| 2 | Identification of a novel mutation in POU3F4 for prenatal diagnosis in a Chinese family with X-linked nonsyndromic hearing loss显示文摘We present the clinical and genetic findings for a Chinese family with X-linked non-syndromic hearing loss in which the affected males showed congenital profound sensorineural hearing impairment. In two affected brothers, the computer tomography of temporal bone showed bilateral dilation of the internal auditory canal with fistulous communication between the lateral canal and the basal cochlear turn, which is consistent with the typical DFNX2 phenotype. A missense mutation (c.647G→A) in the POU3F4 gene caused a substitu- tion from glycine to glutamic acid at position 216 (p.G216E), and this mutation was found to consistently cosegregate with the deafness phenotype in the family. The mutation resulted in the loss of function of the POU3F4 by decreasing the affinity between the protein and DNA, as shown in silico by the structural analysis. Prenatal diagnosis of pregnant proband of this family revealed the c.647G→A muta- tion in DNA extracted from the amniotic fluid surrounding the fetus. | 3 | Meaningful comparison of photocatalytic properties of {001} and {101} faceted anatase TiO2 nanocrystals显示文摘The facet-dependent photocatalytic performance of TiO_2 nanocrystals has been extensively investigated due to their promising applications in renewable energy and environmental fields. However, the intrinsic distinction in the photocatalytic oxidation activities between the {001}and {101} facets of anatase TiO_2 nanocrystals is still unclear and under debate. In this work, a simple photoelectrochemical method was employed to meaningfully quantify the intrinsic photocatalytic activities of {001} and{101} faceted TiO_2 nanocrystal photoanodes. The effective surface areas of photoanodes with different facets were measured based on the monolayer adsorption of phthalic acid on TiO_2 photoanode surface by an ex situ photoelectrochemical method, which were used to normalize the photocurrents obtained from different faceted photoanodes for meaningful comparison of their photocatalytic activities. The results demonstrated that the {001} facets of anatase TiO_2 nanocrystals exhibited much better photocatalytic activity than that of {101} facets of anatase TiO_2 nanocrystals toward photocatalytic oxidation of water and organic compounds with different functional groups(e.g.,–OH, –CHO, –COOH). | 4 | Tauroursodeoxycholic acid(TUDCA) inhibits influenza A viral infection by disrupting viral proton channel M2显示文摘Influenza is a persistent threat to human health and there is a continuing requirement for updating antiinfluenza strategies. Initiated by observations of different endoplasmic reticulum(ER) responses of host to seasonal H1N1 and highly pathogenic avian influenza(HPAI) A H5N1 infections, we identified an alternative antiviral role of tauroursodeoxycholic acid(TUDCA), a clinically available ER stress inhibitor, both in vitro and in vivo. Rather than modulating ER stress in host cells, TUDCA abolished the proton conductivity of viral M2 by disrupting its oligomeric states, which induces inefficient viral infection. We also showed that M2 penetrated cells, whose intracellular uptake depended on its proton channel activity,an effect observed in both TUDCA and M2 inhibitor amantadine. | 5 | Molecular and serological surveillance of Getah virus in the Xinjiang Uygur Autonomous Region,China,2017–2020显示文摘The Getah virus(GETV),a mosquito-borne RNA virus,is widely distributed in Oceania and Asia.GETV is not the only pathogenic to horses,pigs,cattle,foxes and boars,but it can also cause fever in humans.Since its first reported case in Chinese mainland in 2017,the number of GETV-affected provinces has increased to seventeen till now.Therefore,we performed an epidemiologic investigation of GETV in the Xinjiang region,located in northwestern China,during the period of 2017-2020.ELISA was used to analyze 3299 serum samples collected from thoroughbred horse,local horse,sheep,goat,cattle,and pigs,with thoroughbred horse(74.8%),local horse(67.3%),goat(11.7%),sheep(10.0%),cattle(25.1%)and pigs(51.1%)being positive for anti-GETV antibodies.Interestingly,the neutralizing antibody titer in horses was much higher than in other species.Four samples from horses and pigs were positive for GETV according to RT-PCR.Furthermore,from the serum of a local horse,we isolated GETV which was designated as strain XJ-2019-07,and determined its complete genome sequence.From the phylogenetic relationships,it belongs to the Group III lineage.This is the first evidence of GETV associated to domestic animals in Xinjiang.Overall,GETV is prevalent in Xinjiang and probably has been for several years.Since no vaccine against GETV is available in China,detection and monitoring strategies should be improved in horses and pigs,especially imported and farmed,in order to prevent economic losses. | Ning Shi Xiangshu Qiu Xinyu Cao Zhanhai Mai Xiangyu Zhu Nan Li He Zhang Jinyong Zhang Zhuoxin Li Nuerlan Shaya Huijun Lu Ningyi Jin | 2022 | Virologica Sinica2022,37,2: | 5 |
