release time for the relativistic protons was about 06:41 UT, while that for the near-relativistic electrons was about 06:39 UT. | 19 | COVID-19 vaccination intention among healthcare workers in Vietnam显示文摘Objective:To assess the acceptance of coronavirus disease(COVID-19)vaccine among healthcare workers at two general hospitals in Vietnam when it is available.Methods:A cross-sectional study was conducted using a convenience sampling from January to February 2021 among 410 healthcare workers at two general hospitals in Vietnam via a selfadministered questionnaire.A multivariable regression analysis was performed to determine predictors of vaccine acceptance including the demographic factors,COVID-19 knowledge,and vaccine beliefs based on the domains of Health Belief Model.Results:Among 410 healthcare workers,76.10%showed vaccination willingness.Predictors of acceptance were determined that the group reporting as“vaccine acceptance”was more likely to be positive towards the perceived susceptibility and severity of COVID-19(OR 2.45;95%CI 1.48-4.06,P<0.05),perceived benefits of vaccination,and cues to action(OR 4.36;95%CI 2.35-8.09,and OR 5.49;95%CI 2.84-10.61,respectively,all P<0.001),but less likely to have the perceived barriers to vaccination(OR 0.19;95%CI 0.09-0.38;P<0.001)compared with the no acceptance group.Besides,people who had a good knowledge regarding the severity of illness were 3.37 times more likely to have identified as vaccine acceptance(OR 3.37;95%CI 1.04-10.86,P<0.05).The demographic factors were also associated with willingness to receive the vaccine,with participants who were staff and received COVID-19 information from relatives were less likely to accept the vaccine over those who were doctors and not receiving information from relatives(OR 0.36;95%CI 0.13-0.96,and OR 0.37;95%CI 0.17-0.78,respectively,all P<0.05).Conclusions:A rate of willingness to get vaccinated against COVID-19 was relatively high with discrepancies between occupation,receiving information from relatives,knowledge toward the severity of illness,and the elements of Health Belief Model.The findings will provide information for the management authorities to develop relevant interventions to promote COVID-19 vaccination uptake. | Giao Huynh Thien Thuan Tran Han Thi Ngoc Nguyen Le An Pham | 2021 | Asian Pacific Journal of Tropical Medicine2021,14,4: | 3 |
| 20 | 霉酚酸酯联合糖皮质激素诱导治疗过敏性紫癜性肾炎的回顾性研究(英文)显示文摘目的:合并中等量蛋白尿的过敏性紫癜性肾炎的诱导治疗尚无确切方案,本研究通过回顾性比较分析了霉酚酸酯联合糖皮质激素的疗效与安全性。创新点:首次对霉酚酸酯联合糖皮质激素诱导治疗过敏性紫癜性肾炎进行了回顾性研究。方法:回顾性分析2007年1月至2013年6月间在浙江大学附属第一医院肾脏病中心接受‘肾穿刺活检,且经过3个月以上血管紧张素转换酶抑制剂(ACEI)/血管紧张素受体拮抗剂(ARB)治疗后蛋白尿为1.0-3.5g/24h的过敏性紫癜性肾炎患者95例。根据治疗方案分为3组,霉酚酸酯组(33例)在原剂量ACEFARB的基础上加用霉酚酸酯联合低剂量糖皮质激素,糖皮质激素组(31例)在原剂量ACEFARB的基础上加用全剂量糖皮质激素,对照组(31例)维持单用ACEFARB治疗,但可提高其剂量。患者随访观察6~78月(中位观察时间28月),霉酚酸酯组、糖皮质激素组与对照组的蛋白尿缓解率分别为72.7%、64.5%与45.2%(图1),发生副作用分别为17例、30例与6例,糖皮质激素组高脂血症与高血压发生率较高(表3)。结论:霉酚酸酯联合低剂量糖皮质激素可有效诱导缓解过敏性紫癜性肾炎,其缓解率与全剂量糖皮质激素治疗相当,且副作用较少。 | Fei HAN Liang-liang CHEN Ping-ping REN Jing-yun LE Pei-jing CHOONG Hong-ju WANG Ying XU Jiang-hua CHEN | 2015 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2015,16,9: | 3 |
