2A potent PGK1 antagonist reveals PGK1 regulates the production of IL-1βand IL-6显示文摘Glycolytic metabolism enzymes have been implicated in the immunometabolism field through changes in metabolic status. PGK1 is a catalytic enzyme in the glycolytic pathway. Here, we set up a high-throughput screen platform to identify PGK1 inhibitors. DC-PGKI is an ATPcompetitive inhibitor of PGK1 with an affinity of Kd= 99.08 nmol/L. DC-PGKI stabilizes PGK1in vitro and in vivo, and suppresses both glycolytic activity and the kinase function of PGK1. In addition,DC-PGKI unveils that PGK1 regulates production of IL-1β and IL-6 in LPS-stimulated macrophages.Mechanistically, inhibition of PGK1 with DC-PGKI results in NRF2(nuclear factor-erythroid factor 2-related factor 2, NFE2L2) accumulation, then NRF2 translocates to the nucleus and binds to the proximity region of Il-1β and Il-6 genes, and inhibits LPS-induced expression of these genes. DC-PGKI ameliorates colitis in the dextran sulfate sodium(DSS)-induced colitis mouse model. 9Molecular Mechanism of Zhizi Ganjiang Decoction in Regulating Liver Cirrhosis Based on BATMAN-TCM Platform显示文摘[Objectives]To explore the molecular mechanism of Zhizi Ganjiang Decoction in the treatment of liver cirrhosis based on the network pharmacology.[Methods]The BATMAN-TCM server was used to predict the potential targets of Zhizi Ganjiang Decoction,and the GeneCards database searched for targets related to liver cirrhosis;Cytoscape 3.5.1 software was used to construct compound-cirrhosis target interaction network and protein interaction network;then gene ontology(GO)enrichment analysis,Reactome pathway enrichment analysis,and KEGG-based biological pathway enrichment analysis were performed.[Results]The compound-disease-target network of Zhizi Ganjiang Decoction was constructed.The PPI network of Zhizi Ganjiang Decoction targets constructed based on String contained 130 nodes and 689 interaction relationships.The key nodes mainly involved nuclear hormone receptor-like domain superfamily and ligand binding domain,serpentine type 7TM GPCR chemoreceptor and chemoreceptor Srx,rhodopsin family transmembrane receptors,zinc finger NHR/GATA type nuclear hormone receptors and ligand binding domains,PI3-kinase family p85 binding domain,phosphoinositide(PI)3-kinase catalytic domain and other categories.GO enrichment analysis pathway related entries mainly involved steroid hormone receptor activity,nuclear receptor activity,transcription factor activity,direct ligands regulation sequence-specific DNA binding,G protein-coupled amine receptor activity,steroid binding,catecholamine binding,neurotransmitter receptor activity,adrenergic receptor activity,ammonium ion binding,and oxidoreductase activity,etc.KEGG channel analysis mainly involved neuroactive ligand-receptor interaction,aldosterone regulates sodium reabsorption,regulates lipolysis in adipocytes,regulates inflammatory mediators of TRP channels,tyrosine metabolism,calcium signaling pathway,cAMP signaling pathway,and nonalcoholic fatty liver diseases.[Conclusions]The research results suggest that Zhizi Ganjiang Decoction regulates the process of liver cirrhosis mainly through aldosterone regulation of sodium reabsorption,regulation of lipolysis in adipocytes,regulation of inflammatory mediators of TRP channels,calcium signaling pathways,and nonalcoholic fatty liver diseases.This reflects the multi-target and holistic action characteristics of the Chinese herbal compounds,and is expected to provide new ideas for further research on the molecular mechanism of Zhizi Ganjiang Decoction.Mingchen CAO Enze LI Zhiwei YANG Yanlin SHAO Fangang MENG Zenan ZHANG Wenjing LI Mengxiang FANG Yuefen LIU 2023Medicinal Plant2023,14,2:0