| 6 | Evolution of permafrost in Northeast Chinasince the Late Pleistocene显示文摘In Northeast China, permafrost advanced and retreated several times under the influences of fluctuating paleo-climates and paleo-environments since the Late Pleistocene. During the last 60 years, many new data were obtained and studies were conducted on the evolution of permafrost in Northeast China, but so far no systematic summary and review have been made. Based on sedimentary sequences, remains of past permafrost, paleo-flora and-fauna records, and dating data, permafrost evolution since the Late Pleistocene has been analyzed and reconstructed in this paper. Paleo-temperatures reconstructed from the remains of past permafrost and those from paleo-flora and-fauna are compared, and thus the southern limit of permafrost(SLP) in each climate period is inferred by the relationship of the permafrost distribution and the mean annual air/ground temperatures(MAAT/MAGT). Thus, the evolutionary history of permafrost is here divided into five stages:(1) the Late Pleistocene(Last Glaciation, or LG)(65 to 10–8.5 ka), the Last Glaciation Maximum(LGM, 21–13 ka) in particular, the coldest period in the latest history with a cooling of about 6~10 ?C, characterized by extensive occurrences of glaciation, flourishing Mammathas-Coelodonta Faunal Complex(MCFC), widespread aeolian deposits, and significant sea level lowering, and permafrost greatly expanded southwards almost to the coastal plains(37?N–41?N);(2) the Holocene Megathermal Period(HMP, 8.5–7.0 to 4.0–3.0 ka), 3~5 ?C warmer than today, permafrost retreated to about 52°N;(3) the Late Holocene Cold Period(Neoglaciation)(4.0–3.0 to 1.0–0.5 ka), a cooling of 1~3 ?C, some earlier thawed permafrost was refrozen or attached, and the SLP invaded southwards to 46?N;(4) the Little Ice Age(LIA, 500 to 100–150 a), the latest cold period with significant permafrost expansion; and(5) climate warming since the last century, during which Northeast China has undergone extensive permafrost degradation. The frequent and substantial expansions and retreats of permafrost have greatly impacted cold-region environments in Northeast China. North of the SLP during the HMP, or in the present continuous permafrost zone, the existing permafrost was largely formed during the LG and was later overlapped by the permafrost formed in the Neoglaciation. To the south, it was formed in the Neoglaciation. However, many aspects of permafrost evolution still await further investigations, such as data integration, numerical reconstruction, and merging of Chinese permafrost history with those of bordering regions as well as collaboration with related disciplines. | 9 | High-risk phenotypes of genetic disease in a Neonatal Intensive Care Unit population显示文摘To the Editor: Genetic diseases contribute to 35% of deaths during the first year of life and are a significant cause of intensive care.[1] A previous study based on the China Neonatal Genomes Project investigated the genetic causes of early infant deaths and found that >25% of deceased neonates with genetic diagnoses can be cured if diagnosed in time.[2] Therefore, it is crucial to target and diagnose neonates with genetic diseases as early as possible. According to our experience, the typical phenotypes, such as special facial features or multiple congenital anomalies (MCAs), indicate a high risk of genetic disease and lead physicians to perform genetic testing in neonates as early as possible. However, in practice, infants without typical phenotypes typically undergo a long and costly diagnostic process before genetic diagnoses are confirmed. Moreover, a recent survey by the American College of Medical Genetics and Genomics (ACMG) and other national professional organizations indicated that there are insufficient numbers of qualified geneticists to fulfil genetic service needs.[3] The ACMG published the general clinical features for genetic testing indications. For example, patients with phenotypes or family history data that strongly implicate a genetic cause may undergo genetic testing.[1] However, the study indicated that many genetic conditions arise de novo or are inherited with no family history.[1] A previous study attempted to apply the non-phenotype-driven panel approach in neonates admitted to the neonate intensive care unit (NICU).[4] However, at present, the diagnostic yield is only 3.45% (1/29).