10Description of space charge transport in oil‐paper insulation using adaptive time‐stepping transient upstream finite element method显示文摘The charge transport and accumulation in oil-paper can cause the insulation degradation.So far,the most widely used model to simulate space charge transport and accumulation is the bipolar charge transport(BCT)model,which can well simulate the space charge dynamics.However,there are two shortcomings in the algorithms for solving the BCT model.One is that there is almost no use of vectorisation technology,which may increase the complexity of the algorithm,the other is the usage of fixed step size which might bring extra computation cost.In view of this,an adaptive time‐stepping transient up-stream finite element method(FEM)is developed to solve the BCT model considering trapping/detrapping,as well as the recombination phenomenon under DC condition in this article.Then,a vectorisation technology is used in the method to optimise the al-gorithm.Moreover,the adaptive time‐stepping method is introduced in simulation to reduce computation time and calculation amount.Simulation results are obtained by programing and later presented,which are basically consistent with the corresponding experimental results.Therefore,the proposed method is expected to promote the opti-mization design of the oil-paper insulation system.Jiefeng Liu Mingchen Shi Yiyi Zhang Bing Gao Wei Zhang Shuo Liang 2022High Voltage2022,7,1:0
11IMPROVEMENT OF THE PERFORMANCE OF THE DISPENSER CATHODE WITH Re AS A MIDDLE LAYER显示文摘A new type of dispenser cathode with dual-layer (Os-W/Re) is developed. The cathode coated with Os-W/Re shows better emission performance than the cathode coated with Os-W alloy. X-ray Photoelectron Spectroscopy (XPS) spectra demonstrate that ternary alloy coating (Os-W-Re) formed on the surface of the cathode with dual-layer (Os-W/Re) after full activation is the major reason why it has better emission than the cathode with Os-W alloy. The surface of each variety of the cathode is characterized with Scanning Electron Microscope (SEM) before and after activation: the emitting surface of the cathode with Os-W alloy after ageing appeared non-adherence (flaking) in localized areas, which is one of the reasons for non-uniform emission. However, the surface of the cathode with dual-layer (Os-W/Re) does not present film peeling under the same conditions. 13Multifunctional Gd-CuS loaded UCST polymeric micelles for MR/PA imaging-guided chemo-photothermal tumor treatment显示文摘Hepatocellular carcinoma(HCC)is a life-threatening disease for which there is no effective treatment currently.Novel theranostics simultaneously having excellent imaging and therapeutic functions are highly desired in cancer therapy.Herein,we develop the sialic acid(SA)modified polymeric micelles at an upper critical solution temperature(UCST)of 43℃(sialic acid-poly(ethylene glycol)-poly(acrylamide-co-acrylonitrile),SA-PEG-p(AAm-co-AN)),which further encapsulated with doxorubicin(DOX)and Gd-CuS nanoparticles(Gd-CuS NPs)for chemo-photothermal treatment of HCC guided by magnetic resonance(MR)/photoacoustic(PA)dual-mode imaging.The resultant SA-PEG-p(AAm-co-AN)/DOX/Gd-CuS(SPDG)had an excellent photothermal conversion efficiency,enabling SPDG with an instantaneous release behavior of DOX under near-infrared(NIR)irradiation.This study also revealed that SPDG could actively target to HCC,which was due to that SA had a high affinity with E-selectin overexpressed at the tumor site.Moreover,benefiting from the HCC-targeted ability and NIR light-controlled on-demand delivery of DOX,SPDG showed a superior potential in MR/PA dual-mode imaging-guided chemo-photothermal treatment.Overall,our study reveals that the designed SPDG may be used as an ideal multifunctional nanoplatform for cancer theranostics.Yan Du Di Liu Mingchen Sun Gaofeng Shu Jing Qi Yuchan You Yiting Xu Kai Fan Xiaoling Xu Feiyang Jin Jun Wang Qiying Shen Luwen Zhu Xiaoying Ying Jiansong Ji Liming Wu Daren Liu Yongzhong Du 2022Nano Research2022,15,3:0

15Spatial distribution and export of nutrients and metal elements in the subterranean estuary of Daya Bay显示文摘Subterranean estuaries(STE)are important seawater-groundwater mixing zones with complex biogeochemical processes,which play a vital role in the migration and transformation of dissolved materials.In this study,we first investigated the spatial distributions of dissolved inorganic nitrogen(DIN),dissolved inorganic phosphorous(DIP),dissolved inorganic silicon(DSi)and metal elements(As,Ba,Cr,Cu,Fe,Mn,Ni,Pb,and Zn)in STE including upper intertidal,seepage face and subtidal zones.We then estimated submarine groundwater discharge(SGD)and associated nutrient and metal element fluxes.From the generalized Darcy’s law method,SGD was estimated to be 30.13 cm/d,which was about 7 times larger than the inflow(4.16 cm/d).The nutrient and metal fluxes from SGD were estimated to be(5.33±4.99)mmol/(m^(2)·d)for DIN,(0.22±0.03)mmol/(m^(2)·d)for DIP,(16.20±2.05)mmol/(m^(2)·d)for DSi,(1325.06±99.10)μmol/(m^(2)·d)for Fe,(143.41±25.13)μmol/(m^(2)·d)for Mn,(304.06±81.07)μmol/(m^(2)·d)for Zn,(140.21±13.33)μmol/(m^(2)·d)for Cu,(84.49±2.94)μmol/(m^(2)·d)for Pb,(37.38±5.51)μmol/(m^(2)·d)for Ba,(27.88±3.89)μmol/(m^(2)·d)for Cr,(10.10±6.33)μmol/(m^(2)·d)for Ni,and(6.25±3.45)μmol/(m^(2)·d)for As.The nutrient and metal fluxes from SGD were relatively higher than those from the inflow,suggesting that nearshore groundwater acted as the sources of nutrients and metal elements discharging into the sea.The environmental potential pollution of coastal seawater was evaluated by pollution factor index(Pi),comprehensive water quality index(CWQI),and ecological risk index(ERI).Pb mainly caused potential danger of nearshore environment with considerable contamination(Pi=5.78±0.19),heavy pollution(CWQI=4.09)and high ecological risk(ERI=18.00).This study contributed to better understanding the behavior of nutrients and metal elements and improving the sustainable management of STE under the pressure of anthropogenic activities and climate change.Zhaoxi Liu Mingchen Ge Qianqian Wang Xuejing Wang Kai Xiao Gang Li Hailong Li 2023Acta Oceanologica Sinica2023,42,8:0

18Influence of positive DC electric field on rime icing on the insulator and its experimental verification in the natural environment显示文摘The electric field imposes a significant effect on ice accretion on the insulator.The impact of an AC electric field on insulator icing in an artificial climate chamber has been addressed.Compared with AC,DC electric field exerts an attraction on supercooled water droplets,which is more detrimental to the power system.Moreover,the icing on energised insulators is rarely studied in the natural environment.In the present research,an electrical,fluid coupled‐field simulation was carried out on the insulator.The force characteristics and motion deviation features of water droplets at different positive DC electric field strengths were studied.On this basis,rime icing on the insulator energised with different positive DC voltages was investigated in the field.Results show that the charged droplet shifts along the electric field line under a positive DC voltage.The deviation degree of the droplet increases with the increment of the voltage and droplet size and decreases with an increase in wind speed.As the voltage rises,the surface roughness,icing amount and icing length of rime ice‐covered insulator increase.Consequently,compared with that without energisation,the local collision coefficient,icing amount and icing length increase by 17.6%,46.4%and 63.9%,respectively,under a 40 kV DC voltage.Zongyuan Liu Yuyao Hu Xingliang Jiang Lei Chen Mingchen Chi Feiyan Han Chong Zhao 2023High Voltage2023,8,5:0

20Removal of ions from produced water using Powder River Basin coal显示文摘In addition to being used as an energy source,coal also has significant potential for other,more sustainable uses including water treatment.In this study,we present a simple approach to treat water that was produced during oil production and contained a total dissolved solids(TDS)content of over 150 g/L using Powder River Basin(PRB)coal.PRB coal used as packing material in a flow-through column effectively removed 60%–80%of the cations and anions simultaneously.Additionally,71%–92%of the total organic carbon in the produced water was removed as was all of the total suspended solids.The removal mechanisms of both cations and anions were investigated.Cations were removed by ion exchange with protons from oxygen-containing functional groups such as carboxylic and phenolic hydroxyl groups.Anions,mainly Cl−1,appeared to be removed through either the formation of resonance structures as a result of delocalization of electrons within coal molecules or through ion–πinteractions.We propose that coal is a“pseudo-amphoteric”exchange material that can remove cations and anions simultaneously by exchanging ions with both ionized and non-ionized acids that are ubiquitous in coal structure or resonance effect.Zaixing Huang Fangjing Liu Mingchen Tang Yangyan Gao David MBagley Xin He Alexander Goroncy Maohong Fan 2022International Journal of Coal Science & Technology2022,9,1:0