[4] In addition, the economic and ethical issues associated with genomic screening remain challenging. | 11 | Evolution and changes of permafrost on the Qinghai-Tibet Plateau during the Late Quaternary显示文摘Due to the uplift of Qinghai-Tibet Plateau(QTP), the cryosphere gradually developed on the higher mountain summits after the Neocene, becoming widespread during the Late Quaternary. During this time, permafrost on the QTP experienced repeated expansion and degradation. Based on the remains and cross-correlation with other proxy records such as those from glacial landforms, ice-core and paleogeography, the evolution and changes of permafrost and environmental changes on the QTP during the past 150,000 years were deduced and are presented in this paper. At least four obvious cycles of the extensive and intensive development, expansion and decay of permafrost occurred during the periods of 150–130, 80–50, 30–14 and after 10.8 ka B.P.. During the Holocene, fluctuating climatic environments affected the permafrost on the QTP, and the peripheral mountains experienced six periods of discernible permafrost changes:(1) Stable development of permafrost in the early Holocene(10.8 to 8.5–7.0 ka B.P.);(2) Intensive permafrost degradation during the Holocene Megathermal Period(HMP, from 8.5–7.0 to 4.0–3.0 ka B.P.);(3) Permafrost expansion during the early Neoglacial period(ca. 4,000–3,000 to 1,000 a B.P.);(4) Relative degradation during the Medieval Warm Period(MWP, from 1,000 to 500 a B.P.);(5) Expansion of permafrost during the Little Ice Age(LIA, from 500 to 100 a B.P.);(6) Observed and predicted degradation of permafrost during the 20 th and 21 st century. Each period differed greatly in paleoclimate, paleoenvironment, and permafrost distribution, thickness, areal extent, and ground temperatures, as well as in the development of periglacial phenomena. Statistically, closer dating of the onset permafrost formation, more identification of permafrost remains with richer proxy information about paleoenvironment, and more dating information enable higher resolution for paleo-permafrost reconstruction. Based on the scenarios of persistent climate warming of 2.2~2.6 °C in the next 50 years, and in combination of the monitored trends of climate and permafrost changes, and model predictions suggest an accelerated regional degradation of plateau permafrost. Therefore, during the first half of the 21 st century, profound changes in the stability of alpine ecosystems and hydro(geo)logical environments in the source regions of the Yangtze and Yellow rivers may occur. | 13 | Effect of simvastatin on the atherosclerotic plaque stability and the angiogenesis in the atherosclerotic plaque of rabbits显示文摘In this study, the effect of simvastatin on the atherosclerotic plaque stability and the angiogenesis in the atherosclerotic plaque of rabbits was observed. Thirty New Zealand rabbits were randomly divided into the normal control group, the model control group and the simvastatin group, 10 in each group. Rabbits in the normal control group were fed with normal forage, while rabbits in the rest two groups were fed with high fat forage. The balloon injury was performed two weeks later to establish an abdominal aortic atherosclerosis model, and then high fat forage was successively fed to them. Meanwhile, simvastatin at the daily dose of 2.5 mg/kg body weight was administered to rabbits in the simvastatin group. After 6 weeks of successive administration, levels of blood lipids were measured after blood sampling, and the serum high sensitivity C-reactive protein (hsCRP) and matrix metalloproteinase-3,-9 (MMP-3,-9) were detected using enzyme-linked immunosorbent assay. The macroscopically pathological indices of the plaque tissue were observed using hematoxylin and eosin (HE) staining of abdominal aorta specimens under a light microscope, and the plaque area (PA), cross-sectional vascular area (CVA) and correcting plaque area (PA/CVA) were determined quantitatively using imaging software. The protein expressions of the vascular endothelial growth factor (VEGF), factor VIII related antigen (FVIIIRAg), MMP-3 and cluster of differentiation antigen 40 ligand (CD40L) in the plaque were detected with the immunohistochemical method. Compared with the model control group, the levels of VEGF, FVIIIRAg, MMP-3, CD40L protein expression and the serum expression levels of hsCRP, MMP-3, MMP-9 in the simvastatin group were significantly reduced (P<0.05, P<0.01). The ratio PA/CVA in the simvastatin group was more significantly reduced when compared with that in the model control group (P<0.01). The levels of serum total cholesterol (TC), triglyceride (TG) and low-density lipoprotein cholesterol (LDL) were significantly reduced in the simvastatin group when compared with those in the model control group (P<0.05, P<0.01).
